Abstract

背景技术
CN114470084A公开了一种环保高效防治罗非鱼白便的组合物及其制备方法和应用,组合物包括白术、茯苓、黄芪、山药、甘草,制备方法采用复合酶酶解方法和水提醇沉方法相结合,通过复合酶把细胞膜、细胞质内的淀粉多糖、糖蛋白酶解,使得药用多糖更好地释放,再采用水提醇沉的方法把药用多糖浸提、浓缩、沉淀分离出来,最终得到效价高、方便使用、便于保存的提取物。通过从一种中药组合物中提取植物多糖,作为益生元,常温易保存,稳定性好,可用于饲料加工,直达肠道,作为肠道菌群的碳源促进肠道有益菌的增殖、抑制有害菌,从而改善肠道微生态菌群,维持平衡的菌群结构,有利于防治罗非鱼白便和肠炎问题。
CN107594136A公开了一种促进吸收的娃娃鱼饲料,其原料按重量份包括:鱼蛋白粉20-30份,猪肝10-15份,蚯蚓粉10-12份,虾粉8-10份,蛙肉粉6-8份,牛肉膏4-6份,小麦粉10-15份,豆粕6-8份,大豆油6-8份,芝麻油4-6份,复合维生素粉1-2份,复合矿物质粉1-2份,果胶3-5份,羧甲基纤维素6-8份,氯化钙2-3份,益生菌3-5份,海藻酸钠8-10份,添加剂10-15份,复合酶2-4份,柠檬酸0.3-0.5份;其中,添加剂的原料包括:胡萝卜素、芝麻酚、角鲨烯、蒲公英水提物、大青叶水提物、黄芪水提物、白术水提物。能促进肠道消化吸收养分,并缓解肝脏负荷,促进娃娃鱼快速健康生长。
CN104938818A公开了一种保护罗非鱼肠道健康的复方中草药饲料添加剂,其包括有以下重量百分比的组分:黄芪18-25%、野菊花15-18%、金钱草4-7%、草麻黄9-15%、白术16-19%、当归20-30%、甘草5%。其能够提高罗非鱼肠道健康,增加罗非鱼消化吸收能力,提高成活率,降低饵料系数,减少水质污染。
对于鱼类养殖来说,肝胆肠道疾病是非常常见的,是控制养殖成本的一个重要的方面。
所以,本案解决的技术问题是:如何有效的提高鱼类肝胆肠道健康。
发明内容
针对现有技术的不足,本发明的目的在于提供一种水产肠道肝胆修复制剂,该制剂可以有效的提高鱼类免疫力,提高对于转塘鱼、应激鱼的产肠道肝胆的修复功效。
为达到本发明目的,本发明采用以下技术方案:
一种水产肠道肝胆修复制剂,包括主料、生姜提取物、大蒜素、山楂酊;所述主料为白术、黄芪的提取物;
所述主料、生姜提取物、大蒜素、山楂酊的重量比为1000:10~100:10~20:10~50。
在上述的水产肠道肝胆修复制剂中,所述主料的制备方法为:将3000g白术和黄芪以干燥饮片的形式加水煎煮2次,合并煎液,过滤,加入乙醇使醇含量为60%,静置,分取上清液,滤过,回收乙醇,浓缩成为相对密度为1.05~1.1的浓缩膏;所述白术、黄芪的质量比为1:1~3。
