Abstract
本发明涉及一种一种缓解痛风的组合物及其制备方法和应用,属于药物组合物技术领域。该缓解痛风的组合物包括以下重量份的原料制备得到:苋菜30‑40份、玉米须5‑10份、樱桃浓缩液20‑30份、菊芋粉5‑10粉、山芹3‑5份。该组合物具有降低尿酸生成,加速嘌呤分解排泄,缓解局部疼痛作用。且本发明以药食同源纯天然食材为原料,不添加任何香精、色素、甜味剂及防腐剂;产品天然、安全、高效,无毒副作用、无刺激性、无上瘾性;生产中无污染,无废渣、废汽及有害物质产生,实现绿色生产。

技术领域
本发明涉及药物组合物技术领域,特别是涉及一种缓解痛风的组合物及其制备方法和应用。
背景技术
痛风是由慢性嘌呤代谢紊乱引起的一组异质性疾病。临床特征为高尿酸血症、反复发作的急性痛风性关节炎,痛风石和间质性肾炎,且在严重病例中表现为关节畸形和功能障碍。急性痛风性关节炎是痛风患者中最常见的首发症状,急性痛风性关节炎是由于该病患体内的尿酸排泄紊乱异常而引起的单钠尿酸盐沉积阻滞于滑膜、软骨等组织导致的一系列发炎症状,是一种急性发病炎症,患者经常会于清晨或深夜惊醒,疼痛十分剧烈,反应为数小时内局部关节出现红、热及明显压痛并伴有发热、白细胞增多和血沉加快等可反复发作的临床性疾病。
近年随着生活水平的提高,生活节奏日益加快,不良生活习惯如酗酒、过度劳累、受寒,进食含嘌呤食物都会增加急性痛风性关节炎的产生,使痛风发病几率在我国逐年攀高。
目前市场上改善痛风产品都含有不同程度的化学药物成分,对人体或多或少都有相应的副作用。
发明内容
基于此,有必要针对上述问题,提供一种缓解痛风的组合物,具有降低尿酸生成,加速嘌呤分解排泄,缓解局部疼痛作用。
本发明公开了一种缓解痛风的组合物,包括以下重量份的原料制备得到:苋菜30-40份、玉米须5-10份、樱桃浓缩液20-30份、菊芋粉5-10粉、山芹3-5份。
本发明人在前期调研中发现,黄嘌呤氧化酶是嘌呤代谢过程中的关键酶,若能有效抑制黄嘌呤氧化酶活性可以减少黄嘌呤向尿酸的转化,从而降低体内尿酸和超氧自由基的水平,进而达到缓解痛风的目的。
从此理论基础出发,再结合中医理论中认为,痛风是受外部阴寒水湿,侵袭人皮肉筋脉;内因为平素肥甘过度。寒与湿邪相结郁而化热,停留肌肤,病变部位红肿潮热,久则骨蚀。
本发明从上述西医理论出发,结合中医理论选材,选取了多种有利于抑制黄嘌呤氧化酶的天然植物,经高度浓缩萃取提纯,通过营养成分协调增效,实现缓解痛风症状。
具体的,本发明组合物中医组方原理为:苋菜性凉,能清热解毒,除湿利尿。玉米须性平,可利水消肿,轻身。菊芋粉清肝利胆,健胃消食,利尿消肿。三者可以提高尿液排泄量,进而将嘌呤排出体外,改善尿酸值过高的不良表现。樱桃性温味甘,可以补中益气,健脾和胃,加速身体代谢,同时樱桃中含有的樱桃花青素具有抑制肾小管重吸收尿酸的作用。山芹味苦、辛,可以解毒消肿。多种药食同源原料协同合力,互补增效,泄浊化瘀、清热利湿、调脾益肾、培补人体正气、消除关节肿胀、健体养身,以达到缓解痛风的目的。
在其中一个实施例中,该组合物还包括:维生素C1-2份、赤藓糖醇10-15份、小球藻提取物1-3份、硫酸锌0.2-0.5份、氯化钾0.3-0.5份、维生素B6 0.01-0.05份。
上述原料中,维生素C具有很强的还原性,可以修补受损组织,维持免疫功能。赤藓糖醇具有良好的清凉感甜味,热量极低,可在肠道快速吸收,不会被代谢分解,不影响胰岛素分泌水平。小球藻提取物可以修复受损细胞,同时利用自身碱性性质清除尿酸结晶石。硫酸锌、氯化钾可为机体提供必需的锌和钾矿物元素,锌有助于改善食欲和皮肤健康,钾可以促进尿液中钠的排泄。维生素B6有助于蛋白质的代谢和利用。上述原料共同协助苋菜、玉米须、樱桃浓缩液、菊芋粉和山芹等,起到更佳的效果。
在其中一个实施例中,所述樱桃浓缩液通过以下方法制备得到:选取樱桃,通过压榨设备破碎取汁,再经澄清及离心处理,去除粗纤维,得到樱桃汁,浓缩至原始体积的十分之一,即得到樱桃浓缩汁。
