背景技术
乳腺癌是女性最常见的肿瘤疾病[Parkin D.M.,Bray F.,Ferlay J.,Pisani P.,CA Cancer J.Clin.,2005,vol.55,p.74-108]。根据世界卫生组织(WHO)的数据,每年全球乳有209万例患者,每年因乳腺癌而死亡的女性人数则为62.7万[http://www.who.int/news-room/fact-sheets/detail/cancer]。
此部位肿瘤的相当一部分是受雌激素的影响而产生的[Yue W.,Yager J.D.,WangJ.-P.,Jupe E.R.,Santen R.J.,Steroids,2013,vol.78,p.161-170.]。免疫组织化学分析揭示了表达雌激素受体(ER)、孕酮受体(PR)和HER2NEU蛋白质(其使癌具有“赫赛汀敏感性”)受体的乳腺癌。相当一部分肿瘤中带有ER、PR和/或HER2NEU 3+。如果某类型的肿瘤中不带有ER、PR,同时不具有赫赛汀敏感性(ER0,PR0,HER2NEU 0-1),就会被判定为三阴性型(ER-/PR-/HER-2-)。这是最致命的乳腺癌之一,因为它没有可抑制其生长的标靶。目前,世界上还没有治疗此型癌的有效药物。
雌激素通过血液循环以硫酸酯的形式积累在肿瘤中,这些硫酸酯无法与雌激素受体结合,但在转化为游离激素后,它们激活了肿瘤的生长。因此,一种有希望的治疗策略是采用阻碍肿瘤中游离激素的形成的抑制剂。
他莫昔芬是一种属于选择性雌激素受体调节剂的药物,它可以阻断雌激素对于包括胸组织在内的激素依赖性组织的影响。无论是对绝经前还是绝经后的妇女,他莫昔芬都是一种激素治疗的标准。由于施用他莫昔芬而产生的最典型的副作用包括:增加静脉血栓形成的风险,加重心血管疾病(包括心绞痛发作)的病程,促进子宫内膜肿瘤(包括息肉和子宫内膜癌)以及子宫纤维瘤的发展。他莫昔芬同样具有肝毒性。根据戈登·阿米顿(GordonAmidon)等在1995年提出的生物药剂学分类系统(BCS),他莫昔芬属于第二类,代表着低溶解性和高渗透性的药物。
芳香酶抑制剂已经被证明比他莫昔芬更有效。目前,来曲唑和阿那唑等芳香酶抑制剂已经被用于这一领域。它们被BCS系统归属为第一类,也就是具有高溶解度和高渗透性的药物。芳香酶抑制剂的副作用包括骨质疏松症、潮热和头疼。
除上述外,类固醇硫酸脂酶也是一个备受关注的课题,因其能从雌酮硫酸酯的的大池中局部间质生成雌激素。类固醇硫酸脂酶催化雌酮硫酸酯水解产生雌酮,以及催化脱氧表雄酮(DHEA)硫酸酯产生脱氧表雄酮(Dibbelt I.,Biol.Chem.,Hoppe-Seyler,1991,vol.372,p.173-185.;Stein C.,J.Biol.Chem.,1989,vol.264,p.13865-13872.)。
最著名的类固醇硫酸脂酶抑制剂是EMATE——雌酮氨基磺酸酯(Ahmed S.,Curr.Med.Chem.,2002,vol.9,no.2,p.263-273.)。然而,它有一个明显的缺点。伴随着自由配体的释放,具有氨基磺酸基的雌酮硫酸脂酶抑制剂会导致不可逆的酶失活[HowarthN.M.,Purohit A.,Reed M.J.J.Med.Chem.,1994,vol.37,p.219-221.]。特别是,虽然雌酮氨基磺酸酯抑制了雌酮硫酸酯酶,但释放的游离激素会导致强烈的亲子宫活性的出现[Shields-Botella J.et al.,J.Steroid Biochem.Mol.Biol.,2003,vol.84,p.327-335.]。
在WO9933858(А2)-1999-07-08中描述了一组雌酮硫酸酯酶抑制剂,其中下列通式的衍生物可作为雌酮硫酸酯酶抑制剂,用于治疗或预防雌激素依赖性疾病,如乳腺癌。
这些化合物也具有亲子宫活性。
因此,在寻找新的抗癌药物时,必须考虑到雌酮硫酸酯酶抑制剂的氨基磺酸基的载体应该不具有激素活性。
