Abstract

本发明公开了一种改善精液品质的植物提取物复方产品及其生产工艺和应用,所述复方产品由β‑胡萝卜素、虎杖提取物、松树皮提取物、裂壶藻破壁粉组成;进一步的,上述复方产品的配比为:β‑胡萝卜素1‑5份、虎杖提取物1‑5份、松树皮提取物1‑3份、裂壶藻破壁粉1‑3份;进一步优选的,上述复方产品的配比为:β‑胡萝卜素3份、虎杖提取物3份、松树皮提取物2份、裂壶藻破壁粉2份。本发明基于天然植物提取物,副作用小,绿色安全、使用方便,在小鼠实验中,少量添加进饲料中,能明显的提升小鼠精液质量。因此,本品可安全广泛的用于制备相关的药品和保健品,具有良好的市场前景。

一种改善精液品质的植物提取物复方产品及其生产工艺和 应用
技术领域
本发明属于天然植物提取产品领域,具体公开了一种改善精液品质的植物提取物复方产品及其生产工艺和应用。
背景技术
随着社会的发展和环境的恶化,紊乱的作息规律和环境污染对生殖健康的影响成为21世纪人类开展生命科学研究的重要课题和人类健康所面临的巨大挑战。统计显示全球约有15%的夫妇不孕,因男性导致的不孕占不孕症的一半,其中精液品质下降是主要原因之一。环境、饮食、生活习惯的改变对精液品质有很大影响,而研究表明,营养素的补充能发挥积极作用。环境应激和生殖道炎症可导致精液减少,精子密度、活力、授精能力降低,植物提取物具有抗氧化、抗炎、抑菌等多重功效,可有效缓解应激、改善代谢和血液循环、增强细胞活力,因而具有改善精液品质的潜在的改善作用。β-胡萝卜素(β-carotene)是类胡萝卜素中活性最高的维生素A前体物,具有抗氧化、调节免疫等生物学活性,对繁殖性能也具有潜在的改善作用。虎杖富含植物多酚白藜芦醇,具有利湿退黄,清热解毒,散瘀止痛,止咳化痰的功效,传统用于湿热黄疸,淋浊,带下,风湿痹痛,痈肿疮毒,水火烫伤,经闭,症瘕,趺打损伤,肺热咳嗽。松树皮富含植物多酚,其提取物具有强抗氧化和抗炎能力。裂壶藻是一种富含DHA的海洋真菌。在本团队前期研究中,以上4种提取物均表现出改善动物繁殖性能的作用,但单一的提取物效果有限。现有技术中尚未发现上述四种提取物的组合效果的研究。
发明内容
针对上述情况,本发明将以上4种提取物进行复配,旨在形成一种可显著改善精液品质的植物提取物复方产品,同时研究其制备工艺和应用。
本发明的技术方案如下:
一种改善精液品质的植物提取物复方产品,所述复方产品由β-胡萝卜素、虎杖提取物、松树皮提取物、裂壶藻破壁粉组成。
进一步的,上述一种改善精液品质的植物提取物复方产品,所述复方产品组成如下:β-胡萝卜素1-5份、虎杖提取物1-5份、松树皮提取物1-3份、裂壶藻破壁粉1-3份。
进一步的,上述一种改善精液品质的植物提取物复方产品,所述复方产品组成如下:β-胡萝卜素3份、虎杖提取物3份、松树皮提取物2份、裂壶藻破壁粉2份。
进一步的,上述一种改善精液品质的植物提取物复方产品生产工艺,包括以下步骤:
(1)虎杖和松树皮粉碎过20-60目筛,随后采用酶解水提的方式进行提取,所述酶解过程使用酶的种类为:纤维素酶1000-3000U/Kg、半纤维素酶500-1500U/Kg、果胶酶500-1500U/Kg,酶解时间为:1-3h、酶解温度为室温、酶解pH为5-6;
(2)所得提取液过D101大孔树脂柱吸附、洗脱之后浓缩、低温干燥,所得提取物总多酚含量需≧30%;裂壶藻培养后经酶解破壁、冷冻干燥成粉,所述酶解过程使用酶的种类为:纤维素酶2000-4000U/Kg、果胶酶500-1500U/Kg,酶解时间为:1-3h、酶解温度为室温、酶解pH为5-6;
(3)将上述虎杖提取物、松树皮提取物、裂壶藻破壁粉与市售β-胡萝卜素按比例混匀,形成复方产品。。

