背景技术
将锡离子源诸如氟化亚锡添加到洁牙剂组合物中,以递送抗微生物和抗过敏有益效果。然而,由于(i)锡离子与其他洁牙剂组分诸如二氧化硅之间的反应性和(ii)在特定pH处形成不溶性锡化合物,故锡离子源可能难以掺入到洁牙剂组合物中。因此,仅通过在储存期间使锡离子与关键洁牙剂组分之间的相互作用最小化,以使与口腔表面诸如釉质、牙质、牙龈、牙斑和细菌的反应性的锡离子可用性最大化,才可能将锡离子掺入到洁牙剂组合物中。
锡离子溶液的化学不稳定性可通过降低溶液的pH,提供阴离子螯合剂诸如有机酸的阴离子,或提供聚合物螯合剂诸如多磷酸盐或聚羧酸盐来减轻。这些螯合剂中的一些螯合剂反而还可通过与锡离子直接结合来防止锡离子与二氧化硅分子表面的相互作用。然而,如果螯合剂-锡离子相互作用强于锡离子与口腔内表面之间的结合亲和力,则强螯合剂-锡离子相互作用也可能不利于锡离子可用性,此种锡离子递送是对抗口腔疾病诸如牙斑、齿龈炎、恶臭、龋齿、敏感性和牙齿酸蚀等性能的先决条件。因此,即使螯合剂的选择也必须与整个洁牙剂制剂相平衡,以最大限度地提高锡离子可用性。
另外,所选磨料系统可以影响锡离子可用性。例如,钙磨料诸如焦磷酸钙和磷酸二钙可以与由氟化亚锡产生的锡离子相互作用,导致在洁牙剂的使用寿命期间损失高达90%的可用锡离子。从基于磷酸钙的磨料切换到二氧化硅磨料系统可以改善可用锡离子的量,但是由于锡离子与二氧化硅磨料之间的表面相互作用,仍将导致仅最多50%的可用锡离子。50%锡离子可用性仍然显著足以需要添加补充性锡离子源诸如氯化亚锡以补偿锡离子与二氧化硅磨料表面之间的反应性。其他方法包括二氧化硅表面的疏水改性、热处理二氧化硅以去除反应性表面羟基官能团或配制具有最少水或无水的洁牙剂,以防止磨料和游离锡离子之间的反应性。然而,这些用于减轻溶液中游离锡离子的损失并可用于与口腔相互作用的策略中的每一个策略导致成本更高,从而限制了这些方法的商业可行性。因此,需要一种以可承受的成本提供可用锡离子源的洁牙剂组合物。

具体实施方式
本发明涉及包含锡离子源和钙磨料的洁牙剂组合物。另外,本发明涉及包含氟化亚锡和碳酸钙的洁牙剂组合物。
虽然希望在口腔中形成CaF2,但是许多洁牙剂制剂不采用含水洁牙剂组合物中的钙离子和氟离子,因为CaF2可以在储存期间从溶液中沉淀出来,从而消除了源自钙离子或氟离子的任何益处。例如,碳酸钙(CaCO3)是最小化洁牙剂制剂成本的精选磨料。然而,由于游离氟和钙离子之间的不相容性,几乎所有CaCO3基制剂使用单氟磷酸钠作为较低反应性氟化物源。
除了过早CaF2制剂的可能性之外,使用CaCO3与锡离子源组合还可以提高洁牙剂组合物的pH,这可能导致Sn(OH)2从洁牙剂组合物中沉淀出来,从而消除了源自游离锡离子的任何益处。

锡
本发明的洁牙剂组合物包含锡。锡可由锡离子源提供。锡离子源可为任何合适的化合物,当将洁牙剂组合物施用到口腔中时,该化合物可在洁牙剂组合物中提供锡离子和/或将锡离子递送到口腔中。锡离子源可包含一种或多种含锡化合物,诸如氟化亚锡、氯化亚锡、溴化亚锡、碘化亚锡、氧化亚锡、草酸亚锡、硫酸亚锡、硫化亚锡、氟化锡、氯化锡、溴化锡、碘化锡、硫化锡和/或它们的混合物。优选地,锡离子源可包括氟化亚锡、氯化亚锡和/或它们的混合物。

钙磨料
本发明的洁牙剂组合物包含钙。钙可由钙磨料提供。可将磨料添加到洁牙剂制剂中,以有助于去除牙齿上的表面污渍。钙磨料可为任何合适的磨料化合物,当将洁牙剂组合物施用到口腔中时,该磨料化合物可在洁牙剂组合物中提供钙离子和/或将钙离子递送到口腔中。
