Abstract
本发明涉及一种西洋参鲜细胞的简易工业化生产方法,该方法包括以下步骤:配制储备液,装瓶,灭菌,制成固体培养基,在无菌条件下接入西洋参鲜细胞种源,放入培养室静止暗培养,30天左右可在固体培养基上收获西洋参鲜细胞培养物。本发明提供的工艺方法简单,生产周期短,成本低。利用该方法获得的西洋参鲜细胞除含有较高的皂甙成分,不含有新的多糖化合物,SOD,生物有机锗等生物活性物质。

本发明涉及一种植物组织培养技术,特别是西洋参鲜细胞组织培养方法。
西洋参(Panax Quinquefolium L.)主产于美国、加拿大、法国,其特点是性凉,味甘微苦,入心、肺、肾径,有益气养阴、清热生津、调补五脏、大补元气之功效。栽培西洋参,受土壤、气候的限制,条件要求高,产量小且生产周期长,生长4年以上才能收获,价格昂贵。
在应用植物组织和细胞培养技术生产天然药物方面的研究,国内从1964年就已开始,70年代中期昆明植物研究所、医科药物研究所、中国药科大学等单位先后进行了三分三、人参、三七、紫草、西洋参等组织和细胞培养研究,其中以三分三、人参较为成功,但主要是药用成分和药理活性以及建立高产稳产细胞株系的研究,80年代中期,中国药科大学在人参(吉林参)鲜细胞培养方面首次取得成功,并申请了中国专利(CN87102382,2)。继我国人参组织培养技术建立以后,前苏联、日本、美国、印度等国也先后开展了这方面的研究,日本在1973年曾有固体培养人参细胞的报道,1988年2吨罐大量培养获得成功之后,于1989年进行了20吨罐级大量培养的生产试验,正处于产品开发准备时期,但到目前为止,国内外还未见有关西洋参鲜细胞作产品或开发产品的报导。
本发明的目的在于提供一种西洋参鲜细胞组织培养方法,以实现西洋参鲜细胞工业化生产,弥补栽培西洋参生产周期长,产量小,成本高的缺陷,并提高有效物质含量。
本发明所述的西洋参鲜细胞的培养生产方法,经过以下步骤:

(3)灭菌:密封瓶口,将瓶放入高压灭菌装置中,在温度为115-122℃,压力1.0-1.2kg/cm2下灭菌20-30分钟,冷却后形成固体培养基;
(4)接种,培养:在固体培养基上,无菌条件下接入西洋参鲜细胞种源,然后放入培养室静止暗培养,室内温度控制在24-27℃,30天左右,在固体培养基表层取得西洋参鲜细胞培养物。每升储备液可收获西洋参培养物400-500克(干重25克左右)。
将本发明西洋参组培方法所生产的西洋参培养物与中国药品检验所提供的栽培西洋参标准样品对照,薄层分析,培养物与标样斑点一致,培养物的皂甙含量为13.2%,栽培西洋参含量为8%左右;水溶性总糖含量12-18.3%,高于栽培西洋参;SOD含量为340.7-1022.0U/克·干重;氨基酸总量为14.086%,15种氨基酸的含量如下:

本发明生产的西洋参培养物经过动物药理毒性试验,随机抽取健康小白鼠,按10克体重0.2ml计量给药,灌胃10天以上和腹腔注射72小时以上,均未发现任何异常反应,将心、肺、肾、肠、胃、子宫、睾丸等组织固定病理切片检查,均未发现对组织器官的毒性反应。
应用植物组织与细胞培养技术生产的西洋参鲜细胞,不仅有栽培西洋参的药理活性,而且还具有以下优点:
1.生产周期短,成本低。本发明是在人工可控条件下进行大量培养西洋参鲜细胞,每30天左右收获一次,生产率1∶6,生产周期比栽培西洋参缩短4年,生产成本仅为目前栽培市场售价的五分之一;
2.工艺简单,见效快。本发明应用生物工程高技术进行无土、无菌、细胞培养法,生产工艺简单,在缺少资金和植物培养反应器的条件下,可形成规模生产,具有投资少,见效快,消耗少等特点;
3.本方法培养的西洋参鲜细胞与栽培西洋参相比,除含有较高的皂甙成分,还含有新的多糖化合物、过氧化物歧化酶(SOD)、生物有机锗等生理活性物质,对提高机体免疫功能有较强的生理活性。
4.本发明培养的西洋参鲜细胞以其“鲜、嫩、活”的优势,使多种新型生理活性物质在制药医疗、化妆品、食品、营养保健品等方面有着非常广阔的开发前景。
下面提供本发明的实施例。
实施例1:
在400-500ml玻璃罐头瓶中,按附表中所列的试剂进行称量,加入一定量水,配制成储备液;按8克/升标准将琼脂溶解于热水中,再加入配好的储备液,充分搅拌后,趁热装入瓶中,液体厚度为7毫米,待冷却后形成固体薄层;用聚丙烯薄膜封住瓶口,将瓶放入高压灭菌锅中,在温度118℃±2℃,压力1.0-1.2kg/cm2
下灭菌20分钟,形成固体培养基;在该固体培养基上接入西洋参鲜细胞种源,然后放入培养室静止暗培养,室内温度为25℃±2℃;30天左右,在固体培养基上取得西洋参鲜细胞培养物。
实施例2:
按附表中的试剂剂量进行称量,加水配成储备液;按8克/升标准将琼脂溶解于热水中,再加入配好的储备液,充分搅拌后装入300ml锥形瓶中,液体厚度大致为6毫米左右,待冷却后形成固体薄膜;用塑料薄膜封住瓶口,在温度121℃,压力1.2kg/cm2下灭菌消毒20-30分钟,形成固体培养基;在固体培养基上,无菌条件接入西洋参鲜细胞种源,放入培养室静止暗培养35天,可在固体培养基上取得西洋参鲜细胞培养物。
实施例3:
按附表中所列的试剂剂量进行称量,以后各步骤与实施例1相同。

Claims (3)
1.一种西洋参鲜细胞简易工业化生产方法,其特征在于经过以下步骤:
(1)配制储备液;
(2)装瓶:按7-10克/升比例将琼脂溶解于热水中,加入配制好的储备液,充分搅拌后,装入瓶中;
(3)灭菌:密封瓶口,将温度控制在115-122℃,压力控制在1.0-1.2kg/cm2,灭菌20-30分钟,冷却形成固体培养基;
(4)接种,培养:在制成的固体培养基上,无菌条件下接入西洋参鲜细胞种源,然后放入培养室静止暗培养,28-35天,在固体培养基表层收取西洋参培养物。
2.根据权利要求1所述的西洋参鲜细胞简易工业化生产方法,其特征在于储备液的成分及含量如下:
