本发明涉及医药技术领域,公开了一种利咽祛痰颗粒,包括重量比为1:2的玉簪花冻干粉和辅料,所述辅料包括可溶性淀粉和糊精,所述可溶性淀粉和糊精的重量比为5:1。并进一步公开了利咽祛痰颗粒的制备工艺。本发明将玉簪花原药材进行提取,浓缩成清膏,冷冻干燥,制备颗粒剂,具有前体剂型的优点,服用方便、携带方便,避免了虫蛀问题的困扰,口感好、质量稳定,吸收快、显效迅速。通过本发明工艺制备的颗粒,仅仅采用一味主药材玉簪花,即可取得良好的抗炎、镇咳和祛痰效果。

技术领域
本发明涉及医药技术领域,具体涉及一种利咽祛痰颗粒及其制备工艺。
背景技术
玉簪花(Hostaplantaginea)为蒙医习用常用药材,蒙古名为哈斯-哈塔古尔-其其格,查干邦占,查干嘎尔布。为百合科植物玉簪的干燥花。《中华本草》(蒙药卷)记载其性寒,味涩、苦,效软、柔、稀,具有清热、解毒、止咳、利咽喉之功效,主治咽喉肿痛,音哑,肺热,毒热。
传统用药方法为:干燥的玉簪花冲茶引用,但是口感差,携带不便,冲泡不方便,容易虫蛀储藏不便。同时,不仅影响了玉簪花的真正药用部位的充分利用,浪费了药材成本,也影响了真正的治疗效果。
发明内容
为了解决上述现有技术的缺陷和不足,本发明目的在于提供一种利咽祛痰颗粒及其制备工艺。
为实现上述目的,本发明采用的技术方案是:
一种利咽祛痰颗粒,包括重量比为1:2的玉簪花冻干粉和辅料,所述辅料包括可溶性淀粉和糊精,所述可溶性淀粉和糊精的重量比为5:1。
进一步地,所述利咽祛痰颗粒的制备工艺,包括以下步骤:
步骤一、玉簪花提取:将玉簪花用去离子水提取,取滤液,并将滤液蒸发浓缩致密度为1.055g/mL,冷冻干燥后得到玉簪花冻干粉,备用。
步骤二、颗粒的制备:将辅料可溶性淀粉和糊精研磨成细粉,过50目筛备用;将步骤一制备的玉簪花冻干粉与辅料按照重量比1:2,充分混匀,用70%乙醇润湿,制软材,直至能够“手握成团,轻压即散”,经14目筛制粒,60℃干燥3h,整粒,筛除细粉,即得。
优选的,所述步骤一中,将玉簪花用去离子水提取时,料液比为1:40(g/mL)。
优选的,所述步骤一中,玉簪花用去离子水提取前浸泡4小时。
优选的,所述步骤一中,将玉簪花用去离子水提取2次,每次提取时间为4小时。优选的,所述步骤一中,将玉簪花用去离子水提取时,提取温度为100℃。
本发明还涉及所述利咽祛痰颗粒在制备治疗抗炎和/或镇咳和/或祛痰药物中的应用。
与现有技术相比,本发明具有以下有益效果:本发明首次将玉簪花原药材进行提取,浓缩成清膏,冷冻干燥,后制备成颗粒剂,具有前体剂型的优点,服用方便、携带方便,避免了虫蛀问题的困扰,口感好、质量稳定,吸收快、显效迅速,同时也克服前体剂型的不足;通过本发明工艺制备的颗粒,仅仅采用一味主药材玉簪花,即可取得良好的抗炎、镇咳和祛痰效果。