洁牙剂组合物可包含约5%至约70%、约10%至约50%、约10%至约60%、约20%至约50%、约25%至约40%或约1%至约50%的钙磨料。钙磨料可包含一种或多种钙磨料化合物,诸如碳酸钙、沉淀碳酸钙(PCC)、研磨碳酸钙(GCC)、白垩、磷酸二钙、焦磷酸钙和/或它们的混合物。
洁牙剂组合物可以包含附加的非钙磨料,诸如膨润土、硅胶(单独的,并且具有任何结构)、沉淀二氧化硅、无定形沉淀二氧化硅(单独的,并且也具有任何结构)、二氧化硅水合物、珍珠岩、二氧化钛、氧化铝、氧化铝水合物、煅烧氧化铝、硅酸铝、不溶性偏磷酸钠、不溶性偏磷酸钾、不溶性碳酸镁、硅酸锆、微粒热固性树脂和其它合适的磨料材料。可将此类材料引入洁牙剂组合物中以适合目标洁牙剂制剂的抛光特性。洁牙剂组合物可包含按洁牙剂组合物的重量计约5%至约70%、约10%至约50%、约10%至约60%、约20%至约50%、约25%至约40%或约1%至约50%的非钙磨料。
另选地,洁牙剂组合物可基本上不含、实质上不含或不含二氧化硅、氧化铝或任何其他非钙磨料。洁牙剂组合物可包含少于约5%、少于约1%、少于约0.5%、少于约0.1%或0%的非钙磨料,诸如二氧化硅和/或氧化铝。
多元醇
本发明的口腔护理组合物可包含多元醇。多元醇是具有多于一个羟基官能团的有机化合物。多元醇可为在使用前储存口腔护理组合物时可与锡离子弱缔合、相互作用或键合的任何合适的化合物。多元醇可为糖醇、单糖、二糖、多糖或非还原糖。糖醇是一类多元醇,其可通过具有式(CHOH)nH2(其中n=4-6)的糖化合物的氢化获得。优选地,n为5和/或6,因为这些化合物已示出与锡离子源出乎意外地相互作用产生可溶性络合物,如本文所述。

口腔护理组合物可包含一种或多种湿润剂,每种湿润剂的含量为按该口腔护理组合物的重量计约0.01%至约70%、约5%至约50%、约10%至约60%或约20%至约80%。
水
本发明的洁牙剂组合物可为无水制剂、低水制剂或高水制剂。总体而言,口腔护理组合物可包含按组合物的重量计0%至约99%、约5%至约75%、约20%或更多、约30%或更多或者约50%或更多的水。优选地,水为USP水。
在高水洁牙剂制剂中,洁牙剂组合物包含按组合物的重量计约45%至约75%的水。高水洁牙剂组合物可包含按组合物的重量计约45%至约65%、约45%至约55%或约46%至约54%的水。可将水添加到高水洁牙剂制剂中,并且/或者水可因包含其他成分而进入组合物中。
在低水洁牙剂制剂中,洁牙剂组合物包含按组合物的重量计约5%至约45%的水。低水洁牙剂组合物可包含按组合物的重量计约5%至约35%、约10%至约25%或约20%至约25%的水。可将水添加到低水洁牙剂制剂中,并且/或者水可因包含其他成分而进入组合物中。
在无水洁牙剂制剂中,洁牙剂组合物包含按组合物的重量计小于约10%的水。无水洁牙剂组合物包含按组合物的重量计小于约5%、小于约1%或0%的水。可将水添加到无水制剂中,并且/或者水可因包含其他成分而进入洁牙剂组合物中。
洁牙剂组合物还可包含其他口腔可接受载体材料,诸如醇、湿润剂、聚合物、表面活性剂和接受改善剂(诸如调味剂、甜味剂、着色剂和/或凉爽剂)。
pH
