Abstract
本发明公开了一种含羟基酪醇的油包多元醇纳米乳液和制备方法及应用,包括,羟基酪醇、多元醇、乳化剂和油脂,其中,以原料质量为百分百计,所述羟基酪醇含量为0.1%~10%,所述多元醇含量为5%~30%,所述乳化剂含量为5%~20%,所述油脂含量为40%~90%。本发明采用油包多元醇的方式制备含羟基酪醇的乳液,相比常规的油包水或水包油体系,该油包多元醇组合能有效抑制羟基酪醇的氧化和变色:鉴于羟基酪醇优异的抗氧化活性,其有助于提升皮肤屏障,该油包多元醇体系能提高羟基酪醇的功效稳定性,降低了其在在化妆品领域中的应用限制。
Description
技术领域
本发明属于化妆品技术领域,具体涉及到一种含羟基酪醇的油包多元醇纳米乳液和制备方法及应用。
背景技术
皮肤屏障容易受到外界因素的破坏,如紫外照射、接触有害化学品、微生物侵入、过度清洗等,这些因素会诱导细胞中的ROS(活性氧)增加并导致氧化与还原失衡,从而引起氧化应激反应,造成细胞损伤。皮肤屏障被破坏时,水分流失加速,引起皮肤干燥,因此减少ROS带来的刺激对于提升皮肤屏障具有重要意义。
羟基酪醇主要存在于橄榄的果实与枝叶中,归功于两个酚羟基结构,其是具有优异抗氧化活性的天然多酚类化合物,能够通过清除ROS来改善氧化应激反应造成的细胞损伤。然而,这也表明该物质易被氧化,从而活性受到影响,因此其应用受到限制。
羟基酪醇是一种水溶性化合物,但是水易溶解氧和重金属离子,这对于维持抗氧化活性是一种挑战。
发明内容

因此,本发明的目的是,克服现有技术中的不足,提供一种含羟基酪醇的油包多元醇纳米乳液。
为解决上述技术问题,本发明提供了如下技术方案:一种含羟基酪醇的油包多元醇纳米乳液,包括,
羟基酪醇、多元醇、乳化剂和油脂,其中,以原料质量为百分百计,所述羟基酪醇含量为0.1%~10%,所述多元醇含量为10%~30%,所述乳化剂含量为6%~9%,所述油脂含量为60%~90%。
作为本发明所述含羟基酪醇的油包多元醇纳米乳液的一种优选方案,其中:所述纳米乳液体系为无水体系。
作为本发明所述含羟基酪醇的油包多元醇纳米乳液的一种优选方案,其中:所述多元醇为甘油、丙二醇、1,3-丁二醇、山梨醇、戊二醇、己二醇中的一种或几种。
作为本发明所述含羟基酪醇的油包多元醇纳米乳液的一种优选方案,其中:所述乳化剂为聚甘油-2二聚羟基硬脂酸酯、聚甘油-3二异硬脂酸酯、聚乙二醇月桂酸酯、乙二醇单硬脂酸酯、硬脂酸聚氧丙烯酯、甘油单月桂酸酯、聚甘油脂肪酸酯、二甘油单月桂酸酯、山梨醇单月桂酸酯、壬基苯氧基聚乙氧基乙醇、聚甘油硬脂酸酯、聚甘油油酸酯、鲸蜡硬脂基葡糖苷、甘油硬脂酸酯/PEG100硬脂酸酯、单油酸甘油酯、司盘80、硬脂酸甘油酯、大豆磷脂、卵磷脂和鲸蜡基聚二甲基硅氧烷中的一种或几种。

作为本发明所述含羟基酪醇的油包多元醇纳米乳液的一种优选方案,其中:所述多元醇在组合物中的占比为10%~30%。
本发明的再一个目的是,克服现有技术中的不足,提供一种含羟基酪醇的油包多元醇纳米乳液的制备方法。
为解决上述技术问题,本发明提供了如下技术方案:一种含羟基酪醇的油包多元醇纳米乳液的制备方法,包括,
将乳化剂和油脂混合并搅拌均匀,得到透明流动液体A相;
将羟基酪醇和多元醇混合并搅拌均匀至获得透明流动液体B相;
在室温下将B相加到A相中,高速分散混合均匀后,采用高压微通道多相流技术得到含有羟基酪醇的油包多元醇组合物;其中,压力为40~50MPa,均质次数为3~5次。
