Description

Anti-wrinkle firming composition and cosmetic
Technical Field
The invention relates to the technical field of cosmetics, and particularly relates to an anti-wrinkle firming composition and a cosmetic.
Background
The skin of a human body inevitably generates wrinkles as it ages. The generation of skin wrinkles is a dynamic multi-level process, and is mainly related to the deterioration of various functions of the epidermis, dermis, and dermis junction (basement membrane).
Metabolism of skin epidermal cells is gradually slowed down along with the growth of the skin, the generation of epidermal lipid and natural moisturizing factors is reduced, the water storage capacity of a horny layer is reduced, an epidermal layer is thinned, and desquamation, dry lines and fine lines are easily generated on the epidermis.
Collagen I is the most prominent functional protein in the dermis layer, usually associated with collagen III, which lines the collagen fibers of the dermis, providing sufficient nutrition and strength to the dermis, and is also closely related to skin regeneration. In the structure of dermal fibers, elastin, an important protein, is the main component of dermal elastic fibers. The elastic fiber and the collagen fiber jointly form a fiber network structure of the dermis, so that the elasticity, the strength and the flexibility of the skin are endowed. With age, the synthesis of type I and type III collagen and elastin in the dermis decreases, resulting in a substantial loss of function of the collagen and elastic fibers in the dermis, which loses sufficient strength and elasticity in the dermis, resulting in deep wrinkles in the skin.
The collagen IV fixes the epidermis to the true epidermis connecting layer to form a net structure, so that the true epidermis connecting layer is stable in structure; collagen XVII is an important transmembrane protein connecting the epidermal layer and the dermal layer, and is also released as a signal for skin regeneration, and together they constitute a tensile structure of the epidermal connecting layer (basement membrane) and exert an adhesive effect. The synthesis of collagen IV and collagen XVII is reduced, the nutrition permeation between epidermis and dermis is reduced, the true epidermal junction layer (basement membrane) is flattened and even sunken and collapsed, the skin is loosened and loses toughness, and the generation of wrinkles is accelerated.
At present, most anti-wrinkle products in the market mainly focus on the supplement of collagen or elastin in the dermis, and the anti-wrinkle products have relatively single action way and are difficult to achieve quick, continuous and effective improvement effect in the aspect of wrinkle improvement.

Table 1: formulation compositions (%)
Figure BDA0003282496020000061
The preparation methods of the anti-wrinkle firming skin moisturizers of examples 1 to 5 and comparative examples 1 to 4 comprise the following steps:
1. adding water, humectant and thickener into an emulsifying pot according to the weight percentage of the raw materials, stirring and heating to 80-85 ℃, preserving heat and stirring for 30 minutes, and then cooling;
2. cooling to 60 ℃, adding a pH regulator, stirring for 10 minutes until the mixture is uniform, and continuously cooling;
3. cooling to 45 ℃, adding the preservative and the essence, then adding the glycerol glucoside, the hexapeptide-9, the acetyl tetrapeptide-2 and the hydrolyzed collagen, and stirring for 10 minutes until the mixture is uniform, thus obtaining the anti-wrinkle firming skin lotion.
Tests prove that in the embodiment, the humectant can also be one or more of propylene glycol, butanediol, dipropylene glycol, isoprene glycol, 1, 3-propanediol, methyl propylene glycol, polyethylene glycol-32, methyl gluceth-20 and glyceryl polyether-26.
The thickener can also be one or more of ammonium acryloyl dimethyl taurate/VP copolymer, hydroxyethyl cellulose, sodium polyacrylate, sodium acrylate/sodium acryloyldimethyl taurate copolymer, cetearyl alcohol, cetyl alcohol, and behenyl alcohol.
The pH regulator may also be one or more of triethanolamine and inorganic alkali.
Wrinkle resistance test and elasticity test:
20 test subjects aged 30 to 60 years, whose eyes and cheeks were the subject parts (same region and area), and subjects of the same degree of wrinkle level were the subjects, and one side was randomly used as the sample-applied part and the other side was used as the non-applied part, and the applied part was continuously used with the test sample for 28 days.
