Description

Medicinal and edible herbal composition for improving intestinal absorption and immune function
Technical Field
The application belongs to the technical field of medicines, and particularly relates to a medicinal and edible herbal composition for improving intestinal absorption and immune functions.
Background
The gastrointestinal health of modern people is influenced by irregular diet under high living pressure, unreasonable dietary structure caused by dependence on takeout and spicy and hot eating, bad living habits such as overnight overdrawing of bodies, abuse of broad-spectrum antibiotics, food intake containing complex additives, pollutants in the natural environment (contact of toxic chemical substances, pesticide residues in soil, and excessive particulate matters in the atmosphere), and the like.
The gastrointestinal diseases are the most common diseases at present, the total incidence rate accounts for about 20% of the population, and the theory of 'ten nine stomach diseases' is in China. If a person cares about maintaining his gastrointestinal health, the disease is averagely developed over 800 times in a lifetime, and the health cost is a small amount. Gastrointestinal health is important to both the physical health of individuals and the harmonious development of society.
Gastrointestinal health refers to the normal physiological functions of the gastrointestinal tract including digestion, absorption and secretion, and immune function. Traditional Chinese medicine pays attention to gastrointestinal health, and ancient people indicate that the spleen and the stomach are the acquired root. The food can be interpreted by modern scientific language as food intake, transportation and digestion of the gastrointestinal tract, nutrient absorption and waste excretion, and the food is subjected to a series of complex digestion and decomposition processes in the gastrointestinal tract to become small molecular substances which are absorbed by the intestinal tract and processed by the liver to become in-vivo substances for the utilization of the tissues of the whole body. Digestive breakdown of food ingredients in the gastrointestinal tract requires enzymatic reactions involving hydrolases secreted by the pancreas, gastrointestinal glands, bile secreted by the liver, and enterobacterial enzymes. 70-80% of the human immune system is distributed in or around the intestinal tract, and modern researches have proved that the microecology of the gastrointestinal tract plays an important role in regulating the immune system. The gastrointestinal tract is distributed with a large number of flora that constitute the micro-ecology of the gastrointestinal tract. The pre-cause of the dysfunction of the gastrointestinal tract is the disruption of the microecological balance of the gastrointestinal tract. The intestinal tract of a healthy human body is communicated with the outside, and a large number of microorganisms inhabit in the intestinal tract, the types and the numbers of which are relatively stable and balanced when the human body is in a healthy state, are called normal flora or home flora, are indispensable components in the internal environment of the body, and most of the normal flora or the home flora form a symbiotic relationship with the human body in a long-term co-evolution process. On one hand, the human body selectively allows certain microorganisms to be planted in the intestinal tract and provides a proper inhabitation environment for the microorganisms; on the other hand, the microorganisms and the metabolites thereof promote the perfection and influence of the intestinal mucosa immune function and participate in various metabolic functions of the human body. The microecological state of the human gastrointestinal tract can reflect the health state of the gastrointestinal tract function to a certain extent. Disturbances of gastrointestinal tract function are the main cause of gastrointestinal tract organ pathology.
The traditional Chinese medicine and the western medicine have respective treatment advantages in the aspects of regulating the gastrointestinal function and treating the gastrointestinal diseases. Modern medicine suffers from the drawback that it is only effective for certain symptoms of certain gastrointestinal disorders, for example, gastrointestinal discomfort in the face of helicobacter pylori infection is merely bactericidal. The traditional Chinese medicine treatment is usually determined according to syndrome differentiation and typing, has an overall treatment effect on improving the gastrointestinal tract function, and has the characteristics of pertinence, individuation and comprehensiveness. Especially, the traditional Chinese medicine treatment has the effect of relieving the gastrointestinal symptoms of modern medical stranded inadequacy. Taking functional dyspepsia which is common clinically and has no special treatment effect in modern medicine as an example, the traditional Chinese medicine divides the functional dyspepsia into a liver qi attacking stomach type, a diet stagnation type, a spleen and stomach weakness type, cold and heat mutual combination and a qi non-rising type, summarizes symptom signs as gastrointestinal motility dysfunction, choking gag on the upper part, distending pain in the stomach and a constipation on the lower part; if stomach qi does not descend or rise, belching, acid regurgitation, vomiting, heartburn, etc. can occur; spleen qi failing to ascend and descend causes sinking of middle qi, resulting in a sinking and distending stomach, a poor appetite and early fullness, and constipation. Through the methods of soothing liver, regulating qi, relieving stagnation and relieving stuffiness, promoting digestion, relieving stagnation, harmonizing stomach and lowering adverse qi, strengthening spleen, tonifying qi, harmonizing stomach and eliminating dampness.