作为本发明所述含羟基酪醇的油包多元醇纳米乳液的一种优选方案,其中:以原料质量为百分百计,所述羟基酪醇含量为0.1%~10%,所述多元醇含量为10%~30%,所述乳化剂含量为6%~9%,所述油脂含量为60%~90%。
本发明的另一个目的是,克服现有技术中的不足,提供一种含羟基酪醇的油包多元醇纳米乳液在化妆品中的应用,所述的化妆品包括洗面奶、面膜、化妆水、乳液、膏霜、精华中的一种或多种。
本发明有益效果:
本发明提出一种含羟基酪醇的油包多元醇纳米乳液,首次采用油包多元醇的方式制备含羟基酪醇的乳液,相比常规的油包水或水包油体系,该油包多元醇组合能有效抑制羟基酪醇的氧化和变色:鉴于羟基酪醇优异的抗氧化活性,其有助于提升皮肤屏障,该油包多元醇体系能提高羟基酪醇的功效稳定性,降低了其在在化妆品领域中的应用限制。

为使本发明的上述目的、特征和优点能够更加明显易懂,下面结合说明书实施例对本发明的具体实施方式做详细的说明。
在下面的描述中阐述了很多具体细节以便于充分理解本发明,但是本发明还可以采用其他不同于在此描述的其它方式来实施,本领域技术人员可以在不违背本发明内涵的情况下做类似推广,因此本发明不受下面公开的具体实施例的限制。
其次,此处所称的“一个实施例”或“实施例”是指可包含于本发明至少一个实现方式中的特定特征、结构或特性。在本说明书中不同地方出现的“在一个实施例中”并非均指同一个实施例,也不是单独的或选择性的与其他实施例互相排斥的实施例。
实施例1
按照表1中原料配比制备样品,将乳化剂和油脂混合并搅拌均匀得到透明流动液体A相;
将羟基酪醇和多元醇合并搅拌均匀至获得透明流动液体B相;
在室温下将B相加到A相中,高速分散混合均匀后,采用高压微通道多相流技术得到含有羟基酪醇的油包多元醇组合物,其压力为40MPa,均质次数为3次。
表1原料配比表(单位:%)
由表1中可知,样7的多元醇含量为30%,油脂含量为54%,其在50℃保存4周后出现分层;样6的多元醇含量和油脂含量分别为20%和64%,其在50℃保存4周后保持稳定。因此,在乳化剂含量一定的情况下,增加多元醇导致油脂减少,产品的粒径增大,不利于产品的稳定性。
羟基酪醇溶解于多元醇,提高羟基酪醇的浓度同样降低了油脂浓度,并导致产品粒径增加,如表1中的样3、样4和样5(羟基酪醇浓度分别为4%、7%和10%)。根据表1的样9、样10和样11,随着乳化剂含量增加(分别为7%、8%和9%),产品的粒径减小,并且在50℃保存4周后仍保持稳定。相反,减少乳化剂浓度导致产品分层,如表1中的样12(乳化剂为5%)。因此,当羟基酪醇浓度为0.1%~10%时,多元醇的优选浓度为10%~30%,油脂的优选浓度为60%~90%,乳化剂的优选浓度为6%~9%。
实施例2
羟基酪醇:5%
甘油:10%
山梨醇单月桂酸酯:2%
司盘80:4%
辛基十二醇:79%
将山梨醇单月桂酸酯、司盘80和辛基十二醇混合并搅拌均匀得到透明流动液体A相;
将羟基酪醇和甘油混合并搅拌均匀至获得透明流动液体B相;
在室温下将B相加到A相中,高速分散混合均匀得到初乳液。将初乳液分成5份:
(1)采用高压微通道多相流技术,40MPa,分别在循环2次(样1)、3次(样2)、4次(样3)、5次(样4)、6次(样5)后取样。
(2)采用高压微通道多相流技术,20MPa,在循环4次(样6)后取样。
(3)采用高压微通道多相流技术,30MPa,在循环4次(样7)后取样。
(4)采用高压微通道多相流技术,50MPa,在循环4次(样8)后取样。
(5)样品采用高压微通道多相流技术,60MPa,在循环4次(样9)后取样。
