Abstract
本发明要解决的技术问题在于:提供一种肾细胞癌治疗用组合物,并提供一种增加药物向肾细胞癌细胞摄取的组合物或方法。本发明的构成如下:一种含有D‑阿洛糖的肾细胞癌治疗用组合物、或一种增加含有D‑阿洛糖的药物向肾细胞癌细胞摄取的组合物或方法。该组合物是用于对人或人以外的动物中需要肾细胞癌治疗的患者以有效量给药的医药组合物。

技术领域
本发明涉及利用D-阿洛糖被癌细胞摄取的性质而向肾细胞癌细胞选择性输送药物的载体、药物输送方法和用于治疗肾细胞癌的组合物。
背景技术
癌的治疗法大致分为手术疗法、放射线疗法、化学疗法。这些中,化学疗法是将抗癌剂对癌患者给药的疗法,用于为了在手术疗法或放射线疗法的前后提高治愈力的术前、术后的辅助化学疗法、以及无法通过手术疗法或放射线疗法治疗的已转移的癌的治疗。现在,代谢拮抗剂、拓扑异构酶抑制剂、分子靶向药、核酸药剂等抗癌剂已临床实用化,对于几种癌已达到能够期待治愈的程度,但在肾细胞癌的化学疗法中,至今仍未获得令人满意的结果。
抗癌剂往往并不会选择性地作用于肿瘤细胞,还会作用于正常细胞,造成高毒性和副作用。具有高细胞分裂速度的组织特别容易受影响,这种有害的全身毒性限制了能够对癌患者给药的抗癌剂用量,从而限制了效果。并且,由于部分抗癌剂为溶解度低的疏水性,不仅膜透过性低,还会形成不溶解于水性介质的抗癌剂的凝集物,因此在静脉内给药时,有可能在浸透肿瘤之前引起毛细血管的栓塞。
目前,为了将药物选择性地送达肿瘤,已开发例如在胶束、脂质体、微粒、抗体和药物-聚合物结合体等的载体中内包有药物等的药物传递系统(专利文献1)。尽管这样尝试着提高抗癌剂向肿瘤组织的积累能力,但依然难以防止抗癌剂向肝脏、肾脏等正常组织的积累,期待开发出能够将抗癌剂选择性地送达癌细胞的药物载体。
另一方面,在医学领域的稀少糖的应用研究成果中,有以D-阿洛糖作为有效成分的生物体内抗氧化剂(专利文献2)的发明。其是将D-阿洛糖对肝脏癌或皮肤癌的患者给药,利用D-阿洛糖的生物体内抗氧化作用而用作治疗肝脏癌或皮肤癌的组合物的发明。

发明内容
发明要解决的技术问题
本发明的目的在于提供一种利用被癌细胞摄取的性质的D-阿洛糖的新用途,即,提供一种具有被癌细胞摄取的性质的D-阿洛糖作为药物输送载体的新用途。本发明的目的还在于提供一种使用该物输送载体的药物输送方法。而且,本发明的目的还在于提供一种作为对肾细胞癌具有优异抗肿瘤效果的抗癌剂的肾细胞癌治疗用组合物。此外,本发明的目的还在于提供一种能够增加药物向肾细胞癌细胞摄取的肾细胞癌治疗用组合物、以及用于增加药物向肾细胞癌细胞摄取的组合物或方法。
用于解决技术问题的技术方案
本发明的发明人为了解决上述技术问题而进行了深入研究,首次发现了D-阿洛糖被人肾细胞癌细胞特异性摄取并且被摄取的D-阿洛糖抑制肾细胞癌细胞的增殖而具有抗肿瘤活性,从而完成了本发明。并且,通过将能够被人肾细胞癌细胞摄取的D-阿洛糖用作作为抗癌剂的化学疗法剂或核酸药剂所使用的载体,能够增加肾细胞癌细胞对抗癌剂的摄取,从而能够发挥被摄取的D-阿洛糖和抗癌剂双方的抗肿瘤活性。
本发明的要旨在于以下(1)~(4)的利用被肾细胞癌细胞选择性摄取的性质的制剂。
(1)一种利用被肾细胞癌细胞选择性摄取的性质的制剂,其特征在于,包含D-阿洛糖。
(2)如上述(1)所述的制剂,其中,被肾细胞癌细胞摄取的D-阿洛糖发挥抗肿瘤效果。