Early basic prompts of the scientific research team indicate that gastrointestinal tract health is closely related to human immunity, and gastrointestinal tract flora disorder can cause susceptibility to upper respiratory tract infection. The upper respiratory tract infection is caused by virus and bacteria invading human body, and when virus and bacteria invade human body, the immune cells such as lymphocytes and macrophages of the human body are actively mobilized to resist and phagocytize the foreign invaders, so that the course of disease is shortened. 70-80% of the human immune system is distributed in or around the intestinal tract, and gastrointestinal dysfunction affects the systemic immunity. When the viruses exceed the killing ability of lymphocytes and macrophages, the body is invaded by viruses and bacteria. The occurrence of the recurrent respiratory tract infection is directly related to the immunity of the organism, the disease resistance of the body is not strong, and the body cannot resist external pathogenic bacteria, so the recurrent respiratory tract infection is easy to occur.
Through literature search and retrospective review, the traditional Chinese medicine means for treating gastrointestinal diseases has a plurality of schemes and means, and a plurality of classic formulas of the Chinese medicine means are not lacked. For example, Huoxiang Zhengqi pills, which are well known by common people, are commonly used for treating gastrointestinal diseases. The traditional Chinese medicine compositions are found to have certain unsafe factors, are still exemplified by the Huoxiang Zhengqi pills, and the common dosage form of the Huoxiang Zhengqi pills is Huoxiang Zhengqi liquid which contains 40 to 50 percent of alcohol and is not suitable for children and drivers. The pinellia ternate which is one of the components still has the problem of small toxicity after the dosage form improvement, and has challenges in clinical popularization and application and international trade barrier breakthrough. In addition, these classic traditional Chinese medicine compositions have pain spots which are caused by improper medication after being misused as OTC Chinese patent medicine. The traditional Chinese medicine composition has strong curative effect, namely, the traditional Chinese medicine composition has strong medicinal potency, and when the traditional Chinese medicine composition is used correctly, the effect is remarkable, and when the traditional Chinese medicine composition is not used properly, obvious side effects are easy to occur.
The invention solves the pain points and provides a medicinal and edible herbal composition. Consisting of a herbal material meeting the list of notifications about the management of further regulated health food materials (list of articles that are both food and drugs). Su Wen, Wu Chang Zheng Da Lun (plain questions and five often political affairs) considers that "the disease is treated by toxic pathogen, and six of them are removed; treating diseases with common toxicity, seven of them are removed; treating diseases with small toxicity, and removing eight of the medicines; no toxicity, ten remove nine; the cereal, meat, fruit and vegetable are eaten thoroughly. The Chinese medicament has high-efficiency and safe treatment advantages, is the basis of imported products (whether medicaments or food) and has huge development potential.

The intestinal tract is the largest digestive organ and toxin expelling organ of the human body and comprises five parts of small intestine, large intestine, anus, intestinal juice and intestinal flora. The intestinal tract has an absorption function, the small intestine is a nutrient absorber, and the large intestine has an absorption function on water liquid in food residues. The functional disorder of water absorption affects the peristalsis of intestines and stomach, and when the peristalsis of the intestines is weakened, food residues stay in the intestines for a long time, the water absorption is increased, and feces are dry, so that difficult or uncomfortable defecation can be caused. Once people see that the defecation is difficult or uncomfortable, the people are regarded as sub-health but not diseases, the defecation is often delayed and not cured, and chronic habitual constipation is formed, so that the defecation habit is not good after a long time. Pharmacological experimental research results show that after the medicinal and edible herbal composition is used for treatment, the rat intestinal tissue structure is clear, and epithelial cells are arranged in order. The feedback of pharmacological experiments shows that the constipation and other symptoms of children and adults can be effectively improved by taking the medicinal and edible herbal composition.