各样品粒径及不同温度条件下稳定性结果见表2,如下所示:
由上表结果可知:随着压力变大,样品的粒径逐步下降,当到达40MPa时,粒径变化基本平稳;随着循环次数增多,粒径逐步下降,当到达3次时,粒径变化基本平稳。
基于大量实验数据得出,优选的工艺:采用高压微通道多相流技术,压力为40~50MPa,循环次数为3~5次时,在此条件下可以得到粒径范围在200~400nm之间,在-20℃和50℃下可长期放置的含羟基酪醇的油包多元醇纳米乳液。
实施例3
为了表明水对羟基酪醇稳定性的影响,对实施例1中制备的样1和样2在50℃下进行8周的加速氧化实验,样2中含有5%的水。
结果如图1所示,样1下降缓慢,在50℃存储8周后,羟基酪醇的剩余量为92.34%,而样2的剩余量为13.75%。因此认为无水的样1的油包多元醇体系对于维持羟基酪醇的稳定性具有良好的作用。
实施例4
对实施例1中的样1和样2进行DPPH清除实验来表明该体系中羟基酪醇的抗氧化活性稳定性。
步骤如下:
1)将实施例1中的样1和样2产品于50℃进行加速氧化;
2)样品溶液配制:每周取一定量步骤1)中的产品溶解于乙醇并过滤;
3)0.3mMDPPH溶液:称取1.3mgDPPH溶解于50mL甲醇。
4)DPPH抑制测试如下:
样品组:移取100μL样品于96孔板,并加入100μLDPPH溶液,反应30min,用紫外可见分光光度计于517nm处检测吸光度As;
对照组:移取100μL乙醇于96孔板,并加入100μLDPPH溶液,反应30min,于517nm处检测吸光度Ac;
空白组:移取100μL样品溶液或乙醇于96孔板,并加入100μLDPPH溶液,反应30min,于517nm处检测吸光度Asb或Acb;
5)计算DPPH抑制率(I,%):
I/%=[1-(As-Asb)/(Ac-Acb)]*100
DPPH抑制活性/%=In/I0,In和I0分别为第n周和第0周产品的DPPH抑制率。
结果如图2所示,经过8周加速氧化实验后,非水样1的DPPH抑制活性仍保留有91.03%,含5%水的样2仅剩余11.03%,因此认为无水的油包多元醇体系对维持羟基酪醇的抗氧化功效稳定性具有良好的作用。
对比例1
与实施例1中的样1相比,区别在于不添加羟基酪醇。
实施例5
采用对比例1以及实施例1中的样1和样2进行人体屏障保护测试,步骤如下:
1)将对比例1以及实施例1中的样1和样2在50℃下进行8周的加速氧化;
1)测试人员在前臂不同受试部位(3×3cm2)分别涂抹步骤1)的产品,每天使用两次,每次20~25μg,连续使用72h,测试皮肤经皮水分流失和皮肤角质层水分含量;
2)在合格的斑试器中加入3%SLS水溶液(用量:20~25μg),贴敷于步骤1)的受试部位,24h后去除斑试器。去除斑试器0.5h后,测试皮肤经皮水分流失和皮肤角质层水分含量。
受试者:30人,男15人,女15人;年龄为30~40岁。
图3和图4分别为受试者在使用步骤1)产品后的皮肤角质层含水量和皮肤经皮水分流失结果。
由图可知,涂抹样1后,SLS刺激导致的皮肤角质层水分下降和皮肤经皮水分流失增加小于涂抹样2和对比例1产品,对比例1产品的效果和样2相差较小,因此,含有更高浓度羟基酪醇的样1产品相比羟基酪醇失活的样2产品和不含羟基酪醇的对比例1产品能够增强皮肤屏障并提高皮肤对刺激的抵抗能力。
实施例6
化妆品面霜制备
制备工艺:将A相和B相分别在80℃下加热至完全溶解,在高速分散下将A相加入B相,均质5min。待温度降至50℃以下,加入C相并搅拌均匀,即得到产品。