(3)如上述(1)或(2)所述的制剂,其中,D-阿洛糖为D-阿洛糖和/或其衍生物和/或其混合物。
(4)如上述(3)所述的制剂,其中,上述D-阿洛糖的衍生物为选自D-阿洛糖的羰基变成醇基而成的糖醇、D-阿洛糖的醇基氧化而成的糖醛酸、D-阿洛糖的醇基被NH2基取代而成的氨基糖中的D-阿洛糖衍生物。
本发明的要旨在于以下(5)~(11)的向肾细胞癌细胞选择性输送药物的载体。
(5)一种向肾细胞癌细胞选择性输送药物的载体,其特征在于,包含D-阿洛糖。
(6)如上述(5)所述的向肾细胞癌细胞选择性输送药物的载体,其中,被肾细胞癌细胞摄取的D-阿洛糖发挥抗肿瘤效果。
(7)如上述(4)或(5)所述的向肾细胞癌细胞选择性输送药物的载体,其中,D-阿洛糖为D-阿洛糖和/或其衍生物和/或其混合物。
(8)如上述(7)所述的向肾细胞癌细胞选择性输送药物的载体,其中,上述D-阿洛糖的衍生物为选自D-阿洛糖的羰基变成醇基而成的糖醇、D-阿洛糖的醇基氧化而成的糖醛酸、D-阿洛糖的醇基被NH2基取代而成的氨基糖中的D-阿洛糖衍生物。
(9)如上述(5)~(8)中任一项所述的向肾细胞癌细胞选择性输送药物的载体,其中,上述药物包含抗癌剂。
(10)如上述(5)~(9)中任一项所述的向肾细胞癌细胞选择性输送药物的载体,其中,D-阿洛糖和上述药物直接缔合或者经由连接臂通过共价键缔合。
(11)如上述(5)~(10)中任一项所述的向肾细胞癌细胞选择性输送药物的载体,其中,上述药物为放射性同位素、酶、前药活化酶、放射线敏化剂、iRNA、烷基化剂、嘌呤拮抗剂、嘧啶拮抗剂、植物生物碱、嵌入抗生素、代谢拮抗物质、芳香化酶抑制剂、有丝分裂抑制剂、生长因子抑制剂、细胞周期抑制剂或拓扑异构酶抑制剂。
本发明的要旨在于以下(12)~(19)的用于治疗肾细胞癌的组合物。
(12)一种用于治疗肾细胞癌的组合物,其特征在于,含有D-阿洛糖。
(13)如上述(12)所述的用于治疗肾细胞癌的组合物,其中,被肾细胞癌细胞摄取的D-阿洛糖发挥抗肿瘤效果。
(14)如上述(12)或(13)所述的用于治疗肾细胞癌的组合物,其中,D-阿洛糖为D-阿洛糖和/或其衍生物和/或其混合物。
(15)如上述(14)所述的用于治疗肾细胞癌的组合物,其中,上述D-阿洛糖的衍生物为选自D-阿洛糖的羰基变成醇基而成的糖醇、D-阿洛糖的醇基氧化而成的糖醛酸、D-阿洛糖的醇基被NH2基取代而成的氨基糖中的D-阿洛糖衍生物。

本发明的要旨在于以下(20)~(26)的向肾细胞癌细胞输送药物的方法。
(20)一种药物输送方法,其是向肾细胞癌细胞输送药物的方法,其特征在于,将药物载持于药物输送载体上制成药剂,将该药剂应用于肾细胞癌细胞,使该药剂被上述肾细胞癌细胞选择性摄取,由此向该细胞选择性地输送上述药物,其中,上述载体为D-阿洛糖,使上述药剂经由化学键载持于该D-阿洛糖。
(21)如上述(20)所述的药物输送方法,其是利用D-阿洛糖被癌细胞摄取的性质而增加药物向肾细胞癌细胞摄取的方法。
(22)如上述(20)或(21)所述的药物输送方法,其特征在于,上述载体为D-阿洛糖,使上述药剂与该D-阿洛糖直接缔合或者经由连接臂通过共价键缔合并载持而成。
(23)如上述(20)~(22)中任一项所述的药物输送方法,其中,上述药物包含抗癌剂。

发明的效果
