背景技术
血友病B是世界上最常见的遗传性出血性疾病之一,其是由缺乏因子IX(FIX)所引起的,该缺乏可能是由于因子IX蛋白的合成减少或活性降低的缺陷分子造成的。这会导致体内和体外凝血活性降低,并且需要在受影响个体的整个生命中进行广泛的医学监测。如果没有有效的预防措施,复发性关节积血会导致进行性和致残性关节病的发展以及较差的生活质量(Giangrande P.,Expert Opin Pharmacother.2005;6:1517-24)。
可以通过用富含因子IX的外源因子浓缩物替代缺失的凝血因子来治疗血友病B。然而,从血液中产生这样的浓缩物充满了技术困难。尽管从血浆中纯化FIX(血浆衍生的FIX;pdFIX)几乎只产生活性因子IX,但这种从血浆中纯化因子IX是非常困难的,这主要是因为因子IX在血浆中的浓度很低(Andersson,Thrombosis Research 7:451 459(1975))。另外,从血液中纯化需要去除或灭活传染源,例如HIV和HCV。此外,pdFIX具有较短的半衰期并且因此需要经常向患者给药,从而降低了对预防性治疗的依从性。
重组因子IX(rFIX)也是可行的,但与pdFIX具有相同的较短的半衰期和需要频繁给药(例如,每周2-3次进行预防性治疗)的问题。此外,本领域中需要在异源系统中有效表达的FIX序列。
发明内容
本发明的目的在于提供编码因子IX的修饰的多核苷酸,以用于最佳表达人因子IX,进而实现更好的治疗效果。

本发明的一个优点是提供一种在受试者中增加编码因子IX的修饰多核苷酸的表达的方法,包括向有需要的受试者施用本发明的载体,其中相对于包含参考核酸分子的载体,本发明的修饰的多核苷酸的表达增加,并且在受试者的治疗中始终获得比此类参考核酸分子好得多的治疗效果。

图5A-图5B显示了7个构建体以两种载体剂量对小鼠眶后(retro-orbital)给药后的FIX表达情况。如图5B所示,pXLLY14和pXLLY14-5X的表达量显著高于其他6个载体的表达量。
图6A-图6C显示了3个构建体以两种载体剂量在两个时间点对小鼠眶后(retro-orbital)给药后的FIX表达情况。pXLLY027和pXLLY027-5X的表达量显著高于其他两个载体的表达量(图6B:注射后14天;图6C:注射后21天)。
图7A-图7B显示了图3所示的5个构建体包装入AAV8衣壳后,以两种载体剂量(5.0x109 vg/小鼠和2.0x1010 vg/小鼠)在两个时间点(第14天(图7A)和第21天(图7B))对小鼠眶后(retro-orbital)给药后的血浆FIX表达情况。pXLLY027和pXLLY105的表达量显著高于pXLLY11。

