本发明涉及动物饲料技术领域,更具体的说是涉及一种提高仔猪抗氧化活性和机体免疫力的饲料及制备方法。
背景技术
随着人民生活水平的提高,人民对健康有品质的生活的追求日益增加,健康、口感好、营养高的猪肉倍受青睐。但猪生长较慢,出栏周期较长,且在养殖过程中容易因季节变化、缺乏营养、细菌感染等原因而造成食欲不振、消化不良、患病、甚至死亡,造成巨大经济损失。
仔猪成活率低,一直是困扰养猪业健康发展的重要因素之一,更是反映集约化养猪场生产水平的一个重要指标,直接影响到集约化养猪场的经济效益。仔猪的生长发育快、消化系统发育不全、免疫力低下、体温调节能力差等生理特点,同时哺乳期仔猪自身生理条件及环境因素的影响,常导致哺乳仔猪大量死亡,给养殖生产带来严重损失。
因此,如何提供一种适用于仔猪食用,且能够提高仔猪成活率的饲料是本领域技术人员亟需解决的问题。

有鉴于此,本发明提供了一种提高仔猪抗氧化活性和机体免疫力的饲料及其制备方法。

有益效果:本发明中的饲粮复合NRC(2012)营养需要标准,配制成颗粒料后进行饲喂。能够显著降低断奶仔猪腹泻率,改善机体抗氧化活性,同时显著线性提高血清IgA的含量,提高仔猪机体免疫,进而提高仔猪饲养成活率。
优选的,所述甜叶菊提取物中包括以下重量分数的化合物:绿原酸及其类似物质43.3%,奎尼酸1.3%,咖啡酸0.3%,咖啡酸乙酯0.6%,芦丁1.5%,槲皮素0.5%,水分3.40%和粗灰分4.70%。
优选的,所述绿原酸及其类似物质中包括以下质量分数的化合物:异绿原酸A16.66%,异绿原酸B1.62%,异绿原酸C 7.63%,绿原酸13.69%、新绿酸2.00%,隐绿原酸1.68%。

一种提高仔猪抗氧化活性和机体免疫力的饲料的制备方法,包括以下步骤:
(1)按比例称取所有原料,备用;
(2)将所述甜叶菊提取物与预混料混合均匀后,再依次加入玉米、膨化玉米、膨化大豆粉、发酵豆粕、豆粕、鱼粉、低蛋白乳清粉、乳清浓缩蛋白、蔗糖、柠檬酸钙、磷酸氢钙、氯化钠、氨基酸和氯化胆碱混合均匀,然后将物料粉碎至粒径小于0.7mm,备用;
(3)将所述大豆油与步骤(2)中粉碎后的原料加入到混合机内,混合120s,混合变异系数≤2.5%,得到混合料;
(4)将步骤(3)中所述混合料送入调制器中蒸汽熟化,得到混合物;

经由上述的技术方案可知,与现有技术相比,本发明公开提供了一种提高仔猪抗氧化活性和机体免疫力的饲料,其原料配合适用于仔猪消化,能够显著降低断奶仔猪腹泻率,改善机体抗氧化活性,同时显著线性提高血清IgA的含量,提高仔猪机体免疫,进而提高仔猪饲养成活率。

其中,甜叶菊提取物中包括以下重量分数的化合物:绿原酸及其类似物质43.3%,奎尼酸1.3%,咖啡酸0.3%,咖啡酸乙酯0.6%,芦丁1.5%,槲皮素0.5%,水分3.40%和粗灰分4.70%。

一种提高仔猪抗氧化活性和机体免疫力的饲料,与实施例1不同的是,其中的甜叶菊提取物添加量为0.05份。
对比例4
一种提高仔猪抗氧化活性和机体免疫力的饲料,与实施例1不同的是,其中不包括甜叶菊提取物,同时添加0.02份VE。
技术效果:
选用平均初始体重为6.80±0.05kg的21日龄断奶仔猪(杜×长×大)252头,采用完全随机试验设计,分为7个处理组,每组6个重复,每个重复6头猪,公母各半,依次利用实施例1-3和对比例1-4中的饲料喂食。
其中,对比例1中的饲料(风干基础)营养水平如表1所示