关于D-阿洛糖,虽然已知利用生物体内抗氧化作用而对肝脏癌或皮肤癌的抗肿瘤效果,但对肾细胞癌的抗肿瘤效果未被知晓。并且,至今未证明D-阿洛糖被摄取至人癌细胞内。
本发明的发明人首次发现D-阿洛糖被人肾细胞癌细胞摄取,并且被摄取的D-阿洛糖具有抗肿瘤活性。由于D-阿洛糖为水溶性物质,所提膜透过性高,并且因溶解于水性介质而不会形成凝集物。
而且,在人癌细胞中特别是被肾细胞癌细胞特异性摄取,因此,能够用作可将抗癌剂选择性地送达肾细胞癌细胞的药物载体(选择性药物输送载体),能够增加肾细胞癌细胞对抗癌剂的摄取,从而能够使被摄取的D-阿洛糖和抗癌剂双方发挥抗肿瘤活性。
日本的临床病期IV期肾细胞癌的5年存活率为18.1%,依然较低,因此,本发明的含有D-阿洛糖的肾细胞癌治疗用组合物具有能够成为突破性的分子靶向治疗药的可能性。

具体实施方式
D-阿洛糖作为利用被肾细胞癌细胞选择性摄取的性质的制剂使用。被肾细胞癌细胞摄取的D-阿洛糖发挥抗肿瘤效果。并且,D-阿洛糖作为向肾细胞癌细胞选择性输送药物的载体使用,提供向肾细胞癌细胞输送药物的方法。该药物包含抗癌剂,药物直接或经由连接臂通过共价键与D-阿洛糖缔合。另外,本发明为一种含有D-阿洛糖的肾细胞癌治疗用组合物,含有有效量的D-阿洛糖或药理上可接受的盐或/和水合物的制剂。上述的药物输送方法中,使用包含D-阿洛糖的载体,作为载持于该载体的药物,能够使用下述的各种药物,可以根据目的适当选择,但由于所载持的药剂被肾细胞癌细胞选择性摄取,因此优选使用抗癌剂等的药剂。换言之,希望制成以包含D-阿洛糖的载体被癌细胞摄取作为目标而载持抗癌剂的药剂。
本发明中使用的D-阿洛糖是存在量远远少于自然界中大量存在的D-葡萄糖的稀少糖。作为糖的基本单位的单糖(碳原子数为6的单糖:己糖)总共有34种,醛糖为16种、酮糖为8种、糖醇为10种)中,相对于自然界中大量存在的以D-葡萄糖为代表的“天然型单糖”,将自然界中仅微量存在的单糖(醛糖、酮糖)及其衍生物(糖醇)定义为“稀少糖”。现在能够大量生产的稀少糖为D-阿洛酮糖(D-阿卢糖)和D-阿洛糖。D-阿洛糖是分类于六碳糖的醛糖的阿洛糖的D构型体。
作为得到D-阿洛糖的方法,有利用从施氏假单胞菌(Pseudomonas stutzeri)分离的L-鼠李糖异构酶(非专利文献1)由D-阿洛酮糖合成的方法、使D-木糖异构酶作用于含有D-阿洛酮糖的溶液而得到的方法,并且,关于制造高纯度D-阿洛糖,有利用D-阿洛糖的结晶化法的分离法(专利文献3)等。本发明的D-阿洛糖不限于此,也可以是通过化学处理方法异构化而成的等,可以是通过任意方法得到的D-阿洛糖。关于作为D-阿洛糖的原料的D-阿洛酮糖,目前普遍的是对果糖进行酶(差向异构酶)处理而得到的制法,但并不仅限于此,可以是通过使用生产该酶的微生物的制法得到的,也可以是从天然物质中提取的,或者可以直接使用天然物中含有的,还可以是通过化学处理方法异构化而成的。另外,利用酶将D-阿洛酮糖精制的方法是公知的。
对D-阿洛糖的衍生物进行说明。将从某起始化合物通过化学反应使分子结构发生了变换的化合物称为起始化合物的衍生物。作为包含D-阿洛糖的六碳糖的衍生物,普遍的有糖醇(将单糖类还原时,醛基和酮基变成醇基,成为与碳原子同数的多元醇)和糖醛酸(为单糖类的醇基氧化而成的物质,天然的已知D-葡萄糖醛酸、半乳糖醛酸、甘露糖醛酸)、氨基糖(糖分子的OH基被NH2基取代而成的物质,有葡糖胺、软骨糖胺、配糖体等)等,但不限定于此。D-阿洛糖的衍生物为选自D-阿洛糖的羰基变成醇基而成的糖醇、D-阿洛糖的醇基氧化而成的糖醛酸、D-阿洛糖的醇基被NH2基取代而成的氨基糖中的D-阿洛糖衍生物。