针对羟基酪醇易氧化的问题,本发明采用油包多元醇的方式制备羟基酪醇纳米乳液。油包多元醇体系低了羟基酪醇与氧气的接触,提高了羟基酪醇的稳定性。另外,通过优选多元醇、乳化剂和油脂的配比以及优选制备条件来获得体系稳定的含羟基酪醇的油包多元醇纳米乳液,该纳米乳液可使用在化妆品产品体系中,具有广阔的应用前景。
应说明的是,以上实施例仅用以说明本发明的技术方案而非限制,尽管参照较佳实施例对本发明进行了详细说明,本领域的普通技术人员应当理解,可以对本发明的技术方案进行修改或者等同替换,而不脱离本发明技术方案的精神和范围,其均应涵盖在本发明的权利要求范围当中。
Claims (10)
1.一种含羟基酪醇的油包多元醇纳米乳液,其特征在于:包括羟基酪醇、多元醇、乳化剂和油脂,其中,以原料质量为百分百计,所述羟基酪醇含量为0.1%~10%,所述多元醇含量为10%~30%,所述乳化剂含量为6%~9%,所述油脂含量为60%~90%。
2.如权利要求1所述含羟基酪醇的油包多元醇纳米乳液,其特征在于:所述纳米乳液体系为无水体系。
3.如权利要求1所述含羟基酪醇的油包多元醇纳米乳液,其特征在于:所述多元醇为甘油、丙二醇、1,3-丁二醇、山梨醇、戊二醇、己二醇中的一种或几种。
4.如权利要求1所述含羟基酪醇的油包多元醇纳米乳液,其特征在于:所述乳化剂为聚甘油-2二聚羟基硬脂酸酯、聚甘油-3二异硬脂酸酯、聚乙二醇月桂酸酯、乙二醇单硬脂酸酯、硬脂酸聚氧丙烯酯、甘油单月桂酸酯、聚甘油脂肪酸酯、二甘油单月桂酸酯、山梨醇单月桂酸酯、壬基苯氧基聚乙氧基乙醇、聚甘油硬脂酸酯、聚甘油油酸酯、鲸蜡硬脂基葡糖苷、甘油硬脂酸酯/PEG100硬脂酸酯、单油酸甘油酯、司盘80、硬脂酸甘油酯、大豆磷脂、卵磷脂和鲸蜡基聚二甲基硅氧烷中的一种或几种。
5.如权利要求1所述含羟基酪醇的油包多元醇纳米乳液,其特征在于:所述油脂为聚二甲基硅氧烷、环聚二甲基硅氧烷、角鲨烷、白油、辛酸癸酸三甘油酯、棕榈酸乙基己酯、异硬脂基甘油醚、肉豆蔻酸异丙酯、棕榈酸异丙酯、丁二醇二辛酸二癸酸酯、异十六烷、油酸癸酯、异壬酸异壬酯、肉豆蔻醇肉豆蔻酸酯、棕榈酸、硬脂酸、山嵛酸、山嵛醇、肉豆蔻酸、碳酸丙二醇酯、碳酸二辛酯、异十三醇异壬酸酯、亚油酸癸酯山嵛醇、新戊二醇二癸酸酯、苯甲酸异硬脂酸酯、PPG-3肉豆蔻基醚、聚丙二醇-15硬脂基醚、氢化椰油甘油酯类、山茶籽油、牡丹籽油、白池花籽油、葵花籽油、牛油果油脂霍霍巴酯类、硬脂醇、霍霍巴油、杏仁油、橄榄油、椰子油、亚油酸中的一种或几种。
6.如权利要求1所述含羟基酪醇的油包多元醇纳米乳液,其特征在于:所述多元醇在组合物中的占比为10%~30%。
7.如权利要求1~6中任一所述含羟基酪醇的油包多元醇纳米乳液的制备方法,其特征在于:包括,
将乳化剂和油脂混合并搅拌均匀,得到透明流动液体A相;
将羟基酪醇和多元醇混合并搅拌均匀至获得透明流动液体B相;
在室温下将B相加到A相中,高速分散混合均匀后,采用高压微通道多相流技术得到含有羟基酪醇的油包多元醇组合物;其中,压力为40~50MPa,均质次数为3~5次。
8.如权利要求7所述含羟基酪醇的油包多元醇纳米乳液的制备方法,其特征在于:以原料质量为百分百计,所述羟基酪醇含量为0.1%~10%,所述多元醇含量为10%~30%,所述乳化剂含量为6%~9%,所述油脂含量为60%~90%。
9.权利要求1~6中任一所述含羟基酪醇的油包多元醇纳米乳液在化妆品中的应用。
10.如权利要求9所述的应用,其特征在于:所述的化妆品包括洗面奶、面膜、化妆水、乳液、膏霜、精华中的一种或多种。
