技术领域
本发明涉及一种制备枸杞玉米黄素微胶囊的方法,尤其是涉及一种以枸杞多糖复合壁材制备枸杞玉米黄素微胶囊的方法。
背景技术
枸杞是名贵的中药材和滋补品,富含类胡萝卜素类功效物质,主要是玉米黄素及其衍生物类。已证实枸杞玉米黄素及其衍生物具有优异的抗氧化活性,能够缓解由四氯化碳引起的肝脏Kupffer细胞炎症反应和氧化应激损伤,以及继发性胆汁淤积性肝纤维化损伤,能够有效缓解酒精肝、脂肪肝、乙肝或者肝性脑病引起的肝损伤,其肝保护能力与公认的肝脏保护剂水飞蓟素相当。玉米黄素及其衍生物还能够预防白内障、老年黄斑性病变、动脉粥样硬化等疾病但因其分子结构中含有不饱和双健,对高温、部分金属离子、光不稳定性,易发生氧化反应、降解反应等,且为水不溶性物质,在机体内细胞吸收困难,生物利用率低,极大的限制了其应用。
枸杞多糖是一种由高度支化的多糖及蛋白共同组成的大分子物质,分子质量高达10~2300kDa,是天然的微胶囊壁材。已有大量研究表明枸杞多糖具有抗氧化、免疫调节、降血糖、降低血脂功效,以枸杞多糖-乳清分离蛋白-阿拉伯胶为复合壁材制备枸杞玉米黄素微胶囊,在提升枸杞玉米黄素的稳定性、溶解度和生物利用率的同时还可对其功效起到增效作用。
但是目前枸杞玉米黄素由于溶解性、稳定性以及生物利用度低等问题,极大的限制了其功效发挥和应用,目前还没有克服这种缺陷的方法。
发明内容
本发明的目的主要是克服现有技术的不足,提供一种原料独特、制作方法简单、能提高枸杞玉米黄素的稳定性和生物利用率,且其显著提高的抗氧化活性为其进一步的功效作用研究和开发应用奠定了基础的一种以枸杞多糖复合壁材制备枸杞玉米黄素微胶囊的方法。
本发明通过如下方式实现:
一种以枸杞多糖复合壁材制备枸杞玉米黄素微胶囊的方法,其特征在于:该制备方法为先制备枸杞多糖,并以此为原料之一制备复合壁材溶液,然后将浓度为0.5~1.5%的枸杞玉米黄素加入到上述制备好的壁材溶液中,搅拌、混匀并12000r/min均质得乳液,最后将制备的乳液在预冻至-80℃后采用真空冷冻干燥法进行干燥,制得粒径为210~600nm的枸杞玉米黄素微胶囊;
所述枸杞多糖的制备方法为:将枸杞粉碎后用亚临界丁烷萃取除掉色素及脂溶性物质,再经70%乙醇提取除掉多酚、生物碱类物质,最后用水提取、膜过滤制得分子量>5000的枸杞多糖,上述亚临界丁烷萃取温度为50℃,压强为0.5MPa;
所述壁材溶液的制备方法为:分别将枸杞多糖、乳清分离蛋白、阿拉伯胶中加入一定量的水使其质量浓度依次为2~3%,4-6%,2-3%,然后将其混合,在60-70℃搅拌使其溶解,既得壁材溶液;
所述枸杞为干枸杞。
本发明具有如下效果:
1)制备方法简单、独特:本发明提供的方法为先制备枸杞多糖,并以此制备复合壁材溶液,然后将浓度为0.5~1.5%的枸杞玉米黄素加入到上述制备好的壁材溶液中,搅拌、混匀并12000r/min均质得乳液,最后将制备的乳液在预冻至-80℃后采用真空冷冻干燥法进行干燥,制得粒径为210~600nm的枸杞玉米黄素微胶囊,制作工艺简单独特。
2)显著提高枸杞玉米黄素的生物利用率:本发明分别将枸杞多糖、乳清分离蛋白、阿拉伯胶中加入一定量的水使其质量浓度依次为2~3%,4-6%,2-3%,然后将其混合,在60-70℃搅拌使其溶解,先制备壁材溶液,利用枸杞多糖为壁材之一制备枸杞玉米黄素微胶囊,所制备的微胶囊能够有效保护枸杞玉米黄素免受光、热、氧的破坏,显著提高枸杞玉米黄素的生物利用率,对枸杞玉米黄素的进一步研究和应用开发具有重要意义。
3)为枸杞功能性成分的开发利用提供理论依据:本发明提供的方法是将枸杞多糖与枸杞玉米黄素的进行复配,对其抗氧化等功效的协同作用研究将更有利于开发新的、安全和高效的天然抗氧化保健品和相关药物,为枸杞功能性成分的开发利用提供理论依据。