在本发明的配合有D-阿洛糖和/或其衍生物和/或其混合物的治疗用组合物中,组合物含有有效量的D-阿洛糖和/或其衍生物和/或其混合物。所谓“有效量”是指足以满足所意图的目的(例如组织或被检验体中的所希望的生物学或医学应答)的任意量。例如,在本发明中,是抑制肾细胞癌细胞的增殖、将药物送达肾细胞癌细胞内部的量等。
对本发明的D-阿洛糖或其药理上可接受的盐、或/和水合物的制剂进行说明。
除了仅使用D-阿洛糖和/或其衍生物和/或其混合物以外,还可以配合一般的赋形剂、稳定剂、保存剂、结合剂、崩解剂等适当的添加剂,并选择液剂、胶囊剂、颗粒剂、丸剂、散剂、片剂等适当的制剂型进行制剂。作为本发明的肾细胞癌治疗用组合物,能够通过公知的制剂技术制造将有效成分溶解于含有医学上可接受的载体、赋形剂、润滑剂、结合剂等添加物的各种形态,例如溶解于水或各种输液用制剂中的液剂、散剂、颗粒剂、片剂、注射剂、栓剂或外用制剂等。作为非口服剂,可以选择注射剂、点滴剂、外用药剂或栓剂等剂型。作为注射剂,可以例示皮下注射剂、肌肉注射剂或腹腔内注射剂等。制成上述的制剂型的制剂技术是公知的。
在将本发明的D-阿洛糖或其药理上可接受的盐等通过注射给药的情况下,优选为水性注射剂、水性悬浮注射剂、脂肪乳剂或脂质体注射剂等。在水性注射剂或水性悬浮注射剂中,将本发明所涉及的稀少糖D-阿洛糖或其药理上可接受的盐与精制水混合,根据需要添加水溶性或水膨润性高分子、pH调节剂、表面活性剂、渗透压调节剂、防腐剂或保存剂等并混合,根据需要边加热边使之溶解或悬浮,灭菌并充填密封在注射剂容器中,制成水性注射剂或水性悬浮注射剂。水性注射剂可以对静脈内、皮下、肌肉内、皮内或关节腔内等给药。并且,水性悬浮注射剂可以在皮下、肌肉内、皮内或关节腔内等给药。还可以口服给药。
作为水溶性或水膨润性高分子,优选明胶、纤维素衍生物、丙烯酸衍生物、聚维酮、聚乙二醇、聚氨基酸衍生物或多糖体类,明胶类中,优选精制明胶;纤维素衍生物中,优选甲基纤维素、羟丙基甲基纤维素2910、羟丙基甲基纤维素2208、羟丙基甲基纤维素2906、羟丙基纤维素、低取代度羟丙基纤维素、羧甲基纤维素钠;作为丙烯酸衍生物,优选氨基丙烯酸甲基丙烯酸酯共聚物、甲基丙烯酸共聚物;作为聚氨基酸衍生物,优选聚赖氨酸、聚谷氨酸。作为多糖体,特别优选透明质酸、葡聚糖或环糊精。水溶性或水膨润性高分子的添加量因七叶亭、其衍生物或其药理上可接受的盐的性质、量、以及水溶性或水膨润性高分子的性质、分子量、应用部位而不同,但大致可以相对于制剂总量以0.01%~10%的范围使用。
pH调节剂使用对人体无害的酸或碱,表面活性剂可以使用非离子性表面活性剂、阴离子性表面活性剂、两性表面活性剂。另外,渗透压调节剂可以例示氯化钠、葡萄糖等;防腐剂可以例示对羟基苯甲酸酯类,保存剂可以例示抗坏血酸或亚硫酸盐类。它们的使用量没有特别限定,可以以分别能够发挥其作用的范围使用。另外,根据需要,还可以添加盐酸普鲁卡因等局部麻醉剂、苄醇等无痛剂、螯合剂、缓冲剂或水溶性有机溶剂等。
脂肪乳剂通过向适当的油脂中配合乳化剂和D-阿洛糖、或其药理上可接受的盐,添加精制水,根据需要添加水溶性或水膨润性高分子、pH调节剂、表面活性剂、渗透压调节剂、防腐剂或保存剂等,利用适当的乳化装置乳化,灭菌并向注射剂容器中充填密封来制备。