从上述技术方案可以得出,本发明公开的改善精液品质的植物提取物具有如下有益效果:
1、炎症和氧化应激是精子细胞功能障碍的主要原因之一,对精子结构和功能完整有重要影响,本植物提取物复方组合物在实验过程中发现,将本品添加入小鼠日粮中,可明显提高小鼠精子数量和密度,同时改善了小鼠的精子质量。
2、本植物提取物复方组合物绿色安全、使用方便,无需复杂的设备即可实现大规模工业化生产,应用前景广泛。

下面对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其它实施例,都属于本发明保护的范围。
本发明下面实施例中,所用试验小鼠为昆明小鼠,试验基础日粮为市售产品。
本发明实施例中使用的试剂或仪器未注明生产厂商者,均为可以通过市购获得的常规试剂产品。
以下结果为本发明所涉及的植物提取物复方产品代表性试验结果。
实施例1
制备例
(1)虎杖和松树皮粉碎过40目筛,随后采用酶解水提的方式进行提取,所述酶解过程使用酶的种类为:纤维素酶2000U/Kg、半纤维素酶1000U/Kg、果胶酶1000U/Kg,酶解时间为:2h、酶解温度为室温、酶解pH为5.5;
(2)所得提取液过D101大孔树脂柱吸附、洗脱之后浓缩、低温干燥,所得提取物总多酚含量需≧30%;裂壶藻培养后经酶解破壁、冷冻干燥成粉,所述酶解过程使用酶的种类为:纤维素酶3000U/Kg、果胶酶1000U/Kg,酶解时间为:2h、酶解温度为室温、酶解pH为5.5;
(3)将上述虎杖提取物、松树皮提取物、裂壶藻破壁粉与市售β-胡萝卜素按β-胡萝卜素3份、虎杖提取物3份、松树皮提取物2份、裂壶藻破壁粉2份的比例混匀,形成复方产品。
实施例2
实验例
(1)试验日粮
将实施例1制备的植物提取物复方产品按100mg/kg和300mg/kg的比例均匀混合于基础日粮中,制粒并分袋标记,存放于4℃冰箱中待用。
(2)试验设计
选取6周龄雄性昆明小鼠27只,预饲一周后,将小鼠随机分为3组,每组9个重复,正式试验期30d。试验期保证自由采食与饮水,室温保持在18-22℃,12小时光/暗循环(8:00-20:00开灯,20:00-次日8:00黑暗),单笼饲养。各实验组分别为:空白组(CTL):饲喂基础日粮,低剂量组(L):饲喂含100mg/kg植物提取物复方产品的日粮,高剂量组(H):饲喂含300mg/kg植物提取物复方产品的日粮,每3日更换垫料以及饮用水。
(3)样品采集
试验30d后将小鼠麻醉并处死,解剖,采集单侧附睾,放入装有1mL精子获能液的培养皿中,37℃孵育3分钟,进行精液品质检测。
(4)指标测定
小鼠精液检测:利用迈郎鼠精子全自动分析系统(南宁松景天伦生物科技有限公司)对雄鼠精子进行检测与分析。
(5)试验结果
各组小鼠精子数量、密度及精子质量相关结果如表1所示,日粮中添加植物提取物复方产品提高了小鼠精子数量和密度,且提高了精子直线速率、路径速率、曲线速率和鞭打频率,对精子质量有积极作用。由此可见,日粮添加植物提取物复方产品不仅可明显提高小鼠精子数量和密度,同时改善了小鼠的精子质量。

上述实验结果说明:将本品添加入小鼠日粮中,可明显提高小鼠精子数量和密度,同时改善了小鼠的精子质量。

1.一种改善精液品质的植物提取物复方产品,其特征在于,所述复方产品由β-胡萝卜素、虎杖提取物、松树皮提取物、裂壶藻破壁粉组成。
2.根据权利要求1所述的一种改善精液品质的植物提取物复方产品,其特征在于,所述复方产品组成如下:β-胡萝卜素1-5份、虎杖提取物1-5份、松树皮提取物1-3份、裂壶藻破壁粉1-3份。
3.根据权利要求1所述的一种改善精液品质的植物提取物复方产品,其特征在于,所述复方产品组成如下:β-胡萝卜素3份、虎杖提取物3份、松树皮提取物2份、裂壶藻破壁粉2份。
4.如权利要求1-3任一项所述的一种改善精液品质的植物提取物复方产品的生产工艺,其特征在于,包括以下步骤:
(1)虎杖和松树皮粉碎过20-60目筛,随后采用酶解水提的方式进行提取,所述酶解过程使用酶的种类为:纤维素酶1000-3000U/Kg、半纤维素酶500-1500U/Kg、果胶酶500-1500U/Kg,酶解时间为:1-3h、酶解温度为室温、酶解pH为5-6;