The intestinal tract is not only the main site for the digestion and absorption of nutrients, but also the congenital barrier for ensuring the homeostasis of the body. The important link forming the intestinal immunity is the immune barrier, the intestinal tract is the largest immune organ in vivo, the immunity of the intestinal tract is dragged to move the whole body, and the abnormal occurrence of the low immune function of the intestinal tract can cause the digestion and absorption function disorder of nutrient substances, dyspepsia, abdominal distension and pain, the enhancement of the susceptibility to pathogenic microorganisms and the like, thereby causing various diseases. Pharmacological experiment research results show that the normal group (N groups) has clear intestinal tissue structure and orderly arranged goblet cells; the intestinal tissue structure of the model group (group M) collapsed and goblet cells increased compared to the normal group; compared with the model group, the intestinal tissue of the medicinal and edible herbal composition group (KLB group) has compact structure and less goblet cell number. Goblet cell mucus secreting cells, which are closely related to glucose and amino acid uptake, are part of the mucosal barrier of the gut. The maintenance of goblet cells and epithelial cells by the medicinal and edible herbal composition indicates that the medicinal and edible herbal composition can effectively improve the immune function of intestinal tracts. Feedback of pharmacological experiments prompts that children and adults taking the medicinal and edible herbal composition can effectively improve symptoms such as dyspepsia, abdominal distension and pain.
The solid beverage prepared from the medicinal and edible herbal composition has good feedback on practical application, and takes an applicable questionnaire in Beijing area as an example. We find that the symptoms of constipation, fullness after eating and the like are improved after a user who has poor dietary habits drinks the traditional Chinese medicine granules prepared from the medicinal and edible herbal composition. The symptoms of diarrhea, oral ulcer, halitosis and the like are obviously reduced or eliminated after a person who experiences oily or spicy food takes the traditional Chinese medicine granules prepared from the medicinal and edible herbal composition to drink. The experients with fatness and poor self-induction intestinal peristalsis have regular stool, normal appetite and weight reduction tendency after taking the traditional Chinese medicine granules prepared from the medicinal and edible herbal composition.
The above medicinal materials are common foods, and have no toxicity and high safety. The medicinal and edible herbal composition has the effects of regulating intestinal flora, improving intestinal function and maintaining intestinal health. The medicinal and edible herbal composition has good regulating effect on gastrointestinal heat accumulation, poor eating habits and low immunity of children and children, has no heavy bitter taste, has sufficient natural fragrance of the herbal, and can be taken by children and children. Meanwhile, the medicinal and edible herbal composition is also suitable for people with healthy intestinal tracts, has comfortable mouthfeel and can be developed into natural herbal beverages, foods and the like.
The intestinal functions of infants and children are not completely developed, the digestive ability is weak, and in addition, the infants and children are rich in life and have a sense of pleasure, and the children often eat a large amount of hamburgers, fried chicken, chocolate, rice crust and other snacks which are easy to cause gastrointestinal heat accumulation, so that a large amount of infants and children have gastrointestinal heat accumulation symptoms, such as constipation, poor defecation habits, dyspepsia, abdominal distension and pain, halitosis, red tongue, uneasy sleep, easy cold and the like. Eating snacks breaks up normal three meals a day, frost is added on snow to gastrointestinal development, and vicious circle is involved. If parents do not select milk powder for infants, gastrointestinal heat accumulation of babies is also easily caused. Adverse environments such as air pollution also affect the gastrointestinal development of this generation of infants and children. The medicinal and edible herbal composition can treat and improve gastrointestinal heat accumulation symptoms, enhance intestinal functions of infants and children, assist intestinal development and improve autoimmunity of infants and children.