本发明的“药物”、“抗癌剂”是为了治疗而向癌或癌前状态的组织给药的物质,例如可以列举放射性同位素(例如碘-131、镥-177、铼-188、钇-90)、毒素(例如白喉毒素、假单胞菌毒素、蓖麻毒素、白树毒素)、酶、活化前药的酶、放射线敏化剂、干扰RNA、超抗原、抗血管新生剂、烷基化剂、嘌呤拮抗剂、嘧啶拮抗剂、植物生物碱、嵌入抗生素、芳香化酶抑制剂、代谢拮抗物质、有丝分裂抑制剂、生长因子抑制剂、细胞周期抑制剂、拓扑异构酶抑制剂、生物学应答改变因子、抗激素和抗雄激素等。
药物与D-阿洛糖缔合(例如键合、相互作用)后使用。缔合可以是共价键合,也可以是非共价键合。关于D-阿洛糖与药物的缔合,需要在送达肾细胞癌细胞和被摄取到肾细胞癌细胞内之前或其期间不发生解离的充分的强度,可以使用本领域技术人员公知的任意的化学的、生物化学的、通过酶的偶联。
D-阿洛糖与药物的缔合是非共价键合时,可以列举疏水性相互作用、静电相互作用、偶极相互作用、范德瓦尔斯相互作用和氢键;D-阿洛糖与药物的缔合是共价键合时,可以直接键合,或者经由连接臂间接键合。这样的共价键可以利用酰胺、酯、碳-碳、二硫醚、氨基甲酸酯、醚、硫醚、尿素、胺或碳酸酯键来实现。
关于用作医药成分所需的D-阿洛糖的安全性的验证,稀少糖虽然微量但是自然界中存在的单糖,因此能够预见其是安全的。将突变原性、生物分解度试验和3种急性毒性试验(口服急性毒性试验、皮肤一次刺激试验、眼睛一次刺激试验定为最基本的安全性试验,发明人委托指定机构实施D-阿洛糖的基本部分的安全性试验,作为其结果,确认了其安全性没有问题。
本发明的治疗用组合物的对象为包括人在内的动物(人、牛、猪、狗、猫等哺乳类、鸡等鸟类等)。另外,本发明的治疗用组合物作为靶向的癌细胞是肾细胞癌细胞,作为细胞株,可以列举人肾细胞癌细胞株ACHN、Caki-I、Caki-II。
接下来,利用实施例对本发明的详细内容进行说明。本发明并不受这些实施例的任何限定。
实施例1
[使用人膀胱癌细胞株、人前列腺癌细胞株、人肾细胞癌细胞株的稀少糖摄取实验]
使用3种癌的细胞株,分析在培养基中添加的稀少糖向癌细胞内的摄取量。作为稀少糖,使用D-阿洛糖、D-阿洛酮糖(D-阿卢糖)、L-阿洛酮糖(L-阿卢糖)或蒜糖醇共4种,作为不是稀少糖的单糖,使用D-葡萄糖和果糖。
在RPMI-1640培养基中培养各细胞株的细胞,途中将各6种糖分别添加入培养基中并进行培养,在培养后,分析被摄取入细胞内的糖的量。将培养途中在培养基中不添加糖的作为对照。
1)使用细胞株
使用人膀胱癌细胞株(RT112、253J、J82)、人前列腺癌细胞株(LNCa、Du145、PC-3)、人肾细胞癌细胞株(ACHN、Caki-I、Caki-II)。
2)使用培养基
在RPMI-1640(2000mgD-葡萄糖/L)培养基中培养细胞后,向各培养基添加各单糖、稀少糖。具体为RPMI-1640单独(对照)、RPMI-1640+D-葡萄糖、RPMI-1640+L-阿洛酮糖、RPMI-1640+D-阿洛酮糖、RPMI-1640+D-果糖、RPMI-1640+D-阿洛糖、RPMI-1640+D-蒜糖醇共7种,使所添加的糖在培养基中的最终浓度达到10mM,在对照中不添加。
3)培养方法