The PROMIS system analysis was used clinically to assess the percent reduction in tail-eye wrinkle depth, wrinkle area for 7 days, 14 days, 28 days.
The percent increase in skin elasticity was measured on the cheek area for 7, 14, and 28 days using a skin elasticity tester ELASTIMETER ELM 1086.
The anti-wrinkle tightening lotions of examples 1 to 5, the blank examples and comparative examples 1 to 4 were tested according to the anti-wrinkle test and elasticity test methods, and the test results are shown in fig. 1 to 3; in the figure: in the schematic representation of the same test sample, from left to right are the test results for days 7, 14 and 28, respectively.
As can be seen from the test results of fig. 1 to 3, examples 1 to 5 according to the present invention are significantly superior to the blank control group in the wrinkle depth, wrinkle area reduction and skin elasticity improvement through the use test for 7 days to 14 days to 28 days, compared to the blank example.
The effects of comparative examples 1 to 5 and comparative examples 1 to 4 were further analyzed:
comparative example 1 excludes glycerol glucoside, which is an ingredient for promoting epidermal cell moisturizing, regeneration and proliferation, and the addition amount of the remaining hexapeptide-9, acetyl tetrapeptide-2 and hydrolyzed collagen in the composition is increased to 4 times that of example 1 or 2 times that of example 3.
Comparative example 2 was to remove the anti-wrinkle component hexapeptide-9 of the true epidermal junction layer and to increase the amount of the remaining three components of glycerol glucoside, acetyl tetrapeptide-2, and hydrolyzed collagen to 4 times that of example 1 or 2 times that of example 3.
Comparative example 3 excludes hydrolyzed collagen that promotes collagen synthesis in the dermis layer, and the amount of the remaining three components of glycerol glucoside, hexapeptide-9, and acetyl tetrapeptide-2 in the composition was increased to 4 times that of example 1 or 2 times that of example 3.
Comparative example 4 excludes acetyl tetrapeptide-2, which promotes elastin synthesis in the dermis layer, and the amounts of the remaining three components, namely glycerol glucoside, hexapeptide-9 and hydrolyzed collagen, in the composition are increased to 4 times that of example 1 or 2 times that of example 3.
From the results, it can be seen that: the anti-wrinkle and firming composition components of the epidermal layer, the real epidermal connecting layer and the dermal layer in the composition of each embodiment of the invention complement each other in anti-wrinkle mechanism, have the mutual synergistic and mutually-promoting effects, and are obviously superior to each proportion in the aspects of improving wrinkle depth, wrinkle area and skin elasticity.
Comparing example 1 with comparative example 1, comparative example 1 needs to increase the addition amount of the other three components to 4 times or more after removing the glycerol glucoside component which can promote the regeneration and proliferation of epidermal cell moisturizing, and comparative example 1 still fails to achieve the effect of example 1 on the improvement of wrinkle depth, wrinkle area and skin elasticity. The improvement of the wrinkles is not only related to the repair of the dermis, the metabolism of the epidermis is accelerated, and the cell repair and proliferation of the epidermis are promoted, so that the wrinkle-removing cream has very important effects on comprehensive wrinkle removal.
Further comparing example 3 with comparative example 2, the addition amount of glyceryl glucoside, acetyl tetrapeptide-2, and hydrolyzed collagen in comparative example 2 was 2 times of that of example 3, but the nutrition of the epidermal layer and the dermal layer connecting layer could not be rapidly and effectively delivered by permeation due to the lack of hexapeptide-9, which is an effective ingredient capable of repairing the basement membrane, and the true effect of the anti-wrinkle active substance could not be fully exerted.