由于具备广阔的活性范围,这些要求导致基于雌激素的氨基磺酸盐的应用的扩大,特别是可用于治疗复发性乳腺癌[Shah R.,et al.,Sulfatase inhibitors forrecidivist breast cancer treatment:A chemical review,Eur.J.Med.Chem.,2016,vol.114,p.170-190.]。
现有技术还公示了一种化合物3,17αβ-二氨磺酰氧基-7β-甲基-D-增-6-氧杂-雌-1,3,5(10),8,14-五烯,将其作为一种MCF-7乳腺癌细胞的生长抑制剂(RU 2619457,公开于2017年5月16日)。可以将该化合物选为本发明的最接近的现有技术。
发明内容
本发明的技术问题是提供化合物并开发出一种有效的合成方法,这些化合物可在乳腺癌的单药治疗和辅助治疗中用作抗癌试剂,所述乳腺癌包括其最致命的类型如三阴性(ER-/PR-/HER-2-)乳腺癌。
这一问题通过提供抑制雌酮硫酸酯酶的式(1)的化合物(名为3-O-氨磺酰氧基-7β-甲基-O-增-6-氧杂雌-1,3,5(10),8(9)-四烯-17a-酮)作为用于包括三阴性型(ER-/PR-/HER-2-)乳腺癌的初始和进展型乳腺癌的单一和辅助治疗的抗癌试剂的用途而得到解决。
前述问题通过提供一种抗癌试剂的制备方法也得到了解决。该提出的方案的显著优点之一是允许将目标化合物的生产扩大到工业级规模。这一工艺涉及到7β-甲基-6-氧杂-D-增-8α-雌酮甲醚在醋酸中经氢溴酸的酸性水解,水解在氩气环境下进行,再经过乙酸乙酯萃取,水洗,通过饱和氯化钠溶液,干燥;将产生的外消旋7β-甲基-6-氧杂-D-增-8α-雌酮溶解于二甲基甲酰胺中,再加入氨基磺酰氯;反应产物经乙酸乙酯萃取后,用饱和氯化钠溶液洗涤,再使用硫酸钠干燥。
由此产生的抗癌试剂属于第五类危险品(半数致死量(LD50)>500毫克/千克)。已获得数据允许将为抗肿瘤活性研究使用的剂量分类为安全,该药物的治疗有效剂量为0.1-20毫克/千克。
HER-2/neu转基因的雌性FBV/N小鼠的乳腺肿瘤(BT)作为恶性腺瘤,表现出的特征包括雌激素受体(ER)含量低(图1)以及缺乏孕激素受体(PR)。两个恶性腺瘤亚型可加以区分,尤其是ER+,PR-和ER-,PR-[2]。对乳腺肿瘤中ERα的免疫组化分析显示,45个样本中只有10个细胞核呈单阳性染色(图1)。
图1描述了ERα抗体对乳腺肿瘤的染色。视觉表现:辣根过氧化物酶+二氨基联苯胺,放大倍数x100(A)。乳管细胞核和单肿瘤细胞细胞核的特异性染色(B),放大倍数x400。
该模型对CAF(环磷酰胺,阿霉素,5-氟尿嘧啶)治疗敏感,并具有明显的肿瘤生长抑制(TGI)作用,结果表现为平均肿瘤体积的稳定(图2)。实验中,TGI在第14天为63%,在第21天为46%。
图2描述了接受CAF治疗的小鼠在实验开始时的肿瘤平均体积。
因为动物可能会患上多种肿瘤,分析小鼠的平均总肿瘤体积(图3)以及实验开始后肿瘤体积的相对变化(图4)是合理的。肿瘤体积相对增加的曲线图显示出对照组中的该参数的线性变化,这表明了该参数适合用于评估肿瘤对治疗的生物学反应。例如,CAF疗法的治疗近3周后没有使该参数发生任何变化,这表明了疾病的稳定。在第21天,单个肿瘤稳定的发生率约为60%。
本研究的对象为采用常规分类的经过HER-2/neu转基因操作的雌性FVB小鼠,样本来自查尔斯河实验室(意大利)的老鼠银行。研究开始时,动物的年龄为21-41周,体重为25-30克。
实验使用他莫昔芬Hexal(Hexal AG,德国,83607霍尔茨基兴,批号HA1575,17年1月),代表一种药物剂型,其制备为白色或微黄色的包衣双凸圆片,表面均匀光滑,含有20毫克的活性化合物。
居住条件根据《实验动物护理和使用指南》(ILAR publication,1996,NationalAcademy Press,1996)规定的标准设置。