将使用RPMI-1640制作的3.0×104cells/ml的细胞悬浮液在每个直径6cm的培养皿中分别接种5ml(1.5×105cells/培养皿),在含有作为营养源的D-葡萄糖2000mg/l的RPMI-1640培养基中培养24小时。在24小时后变更为上述2)中所记载的分别含有单糖、稀少糖的培养液培养48小时。
4)培养细胞内的糖的分析方法
在48小时的培养后,机械地剥离贴壁的人癌细胞,将包括培养液和悬浮细胞收集在微量离心管中。除去由离心(4℃·1200rpm·5分钟)形成的上清,仅留下细胞沉淀后,以清洗为目的,用5ml的磷酸缓冲生理盐水(PBS)将细胞沉淀制成细胞悬浮液,再次以4℃、1200rpm进行5分钟离心并除去上清。向清洗后的细胞沉淀添加1ml的纯水使其悬浮后,进行超声波破碎(强度40%、30秒),对细胞破碎液中的单糖进行ABEE标记后,利用HPLC进行分析。利用标准曲线从所得到的面积求出细胞的糖含量。关于酮糖,在ABEE标记后确认到2个峰,所以由两个峰的标准曲线求出细胞中的糖含量。

〈实验结果的总结〉
在所有人癌细胞株中显示D-阿洛酮糖和L-阿洛酮糖向细胞内的摄取,与此相对,D-阿洛糖未见向人膀胱癌细胞株中的摄取,而在人前列腺癌细胞株中,仅在3个细胞株中的1个(PC-3)中显示摄取,另一方面,在人肾细胞癌细胞株中,在所有细胞株(ACHN、Caki-I、Caki-II)中显示D-阿洛糖向细胞内的摄取。
实施例2
[以人肾细胞癌细胞株为对象的稀少糖的抗肿瘤效果的解析]
使用实施例1中所使用的3种人肾细胞癌细胞株(ACHN、Caki-I、Caki-II),解析稀少糖的抗肿瘤效果。作为稀少糖,使用L-阿洛酮糖、D-阿洛酮糖、D-阿洛糖、L-果糖、D-甘露糖、L-山梨糖、D-塔格糖、D-半乳糖、D-山梨糖和L-塔格糖共10种,作为不是稀少糖的单糖,使用D-葡萄糖和D-果糖。
[使用培养基]
培养基使用含有1000mg/l的D-葡萄糖的最低必需培养基(MEM),使用调整为各单糖或各稀少糖含有10mM、25mM、50mM的MEM〔图中用(10)、(25)、(50)表示。〕对稀少糖的抗肿瘤效果进行评价。此外,在对照中不向MEM添加糖。
[实验步骤]
用MEM制作分别为5.0×104cells/ml的细胞悬浮液,在96孔板中分别以0.1ml/孔进行分配后,培养24小时。之后将培养液除去,向MEM分别以0.1ml/孔添加含有10mM、25mM、50mM的各单糖或各稀少糖的培养液,培养24小时(#1~#13)。

[细胞存活试验(MTT法)]
MTT法是测定存活的检查法之一,利用伴随四唑盐MTT的还原的不溶性甲色素(蓝色)的显色反应。MTT的还原由线粒体的还原酶琥珀酸-四唑还原酶(succinate-tetrazolium reductase)引起。活细胞中该酶活性高,可确认到显色,而发生包括细胞凋亡的细胞死亡时显色消失。通过用酶标仪测定该显色来测定细胞的存活率。
[细胞存活试验的结果]
将人肾细胞癌细胞的存活率的测定结果示于图1~图3。即,图1表示肾细胞癌细胞株(ACHN)的存活试验的结果,图2表示肾细胞癌细胞株(Caki-I)的存活试验的结果,图3表示肾细胞癌细胞株(Caki-II)的存活试验的结果。如图1~图3所示,在这些3种细胞株中,D-阿洛糖在10mM、25mM、50mM的所有浓度下,使存活率相比于对照显著降低(p<0.05),在所解析的糖中发挥了最强的抗肿瘤效果。
