Abstract
本发明公开了一种调节口腔和咽喉健康的益生菌速溶固体饮料及其制备方法。重量份组分为益生菌5份、草本原料10份、益生元30份、糖醇30份、水果粉20份、食用香精2份、柠檬酸2份、柠檬酸钠1份。经过过筛、称量、混合、沸腾造粒、分装工艺制备完成。本发明添加多种口腔益生菌,帮助平衡口腔微生物组,减少口臭、牙周疾病的发生,对预防咽喉炎、中耳炎、上呼吸道感染等均有效果;复配多种天然草本原料,清热解毒、清咽润喉、滋阴润肺,有助于缓解咽喉不适。本发明通过沸腾造粒技术,制备的产品能够在口腔中迅速崩解,有助于益生菌的释放,帮助益生菌迅速溶解于唾液,有利于益生菌在口腔中定殖。

技术领域:
本发明属于益生菌产品技术领域,特别涉及一种调节口腔和咽喉健康的益生菌速溶固体饮料及其制备方法。
背景技术:
健康成年人口腔内菌群结构丰度广,跨越110个属,12个门,这些错综复杂的微生物共同生活,构成了人体健康的一道屏障,将有害菌抑制在可控程度内,从而建立动态的平衡系统,一旦平衡打破,就会引发各种疾病。近些年的研究表明,口腔疾病并非某一种或几种有害菌所导致,而是口腔内微生态失衡所导致的。在口腔各组织器官如牙齿、龈沟、颊舌部、软硬颚等软组织表面,人们发现了多达600余种细菌、病毒、真菌、支原体及衣原体等微生物,这些微生物统称为口腔微生物组。口腔微生物组生态失衡不仅可诱发多种口腔疾病,如龋齿、牙龈炎、咽喉肿痛、中耳炎、念球菌感染、鼻窦炎等,还与心血管系统疾病、呼吸系统疾病、肿瘤、糖尿病、类风湿性关节炎、早产、神经系统疾病等全身疾病紧密相关。
口腔是整个消化道的起点,因此口腔健康至关重要。保持个人口腔卫生和抑菌剂的使用是预防口腔致病菌的重要手段,然而口腔致病菌寄居在口腔牙菌斑中,牙菌斑的封闭结构保护了牙菌斑中的致病菌不受外界影响,因此那些在牙菌斑中寄居的致病菌难以通过保持口腔卫生、抑菌剂等手段进行有效抑制。变形链球菌是口腔天然菌群中占比最大的链球菌属中的一种,为牙斑的主要成分之一。变形链球菌一层一层地吸附在牙齿上,形成所谓的“生物膜”。它们消耗糖类、产生酸,腐蚀牙齿的釉层,造成牙洞。细菌/牙菌斑堆积引起牙龈发炎,如果不治疗可演变成牙周炎:细菌伸入牙齿根部的骨头并破坏它,最终造成牙齿脱落。
现有的针对口腔健康的益生菌固体饮料,需要在水中冲调后食用,益生菌通过水溶液直接进入胃部,在胃肠道中定殖,由于在口腔停留时间短,无法直接定殖于牙齿表面及唾液中,对口腔的调节作用有限;若直接倒入口中食用,在口腔中溶解性不佳,粉体吸水后成坨状,口感较差,且益生菌无法快速释放,亦无法起到良好的口腔定殖效果。此外有研究表明,现有的针对口腔健康的益生菌对咽喉健康同样具有良好预后作用,因此发明一个调节口腔和咽喉健康的益生菌速溶固体饮料具有较为重要的意义。

本发明的目的在于提供一种调节口腔和咽喉健康的益生菌速溶固体饮料及其制备方法,从而克服上述现有技术中的缺陷。
为实现上述目的,本发明提供了一种调节口腔和咽喉健康的益生菌速溶固体饮料,其重量份组分为:益生菌5份、草本原料10份、益生元30份、糖醇30份、水果粉20份、食用香精2份、柠檬酸2份、柠檬酸钠1份。
优选地,技术方案中,益生菌包括植物乳杆菌CCFM8724、副干酪乳杆菌ET-66、唾液乳酸杆菌AP-32TM、唾液链球菌Blis K12中的至少一种。