在上述的水产肠道肝胆修复制剂中,所述主料、生姜提取物、大蒜素、山楂酊的重量比为1000:20~70:12~16:20~40。
在上述的水产肠道肝胆修复制剂中,将浓缩膏、生姜提取物、大蒜素、山楂酊、辅料混合,制粒;所述辅料为淀粉。
同时,本发明还公开了另外一种形式的水产肠道肝胆修复制剂,包括主料、生姜大蒜混合提取物、山楂酊;所述主料为白术、黄芪的提取物;
所述主料、生姜大蒜混合提取物、山楂酊的重量比为1000:10~100:10~50。
在上述的产肠道肝胆修复制剂中,所述生姜大蒜混合提取物的提取方法为:将新鲜的生姜、大蒜按照5:1~3的比例混合,采用粉碎机粉碎成混合物,采用90-95vol%的乙醇、乙酸乙酯作为提取溶剂提取混合物;乙醇、乙酸乙酯的体积比为6~9:1~4;混合物和提取溶剂的重量比为1:5~10;提取温度低于40℃,提取时间为3~5h;提取结束后经过滤、真空浓缩得到浸膏,将浸膏分散到乙酸乙酯中进行萃取,收集萃取后的油相,减压蒸馏去除乙酸乙酯,得到生姜大蒜混合提取物。
在上述的产肠道肝胆修复制剂中,乙醇、乙酸乙酯的体积比为7~8:2~3;混合物和提取溶剂的重量比为1:6~8。
在上述的产肠道肝胆修复制剂中,所述主料的制备方法为:将3000g白术和黄芪以干燥饮片的形式加水煎煮2次,合并煎液,过滤,加入乙醇使醇含量为60%,静置,分取上清液,滤过,回收乙醇,浓缩成为相对密度为1.05~1.1的浓缩膏所述白术、黄芪的质量比为1:1-3。
在上述的产肠道肝胆修复制剂中,将浓缩膏、生姜大蒜混合提取物、山楂酊、辅料混合,制粒;所述辅料为淀粉。
相对于现有技术,本发明具有以下有益效果:
本发明采用白术、黄芪作为载体药,采用生姜提取物、大蒜素、山楂酊来进行肝胆肠道的修复。
白术黄芪汤为较为有名的中医方剂。出自《宣明论方》卷九。具有清热燥湿,泻火解毒,补血活血,除烦止咳,温中散寒,益气补中,健脾养胃之功效。主治五心烦热,自汗,四肢痿劣,饮食减少,肌瘦昏昧。
当其应用到鱼类养殖中后,白术能增强吞噬功能,并且可提高淋巴细胞转化率,黄芪性甘微温,补中益气,偏补脾阳,可治脾虚泄泻。作为佐剂,生姜提取物含6-姜辣素具有抗氧化活性、抑制血小板聚集、保护胃黏膜等功效;大蒜素具有抗菌、诱食、抗氧化等多重功效(参考中国科学院海洋研究所科技情报研究室.《生物饵料增养殖技术与人工配合饵料》:中国科学院海洋研究所科技情报研究室,2010:219-210),山楂酊富含枸橼酸,其可以提高采食量促进营养物质消化吸收、促进肠道菌群健康、增强机体抗应激和免疫能力;
通过以上组合,可以提高白术、黄芪对于鱼类肠道肝胆健康的改善作用。