洁牙剂组合物可包含一种或多种缓冲剂。如本文所用,缓冲剂是指可用于将洁牙剂组合物的浆料pH调节至大于7、大于7至约14、约7.5或更大、约8或更大、约7.5至约10、大于7至约10、约8至约10的范围内的试剂。这些缓冲剂包括碱金属氢氧化物、碳酸盐、倍半碳酸盐、硼酸盐、硅酸盐、磷酸盐、咪唑以及它们的混合物。具体的缓冲剂包括磷酸一钠、磷酸三钠、氢氧化钠、氢氧化钾、碱金属碳酸盐、碳酸钠、咪唑、焦磷酸盐、柠檬酸和柠檬酸钠。洁牙剂组合物可包含一种或多种缓冲剂,每种缓冲剂的含量为按本发明组合物的重量计约0.1%至约30%、约1%至约10%或约1.5%至约3%。
锌
洁牙剂组合物可包含锌。锌可由锌离子源提供。锌离子源可包含一种或多种含锌化合物,诸如氟化锌、乳酸锌、氧化锌、磷酸锌、氯化锌、乙酸锌、六氟锆酸锌、硫酸锌、酒石酸锌、葡萄糖酸锌、柠檬酸锌、苹果酸锌、甘氨酸锌、焦磷酸锌、偏磷酸锌、草酸锌和/或碳酸锌。
锌离子源可以按洁牙剂组合物的重量计约0.01%至约5%、约0.2%至约1%、约0.5%至约1.5%或约0.3%至约0.6%的量存在于总洁牙剂组合物中。
氟化物
洁牙剂组合物可包含氟化物。氟化物可由氟化物离子源提供。氟化物离子源可包含一种或多种含氟化物的化合物,诸如氟化亚锡、氟化钠、氟化钾、氟化胺、单氟磷酸钠、氟化锌和/或它们的混合物。
氟化物离子源和锡离子源可为相同的化合物,例如,氟化亚锡,其可生成锡离子和氟离子。另外,氟化物离子源和锡离子源可为单独的化合物,诸如当锡离子源为氯化亚锡并且氟化物离子源为单氟磷酸钠或氟化钠时。
氟化物离子源和锌离子源可为相同的化合物,例如,氟化锌,其可生成锌离子和氟离子。另外,氟化物离子源和锌离子源可为单独的化合物,诸如当锌离子源为磷酸锌并且氟化物离子源为氟化亚锡时。
洁牙剂组合物可包含能够提供约50ppm至约5000ppm并且优选地约500ppm至约3000ppm的游离氟离子的氟化物离子源。为了递送所需量的氟离子,氟化物离子源可以按洁牙剂组合物的重量计约0.0025%至约5%、约0.2%至约1%、约0.5%至约1.5%或约0.3%至约0.6%的量存在于总洁牙剂组合物中。
多磷酸盐
洁牙剂组合物可包含多磷酸盐。多磷酸盐可由多磷酸盐源提供。多磷酸盐源可包含一个或多个多磷酸盐分子。多磷酸盐是一类通过正磷酸盐脱水和缩合生成不同链长的直链和环状多磷酸盐而获得的物质。因此,多磷酸盐分子通常以多磷酸盐分子的平均数(n)来标识,如下所述。尽管可存在一些环状衍生物,但通常认为多磷酸盐由主要以线性构型排列的两个或更多个磷酸盐分子组成。
优选的多磷酸盐是那些具有平均两个或更多个磷酸基团的多磷酸盐,以使得有效浓度的表面吸附产生足够的未结合的磷酸根官能团,这增强了阴离子表面电荷以及表面的亲水特性。本发明中优选的是具有下式的直链多磷酸盐:XO(XPO3)nX,其中X为钠、钾、铵或任何其他碱金属阳离子,并且n平均为约2至约21。碱土金属阳离子(诸如钙)不是优选的,因为它们趋于由包含氟离子和碱土金属阳离子的水溶液形成不溶性氟化物盐。因此,本文公开的洁牙剂组合物可不含或基本上不含焦磷酸钙。
合适的多磷酸盐分子的一些示例包括例如焦磷酸盐(n=2)、三聚磷酸盐(n=3)、四聚磷酸盐(n=4)、多磷酸钠磷(n=6)、六聚磷酸盐(n=13)、苯多磷酸盐(n=14)、六偏磷酸盐(n=21),其还被称为Glass H.多磷酸盐,可包括由FMC Corporation、ICLPerformance Products和/或Astaris生产的那些多磷酸盐化合物。