所述菊芋粉通过以下方法制备得到:以菊芋根状茎为原料,去除蛋白,胶质,粗纤维和矿物质,以水为溶剂进行提取,提取液进行离子交换,并以孔径为1-3μm的膜过滤,滤液喷雾干燥,得到菊芋粉;
所述小球藻提取物通过以下方法制备得到:取小球藻,以水为溶剂进行提取,得到小球藻悬浮液,加入果胶酶、纤维素酶、蛋白酶中的至少一种,经温育酶解处理,酶解液通过喷雾干燥,得到小球藻提取物。
在其中一个实施例中,所述菊芋粉通过以下方法制备得到:通过电渗析方法去除蛋白,胶质,粗纤维和矿物质,加入水后于50-60℃温度下提取1-2小时,提取液依次经过强酸阳离子交换树脂柱(例如:型号为D001的树脂柱),强碱性阴离子交换树脂柱(例如:型号为D290的树脂柱),弱碱性阴离子交换树脂柱(例如:型号为D311型的树脂柱),得到pH值为6.0-7.0且无色透明的洗脱液,将该洗脱液通过孔径为1-3μm的超滤膜过滤,滤液经浓缩喷雾干燥,即得;
所述小球藻提取物通过以下方法制备得到:取小球藻,以水为溶剂进行提取,得到小球藻悬浮液,按照果胶酶:纤维素酶:蛋白酶:小球藻为30000u:10000u:30000u:1g的量加入酶,在60-70℃pH7.5-8.5条件下酶解5-6h,酶解液通过喷雾干燥,得到小球藻提取物。
本发明还公开了上述的缓解痛风的组合物的制备方法,包括以下步骤:
洗涤:选取原料中固体组分,进行洗涤;
提取:取上述洗涤后原料,加入水,进行微波提取,得到提取液,备用;
纯化:取上述提取液,离心后取上清液,以大孔树脂进行纯化,得到纯化液,备用;
混合:取上述得到的纯化液,加入配方中其它原料,混合均匀,即得。
在其中一个实施例中,所述洗涤步骤中,将原料中苋菜、玉米须和山芹,置于上下震动幅度为6±3mm的水平运动震动筛上,进行动态喷淋清洗,振动筛水平运行速率3±1m/min,喷淋水流速8±2m/s。
在其中一个实施例中,所述提取步骤中,按照10±5ml水/1g原料的量加入水,浸泡后按照180±20W/kg的微波功率,在90-100℃条件下进行提取1±0.5h。
采用微波提取是一种高频电磁波穿透萃取媒介,到瞬间到达被萃取物料的内部,微波能迅速转化为热能使细胞内部温度快速上升,当细胞内部压力超过细胞壁承受能力,细胞破裂,细胞内有效成分自由流出,在较低的温度条件下溶解于萃取媒质再通过进一步的过滤和分离,便获得萃取物料。在微波辐射作用下被萃取物料成分加速向萃取溶剂界面扩散,从而使萃取速率提高数倍,同时还降低了萃取温度,最大限度的保证了萃取的质量。
在其中一个实施例中,所述纯化步骤中,离心条件为:6000±1000r/min的转速离心30±10min,将所述上清液浓度调整为0.3±0.1g原料/ml,再上样至大孔树脂。
在其中一个实施例中,所述纯化步骤中,所述大孔树脂进行纯化的具体步骤如下:
以AB-8型大孔树脂进行纯化,上样速度为2±0.5BV/h,静置吸附2±1h,用浓度为0.5%-0.7%的盐酸溶液洗脱4-6BV,再用体积百分浓度为50±10%的乙醇洗脱4-5BV,收集洗脱液,将回收乙醇后的洗脱液依次用4±1倍量的乙酸乙酯和4±1倍量的水饱和正丁醇萃取3次,将水相萃取液过滤,得到纯化液。
在其中一个实施例中,所述混合步骤中,采用以下方法进行混合:在压力为25-45MPa、温度为45-55℃的条件下进行均质化处理。
本发明还公开了上述的缓解痛风的组合物在制备用于预防和/或治疗痛风的药物中的应用,或在制备用于缓解痛风的保健品中的应用。
在其中一个实施例中,将上述缓解痛风的组合物制备为液态的食品剂型。

以下实施例所用原料,如非特别说明,均为市售可得;以下实施例所用方法,如非特别说明,均为常规方法可实现。
实施例1
一种缓解痛风的组合物,通过以下方法制备得到:
(一)选料
按照以下重量份选料:
取苋菜35份、玉米须7份、樱桃浓缩液25份、菊芋粉7份、山芹4份、维生素C1.5份、赤藓糖醇12份、小球藻提取物1.5份、硫酸锌0.35份、氯化钾0.4份、维生素B6为0.03份。