In comparative example 3, the addition amount of the three components of glycerol glucoside, hexapeptide-9 and acetyl tetrapeptide-2 was increased to 2 times that of example 3, but the effects of example 3 on the improvement of wrinkle depth, wrinkle area and skin elasticity of comparative example 3 were not achieved due to the lack of hydrolyzed collagen which can promote the synthesis of collagen in the dermis layer. Similarly, the addition amount of the three components of glycerol glucoside, hexapeptide-9 and hydrolyzed collagen in comparative example 4 was increased to 2 times that of example 3, but the improvement of wrinkle depth, wrinkle area and skin elasticity in comparative example 4 due to the absence of acetyl tetrapeptide-2, which promotes the synthesis of elastin in the dermis, was also not achieved with the effect of example 3. The improvement of the wrinkles is not only related to the repair of the epidermis layer and the basement membrane and promotes the synthesis of collagen and elastin in the dermis layer, but also plays a very important role in overall wrinkle resistance.
In consideration of the specificity of the molecular structure of glycerol glucoside, the present invention further uses glycerol, sucrose, and polysaccharides such as β -glucan, mannan, etc. in place of glycerol glucoside in equivalent amount in example 3.
Example 3, comparative examples 5 to 8
The cosmetics of example 3 and comparative examples 5 to 8, specifically, anti-wrinkle and tightening skin lotions were formulated according to the formulation compositions in table 2 below.
Table 2: formulation composition (%)
Figure BDA0003282496020000091
Figure BDA0003282496020000101
The preparation method of the anti-wrinkle firming skin moisturizer is the same as the preparation method of the anti-wrinkle firming skin moisturizer in the embodiment 1.
The anti-wrinkle tightening lotions of example 3 and comparative examples 5 to 8 were tested according to the anti-wrinkle test and elasticity test methods, and the test results are shown in fig. 4 to 6; in the figure: in the schematic representation of the same test sample, from left to right are the test results for days 7, 14 and 28, respectively.
The results of the tests according to fig. 4-6 can be found: comparative example 5, which replaces glycerol glucoside with 2% glycerol in equal amounts, showed a significant reduction in the improvement of wrinkle depth, wrinkle area, and skin elasticity, compared to example 3. Although glycerin has a good water-retaining property, it does not have a significant effect of improving wrinkles and increasing skin elasticity.
Further, it can be seen that comparative examples 6 to 8 equally replace glyceroglucoside with sugars such as sucrose, β -glucan, mannan, etc., respectively, while the β -glucan of comparative example 7 also exhibits certain anti-wrinkle and skin elasticity improving effects. This is because β -glucan is also able to give the stratum corneum direct and effective nutritional supplementation, which increases the regenerative capacity of damaged skin cells. However, the effect of β -glucan on improving wrinkle area, wrinkle depth, and skin elasticity is still different from that of the present invention when added in the same amount, and this is probably because the molecular space structure of the present invention is smaller and more easily absorbed and utilized by the skin than β -glucan.
On the basis of the embodiment 3, the invention further screens and compares the influence of the anti-wrinkle polypeptides commonly used in the market on the test result. Example 3 and comparative examples 9-11 are specifically as follows:
comparative example 9, acetyl tetrapeptide-2 of example 3 of the present invention was equally replaced with palmitoyl tetrapeptide-5;
comparative example 10, acetyl hexapeptide-8 was used in place of acetyl tetrapeptide-2 in example 3 of the present invention in equal amounts;
comparative example 11, acetyl octapeptide-3 was used in place of acetyl tetrapeptide-2 in inventive example 3 in equal amounts.
Example 3, comparative examples 9 to 11
The cosmetics of example 3 and comparative examples 9 to 11, specifically, anti-wrinkle tightening lotions, were formulated according to the formulation compositions in table 3 below.
Figure BDA0003282496020000111
The preparation method of the anti-wrinkle firming skin moisturizer is the same as the preparation method of the anti-wrinkle firming skin moisturizer in the embodiment 1.
The anti-wrinkle tightening lotions of example 3 and comparative examples 9 to 11 were tested according to the anti-wrinkle test and elasticity test methods, and the test results are shown in fig. 7 to 9; in the figure: in the schematic representation of the same test sample, from left to right are the test results for days 7, 14 and 28, respectively.