洁牙剂组合物可包含按该洁牙剂组合物的重量计约0.01%至约15%、约0.1%至约10%、约0.5%至约5%、约1%至约20%或约10%或更少的多磷酸盐源。
湿润剂
洁牙剂组合物可包含湿润剂,具有低含量的湿润剂,或不含湿润剂。湿润剂用于增加口腔护理组合物或洁牙剂的稠度或“口感”以及防止洁牙剂变干。合适的湿润剂包括聚乙二醇(具有多种不同的分子量)、丙二醇、甘油(glycerin、glycerol)、赤藓醇、木糖醇、山梨醇、甘露糖醇、丁二醇、乳糖醇、氢化淀粉水解物和/或它们的混合物。洁牙剂组合物可包含一种或多种湿润剂,每种湿润剂的含量为按该洁牙剂组合物的重量计0%至约70%、约5%至约50%、约10%至约60%或约20%至约80%。
表面活性剂
洁牙剂组合物可包含一种或多种表面活性剂。表面活性剂可用于使组合物在美容上更可接受。表面活性剂优选地为赋予组合物去污和发泡性质的去污材料。合适的表面活性剂是安全有效量的阴离子、阳离子、非离子、两性离子、两性甜菜碱表面活性剂,诸如:月桂基硫酸钠、月桂基羟乙基磺酸钠、月桂酰甲基羟乙基磺酸钠、椰油基谷氨酸钠、十二烷基苯磺酸钠、月桂酰肌氨酸、肉豆寇酰肌氨酸、棕榈酰肌氨酸、硬脂酰肌氨酸和油酰肌氨酸的碱金属盐或铵盐、聚氧乙烯山梨醇单硬脂酸酯、异硬脂酸酯和月桂酸酯、月桂基磺基乙酸钠、N-月桂酰肌氨酸、N-月桂酰基、N-肉豆蔻酰基或N-棕榈酰基肌氨酸的钠盐、钾盐和乙醇胺盐,烷基酚的聚环氧乙烷缩合物、椰油酰氨基丙基甜菜碱、月桂酰氨基丙基甜菜碱、棕榈基甜菜碱、椰油基谷氨酸钠等。月桂基硫酸钠是优选的表面活性剂。洁牙剂组合物可包含一种或多种表面活性剂,每种表面活性剂的含量为按洁牙剂组合物的重量计约0.01%至约15%、约0.3%至约10%或约0.3%至约2.5%。
增稠剂
洁牙剂组合物可包含一种或多种增稠剂。增稠剂可用于洁牙剂组合物中以提供凝胶状结构使牙膏稳定防止相分离。合适的增稠剂包括多糖、聚合物和/或二氧化硅增稠剂。多糖的一些非限制性示例包括淀粉;淀粉甘油;树胶,诸如卡拉牙树胶(刺梧桐树胶)、黄蓍胶、阿拉伯胶、茄替胶、阿拉伯树胶、黄原胶、瓜尔胶和纤维素胶;硅酸镁铝(胶体硅酸镁铝);角叉菜胶;藻酸钠;琼脂;果胶;明胶;纤维素化合物,诸如纤维素、羧甲基纤维素、羟乙基纤维素、羟丙基纤维素、羟甲基纤维素、羟甲基羧丙基纤维素、甲基纤维素、乙基纤维素和硫酸化纤维素;天然和合成粘土,例如锂蒙脱石粘土;以及它们的混合物。
增稠剂可包括多糖。适用于本文的多糖包括角叉菜胶、结冷胶、刺槐豆胶、黄原胶、卡波姆、泊洛沙姆、改性纤维素、以及它们的混合物。角叉菜胶是衍生自海藻的多糖。存在几种类型的角叉菜胶,这些角叉菜胶可通过它们的海藻源和/或通过它们的硫酸化程度和位置来区分。增稠剂可包括κ-角叉菜胶、改性的κ-角叉菜胶、ι-角叉菜胶、改性的ι-角叉菜胶、λ-角叉菜胶、以及它们的混合物。适用于本文的角叉菜胶包括可以系列名称“Viscarin”从FMC公司(FMC Company)商购获得的那些,包括但不限于Viscarin TP 329、Viscarin TP388和Viscarin TP 389。
增稠剂可包含一种或多种聚合物。聚合物可为聚乙二醇(PEG)、聚乙烯吡咯烷酮(PVP)、聚丙烯酸、来源于至少一种丙烯酸单体的聚合物、马来酸酐和甲基乙烯基醚的共聚物、交联的聚丙烯酸聚合物,其具有占洁牙剂组合物的各种重量百分比以及各种范围的平均分子范围。