在适用于实验室啮齿类动物的单个T2型笼子中,动物被分组放置在单独的房间内(每组不超过5只动物)。笼子尺寸为268毫米x215毫米x141毫米(底面积370平方厘米),均配有聚碳酸酯盆,带饲料容器的不锈钢盖,以及饮水瓶分隔器。
笼垫使用无尘木屑制成。
实验动物可以不受限制地获得为实验室啮齿动物准备的颗粒饲料。
饮用水容纳在由TECNIPLAST制造的标准190毫升饮水瓶中供自由饮用,瓶子采用耐高温聚砜制造,具有硅环和AISI316不锈钢金属盖。
房间温度保持在20-26摄氏度的水平,相对湿度为50-70%。采用荧光灯人工照明,光周期设置为昼夜12:12小时。
笼子上方有一张识别卡,上面写有笼子/卡号、实验组编号、动物个体编号、动物数量和性别、研究编号、负责研究员的姓名。每个动物的耳廓夹有适用于小型实验室啮齿类动物的金属标记,由美国Kent-Scientific公司的镍合金制成(标记上有制造商冲压的三位数字)。
实验结束后使用二氧化碳对小鼠进行安乐死。所有动物尸体都经过解剖,并进行了微观描述。
药物的半数致死量在经HER-2/neu转基因操作的FVB小鼠中通过OECD 423实验进行测定(OECD化学品测试指南,急性口服毒性-急性毒性类方法)。
此项试验使用了12只雌性小鼠。
在第一步,3只小鼠按照300毫克/千克的剂量被给予药物。研究药物通过金属防损伤饲喂器灌胃(i.g.)给药(该程序相当于临床环境下的口服给药)。为进行灌胃给药,将药物的活性化合物与橄榄油混合,制成所需浓度的悬浮液,给药剂量为每10克动物体重0.1毫升悬浮液。给药剂型是现场配置的。
由于没有动物在以300毫克/千克的剂量给药后死亡,另3只动物被给予了相同剂量的药物,同样没有发生死亡。因此,以2000毫克/千克的剂量分2次进一步给药,每次3只动物。
对体重的临床观察和登记根据表一所示方案进行。动物每天检查2次,给药后60分钟对动物分别进行医学检查,前4小时给予特别关注,24小时后进行周期性检查,在第二天进行检查,然后每周一次检查,持续到第14天。每只动物都在观察者手持的笼子内被仔细检查。记录下动物的整体状况,如行为的特征,运动活动的强度和性质,皮毛和皮肤的外观。
动物的体重使用经过验证的高速电子实验室天平进行记录。天平生产商为OhausScout Pro(美国),其最大负载为2000克,测量步长为0.1克。
第15天,动物被执行安乐死,随后进行解剖分析,记录下宏观变化。这些数据被记录在一份特殊的单独尸检表格中。
在实验结束时,对所有实验动物进行病理形态学检查。对被安乐死的动物仔细检查其外部的病理变化。检查胸腔和腹腔,对内脏进行宏观分析。并没有对实验动物的器官和组织进行显微分析,是因为在整个观察期间没有动物死亡的记录。此外,宏观内脏分析未发现任何病理改变。

研究药物通过金属防损伤饲喂器灌胃(i.g.)给药(该程序相当于临床环境下的口服给药)。给药分为单次每日剂量(20毫克/千克体重)和5倍每日剂量即100毫克/千克体重。现场配置用于给药的药物的橄榄油溶液(Global Village Clasico品牌的添加有未精制的特级初榨橄榄油的精制橄榄油,批次L:183351116,有效期至2020年4月26日,西班牙BAIEO公司生产)。给药量为每10克小鼠体重0.1毫升(每只20克的小鼠给予0.2毫升的即用型制剂)。在随机分组24小时后开始给药,给药持续时间为27-28天。
参考药物他莫昔芬同样使用金属防损伤饲喂器给药(该程序相当于临床环境下的口服给药),根据临床剂量计算,每日给药剂量为4.0毫克/千克[5]。此剂量相当于20毫克/千克的人的每日剂量。将他莫昔芬药片在捣具中捣碎,再将得到的粉末使用40毫升橄榄油制备悬浮液,给药量为0.08毫升/10克小鼠体重,给药持续时间与试验药物组相同,持续27-28天。