实施例1
步骤1:将1500g白术和1500g黄芪以干燥饮片的形式加水煎煮2次,合并煎液,过滤,加入乙醇使醇含量为60%,静置,分取上清液,滤过,回收乙醇,浓缩成为相对密度为1.05~1.1的浓缩膏;
步骤2:取浓缩膏、生姜提取物、大蒜素、山楂酊、淀粉,进行制粒;
浓缩膏、生姜提取物、大蒜素、山楂酊的重量比分别为:1000:50:15:30;
淀粉用量相当于浓缩膏、生姜提取物、大蒜素、山楂酊总重的2倍。
命名为样品1。
实施例2
步骤1:将1500g白术和1500g黄芪以干燥饮片的形式加水煎煮2次,合并煎液,过滤,加入乙醇使醇含量为60%,静置,分取上清液,滤过,回收乙醇,浓缩成为相对密度为1.05~1.1的浓缩膏;
步骤2:取浓缩膏、生姜提取物、大蒜素、山楂酊、淀粉,进行制粒;

生姜提取物规格:粉末状,含1wt%姜辣素;
大蒜素:有效成分≥99%;
山楂酊:由湖北朗博万生物医药有限公司提供,浸膏。
实施例4
步骤1:将1500g白术和1500g黄芪以干燥饮片的形式加水煎煮2次,合并煎液,过滤,加入乙醇使醇含量为60%,静置,分取上清液,滤过,回收乙醇,浓缩成为相对密度为1.05~1.1的浓缩膏;
步骤2:将新鲜的生姜、大蒜按照5:2的重量比例混合,采用粉碎机粉碎成混合物,采用95vol%的乙醇、乙酸乙酯作为提取溶剂提取混合物;
乙醇、乙酸乙酯的体积比为8:2;混合物和提取溶剂的重量比为1:8;提取温度25℃,提取时间为4h;
提取结束后经过滤、真空浓缩得到浸膏,将浸膏分散到乙酸乙酯中进行萃取,收集萃取后的油相,减压蒸馏去除乙酸乙酯,得到生姜大蒜混合提取物。浸膏和乙酸乙酯的重量比为1:10。
步骤3:将步骤1的浓缩膏、生姜大蒜混合提取物、山楂酊的重量比为1000:75:20的比例混合,并加入淀粉制粒,淀粉的用量为浓缩膏、生姜大蒜混合提取物、山楂酊总重的2倍。
命名为样品2。
实施例5
步骤1:将1500g白术和1500g黄芪以干燥饮片的形式加水煎煮2次,合并煎液,过滤,加入乙醇使醇含量为60%,静置,分取上清液,滤过,回收乙醇,浓缩成为相对密度为1.05~1.1的浓缩膏;
步骤2:将新鲜的生姜、大蒜按照5:3的重量比例混合,采用粉碎机粉碎成混合物,采用90vol%的乙醇、乙酸乙酯作为提取溶剂提取混合物;
乙醇、乙酸乙酯的体积比为6:4;混合物和提取溶剂的重量比为1:10;提取温度25℃,提取时间为4h;
提取结束后经过滤、真空浓缩得到浸膏,将浸膏分散到乙酸乙酯中进行萃取,收集萃取后的油相,减压蒸馏去除乙酸乙酯,得到生姜大蒜混合提取物。浸膏和乙酸乙酯的重量比为1:10。
步骤3:将步骤1的浓缩膏、生姜大蒜混合提取物、山楂酊的重量比为1000:50:50的比例混合,并加入淀粉制粒,淀粉的用量为浓缩膏、生姜大蒜混合提取物、山楂酊总重的2倍。
实施例6
步骤1:将1500g白术和1500g黄芪以干燥饮片的形式加水煎煮2次,合并煎液,过滤,加入乙醇使醇含量为60%,静置,分取上清液,滤过,回收乙醇,浓缩成为相对密度为1.05~1.1的浓缩膏;

养殖实验
在广东省某养殖场进行养殖实验,实验所用的养殖对象为200g±10g的罗非鱼苗960尾,分为8组(A组~H组),每组4个重复,每个重复30尾,在20个室内循环流水过滤养殖池中进行养殖;水温平均为28.0±2.0℃,水中溶氧大于7mg/L,氨氮低于0.5mg/L,每天光照时间为12h;按鱼体重的3%作为每日投饲量,每天上午9h和下午4h分2次投喂,按日增重2%计总体重,每7天调整一次投饲量,试验期为6周;养殖周期内无罗非鱼死亡。
投喂的基础饲料采用某品牌罗飞浮水饲料,每吨饲料加入1.5kg水产肠道肝胆修复制剂。
A组为空白组,仅投喂基础饲料;
B组为基础饲料+样品1;
C组为基础饲料+样品2;
D组为基础饲料+样品3;
E组为基础饲料+样品4;
F组为基础饲料+样品5;
G组为基础饲料+样品6;
H组为基础饲料+样品7;
养殖周期结束后,各养殖池中无鱼苗死亡;对各养殖池的鱼苗进行称重,计算平均日增重、肝胆综合症发生情况、肠炎发生情况和白便情况。
平均日增重:计算实验开始和结束的时候的重量差值,再除以实验天数;
肝胆综合症的判定:病鱼尾鳍、背鳍的鳍条尖发白,体表其他无任何明显症状,病鱼的口腔及眼球周围均无明显症状;将活的病鱼提起,有少量血液从鳃盖下方流出鳃部有大量的淤血,黏液增多,鳃丝充血;肝脏轻触易碎,肿大,有点状或块状出血或瘀血,色泽变淡,呈土黄色,或发白,形成黄白相间的“花肝”;肠道外壁无明显症状,但内壁却严重充血,肠道弹性很差,肠道中有少量的食物。
肠炎的判定:病鱼游动缓慢或呆立不动,体色灰暗,离群独游,食欲减退,慢游水边或浮于水面。病鱼腹部膨大并带有红斑,肛门红肿,呈紫红色,轻压腹部有血和黄色黏液外流。剖开腹部,可见肠管充血腐烂,多脓液,局部发炎,充满黏液,无食物,肠壁弹性差,剪开肠道发现肠壁变白、较薄,肠道的内黏膜脱落现象严重;
白便率的判定:观察养殖区域内鱼类排便情况,计算养殖过程中出现白便的鱼的条数。