[考察]
在癌细胞中线粒体内的ATP产生被抑制。在癌细胞中,不使用氧而从葡萄糖(glucose)产生ATP的称为“糖酵解”的代谢系统亢进。糖酵解在细胞质中进行。癌细胞抑制线粒体内的氧呼吸的理由有几种。
一种是由于作为合成细胞构成成分的材料需要大量的葡萄糖。细胞为了分裂并增加数量,需要新生成核酸、细胞膜、蛋白质等细胞构成成分。细胞能够通过糖酵解系统或从该路径衍生的各种细胞内代谢路径从葡萄糖生成核酸、脂质或氨基酸。在线粒体中使用氧将葡萄糖全部用于ATP产生时,就会使制作细胞的材料消失。
并且,线粒体内的氧呼吸增加活性氧的产生。活性氧对细胞造成损伤,抑制增殖或转移,成为引起细胞死亡的原因。据认为,癌细胞抑制线粒体内的氧的利用,以不增加活性氧的产生。对于癌细胞而言,抑制线粒体内的使用氧的代谢则有利于存活和增殖。
已知在癌细胞中的情况下,如果提高癌细胞的线粒体的活性,则能够抑制增殖和转移,引起细胞死亡。这是由于在葡萄糖完全被分解时,用于增加细胞的材料不足,氧呼吸的亢进增加活性氧的产生,通过活性氧造成的损伤使癌细胞自灭的缘故。换言之,据认为活化细胞的线粒体的治疗法能够在提高正常细胞的工作的同时仅使癌细胞灭亡。
启示了D-阿洛糖通过被人肾细胞癌细胞特异性摄取,能够提高线粒体的活性且发挥抗肿瘤效果。
实施例3
[肾细胞癌异种移植小鼠的制作方法]
本实施例的实验使用2种人肾细胞癌细胞株(Caki-1、ACHN)。使用含有10%胎牛血清、以及HEPES缓冲液和青霉素-链霉素(penicillin-streptmycin)的RPMI-1640培养液(2000mg D-葡萄糖/L),在37℃且5%CO2的湿润环境下培养各细胞。
关于肾细胞癌异种移植小鼠模型,将培养细胞用MEM培养液调整成1.0×105cells/mL的细胞悬浮液,向雌性无胸腺裸鼠(Female athymic nude mice)(BALB/c-nu/nu,6周龄)的大腿部皮下组织注入0.1mL,在其1周后确认肿瘤体积为200mm3以上,用于之后的实验。[向肾细胞癌异种移植小鼠模型腹腔内注入D-阿洛糖后的肿瘤中D-阿洛糖浓度的推移]
用生理盐水将稀少糖D-阿洛糖以400mg/kg/0.4mL制成溶液,使用27G针向2种人肾细胞癌细胞株(Caki-I、ACHN)的异种移植小鼠模型的腹腔内注入。在D-阿洛糖给药前、给药后第1小时、第2小时、和第4小时从小鼠摘除肿瘤。肿瘤在1mL PBS中进行超声波破碎,对通过3000转5分钟的离心分离而得到的上清中的单糖进行ABEE(4-aminоbenzоic acid ethylester:4-氨基苯甲酸乙酯)标记,利用HPLC(high pefоrmance liquid chrоmatоgraphy,高效液相色谱)对D-阿洛糖进行定量分析。图4表示向肾细胞癌异种移植小鼠模型腹腔内注入D-阿洛糖后的肿瘤中D-阿洛糖浓度的推移。如图4所示,向肾细胞癌异种移植小鼠模型腹腔内注入D-阿洛糖后1小时内,在Caki-1和ACHN肿瘤中均测定到D-阿洛糖,在Caki-1中在腹腔内注入后的第1小时、在ACHN中在第2小时后达到最高值,以后的D-阿洛糖肿瘤内浓度下降。
[由于腹腔内注入D-阿洛糖导致的肾细胞癌异种移植小鼠模型中肿瘤体积的推移]