对照组给予的是橄榄油(安慰剂)。
评价标准包括临床观察数据,动物体重,死亡时间(如果适用),肿瘤随时间的增长,病理形态学检查数据(通过尸检核实肿瘤,通过内脏器官变化的宏观表现来评估毒性效应)。
在第一次给药前记录动物体重,然后每周两次,使用的是经过验证的高速电子实验室天平。天平生产商为Ohaus Scout Pro(美国),其最大负载为2000克,测量步长为0.1克。
在称重过程中,每周一次地测量动物的宏观上可检测到的肿瘤结节。对每个结节记录两项线性尺寸,包括最大尺寸和相对其成直角的最大尺寸。肿瘤结节的最大尺寸被记录为长度(a),第二个尺寸被记录为宽度(b)。肿瘤的体积按照如下公式进行计算:V=(ab)2/2。
通过记录每只小鼠的平均肿瘤体积的变化、肿瘤生长抑制(TGI),以及平均总肿瘤体积及其随时间的变化来评估治疗的效果。
肿瘤生长抑制百分比按照如下公式进行计算:TGI=(V对照-V实验)/V对照×100(%),其中V对照表示对照组中的平均肿瘤体积,V实验表示实验组的平均肿瘤体积。
来自各个表格的主要数据被录入到微软Excel2007工作簿中。计算所有定量数据的组算数平均值(M)和标准偏差(m)。统计学分析使用GraphPad Prism6.0统计软件进行。各组间的差异采用ANOVA回归分析和费希尔精确检验进行评估。
在对急性毒性进行评估时,在第一个60分钟内对动物进行连续观察,在随后3小时内每小时观察一次,在24小时内再观察一次。在第二天进行两次观察。之后则每天进行一次观察。各动物在给药活性化合物后进行临床检查,每隔一天一次,以及每周进行一次。每只动物都在观察者手持的笼子内被仔细检查。记录下动物的整体状况,如行为的特征,运动活动的强度和性质,皮毛和皮肤的外观。
在实验过程中,当给药剂量为300毫克/千克时,没有发生动物死亡。在整个观察期间,也没有动物表现出可见的中毒迹象。动物的外观和行为同平常一样。当用手拿起时,这些动物表现出常规的微弱反应。在给药时及给药不久后,没有发现动物发出声音。
对动物的一般状况和行为反应的每日监测表明,试验药物的单次口服给药对于暴露在毒性中的实验动物的一般状况和活动没有影响。
体重见表2和表3。

由于给药剂量在300毫克/千克时没有动物死亡,随后设定为以2000毫克/千克的剂量给药。分两步给药,间隔时间为3小时,每次将浓度为100毫克/毫升的药物按照0.1毫升/10克体重的剂量给药。因为油悬浮液的浓度越高就越稠,因此排除了灌胃给药。按2000毫克/千克的剂量给药后未观察到中毒的临床迹象。无论哪一组,动物睑裂的宽度在整个观察期间几乎都没有发生变化。未检出病理性眼睛分泌物。鼻子呈粉红色,中度湿润,无病理性分泌物。所有的老鼠的皮毛都整齐而有光泽,没有秃斑。相比初始体重,第二天动物体重下降3%,第七天体重下降4%(表3)。观察期结束时,平均体重与初始体重无差异。
在第15天对这些动物实施了安乐死。经宏观分析,未发现内部器官的变化。
尸检数据如下:
动物的皮毛外观整洁,有光泽,未见秃班。动物的营养状况令人满意。
各自然孔道无病理性分泌物。
下颌下淋巴结呈圆形,浅粉红色,中度致密。唾液腺形态正常,颜色浅黄,中度致密。
腹膜光滑光亮,腔内无游离液体。
脾脏未见肿大,致密,包膜光滑有光泽。胰腺呈浅粉红色,呈叶状结构。
肝脏的大小和形状未见改变。肝包膜有光泽。肝组织呈褐色,质地中等致密。
肾脏致密,包膜光滑有光泽,肾脏周围有中等程度的脂肪组织外生长。
卵巢未见肿大,表面光亮。
胃内有折叠的有光泽的黏膜,腔内有少量粘液和食物内容物。
胸膜光滑光亮,胸腔内无游离液。胸腺呈三角形,颜色发白。肺呈浅粉色,透气。
因此,根据OECD 243测试,该药物可归入第五类或非定级类,其半数致死量大于或等于5000毫克/千克,相当于根据GOST 12.1.007-76的低危害化合物类别。
动物令人满意地耐受给药,未观察到中毒的临床迹象。在给药第一周结束时,动物体重有所下降。对照组也是如此,这可能是口服给药所带来的压力导致的。随后观察到体重增加,这可能也与小鼠的肿瘤淋巴结的体积的增加有关。
在实验结束时的尸检后,没有发现内脏器官受到毒性损害的具体迹象。在所有小鼠组中,在小肠上部区域的壁上存在脂肪浸润,这可能是油给药导致的。

实验开始时肿瘤平均体积的变化结果见图5和表5。
在整个观察期间,所有组的平均肿瘤体积都在增加。在第27天,20毫克/千克和100毫克/千克的组的TGI分别为9%和21%。该活性与参考药物他莫昔芬相似,后者在观察期结束时TGI为35%(与对照组相比p<0.05)。

如观察期结束时测量的,观察期间出现的新肿瘤的平均体积在组间没有统计学上的显著差异,但各实验组该参数均有下降趋势(表6)。因此,在对照-1组中,新生肿瘤的平均体积为0.33±0.08立方厘米;在20毫克/千克组为0.27±0.05立方厘米;在100毫克/千克组为0.20±0.05立方厘米,与对照组相比分别降低了18%和39%。他莫昔芬-1组的该参数为0.16±0.05立方厘米,比对照组低52%。
在实验第27天,除他莫昔芬-1组的TGI存在统计学显著性,实验组和对照组的测量参数均无统计学显著性的差异。在以100毫克/千克剂量的药物治疗的各组中,肿瘤生长速度逐渐下降。

疾病稳定百分比(%)是一个与RECIST标准相似的参数,反映肿瘤体积到观察期结束时相比治疗开始时的增加量未超过20%的肿瘤的发生率。
在实验的第1天和第6天,实验组的肿瘤平均体积均大于对照组,而在第13天至第27天出现了下滑(表7,图6),这证实了剂量为100毫克/千克的当前药物与参考药物他莫昔芬的抗肿瘤作用。

对小鼠平均肿瘤总体积相对变化的评估表明,该参数在对照组中呈线性变化(对照-1组),并不断增加。在所有实验组中,包括采用参考药物治疗的组(表7,图8),这一参数都出现了具有统计学显著性的降低,这证实了目前正在研究的药物对肿瘤生长有调节作用,尽管它没有导致肿瘤总数的减少。

为了评估药物的激素作用,测量了无子宫角的子宫体的重量(表9)。该器官的重量与对照组相比没有差别,这意味着药物对子宫并无激素作用。

结果表明,该药物在抗肿瘤活性方面与参考药物相似。
在经过HER-2/neu转基因操作的雌性FBV/N小鼠的多乳腺肿瘤模型上测试的药物TGI有效性,在20毫克/千克和100毫克/千克的剂量下,分别为9%和21%。该活性类似于参考药物他莫昔芬(4.0毫克/千克),其TGI在观察期结束时为35%。
结果表明,目前的药物没有亲子宫活性。

具体实施方式
通过下述公开的执行发明的实施例对本发明进行进一步的说明。
实施例1
式(1)的3-O-氨磺酰氧基-7β-甲基-D-增-6-氧杂-8α-雌-1,3,5(10)-三烯-17a-酮的合成。
在室温下,向300毫克式(2)的外消旋7β-甲基6-氧杂-D-增-8α-雌酮的甲基醚的15毫升冰醋酸的溶液中,加入2毫升40%氢溴酸。将获得的反应混合物煮沸7小时,随后倒入水中,再用乙酸乙酯萃取。将有机层进行水洗,通过饱和氯化钠溶液,再使用硫酸钠干燥。使用旋转蒸发器对溶剂进行蒸馏。产率为174毫克(62%)的外消旋7β-甲基-D-增-6-氧杂-8α-雌酮(式3),其熔点为231-232摄氏度(根据WO2009059806,熔点为230-231摄氏度)。
元素分析,%:碳76.14;氢8.17。C19H24O3。计算,%:碳75.97;氢8.05。
将100毫克式(3)化合物的1毫升的二甲基甲酰胺溶液中,加入12毫克的氨基磺酰氯。2小时后,将反应混合物倒入6毫升饱和氯化钠溶液,再用乙酸乙酯萃取所得反应产物。把有机相利用饱和氯化钠溶液搅拌,再使用硫酸钠干燥。产率为93毫克(74%)的式(1)的目标产物,熔点,195-197摄氏度。核磁共振光谱碳-13(DMSO-d6),δ,ppm:214.9,153.5,149.8,125.6,130.6,115.0,111.7,71.6,47.4,44.8,40.7,37.9,34.8,32.9,27.4,26.9,24.7,20.0,19.6。质谱,m/z(Irel,%):379(100),331(18),316(6),305(11),278(62),270(72),251(48),225(32),211(43),199(55),185(22),171(30)。元素分析,%:碳60.12;氢6.48;氮3.56。C19H25NO5S。计算,%:碳60.14;氢6.64;氮3.69。
实施例2
将细胞接种到卡雷尔瓶,每瓶50x104个细胞。为研究硫酸酯酶抑制条件下细胞的增殖活性,在接种24小时后,用含有硫酸酯酶抑制剂的培养基替换原培养基,使其最终浓度为50微克/毫升,然后将这些肿瘤细胞系进行不同时间(24-72小时)的培养。将抑制剂溶于二甲基亚砜(DMSO)。培养基中DMSO的终浓度不超过0.5%。为了排除DMSO的细胞毒性作用,还制备了不含硫酸酯酶抑制剂的含DMSO的对照样品。为了排除化合物的非特异性有害影响,还使用了正常的人类皮肤成纤维细胞。然后用维尔烯-胰蛋白酶溶液(Biolot)分离细胞,将其置于包含新鲜完全培养基的卡雷尔小瓶中。当未处理对照细胞达到培养瓶表面积单位最大细胞密度(单层)时,细胞进行计数,它们的数量设定为100%。所有暴露于硫酸酯酶抑制剂的研究的细胞培养物的增殖活性会进行三次测定。
获得的氨基磺酸酯对传代人MCF-7细胞培养物(乳腺腺癌)增殖的影响的研究表明,20微克/毫升浓度的式(1)化合物完全阻断了肿瘤细胞的增殖,但它对不具备雌激素受体的人类皮肤成纤维细胞的生长并没有产生影响。肿瘤细胞的增殖被抑制的程度与在他莫昔芬作用下相仿,他莫昔芬已在临床实践中使用了30多年。这是非常重要的,因为氨基磺酸酯和他莫西芬有不同的作用机制。实验数据采用“STATISTICA 5.0”统计软件包进行分析。

研究表明,要求保护的试剂没有亲子宫活性,具有降低胆固醇的作用,但不提高血清中甘油三酯的水平。后者很重要,因为血清中高甘油三酯水平与心血管疾病的风险相关。
对经HER-2/neu(ER-/PR-/HER2+)转基因操作的携带乳腺肿瘤的FVB小鼠的实验表明,该试剂抑制肿瘤生长的程度与得到临床批准的他莫昔芬相同,这表明它们可以联合用于治疗乳腺癌(可能会降低副作用)。
在三阴性乳腺癌的MDA-MB-231细胞系上,该化合物也抑制了细胞生长,半抑制浓度为4.8μM,与得到临床批准的化疗药物依托泊苷类似。
结果表明,与未受治疗组相比,使用本药物对已发生腋淋巴结转移扩散的患者进行化疗,能可靠地将无复发生存率增加11%,与他莫昔芬治疗组相比,可提高总体生存率。
在具有三阴性乳腺癌的传代人类肿瘤的动物模型上,该药物显示出比获得临床批准的他莫昔芬和来曲唑之类的芳香化酶抑制剂更好的效果。
工业实用性
实验结果表明,本化合物具有抗三阴性乳腺癌的活性,无毒,具有降胆固醇作用,对血清中甘油三酯水平无影响,对子宫内膜无不良影响,无对于子宫的作用。
鉴于上述特性,本化合物可用于:对绝经后女性早期激素阳性乳腺癌的单药治疗和辅助治疗;对三阴性乳腺癌的治疗;对已接受过2-3年他莫昔芬治疗的绝经后女性的乳腺癌的单药治疗和辅助治疗;对晚期乳腺癌的治疗。
Claims (3)
2.根据权利要求1的用途,其中使用3-O-氨磺酰氧基-7β-甲基-D-增-6-氧杂-8α-雌-1,3,5(10)-四烯-17a-酮对患有早期或晚期乳腺癌以及接受过2-3年他莫昔芬治疗的女性患者进行口服给药。
3.如权利要求1所述的抗癌试剂的制备方法,该方法包括在醋酸中经氢溴酸对7β-甲基-6-氧杂-D-增-8α-雌酮甲基醚进行酸性水解,其特征在于,在氩气环境下进行水解,经乙酸乙酯萃取,经水和饱和氯化钠溶液搅拌,干燥;产生的外消旋7β-甲基-6-氧杂-D-增-8α-雌酮经二甲基甲酰胺溶解后加入氨基磺酰氯;再用乙酸乙酯萃取反应产物。