The composition can be made into decoction, extract, powder, Chinese medicinal granule, decoction, pill, powder, unguent, pellet, tablet, lozenge, colloid, yeast, dripping pill, soft capsule, superfine powder, colloid, injection, etc. The most common decoction is prepared by cleaning the traditional Chinese medicines, soaking for one hour, adding water for decocting, boiling over strong fire, decocting over slow fire for half an hour, and pouring out the filter residue. One dose is taken in the morning and at night. The composition is also suitable for infants. 1/6 for newborn children, 1/3-1/2 for infants, 2/3 for infants and toddlers, and 2/3 for children of school age.
Has the advantages that:
(1) the health-care food can achieve the effect of maintaining the health of the intestinal tract by adjusting the intestinal flora and improving the intestinal function, has the advantage of 'preventing the disease first', and maintains the health of the intestinal tract so as to improve the immunity of a human body.
(2) The pharmacological experiment result shows that: the maintenance of the medicinal and edible herbal composition on goblet cells and epithelial cells shows that the medicinal and edible herbal composition can effectively improve the absorption function and the immune function of intestinal tracts.

SPF grade SD rats 30, male, 15-day-old, 40 ± 10g in weight, purchased from sbeff (beijing) biotechnology limited, license No.: SCXK [ Kn ] 2016-. The feed is raised in an animal house of the college of traditional Chinese medicine of Beijing university of traditional Chinese medicine, the environment is shielded, the temperature is 22-25 ℃, the humidity is 50-60%, water is freely fed, and the natural illumination is performed. The animal experimental protocol was approved by the animal research committee of the university of traditional Chinese medicine, Beijing in advance.
Normal group (N group) normal diet. The model group is fed with special high-heat feed and red star milk powder solution which are uniformly purchased, produced and controlled by the Schbefu (Beijing) biotechnology limited company for 6 days to form a gastrointestinal heat accumulation model. The medicinal and edible herbal composition group (KLB group) is used for intragastric gavage of the medicinal and edible herbal composition granular aqueous solution on the basis of molding.
Milk solution: dissolving red star whole sweet milk powder in pure water to prepare 52% milk solution, wherein the stomach filling amount is 1mL/100g each time, and the milk solution is taken once a day.
Medicinal and edible herbal composition granular water solution: the medicinal and edible herbal composition granules which are equivalent to 10 grams of clinical decoction pieces of dandelion, 10 grams of raw hawthorn, 10 grams of vinegar chicken's gizzard-membrane, 10 grams of malt, 10 grams of fried radish seed, 10 grams of poria cocos and 6 grams of daidai flower are purchased from Beijing kang rentang pharmaceutical industry Co., Ltd, and are non-additive full-ingredient traditional Chinese medicine formula granules. The Chinese medicinal granule is dissolved in pure water to prepare a solution with the concentration of 1.32g/ml twice a day.
Water was withheld on day 6 at night, blood was taken from the abdominal aorta after anesthesia on day 7 morning and colon tissue was collected and fixed in 10% formalin for at least 24h for further histopathological assessment, specifically 3cm above the cecum end. Rat intestinal tissue was fixed with sufficient 10% formalin for 24h, and tissue sections were prepared by material extraction, dehydration, wax immersion, embedding, sectioning, HE staining, and the like.
Experimental study results show that the normal group (N group) has clear intestinal tissue structure, and goblet cells and epithelial cells are arranged in order; the intestinal tissue structure of the model group (group M) collapsed and goblet cells increased compared to the normal group; compared with the model group, the intestine tissue of the medicinal and edible herbal composition group (KLB group) has compact structure and less goblet cell number, and the structure is shown in figure 1. Goblet cell mucus secreting cells, which are closely related to glucose and amino acid uptake, are part of the mucosal barrier of the gut. The intestinal epithelium is the site of maximum nutrient and fluid absorption within the body and must provide a tight barrier to prevent the invasion of pathogens. The maintenance of the medicinal and edible herbal composition on goblet cells and epithelial cells shows that the medicinal and edible herbal composition can effectively improve the absorption function and the immune function of intestinal tracts.