下面结合具体实施例对本发明进行详细说明。以下实施例将有助于本领域的技术人员进一步理解本发明,但不以任何形式限制本发明。应当指出的是,对本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进。这些都属于本发明的保护范围。
以下实施例所用的玉簪花采自辽宁阜新蒙古族自治县佛寺镇(海拔高度:272米,经度:121.334526,纬度:41.927155,气压:98KPa)。
实施例1
玉簪花提取单因素实验:影响玉簪花提取工艺的主要因素为料液比、提取时间,对此二因素依次进行考察,单因素实验设计及结果见表1、表2。
玉簪花提取方法:称取取玉簪花20g,去离子水提取,取滤液,水浴锅蒸发浓缩致密度1.055g/mL,冷冻干燥,备用。

实施例2
一种利咽祛痰颗粒,包括重量比为1:2的玉簪花冻干粉和辅料,所述辅料包括可溶性淀粉和糊精,所述可溶性淀粉和糊精的重量比为5:1。
所述利咽祛痰颗粒的制备工艺,如图1所示,包括以下步骤:
步骤一、玉簪花提取:采用正交实验设计提取,考察料液比、提取次数、浸泡时间、提取时间。根据结果显示,实验8的提取率最高,即料液比1:40,提取2次,浸泡4h,提取8h。具体为称取玉簪花原药材20g,去离子水提取,料液比为1:40(g/mL),100℃,提取2次,每次提取4小时,取滤液,水浴锅蒸发浓缩致密度1.055g/mL,冷冻干燥得到半成品玉簪花冻干粉,备用,浸出物测定干膏得率48.18%。如下表3和表4:

步骤二、颗粒的制备:将辅料可溶性淀粉、糊精研磨成细粉,过50目筛备用,称取玉簪花冻干粉6g,加入可溶性淀粉10g和糊精2g,采用等量递增法充分混匀,用70%乙醇润湿,制软材,不断搓捏混匀至“手握成团,轻压即散”,机械挤压得到颗粒初品,通过制粒筛。经14目筛制粒,60℃干燥3h,整粒,筛除细粉,得到玉簪花颗粒剂。质量检测,所得颗粒,能过5号筛与不能过1号筛的颗粒质量为11.97%,达到中国药典考察成型标准。