增稠剂可包括无机增稠剂。合适的无机增稠剂的一些非限制性示例包括胶态硅酸镁铝、二氧化硅增稠剂。可用二氧化硅增稠剂的非限制性示例包括例如无定形沉淀二氧化硅,诸如165二氧化硅。其他非限制性二氧化硅增稠剂包括153、163和167以及177和265二氧化硅产品(均购自赢创公司(EvonikCorporation)),以及热解法二氧化硅。
洁牙剂组合物可包含0.01%至约15%、0.1%至约10%、约0.2%至约5%或约0.5%至约2%的一种或多种增稠剂。
螯合剂
洁牙剂组合物可包含一种或多种螯合剂,该一种或多种螯合剂具有小于1000的分子量(MW)。如本文所用,术语“螯合剂”是指具有至少两个能够结合二价金属离子的基团的二齿或多齿配体。本文所用的那些螯合剂通常还可与亚锡离子形成水溶性络合物。包含二氧化硅磨料的洁牙剂组合物通常包含一种或多种螯合剂以可逆地结合锡离子,从而防止锡离子不可逆地结合到二氧化硅颗粒的表面。本文适宜的螯合剂包括C2-C6二羧酸和三羧酸,诸如琥珀酸、苹果酸、酒石酸和柠檬酸;被羟基取代的C3-C6一元羧酸,诸如葡糖酸;吡啶甲酸;氨基酸,诸如甘氨酸;植酸、它们的盐以及它们的混合物。优选地,螯合剂为葡萄糖酸钠或柠檬酸盐。

其它成分
洁牙剂组合物可包含多种其他成分,诸如调味剂、甜味剂、着色剂、防腐剂、缓冲剂或适用于口腔护理组合物中的其他成分,如下所述。
还可将调味剂添加到洁牙剂组合物中。合适的调味剂包括冬青油、胡椒薄荷油、留兰香油、丁香芽油、薄荷醇、对丙烯基茴香醚、水杨酸甲酯、桉油精、肉桂、乙酸1-薄荷酯、鼠尾草、丁子香酚、欧芹油、羟苯基丁酮、α-紫罗兰酮、甘牛至草、柠檬、橙、丙烯基乙基愈创木酚、桂皮、香兰素、乙基香兰素、天芥菜精、4-顺式-庚烯醛、丁二酮、对叔丁基苯乙酸甲酯、以及它们的混合物。冷却剂也可以是风味剂体系的一部分。本发明组合物中优选的冷却剂是对孟烷氨基甲酰试剂,诸如N-乙基-对孟烷-3-甲酰胺(商业上称为“WS-3”)或N-(乙氧羰基甲基)-3-对薄荷烷甲酰胺(商业上称为“WS-5”)以及它们的混合物。风味剂体系通常以按洁牙剂组合物的重量计约0.001%至约5%的含量用于组合物中。这些调味剂通常包含醛、酮、酯、酚、酸以及脂族、芳族和其他醇的混合物。
可将甜味剂添加到口腔护理组合物中以赋予产品令人愉悦的味道。合适的甜味剂包括糖精(如糖精钠、糖精钾或糖精钙)、环磺酸盐(如钠盐、钾盐或钙盐)、安赛蜜-K、非洲甜果素、新橙皮苷二氢查耳酮、氨化甘草素、右旋糖、左旋糖、蔗糖、甘露糖、三氯蔗糖、甜菊糖和葡萄糖。
添加着色剂以改善产品的美学外观。合适的着色剂包括但不限于由相应监管机构诸如FDA批准的那些着色剂和欧洲食品和药品指令中列出的那些着色剂,并且包括色素诸如TiO2,以及颜色诸如FD&C和D&C染料。
还可将防腐剂添加到口腔护理组合物中以防止细菌生长。可以安全有效量添加被批准用于口腔组合物中的合适防腐剂,诸如对羟基苯甲酸甲酯、对羟基苯甲酸丙酯、苯甲酸和苯甲酸钠。
二氧化钛也可添加到本发明的组合物中。二氧化钛是能给组合物增加不透明度的白色粉末。二氧化钛通常占按洁牙剂组合物的重量计约0.25%至约5%。
口腔护理组合物中可使用其他成分,诸如脱敏剂、康复剂、其他防龋剂、螯合剂/多价螯合剂、维生素、氨基酸、蛋白质、其他抗牙斑/抗牙结石剂、遮光剂、抗生素、抗酶、酶、pH控制剂、氧化剂、抗氧化剂等。