结果分析:
1.通过上述分析可见,B组优于F至G组,其对于肝胆综合症、肠炎和白便控制均明显,一般来说,罗非鱼的抗病能力比较强,其采食量大,最容易发生的就是白便和肠炎,一般来说,罗非鱼发生了肠炎就容易产生白便,一般来说白便为肠道黏膜的脱落物,但是导致肠道黏膜的脱落的原因不仅仅是肠炎,其消化功能不佳也可能导致肠道黏膜的脱落。
通过实验可以发现,在F至G组中,分别缺失了大蒜素、山楂酊、生姜提取物,其白便现象没有完全抑制,说明生姜提取物、大蒜素、山楂酊基于其各自不同的作用,有着较为明显的功能侧重,其中大蒜素具有抗菌等作用,其对于肠炎等肠道疾病控制较有优势,山楂酊对于肠道健康和肝胆健康具有贡献,但是其需要在大蒜素、生姜提取物两者具有抗氧化、抗菌类物质的存在下才能表现最优的效果。
2.通过B组、A组、F至G组的对比可见,白术、黄芪对于鱼类的肠道健康、肝胆健康有着重要的作用。
3.通过B组、C组来看,采用复合提取的方式能够提高大蒜类提取物、生姜类提取物的药用效果,提高罗非鱼的日增重;在生姜中含有较多的油溶性的姜油、姜辣素,姜油能够提高大蒜素中有效成分的溶出,同时,姜油具有较好的抗氧化性,对大蒜素具有较好的稳定促进作用,抑制单独提取大蒜素导致的大蒜素不稳定。
通过C组、D组、E组,合适的提取溶剂是实现较好的药用效果的有效保证;采用乙醇和乙酸乙酯的配合,可提高极性油溶性产物的溶出,同时采用乙酸乙酯提高油溶性物质的溶出效率;通过两者配合,可以超过采用市购提取物的效果,并且该提取方法为传统的油溶性提取方法,适于厂家低成本生产。

Claims (6)
1.一种水产肠道肝胆修复制剂,其特征在于,包括主料、生姜提取物、大蒜素、山楂酊;所述主料为白术、黄芪的提取物;
所述主料、生姜提取物、大蒜素、山楂酊的重量比为1000:10~100:10~20:10~50;
所述主料的制备方法为:将3000g白术和黄芪以干燥饮片的形式加水煎煮2次,合并煎液,过滤,加入乙醇使醇含量为60%,静置,分取上清液,滤过,回收乙醇,浓缩成为相对密度为1.05~1.1的浓缩膏;所述白术、黄芪的质量比为1:1~3。
2.根据权利要求1所述的水产肠道肝胆修复制剂,其特征在于,所述主料、生姜提取物、大蒜素、山楂酊的重量比为1000:20~70:12~16:20~40。
3.根据权利要求1所述的水产肠道肝胆修复制剂,其特征在于,将浓缩膏、生姜提取物、大蒜素、山楂酊、辅料混合,制粒得到水产肠道肝胆修复制剂;所述辅料为淀粉。
4.一种水产肠道肝胆修复制剂,其特征在于,包括主料、生姜大蒜混合提取物、山楂酊;所述主料为白术、黄芪的提取物;
所述主料、生姜大蒜混合提取物、山楂酊的重量比为1000:10~100:10~50;
所述生姜大蒜混合提取物的提取方法为:将新鲜的生姜、大蒜按照5:1~3的重量比例混合,采用粉碎机粉碎成混合物,采用90-95vol%的乙醇、乙酸乙酯作为提取溶剂提取混合物;乙醇、乙酸乙酯的体积比为6~9:1~4;混合物和提取溶剂的重量比为1:5~10;提取温度低于40℃,提取时间为3~5h;提取结束后经过滤、真空浓缩得到浸膏,将浸膏分散到乙酸乙酯中进行萃取,收集萃取后的油相,减压蒸馏去除乙酸乙酯,得到生姜大蒜混合提取物;

5.根据权利要求4所述的水产肠道肝胆修复制剂,其特征在于,乙醇、乙酸乙酯的体积比为7~8:2~3;混合物和提取溶剂的重量比为1:6~8。
6.根据权利要求5所述的水产肠道肝胆修复制剂,其特征在于,将浓缩膏、生姜大蒜混合提取物、山楂酊、辅料混合,制粒得到水产肠道肝胆修复制剂;所述辅料为淀粉。