实施例3
玉簪花颗粒剂的抗炎实验。
实验方法:如图2所示,选取KM小鼠32只,雌雄各半,恒温恒湿条件下适应饲养3d。将小鼠随机分成空白组、模型组、对照组(玉簪清咽十五味丸0.83g/kg)、实验组(实施例1制备的玉簪化颗粒3.87g/kg,按含药材干膏量计算为1.29g/kg)。各组连续灌胃7d。末次给予相应药物1h后,致炎物质诱导法建立小鼠耳部炎症肿胀模型(致炎物质配制:巴豆油0.2mL、无水乙醚7.3mL、无水乙醇2mL、蒸馏水0.5mL,充分混合),2h后脱臼处死小鼠,沿耳廓基线剪下两耳,用打孔器沿耳廓相同部位冲下左右耳片,分别称重,以左右耳片重量之差作为肿胀程度,计算肿胀抑制率。进行统计SPS19.0分析,考察玉簪花颗粒抗炎疗效。
结果分析:与空白组比较,模型组肿胀率升高,有显著差异(P<0.01),表明诱导肿胀型可靠。与模型组比较,对照组、实验组肿胀率下降,有显著差异(P<0.05),表明玉簪花颗粒抗炎疗效显著,具体见表8。
注:##与空白组比较P<0.01,*与模型组比较P<0.05。
结论:玉簪花颗粒抗炎疗效显著,与方剂玉簪清咽十五味丸无显著差异。
实施例4
玉簪花颗粒剂的镇咳实验。
实验方法:如图2所示,选取KM小鼠32只,雌雄各半,恒温恒湿条件下适应饲养3d。将小鼠随机分成空白组、模型组、对照组(玉簪清咽十五味丸0.83g/kg)、实验组(实施例1制备的玉簪化颗粒3.87g/kg,按含药材干膏量计算为1.29g/kg)。各组连续灌胃7d。末次给予相应药物1h后,进行氨水引咳试验,观察各组小鼠咳嗽的潜伏期及其2min内的咳嗽次数。进行统计SPS19.0分析,考察玉簪花颗粒镇咳疗效。
结果分析:与空白组比较,模型组咳嗽次数增多,有显著差异(P<0.01),表明诱咳模型可靠。与模型组比较,对照组、实验组咳嗽潜伏期延长、且咳嗽次数减少,有显著差异(P<0.05),表明玉簪花颗粒镇咳疗效显著,见表9。
注:##与空白组比较P<0.01,*与模型组比较P<0.05。
结论:玉簪花颗粒镇咳疗效显著,与方剂玉簪清咽十五味丸无显著差异。
实施例5
玉簪花颗粒剂的祛痰实验。
实验方法:如图2所示,选取KM小鼠32只,雌雄各半,恒温恒湿条件下适应饲养3d。将小鼠随机分成空白组、模型组、对照组(玉簪清咽十五味丸0.83g/kg)、实验组(实施例1制备的玉簪化颗粒3.87g/kg,按含药材干膏量计算为1.29g/kg)。各组连续灌胃7d。末次给予相应药物1h后,每只小鼠腹腔注射5%酚红0.6mL(除空白组外),30min后处死动物,剥离气管,剪下甲状腺软骨至气管分支处的一段,置有1mL 5%NaHCO3的离心管中,用紫外分光光度计于550nm波长下测定吸光度值(OD值)。以空白组为校准,与模型组比较,监测各组酚红排泄OD值,OD值越高表明酚红排泄越高,即排痰越多。组间比较,进行SPS19.0统计分析,考察玉簪花颗粒祛痰疗效。
结果:与空白组比较,模型组酚红排泄OD值明显升高,有显著差异(P<0.01),表明该方法可行;与模型组比较,对照组、实验组酚红排泄OD值明显升高,有显著差异(P<0.01),表明酚红排泄增强,进一步表明玉簪花颗粒祛痰疗效显著,见表10。
注:##与空白组比较P<0.01,*与模型组比较P<0.05。
结论:玉簪花颗粒祛痰疗效显著,与方剂玉簪清咽十五味丸无显著差异。

Claims (6)
1.一种利咽祛痰颗粒,其特征在于,包括重量比为1:2的玉簪花冻干粉和辅料,所述辅料包括可溶性淀粉和糊精,所述可溶性淀粉和糊精的重量比为5:1;
所述利咽祛痰颗粒的制备工艺,包括以下步骤:
步骤一、玉簪花提取:将玉簪花用去离子水提取,取滤液,并将滤液蒸发浓缩致密度为1.055g/mL,冷冻干燥后得到玉簪花冻干粉,备用;
步骤二、颗粒的制备:将辅料可溶性淀粉和糊精研磨成细粉,过50目筛备用;将步骤一制备的玉簪花冻干粉与辅料按照重量比1:2,充分混匀,用70%乙醇润湿,制软材,直至能够“手握成团,轻压即散”,经14目筛制粒,60℃干燥3h,整粒,筛除细粉,即得。
2.根据权利要求1所述利咽祛痰颗粒,其特征在于,所述步骤一中,将玉簪花用去离子水提取时,料液比为1g:40ml。
3.根据权利要求1所述利咽祛痰颗粒,其特征在于,所述步骤一中,玉簪花用去离子水提取前浸泡4小时。
4.根据权利要求1所述利咽祛痰颗粒,其特征在于,所述步骤一中,将玉簪花用去离子水提取2次,每次提取时间为4小时。
5.根据权利要求1所述利咽祛痰颗粒,其特征在于,所述步骤一中,将玉簪花用去离子水提取时,提取温度为100℃。
6.如权利要求1~5任一项所述利咽祛痰颗粒在制备治疗抗炎和/或镇咳和/或祛痰药物中的应用。