离子可用性

实施例
以下实施例进一步说明了本发明,这些实施例不应以任何方式被理解为对本发明的范围施加限制。在阅读本文的说明书之后,在不脱离本发明的精神或所附权利要求书的范围的下,可向本领域的普通技术人员提出它们自己的各种其他方面、修改和其等同形式。
实验方法
锡离子可用性
此方法适用于测定来自水性浆料上清液中约5ppm至约5,000ppm Sn的口腔护理牙膏或洁牙剂组合物中的可溶性锡。通过将1份牙膏与3份水混合来制备浆料。对于每种测量的牙膏,对浆料的等分试样进行酸消化、稀释、并且通过电感耦合等离子体光学发射光谱(ICP-OES)进行分析。结果在此报告为牙膏和/或洁牙剂的纯水相中的ppm。
在分析开始之前制备几种标准物和试剂。通过使用带刻度的量筒将100mL浓缩的HCl和浓缩的HNO3分别转移到含有约1L超纯18MΩ(DI)水的2L容量瓶中,制备5%盐酸/5%硝酸冲洗溶液。使溶液涡旋以混合并稀释至带刻度的烧瓶的标记,然后通过重复的烧瓶倒置来充分混合。
购买1000mg/L锡和1000mg/L镓标准溶液(Sigma Aldrich,德国达姆施塔特默克集团(Merck KGaA,Darmstadt,Germany)),用于制备根据表1的标准溶液。使用移液管将准确量的标准物转移到50mL容量瓶中,同时将带刻度的量筒用于浓缩酸。转移后,将容量瓶用DI水填充到刻度线并充分混合。

通过将2.00克样品称重到含有10个玻璃珠的配衡圆底38mL离心管中制备浆料。将重量记录为最少0.001g。在即将浆化之前,将6.0mL DI水转移到管中。将管封盖并放置在涡旋机上,将样品以1200rpm混合60分钟。在完成混合循环后立即从涡旋机中取出管并将其置于离心机中。将它们以21,000个相对离心力(RCF)离心10分钟。离心完成后立即移除管,通过缓慢翻转三次轻轻混合上清液,确保离心管底部的固体塞不受干扰,然后倾析样品。然后将上清液倾析到15mL螺旋盖样品管中,确保大部分上清液被转移。
然后通过将上清液的0.5mL等分试样精确称重(精确到0.001g)到50mL Falcon管中,将上清液样品消化。然后加入2.5mL浓缩的HCl和HNO3。用聚丙烯表面皿覆盖管,并在90℃下放置在预热的块消化器中30分钟。将样品从加热中取出,用ID水(每次约1mL)冲洗表面皿三次,并将冲洗液加入消化的上清液。将镓标准物(0.2mL)移液到消化的上清液中,然后用DI水将上清液样品稀释至50mL。封盖管并进行混合。使用0.5mL DI水代替上清液以相同的方式制备消化方法空白。如果制备的样品多于一次性装入热块的数量,则为每组热块消解准备并分析方法空白。
ICP-OES(Perkin-Elmer 8300,美国马萨诸塞州沃尔瑟姆(Waltham,MA,USA))由经过训练并且合格的操作员进行操作,该操作员具有运行仪器并准确测定口腔护理组合物中锡的量的能力。根据制造商的说明,基于模型和配置选择ICP-OES操作参数。根据以下协议分析样品:
1.根据制造商指南预热和优化ICP-OES。执行建议的系统检查。在进行分析之前,通过样品引入系统运行HCl/HNO3冲洗溶液,将系统调节30分钟。
2.将用于在制造商推荐的波长、积分时间和观察模式下使用镓内标物确定锡的方法加载到操作计算机中。
3.5%HCl/5%HNO3冲洗溶液用于在每个空白、标准或测试溶液的分析之间冲洗样品引入系统。