The results of the tests according to FIGS. 7-9 may give: different kinds of polypeptides show certain wrinkle improvement and skin elasticity improvement effects in practical tests, but the specific wrinkle improvement effects are different.
Further comparison shows that comparative example 9, which is also palmitoyl tetrapeptide-5, which is a tetrapeptide, has a good effect of improving skin elasticity, but has a certain difference from acetyl tetrapeptide-2 of the present invention. While the same acetylated peptides such as acetyl hexapeptide-8 and acetyl octapeptide-3 have certain wrinkle area and depth improving effect after being added in equal amount, when the product is used for 28 days, the effect of improving skin elasticity is very limited in comparative examples 10 and 11 when the product is used for 14 to 28 days, compared with example 3 of the present invention, probably because the acetyl hexapeptide-8 and acetyl octapeptide-3 mainly aim at improving expression lines caused by muscle contraction, while the acetyl tetrapeptide-2 of the present invention can not only induce type I collagen synthesis, but most importantly promote the synthesis of dermal elastin, further repair dermal elastic fibers, increase the space between elastic fibers, make the skin more plump, improve skin firmness, and restore skin elasticity.
Through the screening and comparative tests of different kinds of polypeptides, the acetyl tetrapeptide-2 used in the invention focuses on improving the elastic fiber structure of the dermis, has better skin elasticity improving effect than other polypeptides, and forms an overall synergistic anti-wrinkle scheme with other three active substances (glycerol glucoside, hexapeptide-9 and hydrolyzed collagen) in the invention on an anti-wrinkle mechanism, and the components supplement each other and have irreplaceability.
Further, the present invention further compares the effect of collagen of different molecular weights on the test effect on the basis of example 3.
Comparative example 12 the hydrolyzed collagen of example 3 of the invention was replaced by an equal amount of soluble collagen (molecular weight of about 50000 Da).
Comparative example 13, an equivalent amount of a soluble collagen polymer (molecular weight > 100000Da) was used in place of the hydrolyzed collagen of example 3 of the present invention.
Example 3, comparative examples 12 to 13
The cosmetics of example 3 and comparative examples 12 to 13, specifically, anti-wrinkle tightening lotions, were formulated according to the formulation compositions in table 4 below.
Figure BDA0003282496020000121
Figure BDA0003282496020000131
The preparation method of the anti-wrinkle firming skin moisturizer is the same as the preparation method of the anti-wrinkle firming skin moisturizer in the embodiment 1.
The anti-wrinkle tightening lotions of example 3 and comparative examples 12 to 13 were tested according to the anti-wrinkle test and elasticity test methods, and the test results are shown in fig. 10 to 12; in the figure: in the schematic representation of the same test sample, from left to right are the test results for days 7, 14 and 28, respectively.
The results of the tests according to FIGS. 10-12 may give: in the invention, the collagen with small molecular weight has better effect of improving skin wrinkles and skin elasticity under the condition of the same addition amount. Compared with soluble collagen with medium molecules and high-molecular soluble collagen polymers, the hydrolyzed collagen with smaller molecular weight is used in the invention, can quickly permeate from the epidermal layer to the dermal layer, promotes the synthesis of type I and type III collagen, carries out continuous nutrition supplement for the dermal layer, and has more remarkable effect on the recovery of the collagen fiber structure of the dermal layer.
In conclusion, the invention starts from the mechanism of wrinkle generation, specifically selects and compares various anti-wrinkle active substances from a plurality of surfaces such as an epidermal layer, a true epidermal connecting layer, a dermal layer and the like, further screens out a comprehensive anti-wrinkle compact composition with more comprehensive overall mechanism and more obvious effect through component optimization, and can achieve the effects of improving wrinkles and skin elasticity more quickly, continuously and effectively.
Furthermore, the anti-wrinkle tightening skin lotion can be prepared into cosmetics in other dosage forms, such as anti-wrinkle tightening skin lotion, anti-wrinkle tightening skin cream, anti-wrinkle tightening skin gel, anti-wrinkle tightening skin spray and the like.