将肾细胞癌异种移植小鼠模型的肿瘤大小达到200mm3以上的日期作为day0,从day1起开始向腹腔内注入含有D-阿洛糖的溶液。所注入的溶液是0.4mL的生理盐水、将100mg/kg或400mg/kg的D-阿洛糖溶解于0.4mL的生理盐水而成的溶液,向分为3组(对照、D-阿洛糖100mg/kg、D-阿洛糖400mg/kg)的小鼠的腹腔内以每天1次共注入5周,一周2次经时性测定小鼠的体重和肿瘤的长短径。其中,肿瘤体积以肿瘤的长径×短径×短径×0.5算出。向腹腔内的注入开始后第35天,从小鼠中与肿瘤一同摘出肝脏和肾脏,用于之后的实验。
图5中表示由于腹腔内注入D-阿洛糖导致的肾细胞癌异种移植小鼠模型中肿瘤体积的推移。
如图5所示,在使用ACHN制作的异种移植小鼠模型的肿瘤体积评价中,D-阿洛糖400mg/kg组中,在第13天以后相比于对照组显著小。使用Caki-1制作的异种移植小鼠模型中在D-阿洛糖400mg/kg组中在第16天以后、并且在D-阿洛糖100mg/kg组中在第31天以后的肿瘤体积相比于对照组显著小,此外,D-阿洛糖400mg/kg组的第16天以后的肿瘤体积相比于给药前的肿瘤体积也显著小,得到不仅能够期待肿瘤增殖抑制效果,而根据D-阿洛糖的给药量还能够期待缩小效果(Mann-Whitney U test:曼-惠特尼U检验)的结果。
[肾细胞癌异种移植小鼠模型中D-阿洛糖的抗肿瘤效果:坏死率和核分裂数]
将摘出的肿瘤、肾脏和肝脏用4%多聚甲醛·磷酸缓冲液固定组织,用石蜡包埋后以4μm厚度进行薄切。将薄切切片进行苏木精-伊红染色,由病理医专家进行评价。通过坏死率(%)和核分裂数(Cells/HPF)像评价D-阿洛糖对肿瘤组织造成的影响。另外,坏死率使用图像记录软件Nikоn NIS-Elements D进行计测,舍去小数点以后的数值表示。另一方面,核分裂数利用3个视野的平均值算出,并且同样舍去小数点以后的数值。表10中表示肾细胞癌异种移植小鼠模型中的D-阿洛糖的抗肿瘤效果:坏死率和核分裂数。

结果,在使用Caki-1和ACHN制作的肾细胞癌异种移植小鼠模型的肿瘤组织中,依赖于D-阿洛糖的给药量,坏死率增加,核分裂数减少。
图6中表示D-阿洛糖对肾细胞癌异种移植小鼠模型的影响:体重推移的曲线图。小鼠体重在3组之间未见显著差异(Mann-Whitney Utest:曼-惠特尼U检验)。
图7中示出肾细胞癌异种移植小鼠模型中的D-阿洛糖的影响:表示肾脏组织的变化的显微镜照片(×100)。未见D-阿洛糖对肾脏组织的影响(由病理专家进行诊断)。
图8中示出肾细胞癌异种移植小鼠模型中的D-阿洛糖的影响:表示肝脏组织的变化的显微镜照片(×100)。未见D-阿洛糖对肝脏组织的影响(由病理专家进行诊断)。
产业上的利用可能性
本发明的发明人发现D-阿洛糖被人癌细胞中的人肾细胞癌细胞特异性摄取并且被摄取的D-阿洛糖具有抗肿瘤活性。由于D-阿洛糖是水溶性,所以膜透过性高,并且由于溶解于水性介质而不形成凝集物。
而且,由于特别是被肾细胞癌细胞特异性摄取,能够用作可将抗癌剂选择性地送达肾细胞癌细胞的药物载体,能够增加肾细胞癌细胞对抗癌剂的摄取,能够发挥被摄取的D-阿洛糖和抗癌剂双方的抗肿瘤活性,因此可期待作为对于肾细胞癌的突破性的分子靶向治疗药。

所述D-阿洛糖的衍生物为选自D-阿洛糖的羰基变成醇基而成的糖醇、D-阿洛糖的醇基氧化而成的糖醛酸、D-阿洛糖的醇基被NH2基取代而成的氨基糖中的D-阿洛糖衍生物。