4.在分析期间,记录所有溶液的三至五个读数。
5.分析校准空白。
6.测量10ppm Sn标准物。
7.测量5ppm Sn标准物。
8.测量0.5ppm LLOQ锡标准物。
9.测量方法空白。
10.测量测试溶液。
11.在每个第六测试溶液之后和最后一个样品之后重新测量5ppm Sn标准物。制备足够的标准物以完成分析。
12.在样品分析结束时测量0.5ppm LLOQ锡标准物。
如果10ppm锡标准物和5ppm锡标准物的重复读数的相对标准偏差%小于约3%,则认为分析成功。5ppm检查标准物在其值的96%-104%内。LLOQ在其值的75%-125%内。方法空白显示比LLOQ样品更低的锡信号强度。在每个分析的溶液中的内标物的回收在其值的90%-130%内。
可溶性锡根据下式确定:
此处的结果报告为添加的Sn的百分比。
氟离子可用性
在加速稳定性实验中,制剂A至E储存在40℃下。在1、2和/或3个月分析每种制剂的样品。在40℃下储存1个月的样品将预期接近在室温(约22-23℃)下储存至少8个月的等效样品。
通过在水中制备所选洁牙剂组合物的1:3浆料(1份糊剂加3份水)来确定氟离子可用性。将洁牙剂浆料离心,并分离上清液。在单独的容器中混合等分的洁牙剂上清液和离子强度调节剂缓冲液TISAB IV(RICCA,目录号8673-32)。通过将毫伏值转换为相对于标准氟离子浓度校准曲线的氟离子浓度,利用氟离子选择性电极(VWR Symphony,目录号14002-788)测量氟离子浓度。
表2示出了测试的洁牙剂组合物。配方A和B是本发明实施例,其具有钙磨料、碳酸钙。配方A是低水洁牙剂组合物,并且包括氯化亚锡作为锡离子源。配方B是无水洁牙剂组合物,并且包括氟化亚锡作为锡离子源。
制剂C、D和E是具有二氧化硅磨料、二氧化硅水合物的比较实施例。由于基于溶液中二氧化硅磨料与锡离子之间的相互作用预期锡离子可用性较低,制剂C、D和E为低至中度洁牙剂组合物,含有氟化亚锡和氯化亚锡的组合作为另外的锡离子源。

表3示出了配方A的加速稳定性研究之后的可用锡离子百分比。最初,配方A具有8.61的浆料pH和85%的理论锡离子量或加入组合物中的锡离子的量。在40℃下储存1个月和3个月之后,锡离子可用性分别降至83%和81%。组合物在40℃下储存1个月预期组合物在室温(约22-23℃)下的稳定性至少为8个月。因此,如果在室温(约22-23℃)下储存,锡离子在包含诸如碳酸钙的钙磨料的组合物中稳定至少24个月。

表4示出了配方B的加速稳定性研究之后的可用锡离子和可用氟离子的百分比。最初,配方B具有7.38的浆料pH和83%的理论锡离子量或加入组合物中的锡离子的量。在1个月、2个月和3个月之后,锡离子可用性分别是85%、86%和97%,如上所述,在室温(约22-23℃)下预计将为8个月、16个月和24个月。因此,锡离子在包含诸如碳酸钙的钙磨料的组合物中是稳定的。
最初,配方B具有93%的理论氟离子量或加入组合物中的氟离子的量。在1个月、2个月和3个月之后,氟离子可用性分别是84%、84%和85%。因此,氟离子在包含诸如碳酸钙的钙磨料的组合物中出乎意料地稳定。