一种调节口腔和咽喉健康的益生菌速溶固体饮料的制备方法,其步骤为:(1)将草本原料、益生元、糖醇、水果粉、食用香精、柠檬酸、柠檬酸钠分别过20-40目筛;
(2)步骤(1)中过筛的原料按照重量份称取草本原料10份、益生元30份、糖醇30份、水果粉20份、食用香精2份、柠檬酸2份、柠檬酸钠1份,以等量递增法将上述过筛后的原料混合均匀;
(3)将步骤(2)所得混合原料投入混合机,在混合频率10-20r/min速度下,混合35分钟,确保原料混合均匀,无肉眼可见杂质、抱团;
(4)将步骤(3)中所得混合原料装入倒锥形流化室,在风机频率20-60HZ、内枪压力0.1-0.5Mpa、外枪压力0.2-0.5Mpa、进水转速20-300rmp条件下,以水为粘合剂,采用沸腾造粒工艺进行造粒;
(5)待物料沸腾状态变缓,到达造粒临界点时,关闭粘合剂蠕动泵,并对物料进行加热烘干,加热温度为40-60℃,加热后物料水水分≤3%;
(6)待物料冷却至室温,过筛整粒,筛选20目合格颗粒,分装并做好标识待用;
(7)称取5份重量份的益生菌,将益生菌粉与步骤(6)所得合格造粒体按照等量递增法2-4次投入混合机,在混合频率10-20r/min速度下进行混合,混合35分钟,确保原料混合均匀,无肉眼可见杂质、抱团;
(8)用复合膜包装机分装成2g/袋,产品采用铝箔复合膜或者聚乙烯药品包装用复合膜包装。

实施例1
一种调节口腔和咽喉健康的益生菌速溶固体饮料,其重量份组分为:植物乳杆菌CCFM8724和副干酪乳杆菌ET-66混合物5份、罗汉果粉10份、低聚果糖30份、麦芽糖醇30份、柠檬粉20份、柠檬香精2份、柠檬酸2份、柠檬酸钠1份。

一种耐胃酸的养胃益生菌固体饮料,其重量份组分为:唾液乳酸杆菌AP-32TM 5份、蒲公英粉和胖大海粉混合物10份、低聚半乳糖30份、木糖醇30份、西柚粉20份、玫瑰香精2份、柠檬酸2份、柠檬酸钠1份。
一种调节口腔和咽喉健康的益生菌速溶固体饮料的制备方法,在D级洁净生产区、温度18-25℃、相对湿度RH≤50%环境下进行,其步骤为:(1)将蒲公英粉和胖大海粉混合物、低聚半乳糖、木糖醇、西柚粉、玫瑰香精、柠檬酸、柠檬酸钠分别过20-40目筛;
(2)步骤(1)中过筛的原料按照重量份称取蒲公英粉和胖大海粉混合物10份、低聚半乳糖30份、木糖醇30份、西柚粉20份、玫瑰香精2份、柠檬酸2份、柠檬酸钠1份,以等量递增法将上述过筛后的原料混合均匀;
(3)将步骤(2)所得混合原料投入混合机,在混合频率10-20r/min速度下,混合35分钟,确保原料混合均匀,无肉眼可见杂质、抱团;
(4)将步骤(3)中所得混合原料装入倒锥形流化室,在风机频率20-60HZ、内枪压力0.1-0.5Mpa、外枪压力0.2-0.5Mpa、进水转速20-300rmp条件下,以水为粘合剂,采用沸腾造粒工艺进行造粒;

实施例3

对比试验
试验过程:选择健康成年人,男女比例1:1,食用本发明的产品进行口腔日常护理,调研口腔中变异链球菌、总生菌、厌氧益生菌及兼性厌氧益生菌的变化。
受试对象:100位健康成年人(18岁以上)。
试验方法:
1.将100位健康成年人群分成2组,分别为安慰剂组50人(A)和测试组50人(B);
2.A组食用麦芽糊精,B组食用本发明实施例1提供的速溶益生菌固体饮料,连续食用四周(28d),每天分为早中晚三次食用,于第4周停止服用,样本数据第0、2、4、6周采集一次;
3.收集受试者牙齿菌样本:受试者以棉花刮棒刮取每颗牙齿内外侧,作为牙齿细菌采样样本。测试受试者牙齿及口腔中变形链球菌、总生菌、厌氧益生菌及兼性厌氧益生菌数量,具体结果如表1、表2、表3、表4。