饲喂方法如下:
试验猪舍内安装红外保温灯,将猪舍内环境温度维持在26~28℃。每个重复6头猪在同一个圈内,共用一个乳头饮水器及料槽。试验猪每天饲喂3次(7:00,12:00,17:00),添料量以料槽内有少量剩料为准,每天准确记录每个重复的饲料添加、剩余、损耗量及料槽内清理的霉变料量。试验开始及整个试验期间每两周进行一次体重称量,试验为期42d。试验前,清洗料槽和水槽,对猪舍进行彻底消毒,对试验猪进行常规免疫与驱虫。所有试验猪均自由饮水和采食。观察试验猪临床状况,记录腹泻及死亡情况。
试验过程中,要求饲养管理人员认真负责,有责任心,严格按照试验要求进行饲养管理,认真填写有关表格;定期冲栏,并进行正常消毒;饲料生产按标准进行,不受潮,不结块,无霉变,不添加任何抗生素和促生长剂;饮水水质良好,无污染。
统计分析
试验数据采用Excel 2007整理后,用SPSS(18.0)软件进行统计分析,数据均以平均值±标准误表示。评估组间差异前,采用Shapiro-Wilk检验数据的正态性。当变量数据呈非正态分布时,采用Kruskal-Wallis one-wayANOVA分析并用FDR进行多重校正,当数据呈正态分布时,采用one-wayANOVA分析并用LSD进行检验。对各变量用回归分析的曲线估计模型建立对甜叶菊提取物添加水平响应的线性模型和二次曲线模型,以P<0.05为差异显著,0.05≤P<0.1表示有显著趋势。
1)饲料对仔猪生产性能影响试验:
在试验开始、试验每两周和试验结束时以个体为单位对所有试验猪进行空腹称重,试验过程中准确记录每个重复的采食量,记录猪只的腹泻情况及死亡情况,以计算平均日增重、平均日采食量、料肉比、腹泻率和死亡数。腹泻率:由两位固定人员在试验期每天早晚两次观察仔猪粪便情况,记录每个重复腹泻仔猪头次。腹泻评分参考表2,当腹泻评分为2或以上,认为仔猪发生腹泻,腹泻评分见下表2。ADFI=试验期采食量/(试验天数×仔猪头数);ADG=试验期增重/试验天数;F/G=ADFI/ADG。腹泻率(%)=总腹泻头数/(仔猪头数×试验天数)×100%。

试验结果:
如表3-4所示,在1~28d,断奶仔猪料重比随甜叶菊提取物添加量增加呈现显著的二次关系(P<0.05),即先降低后升高,其中甜叶菊提取物添加水平达到300mg/kg时,料重比最低。并且,甜叶菊提取物的添加可显著降低(P<0.05)断奶仔猪腹泻,尤其在甜叶菊添加量达到500mg/kg组腹泻率最低。
在1~42d,料重比随甜叶菊提取物添加量的增加有二次关系的趋势(P=0.050),在添加量为200mg/kg时达到最低。甜叶菊提取物可显著降低断奶仔猪腹泻率(P<0.05)。

2)饲料对仔猪抗氧化能力影响
总抗氧化能力(TAOC)采用ABTS法测定(货号:A015-2-1),丙二醛(MDA)采用TBA法测定(货号:A003-2),过氧化氢酶(CAT)采用钼酸铵法测定(货号:A007-1-1),总超氧化物岐化酶(T-SOD)活性采用羟胺法测定(货号:A001-1),谷胱甘肽过氧化物酶(GSH-Px)活性采用二硫代二硝基苯甲酸法测定(货号:A005),各试剂盒均购自南京建成生物工程研究所,参照试剂盒说明书测定。D-乳酸(D-LA)采用双抗体夹心法测定、脂多糖(LPS)采用双抗体夹心法测定、脂多糖结合蛋白(LBP)采用双抗体夹心法测定,以上试剂盒均购自北京方程生物科技有限公司,参照试剂盒说明书测定。8-羟基脱氧鸟苷(8-OHdG)采用双抗体夹心法测定(货号:ml320071),试剂盒购自上海酶联生物科技有限公司,参照试剂盒说明书测定。
肝脏样品先用生理盐水匀浆离心,得10%匀浆液用于检测。肝脏匀浆液用ThermoBCAkit检测其中蛋白量,抗氧化活性均以每毫克蛋白表示。
试验结果:
如表5和6所示试验第28d,添加甜叶菊提取物及VE均显著降低(P<0.05)血清MDA含量。添加200mg~500mg/kg甜叶菊提取物及VE可显著提高(P<0.05)血清T-SOD活性,且血清T-SOD活性随甜叶菊提取物添加水平提高呈现显著的二次线性关系(P<0.05)。添加400mg/kg、500mg/kg甜叶菊提取物可显著提高(P<0.05)血清中GSH-px酶活,并且血清MDA含量、T-AOC含量、CAT活性、GSH-px活性随甜叶菊提取物添加水平提高呈现显著的线性及二次关系(P<0.05)。
试验第42d,添加甜叶菊提取物及VE均显著降低(P<0.05)血清中MDA含量。添加甜叶菊提取物及VE均显著提高血清T-SOD酶活(P<0.05)。添加100~400mg/kg的甜叶菊提取物及VE显著提高血清CAT酶活(P<0.05),且血清CAT活性与甜叶菊提取物添加水平呈现显著的二次关系(P<0.05)。添加200~500mg/kg甜叶菊提取物显著提高(P<0.05)血清GSH-px活性,并且血清MDA含量、T-AOC含量、T-SOD活性、GSH-px活性随甜叶菊提取物添加水平提高呈现显著的线性及二次关系(P<0.05)。