表5示出了配方C、D和E的加速稳定性研究之后的可用锡离子百分比。最初,配方C具有8.18的pH浆料和30%的理论锡离子量或加入组合物中的锡离子的量。在1个月和2个月之后,锡离子可用性分别是18%和18%。最初,配方D具有8.00的浆料pH和39%的理论锡离子量或加入组合物中的锡离子的量。在1个月和2个月之后,锡离子可用性分别是24%和19%。最初,配方E具有9.18的浆料pH和64%的理论锡离子量或加入组合物中的锡离子的量。在1个月和2个月之后,锡离子可用性分别是43%和36%。如表5所示,即使在室温(初始值)下,锡离子也有显著损失。在40C下1个月后,锡离子的损失继续。配方D和E的相对较高值是由于螯合剂葡萄糖酸盐和焦磷酸盐的锡离子的更好结合,这有助于使与二氧化硅的反应性最小化并且通过与锡离子结合来稳定锡离子,防止氧化和水解。
重要的是,在包含碳酸钙磨料(即配方A和配方B)的组合物中,锡离子可用性比包含二氧化硅磨料(即配方C至E)的组合物更高。表3和表4示出了理论锡离子量的至少约80%的锡离子可用性。相比之下,在40℃加速稳定两个月后,配方C至E的锡离子可用性仅为理论锡量的18%至36%。因此,用碳酸钙磨料替换二氧化硅磨料可提高锡离子可用性高达450%(从18%到86%)。因此,表2至5中的发现表明,当与钙磨料组合时,亚锡离子在大于7的pH下是化学稳定的。另外,与二氧化硅磨料不同,亚锡离子对诸如碳酸钙的钙磨料不反应。具体地,表3和表4中的结果表明亚锡离子在包含碳酸钙的组合物中具有出色的稳定性。
虽然预期来自钙磨料的钙离子之间的相互作用将与来自氟化物离子源的可用氟离子反应并减少其量,但包含碳酸钙和氟化亚锡的配方B在40℃下加速稳定3个月后,氟离子可用性最初为93%,之后为85%。

Claims (9)
1.一种洁牙剂组合物,其包含:
(a) 按所述洁牙剂组合物的重量计约0.0025%至约5%的锡,优选地其中所述锡包括氟化亚锡、氯化亚锡或它们的组合;
(b) 按所述洁牙剂组合物的重量计约10%至约50%的钙磨料,优选地其中所述钙磨料为碳酸钙;和
(c) 按所述洁牙剂组合物的重量计最多约45%,或优选地小于约5%的水,
其中所述洁牙剂组合物具有大于7的pH。
2.根据权利要求1所述的洁牙剂组合物,其中所述洁牙剂组合物在40℃下三个月之后具有约70%或更高的可用锡离子。
3.根据权利要求1或2所述的洁牙剂组合物,其中所述洁牙剂组合物包含按所述洁牙剂组合物的重量计约0.01%至约5%的锌。
4.根据权利要求1至3中任一项所述的洁牙剂组合物,其中所述洁牙剂组合物包含二氧化硅磨料。
5.根据权利要求1至4中任一项所述的洁牙剂组合物,其中所述洁牙剂组合物包含约20%至约80%的湿润剂,优选地其中所述湿润剂包括甘油、山梨醇、赤藓醇、木糖醇、丁二醇、丙二醇、聚乙二醇或它们的组合。
6.根据权利要求1至5中任一项所述的洁牙剂组合物,其中所述洁牙剂组合物包含按所述洁牙剂组合物的重量计约0.01%至约15%的一种或多种表面活性剂。
7.根据权利要求1至6中任一项所述的洁牙剂组合物,其中所述洁牙剂组合物包含按所述洁牙剂组合物的重量计约1%或更少的二氧化硅。
8.根据权利要求1至7中任一项所述的洁牙剂组合物,其中所述洁牙剂组合物包含增稠剂,优选地其中所述增稠剂包括黄原胶、角叉菜胶、聚乙二醇、交联的聚丙烯酸聚合物、聚乙烯吡咯烷酮、脂肪醇或它们的组合。
9.根据权利要求33所述的洁牙剂组合物,其中所述洁牙剂组合物包含按所述洁牙剂组合物的重量计约0.01%至约5%的氟化物,优选地其中所述氟化物包括氟化钠、氟化亚锡、单氟磷酸钠、氟化胺、氟化锌或它们的组合。
