Abstract

本发明公开了一种涂层支架及其制备方法,该涂层支架呈网状结构,构成网状结构的部件由三层结构组成,内层为支架层、中间层为抗凝血改性层、最外层为纳米短肽涂层;该涂层支架不仅能抗凝血,起到防止血栓的作用,而且能够促进血管细胞的快速内皮化,能有效的促进血管病变区域的细胞再生;且整个生产工艺简单、对环境无污染、成本低。

一种涂层支架及其制备方法
技术领域
本发明属于医疗器械技术领域,尤其涉及一种涂层支架及其制备方法。
背景技术
随着经济技术的发展,人们的生活水平越来越高,人们生活方式的改变,从而导致心脑血管疾病等非常频发,在目前和以后相当长的时间内心血管疾病都会是人类死亡的首要因素。当前对心血管疾病的主要治疗手段有药物治疗、外科手术治疗和介入治疗等。而血管支架介入术因其具有创伤小、术后即刻效果显著、术后可较长时间发挥效果等特点成为目前临床上应对心血管疾病常用的方法之一。在30多年的研发使用过程中,血管支架由最初的经皮球囊成形术,经历了药物洗脱支架,全降解支架以及各种新型功能型支架。然而,虽然历经更迭,目前的血管支架仍面临植入后再狭窄和晚期血栓的问题,血管支架表面改性研究任重而道远。
一般来说,心血管支架由不锈钢、钛及其合金、NiTi合金以及其它材料制成,但这些材料的血液相容性很差,植入后容易引发血栓形成,从而引发更大的风险;同时,随着现代医学技术的发展,具有促进心血管内皮细胞的快速生长已成为治疗心血管疾病的金标准。
因此,设计一种支架涂层以实现抗凝血性能与生物活性功能分子的组合,一方面能满足支架抗凝血的性能,另一方面还能保持生物活性分子的生物活性,促进血管细胞的快速内皮化,对于进一步提高支架疗效、减少并发症并改善患者的生存质量意义重大。
发明内容
本发明的目的是克服现有技术中存在的不足,提供一种涂层支架,其在介入治疗中,具有很好的抗血栓和促进心血管支架的内皮化性能。该制备工艺简单、成本低,且可用于自动化生产。大大提高了生产效率和经济成本。
为实现上述目的,本发明的技术方案是提供一种涂层支架,所述的涂层支架呈网状结构,所述的网状结构构成部件由三层结构组成,内层为支架层、中间层为抗凝血改性层、最外层为纳米短肽涂层,所述的纳米短肽涂层为不连续排列;所述的涂层支架制备方法的具体步骤如下:
(1)支架的加工:支架可经编织机编织而成,也可由管状材质经切割而成;
(2)等离子体沉积构建支架表面改性:对加工完成的支架进行氧等离子体辉光放电处理:放电功率为20-300W,处理时间范围为2-45min,氧气流量为100ml/min~800ml/min;将氧处理后的支架与多元醇或含羧酸官能团的物质进行反应;反应完成后继续进行羧基活化处理:最后再将活化后的支架进行抗凝血物质的等离子沉积改性:等离子沉积温度为10~180℃,辉光放电沉积时间为2min-60min,放电功率为50-300W;
(3)短肽涂层:对表面修饰改性的抗凝血改性层,继续进行短肽的间断不连续的喷涂,使短肽利用共价键与部分抗凝血涂层进行反应。
进一步地,所述支架网络结构的内层由直径为0.01mm~0.08mm的金属或高分子丝构成;所述涂层支架直径为1.0~30.0mm。
进一步地,所述的抗凝血改性层为肝素、聚乙二醇、磷酸胆碱、羟乙基纤维素、壳聚糖及其衍生物、白蛋白中的一种或几种的混合物。
进一步地,所述的纳米短肽涂层为具有促进血管内膜再生的纳米短肽RGD、YIGSR、REDV、CAG、SVVYGLR、RADA-16中的一种或几种混合物。
进一步地,所述的抗凝血改性层的厚度不大于200nm;所述的纳米短肽涂层的厚度为10-200nm。
与现有技术相比,本发明具有的有益效果:该涂层支架不仅能抗凝血,起到防止血栓的作用,而且能够促进血管细胞的快速内皮化,能有效的促进血管病变区域的细胞再生;且整个生产工艺简单、对环境无污染、成本低。