由以上数据可以看出,安慰剂组食用麦芽糊精后,口腔中的变形链球菌、总生菌、厌氧益生菌及兼性厌氧益生菌数量均没有显著变化;测试组在每日早中晚三次食用速溶益生菌固体饮料后,0-4周口腔中变形链球菌数量及总生菌数量显著下降(p<0.01),厌氧益生菌及兼性厌氧益生菌数量显著上升(p<0.01);停止食用速溶益生菌固体饮料2周后(第6周),变形链球菌数量及总生菌数量较第0周显著下降(p<0.01),厌氧益生菌及兼性厌氧益生菌数量较第0周显著上升(p<0.01)。由此数据可说明,本发明中的益生菌速溶固体饮料可以有效减少口腔中变形链球菌及总生菌数量,增加口腔中厌氧益生菌及兼性厌氧益生菌数量,帮助平衡口腔微生物组,降低口腔微生物组引起的口腔健康问题风险;且停止食用后,口腔中变形链球菌及总生菌数量依旧显著降低(p<0.01),厌氧及兼性厌氧益生菌数量依旧显著上升(p<0.01),说明本发明中的益生菌速溶固体饮料能够使益生菌在口腔中起到良好的定植作用。

2.根据权利要求1所述的调节口腔和咽喉健康的益生菌速溶固体饮料,其特征在于:益生菌包括植物乳杆菌CCFM8724、副干酪乳杆菌ET-66、唾液乳酸杆菌AP-32TM、唾液链球菌Blis K12中的至少一种。
3.根据权利要求1所述的调节口腔和咽喉健康的益生菌速溶固体饮料,其特征在于:草本原料包括蒲公英粉、罗汉果粉、金银花粉、甘草粉、胖大海粉中的至少一种。

6.根据权利要求1所述的调节口腔和咽喉健康的益生菌速溶固体饮料,其特征在于:水果粉包括柠檬粉、青桔粉、西柚粉中的至少一种。
7.根据权利要求1所述的调节口腔和咽喉健康的益生菌速溶固体饮料,其特征在于:食用香精包括柠檬香精、青桔香精、薄荷香精、桃香精、玫瑰香精、荔枝香精、蓝莓香精、蔓越莓香精中的至少一种。
8.一种调节口腔和咽喉健康的益生菌速溶固体饮料的制备方法,其步骤为:(1)将草本原料、益生元、糖醇、水果粉、食用香精、柠檬酸、柠檬酸钠分别过20-40目筛;
(2)步骤(1)中过筛的原料按照重量份称取草本原料10份、益生元30份、糖醇30份、水果粉20份、食用香精2份、柠檬酸2份、柠檬酸钠1份,以等量递增法将上述过筛后的原料混合均匀;
(3)将步骤(2)所得混合原料投入混合机,在混合频率10-20r/min速度下,混合35分钟,确保原料混合均匀,无肉眼可见杂质、抱团;
(4)将步骤(3)中所得混合原料装入倒锥形流化室,在风机频率20-60HZ、内枪压力0.1-0.5Mpa、外枪压力0.2-0.5Mpa、进水转速20-300rmp条件下,以水为粘合剂,采用沸腾造粒工艺进行造粒;
(5)待物料沸腾状态变缓,到达造粒临界点时,关闭粘合剂蠕动泵,并对物料进行加热烘干,加热温度为40-60℃,加热后物料水水分≤3%;
(6)待物料冷却至室温,过筛整粒,筛选20目合格颗粒,分装并做好标识待用;
(7)称取5份重量份的益生菌,将益生菌粉与步骤(6)所得合格造粒体按照等量递增法2-4次投入混合机,在混合频率10-20r/min速度下进行混合,混合35分钟,确保原料混合均匀,无肉眼可见杂质、抱团;
(8)用复合膜包装机分装成2g/袋,产品采用铝箔复合膜或者聚乙烯药品包装用复合膜包装。
9.一种根据权利要求8所述的调节口腔和咽喉健康的益生菌速溶固体饮料的制备方法,其特征在于:步骤(1)-(8)均在D级洁净生产区、温度18-25℃、相对湿度RH≤50%环境下进行。