如表7所示,添加甜叶菊提取物及VE均可显著降低(P<0.05)肝脏MDA、8-OhdG含量。添加300~400mg/kg甜叶菊提取物及VE均可显著提高(P<0.05)肝脏CAT活性。肝脏GSH-px活性随甜叶菊提取物添加水平提高呈现显著的二次关系(P<0.05),添加300mg/kg达到最高(P<0.05),即先增高后降低。添加100~300mg/kg甜叶菊提取物显著降低(P<0.05)肝脏D-LA含量,添加500mg/kg甜叶菊提取物显著升高(P<0.05)肝脏D-LA含量。添加100~400mg/kg甜叶菊提取物显著降低(P<0.05)肝脏LPS含量。添加500mg/kg显著升高(P<0.05)LPS、LBP含量。且肝脏MDA、8-OhdG、D-LA、LBP含量及T-SOD、CAT活性随甜叶菊提取物添加水平提高呈现显著的线性及二次关系(P<0.05)。

3)饲料对仔猪免疫功能影响
血清免疫球蛋白IgA,IgG,IgM均采用北京方程生物科技有限公司试剂盒检测,参照试剂盒说明书测定。肠道粘膜炎症因子IL-1(货号:ml002322-2),IL-8(货号:ml002320-2),IL-10(货号:ml002319-2),IL-22(货号:ml694870-2),TNF-α(货号:ml002360-2),(货号:ml257170-2)采用试剂盒检测,试剂盒均购自上海酶联生物科技有限公司,参照试剂盒说明书测定。
试验结果:
如表9-11所示,添加甜叶菊提取物可显著线性提高血清IgA的含量(P<0.05),且血清IgA含量随甜叶菊提取物添加量提高呈现显著的二次关系(P<0.05)。添加甜叶菊提取物对血清IgG、IgM含量,空肠及回肠粘膜中炎症因子IL-1、IL-8、IL-10、IL-22、TNF-α均无显著影响(P>0.05)。

由以上试验例可以看出:
1)在本实验条件下,试验1~28d,断奶仔猪料重比随甜叶菊提取物添加水平提高呈现显著的二次关系,在300mg/kg时料重比最低。试验1~42d,断奶仔猪料重比随甜叶菊提取物添加水平增加有二次变化的趋势,200mg/kg时达到最低。添加甜叶菊提取物可降低断奶仔猪料重比,显著降低断奶仔猪腹泻率。
2)添加甜叶菊提取物可显著降低血清、肝脏MDA含量,特别的,添加200~400mg/kg甜叶菊提取物显著提高血清T-SOD、CAT、GSH-px活性,并降低肝脏中8-OhdG、D-LA、LPS含量。添加甜叶菊提取可显著改善机体抗氧化活性。
3)添加甜叶菊提取物可显著线性提高血清IgA的含量,对机体免疫有一定改善作用。

6.根据权利要求1-5任一所述一种提高仔猪抗氧化活性和机体免疫力的饲料的制备方法,其特征在于,包括以下步骤:
(1)按比例称取所有原料,备用;
(2)将所述甜叶菊提取物与预混料混合均匀后,再依次加入玉米、膨化玉米、膨化大豆粉、发酵豆粕、豆粕、鱼粉、低蛋白乳清粉、乳清浓缩蛋白、蔗糖、柠檬酸钙、磷酸氢钙、氯化钠、氨基酸和氯化胆碱混合均匀,然后将物料粉碎至粒径小于0.7mm,备用;
(3)将所述大豆油与步骤(2)中粉碎后的原料加入到混合机内,混合120s,混合变异系数≤2.5%,得到混合料;
(4)将步骤(3)中所述混合料送入调制器中蒸汽熟化,得到混合物;
(5)将步骤(4)中所述混合物造粒后冷却至室温,得到饲料颗粒;
(6)将步骤4所得颗粒称量包装,即完成制备。
7.如权利要求1所述的一种提高仔猪抗氧化活性和机体免疫力的饲料的制备方法,其特征在于,步骤(4)中所述蒸汽熟化温度为85℃,熟化时间为15s。
8.如权利要求1所述的一种提高仔猪抗氧化活性和机体免疫力的饲料的制备方法,其特征在于,步骤(5)中所述造粒颗粒尺寸为:直径2.3~2.4mm,长度0.7~1.0cm。
9.如权利要求1所述的一种提高仔猪抗氧化活性和机体免疫力的饲料的制备方法,其特征在于,步骤(5)中所述冷却速度为7℃/min。