具体实施方式
为使本发明的上述目的、特征和优点能够更加明显易懂,下面结合附图对本发明的具体实施方式做详细的说明。在详述本发明实施例时,为了便于说明,示意图不依一般比例局部放大,不应以此作为对本发明的限定。
实施例1
如图1、2及3所示,本发明专利的涂层支架呈网络结构,其网络结构的构成部件由三层结构组成,内层为支架层、中间层为抗凝血改性层、最外层为纳米短肽涂层;所述支架网络结构的内层由直径为0.01mm的金属或高分子丝构成;所述涂层支架直径为3.0mm。
如图3、4所示,本发明专利的涂层支架的由三层结构组成,中间层为抗凝血改性层,其所述的抗凝血改性层为肝素、聚乙二醇、磷酸胆碱、羟乙基纤维素、壳聚糖及其衍生物、白蛋白中的一种或几种的混合物;所述的抗凝血改性层的厚度为10nm。所述的纳米短肽涂层为具有促进血管内膜再生的纳米短肽RGD、YIGSR、REDV、CAG、SVVYGLR、RADA-16中的一种或几种的混合物。所述的纳米短肽涂层的厚度为20nm。
本发明的涂层支架与导入鞘、输送导丝、微导管以及导引导管等配合使用,均为本领域技术人员公知的现有技术。
本发明的涂层支架具有收缩性,输送状态下收缩固定于微导管内,自由释放状态下外形如图1所示;而在病变区域血管内的释放状态如图2所示;支架在显影点的影响下,精准放置于病变区域,其中中间层的抗凝血改性层可以有效的防止动脉瘤处脑动脉血管壁的血栓形成;而不连段分布的外层纳米短肽层可以很好的促进血管壁内皮细胞的增长。
实施例2
对如图1所示的涂层支架进行如下工艺的制备:支架网络结构经编织机编织而成;首先,对加工完成的支架网络结构进行氧等离子体辉光放电处理:放电功率为100W,处理时间为30min,氧气流量为500ml/min;将氧处理后的支架与二羧酸聚乙二醇进行反应;反应完成后继续利用N-羟基丁二酰亚胺和碳二亚胺盐酸盐进行羧基活化处理:再将活化后的支架进行抗凝血物质磷酸胆碱的等离子沉积改性:等离子沉积温度为70℃,辉光放电沉积时间为45min,放电功率为300W。最后,对表面修饰改性的抗凝血改性层,继续进行短肽RGD的间断不连续的喷涂,使短肽RGD利用共价键与部分抗凝血涂层进行反应。
实施例3
对如图1所示的涂层支架进行如下工艺的制备:支架网络结构为镍钛管经切割而成;首先,对加工完成的支架网络结构进行氧等离子体辉光放电处理:放电功率为20W,处理时间为30min,氧气流量为100ml/min;将氧处理后的支架与聚丙烯酸进行反应;反应完成后继续利用N-羟基丁二酰亚胺和碳二亚胺盐酸盐进行羧基活化处理:再将活化后的支架进行抗凝血物质肝素的等离子沉积改性:等离子沉积温度为105℃,辉光放电沉积时间为20min,放电功率为200W。最后,对表面修饰改性的抗凝血改性层,继续进行短肽RADA-16的间断不连续的喷涂,使短肽RADA-16利用共价键与部分抗凝血涂层进行反应。

Claims (5)

1.一种涂层支架(1),所述的涂层支架呈网状结构(2),其特征在于:所述的网状结构由三层结构组成,内层为支架层(3)、中间层为抗凝血改性层(4)、最外层为纳米短肽涂层(5);所述的抗凝血改性层的厚度不大于200nm;所述的纳米短肽涂层为不连续的排列;所述涂层支架的制备方法的具体步骤如下:
(1)支架层的加工:支架层经编织机编织而成;
(2)等离子体沉积构建抗凝血改性层:对加工完成的支架层进行氧等离子体辉光放电处理:放电功率为20-300W,处理时间为2-30min,氧气流量为100ml/min~800ml/min;将氧处理后的支架层与多元醇或含羧酸官能团的物质进行反应;反应完成后继续进行羧基活化处理:最后再将活化后的支架层进行抗凝血物质的等离子沉积改性:等离子沉积温度为10~180℃,辉光放电沉积时间为2min-45min,放电功率为50-300W;
(3)短肽涂层:对表面修饰改性的抗凝血改性层,继续进行短肽的间断不连续的喷涂,使短肽利用共价键与抗凝血改性层进行反应。
2.根据权利要求1所述的涂层支架,其特征在于:所述网状结构的内层由直径为0.01mm~0.60mm的金属或高分子丝构成;所述涂层支架直径为1.0~30.0mm。
3.根据权利要求1所述的涂层支架,其特证在于:所述的抗凝血改性层为肝素、聚乙二醇、磷酸胆碱、羟乙基纤维素、壳聚糖及壳聚糖衍生物、白蛋白中的一种或几种的混合物。
4.根据权利要求1所述的涂层支架,其特征在于:所述的纳米短肽涂层为具有促进血管内膜再生的纳米短肽RGD、YIGSR、REDV、CAG、SVVYGLR中的一种或几种混合物。
5.根据权利要求1所述的涂层支架,其特征在于所述的纳米短肽涂层的厚度为10-200nm。