实施例1.通过GENSCRIPT OPTIMUMGENETM进行密码子优化
使用GENSCRIPT OPTIMUMGENETM密码子优化技术(GenScript Corp.,New Jersey,USA)对因子IX核苷酸序列进行密码子优化。GENSCRIPT OPTIMUMGENETM密码子优化技术描述于Burgess-Brown et al.,Protein Expr Purif.59(1):94-102(2008)。
密码子使用根据人类偏好进行调整,人类密码子适应指数(codon adaptionindex,CAI)从0.72(野生型人因子IX)变为0.87(密码子优化的因子IX)。G/C含量从41.17%增加到51.37%,并且去除了60bp窗口中G/C含量的峰值。此外,优化的因子IX序列经过调整,避开了多个位点,其包括mRNA二级结构、隐蔽剪接位点、未成熟的PolyA位点、内部chi位点、核糖体位点、CpG岛、RNA不稳定基序、重复序列(同向重复、反向重复和Dyad重复),以及可能干扰克隆的限制性位点。
所获得的人因子IX基因的优化后的序列用于图1中的构建体。图1的构建体中包含了SEQ ID NO:16(cohFIX-CDS14),SEQ ID NO:19(cohFIX-CD21),和SEQ ID NO:21(cohFIX-CDS52)。
实施例2.表达构建体
图1中的所有构建体均以pXLLY11载体的骨架制造。合成并通过测序确认了pXLLY11载体(GeneScript,China),该载体包含5’反向末端重复(inverted terminalrepeat),即5’-ITR(SEQ ID NO:1)、ApoE肝调控区的增强子,即ApoE-HCR(SEQ ID NO:2)、人alpha-1抗胰蛋白酶(hAAT)启动子(SEQ ID NO:3)、修饰的SV40内含子(SEQ ID NO:4)、Kozak序列(SEQ ID NO:5)、SV40晚期polyA信号序列(SEQ ID NO:7)、删除了末端解析位点(resolution site)的3’反向末端重复,即3’ΔITR(SEQ ID NO:8),以及用于选择的氨苄青霉素抗性基因。pXLLY11含有cohFIX-CDS11(SEQ ID NO:15)。图2示出了pXLLY11的图谱,并且pXLLY11的序列如SEQ ID NO:28所示。pXLLY11是一种用于血友病B临床试验的自我互补载体。采用标准分子克隆技术,所有构建体均通过DNA测序确认。
在pXLLY11骨架的基础上构建了基础载体pXLLY23,该pXLLY23使用天然密码子并驱动野生型因子IX的表达(SEQ ID NO:20)。所构建的表达构建体还包括:1)pXLLY23,其使用天然密码子并驱动野生型因子IX的表达;2)pXLLY14(载体序列为SEQ ID NO:30)、pXLLY21和pXLLY52,其分别使用密码子优化的序列cohFIX-CDS14(SEQ ID NO:16),cohFIX-CDS21(SEQ ID NO:19)或cohFIX-CDS52(SEQ ID NO:21)并驱动R338L因子IX变体的表达;3)pXLLY19和pXLLY20,其分别包括SEQ ID NO:17和SEQ ID NO:18序列,并驱动R338L因子IX变体的表达。
实施例3.重组质粒的构建
在GenScript合成并通过测序确认了质粒pAAV.TBG.PI.eGFP.WPRE.bGH(Addgene,plasmid#105535),并将其用作AAV骨架构建体。将一段包含增强子ApoE-HCR(SEQ ID NO:10)、hAAT启动子(SEQ ID NO:11)、5’UTR(SEQ ID NO:32)、FIX编码序列,其包含外显子1(SEQ ID NO:22),第一内含子(也称为内含子I;序列如SEQ ID NO:12所示),外显子2-8(SEQID NO:23),以及牛生长激素polyA信号序列(SEQ ID NO:13)的3893bp长的片段在克隆位点PmeI和XhoI处插入到载体pAAV.TBG.PI.eGFP.WPRE.bGH中,以产生pXLLY13,并且pXLLY13的序列如SEQ ID NO:29所示。图3示出了pXLLY13和pXLLY27的示意图。

构建体pXLLY096(SEQ ID NO:33)是在pXLLY14载体的基础上制备的,其中pXLLY096包含5’-ITR(SEQ ID NO:9)、增强子3XSERP&TTRe(SEQ ID NO:38)、TTRm&TTRm5’U启动子(SEQ ID NO:39)、MVM内含子(SEQ ID NO:42)、FIX编码序列(SEQ ID NO:47)、kozak序列(SEQ ID NO:5)、WPRE3序列(SEQ ID NO:43)、SV0LpAUE ployA信号序列(SEQ ID NO:34)、3’-ITR(SEQ ID NO:14),和用于选择的氨苄青霉素抗性基因。FIX编码序列如SEQ IDNO:47所示,其包含外显子1(SEQ ID NO:44),第一内含子(有时也称为内含子I;序列如SEQID NO:45所示),和外显子2-8(SEQ ID NO:46)。采用标准分子克隆技术,所有构建体均通过DNA测序确认。

实施例4.HepG2细胞中密码子优化的表达分析
将图4所示的5个构建体(pXLLY11、pXLLY027、pXLLY096、pXLLY105、pXLLY120)包装入AAV8衣壳后,以2种剂量(MOI=1.0x104 vg/细胞(图4A)和MOI=3.0x104 vg/细胞(图4B))转染HepG2细胞并培养,并分析了FIX的表达量。从图4A-4B可见,pXLLY027,pXLLY096,pXLLY105和pXLLY120的FIX表达量显著高于pXLLY11的表达量。
实施例5.C57BL/6小鼠中密码子优化的表达分析
使用三质粒转染法(triple transfection method)在293T细胞中生产重组AAV病毒,通过碘克沙醇梯度超速离心进行纯化,并用液滴数字PCR(ddPCR)进行滴度测定。为了确定密码子优化是否增加了因子IX蛋白的表达,提供了AAV8-pXLLY14、AAV8-pXLLY19、AAV8-pXLLY20、AAV8-pXLLY21、AAV8-pXLLY052,或作为对照的AAV8-pXLLY11和AAV8-pXLLY23,并将它们经由框后注射(retro-orbital injection)的方式转入C57BL/6小鼠。然后,通过FIXELISA监控注射后小鼠中FIX蛋白的水平。
向体重为20-35g,平均12周龄的C57BL/6小鼠框后注射AAV8-pXLLY14、AAV8-pXLLY19、AAV8-pXLLY20、AAV8-pXLLY21、AAV8-pXLLY052,或作为对照的AAV8-pXLLY11和AAV8-pXLLY23。注射液由在0.9%无菌盐水溶液中的低剂量的1.0x1011 vg/小鼠和高剂量的5.0x1011 vg/小鼠的DNase抗性颗粒组成,总体积为100μL。快速进行注射,注射完整的100μl病毒溶液不超过4-7秒。注射后密切监测小鼠两小时,或直到恢复正常活动。在注射后的第14天,通过眶后采血收集样品(如图5A所示);制备血浆并储存在-80℃以供进一步分析。
使用人F9 ELISA试剂盒(LifeSpan Biosciences,Inc),通过FIX ELISA测定法测量给药小鼠血浆中的因子IX蛋白水平。如图5B所示,与pXLLY11相比,pXLLY14在低剂量和高剂量下均显示出显著的3到10倍的FIX表达水平,这表明密码子优化的序列cohFIX-CDS14(SEQ ID NO:16)是hFIX密码子优化序列的绝佳选择。
实施例6.C57BL/6小鼠中密码子优化的表达分析