4)枸杞玉米黄素由于溶解性、稳定性以及生物利用度低等问题,极大的限制了其功效发挥和应用,微胶囊包埋能有效的改善特性。因此,本发明以枸杞多糖-乳清分离蛋白-阿拉伯胶复合物的微胶囊负载枸杞玉米黄素,目的是提高枸杞玉米黄素的稳定性、生物利用率,为其进一步的功效作用研究和开发应用奠定基础,且以活性物质枸杞多糖为壁材之一,制得微胶囊的抗氧化活性显著高于枸杞玉米黄素。

实施例一:一种以枸杞多糖复合壁材制备枸杞玉米黄素微胶囊的方法,该制备方法为:分别将枸杞多糖、乳清分离蛋白、阿拉伯胶中加入一定量的水使其质量浓度依次为2%、5%、3%,然后将其混合,在60℃搅拌使其溶解,既得壁材溶液,然后将浓度为0.5~1.5%的枸杞玉米黄素加入到上述制备好的壁材溶液中,搅拌、混匀并12000r/min均质得乳液,最后将制备的乳液在预冻至-80℃后采用真空冷冻干燥法进行干燥,制得粒径为210~600nm的枸杞玉米黄素微胶囊。
实施例二:一种以枸杞多糖复合壁材制备枸杞玉米黄素微胶囊的方法,该制备方法为:分别将枸杞多糖、乳清分离蛋白、阿拉伯胶中加入一定量的水使其质量浓度依次为3%、4%、2%,然后将其混合,在65℃搅拌使其溶解,既得壁材溶液,然后将浓度为0.5~1.5%的枸杞玉米黄素加入到上述制备好的壁材溶液中,搅拌、混匀并12000r/min均质得乳液,最后将制备的乳液在预冻至-80℃后采用真空冷冻干燥法进行干燥,制得粒径为210~600nm的枸杞玉米黄素微胶囊。
实施例三:一种以枸杞多糖复合壁材制备枸杞玉米黄素微胶囊的方法,该制备方法为:分别将枸杞多糖、乳清分离蛋白、阿拉伯胶中加入一定量的水使其质量浓度依次为2.5%、6%、2.5%,然后将其混合,在70℃搅拌使其溶解,既得壁材溶液,然后将浓度为0.5~1.5%的枸杞玉米黄素加入到上述制备好的壁材溶液中,搅拌、混匀并12000r/min均质得乳液,最后将制备的乳液在预冻至-80℃后采用真空冷冻干燥法进行干燥,制得粒径为210~600nm的枸杞玉米黄素微胶囊。
实施例四:一种以枸杞多糖复合壁材制备枸杞玉米黄素微胶囊的方法,该制备方法为:分别将枸杞多糖、乳清分离蛋白、阿拉伯胶中加入一定量的水使其质量浓度依次为2.1%、5.5%、2.8%,然后将其混合,在70℃搅拌使其溶解,既得壁材溶液,然后将浓度为0.5~1.5%的枸杞玉米黄素加入到上述制备好的壁材溶液中,搅拌、混匀并12000r/min均质得乳液,最后将制备的乳液在预冻至-80℃后采用真空冷冻干燥法进行干燥,制得粒径为210~600nm的枸杞玉米黄素微胶囊。
上述枸杞多糖的制备方法为:将枸杞粉碎后用亚临界丁烷萃取除掉色素及脂溶性物质,再经70%乙醇提取除掉多酚、生物碱类物质,最后用水提取、膜过滤制得分子量>5000的枸杞多糖,上述亚临界丁烷萃取温度为50℃,压强为0.5MPa。
上述枸杞为干枸杞。
Claims (4)
1.一种以枸杞多糖复合壁材制备枸杞玉米黄素微胶囊的方法,其特征在于:该制备方法为先制备枸杞多糖,并以此为原料之一制备复合壁材溶液,然后将浓度为0.5~1.5%的枸杞玉米黄素加入到上述制备好的壁材溶液中,搅拌、混匀并12000r/min均质得乳液,最后将制备的乳液在预冻至-80℃后采用真空冷冻干燥法进行干燥,制得粒径为210~600nm的枸杞玉米黄素微胶囊。
2.如权利要求1所述的一种以枸杞多糖复合壁材制备枸杞玉米黄素微胶囊的方法,其特征在于:所述枸杞多糖的制备方法为:将枸杞粉碎后用亚临界丁烷萃取除掉色素及脂溶性物质,再经70%乙醇提取除掉多酚、生物碱类物质,最后用水提取、膜过滤制得分子量>5000的枸杞多糖,上述亚临界丁烷萃取温度为50℃,压强为0.5MPa。
3.如权利要求1所述的一种以枸杞多糖复合壁材制备枸杞玉米黄素微胶囊的方法,其特征在于:所述壁材溶液的制备方法为:分别将枸杞多糖、乳清分离蛋白、阿拉伯胶中加入一定量的水使其质量浓度依次为2~3%,4-6%,2-3%,然后将其混合,在60-70℃搅拌使其溶解,既得壁材溶液。
4.如权利要求1所述的一种以枸杞多糖复合壁材制备枸杞玉米黄素微胶囊的方法,其特征在于:所述枸杞为干枸杞。
