Description
Technical Field
The invention relates to the field of insect feeding, and particularly relates to an artificial feed for spruce flower monochamus alternatus larvae and a preparation method thereof.
Background
The spruce champion (Monochamus saltutorius) is a trunk-boring pest which mainly harms Pinus koraiensis, Pinus sylvestris, spruce and other pinaceae plants, more importantly, a main propagation medium of pine wood nematodes (Bursaphelenchus xylophilus) in intermediate temperature zones, the research reports on the champion are few at present, many related basic problems need to be researched definitely, and the method is not slow and has a great demand for testing insects.
The larvae and pupae stage of the spruce champion mainly move in xylem, the adult stage mainly moves on the upper part of the crown, and the collection difficulty is high. And forest collection is limited by climate, quantity, age and vitality, and the requirements of the test on the conditions of the age, the insect state and the like of the test longicorn cannot be guaranteed. Therefore, it is necessary to research the indoor artificial feeding method so as to provide materials for relevant experimental research.
Disclosure of Invention
Therefore, the invention provides an artificial feed for spruce flower monochamus larva and a preparation method thereof, and aims to solve the problem that the spruce flower monochamus larva cannot be further researched due to the lack of test insect materials in the prior art.
In order to achieve the above purpose, the invention provides the following technical scheme:
according to one aspect of the invention, the artificial feed for the spruce flower monochamus alternatus larva comprises Korean pine sawdust, agar, wheat germ powder, cane sugar, sodium caseinate, yeast powder, ascorbic acid, wegener salt, potassium sorbate, choline chloride and distilled water.
Further, the feed comprises 50-100 parts of Korean pine sawdust, 20-40 parts of agar, 12.5-30 parts of wheat germ powder, 10-30 parts of sucrose, 10-30 parts of sodium caseinate, 6-15 parts of yeast powder, 2-4 parts of ascorbic acid, 0.5-1 part of Webster salt, 0.5-1 part of potassium sorbate, 0.5-1 part of choline chloride and 800 parts of 400-ion water.
According to another aspect of the invention, a preparation method of an artificial feed for spruce flower monochamus alternatus larvae is provided, wherein the method comprises the following steps:
step one, processing Korean pine sawdust and then sieving;
adding distilled water and prepared red pine sawdust, agar, wheat germ powder, cane sugar, sodium caseinate, yeast powder, ascorbic acid, wechsler's salt, potassium sorbate and choline chloride into a container, and fully stirring to uniformly mix the materials;
and step three, naturally cooling the sterilized spruce flower monochamus longissimus larvae to obtain the artificial feed for the spruce flower monochamus longissimus larvae.
Further, in the first step, the method for processing the sawdust of the Korean pine is natural air drying.
Further, in the first step, the mesh number of the sieve is 20 meshes.
Further, in the third step, the high-pressure sterilization condition is 121 ℃ high-pressure sterilization pot.
Further, in the third step, the time for autoclaving is 20 minutes.
Further, in the third step, the temperature for low-temperature preservation is 4 ℃.
The invention has the following advantages:
the feed for artificially feeding spruce flower monochamus alternatus larvae, which is prepared by the invention, has low raw material cost and a simple preparation method. The survival rate of the larvae fed by the feed is up to 78.00 percent, the development time is short, the development period from egg development to eclosion is about 120 days, and the development time is obviously shortened compared with the development time under the natural environment of forests, so that the method is favorable for realizing the successive generation feeding of the spruce thyestillageblerifacldermann larvae under the artificial temperature control, and the test population supply of the spruce thyestillageblerifacldermann larvae with different insect ages and different insect states is ensured.