Claims (8)

1.编码人因子IX的多核苷酸,其包含如SEQ ID NO:27、SEQ ID NO:16、SEQ ID NO:26或SEQ ID NO:47所示序列,
其中,任选地,所述多核苷酸还包含位于所述编码人因子IX的多核苷酸的5’端的5’UTR序列,其中任选地,所述5’UTR序列如SEQ ID NO:32或SEQ ID NO:42所示。
2.表达盒,其包含权利要求1所述的多核苷酸,
其中任选地,所述表达盒还包含表达调控元件和一种或多种腺相关病毒(AAV)反向末端重复(ITR),
其中任选地,所述表达调控元件可操作地连接到编码人因子IX的序列,其中所述表达调控元件包含组成型、可调节的调控元件,或组织特异性表达调控元件;
其中任选地,所述表达调控元件包含以下(a)或(b)任一:
(a)hAAT启动子和/或载脂蛋白E(ApoE)HCR增强子,或者
(b)包含SEQ ID NO:39的TTRm启动子和/或包含SEQ ID NO:38的3XSERP&TTRe增强子;
其中任选地,所述表达盒还包含位于所述编码人因子IX的多核苷酸的3’端的聚腺苷酸化信号序列,任选地所述聚腺苷酸化信号序列包含牛生长激素聚腺苷酸化信号序列或SV40晚期聚腺苷酸化信号序列,其中任选地,所述聚腺苷酸化信号序列包含如SEQ ID NO:13、SEQ ID NO:7或SEQ ID NO:34所示序列;
其中任选地,所述表达盒还包含位于所述编码人因子IX的多核苷酸的5’端的Kozak序列,任选地,所述Kozak序列包含如SEQ ID NO:5或SEQ ID NO:6所示序列;
其中任选地,所述表达盒还包含位于所述Kozak序列5’端的修饰的SV40内含子序列,任选地,所述修饰的SV40内含子包含如SEQ ID NO:4所示序列;
其中任选地,所述表达盒还包含表达调控元件,所述表达调控元件包含增强子,所述增强子包含如SEQ ID NO:10,SEQ ID NO:2或SEQ ID NO:38所示序列;
其中任选地,所述表达盒还包含表达调控元件,所述表达调控元件包含启动子,所述启动子包含如SEQ ID NO:11,SEQ ID NO:3,或SEQ ID NO:39所示序列;
其中任选地,所述AAV ITR包含左ITR(例如包含SEQ ID NO:9或SEQ ID NO:1所示序列)和右ITR(例如包含SEQ ID NO:14或SEQ ID NO:8所示序列)。
3.病毒载体,其包含权利要求2所述的表达盒,
其中优选地,所述病毒载体是腺相关病毒(AAV)载体;
其中更优选地,所述AAV载体为选自以下血清型的AAV载体:AAV1、AAV2、AAV3、AAV4、AAV5、AAV6、AAV7、AAV8、AAV9、AAV10,或AAV11;
其中最优选地,所述AAV载体为AAV8。
4.重组腺相关病毒载体,其从5’至3’方向包含以下元件:
(a)第一AAV2 ITR(例如如SEQ ID NO:9所示序列),
(b)ApoE HCR-1增强子(例如如SEQ ID NO:10所示序列),
(c)hAAT启动子(例如如SEQ ID NO:11所示序列),
(d)5’UTR序列(例如如SEQ ID NO:32所示序列),
(e)编码人因子IX的多核苷酸,所述多核苷酸包含如SEQ ID NO:26或SEQ ID NO:27所示序列,
(f)牛生长激素聚腺苷酸化信号序列(例如如SEQ ID NO:13所示序列),和
(g)第二AAV2 ITR(例如如SEQ ID NO:14所示序列);
其中任选地,所述重组病毒载体为pXLLY027,其序列为SEQ ID NO:31所示;
或者重组腺相关病毒载体,其从5’至3’方向包含以下元件:
(a)第一AAV2 ITR(例如如SEQ ID NO:1所示序列),
(b)ApoE HCR-1增强子(例如如SEQ ID NO:2所示序列),
(c)hAAT启动子(例如如SEQ ID NO:3所示序列),