其中,樱桃浓缩液通过以下方法制备得到:选取优质、外表完整的新鲜樱桃,清洗去除杂质后,通过压榨设备进行破碎取汁,再经澄清及离心处理,去掉果皮及果肉中粗纤维,得到纯净的樱桃汁,经高效浓缩器浓缩至十分之一,即得到樱桃浓缩汁。
菊芋粉通过以下方法制备得到:以菊芋根状茎为原料,通过电渗析方法去除菊芋根状茎的蛋白,胶质,粗纤维和矿物质,加入水后于50-60℃温度下提取1-2小时,50-60℃下,提取液首先经过型号为D001的强酸阳离子交换树脂柱,控制流速为1.5-1.8L/h,再经过型号为D290的强碱性阴离子交换树脂柱,控制流速为1.1-1.4L/h,最后再经过型号为D311型的弱碱性阴离子交换树脂柱,控制流速为1.0-1.2L/h,得到pH值为6.0-7.0且无色透明的洗脱液,将该洗脱液通过孔径为1-3μm的超滤膜过滤,滤液经浓缩喷雾干燥,得到高纯度高聚合度菊芋粉。
小球藻提取物通过以下方法制备得到:取小球藻,以水为溶剂进行提取,得到小球藻悬浮液,按照果胶酶:纤维素酶:蛋白酶:小球藻为30000u:10000u:30000u:1g的量加入酶,在60-70℃pH7.5-8.5条件下酶解5-6h,酶解液通过喷雾干燥,得到小球藻提取物。
上述酶的配制,根据市售产品标注的活性单位unit进行计算称量并配合。
(二)洗涤
取配料中苋菜、玉米须、山芹,挑选去除不可食用的杂质及霉变部分,按配方量称重,置于开放的上下震动幅度为6mm的水平运动震动筛上,进行动态喷淋清洗,振动筛水平运行速率3m/min,喷淋水流速8m/s。
(三)提取
步骤(二)进行喷淋洗涤后的原料已经变得非常柔软,将步骤(二)获得的原料按质量份数比1g:10ml水的量加入饮用水,搅拌均匀,浸泡4h后,于微波功率180W/kg物料、温度达到100℃条件下,立即进入微波震荡提取和杀菌处理阶段,微波处理1h,促进物料中有效成分的溶出,同时杀灭有害微生物,得到提取液备用;
(四)纯化
将步骤(三)得到的提取液离心30min,转速为6000r/min。取其上清液,调整浓度为0.3g原料/ml,以AB-8型大孔树脂纯化,上样速度为2BV/h,静置吸附2h,用质量百分浓度为0.6%的盐酸溶液洗脱5BV,用体积百分浓度为50%的乙醇水溶液洗脱4BV,收集乙醇水溶液的洗脱液,回收乙醇,余下水性溶液再依次用4倍量的乙酸乙酯和水饱和正丁醇分别萃取3次,将萃取液过滤,得到纯化液。
(五)混合
无菌条件下将经步骤(四)处理得到的纯化液常温冷却至25℃,加入按(一)的配方准备的维生素C和赤藓糖醇、菊芋粉、樱桃浓缩液、小球藻提取物、硫酸锌、氯化钾、维生素B6,剪切均匀后,在压力25MPa、温度为46℃条件下进行均质化处理,使调配液均匀润滑、口感细腻、具有特殊的芳香;
(六)无菌灌装
将步骤(五)得到的产物无菌条件下灌入经过杀菌处理的容器中、封口,贮存。
(七)高温杀菌
将步骤(六)得到的成品放置于压力蒸汽灭菌器进行高温杀菌,杀菌后贮存。

一种缓解痛风的组合物,与实施例1中的组合物基本相同,区别仅在于:

实验例
取上述实施例和对比例制备得到的产品,进行小鼠血尿酸数据指标对比实验,确定其有效性,具体方法如下:
1.取昆明雄鼠,体重(25±5)g,随机分组,分别为空白组(给予生理盐水)、建模组。
2.空白组小鼠每天按照等体积给予生理盐水灌胃,进行饲养。
3.建模组按照每天20g·kg-1酵母膏灌胃,制备成高尿酸血症模型。
实验期间每日早间称其体重。建模组通过酵母膏灌胃的方法,给予小鼠大量嘌呤性物质,使小鼠血尿素值升高,嘌呤代谢紊乱,从而达到短时间内小鼠高尿酸水平的目的。
4.空白组、建模组小鼠连续7日灌胃后,于第8日早晨分别取血测定每只小鼠尿酸水平。根据血尿酸水平测试结果,从空白组平均值±30区域中选出10只小鼠,从建模组平均值±30区域中选出60只小鼠进行继续实验。
5.将建模组小鼠随机分为模型组和样品组一至五,每组10只。
模型组继续按照酵母膏灌胃饲养,样品组一给予每只小鼠实施例1制备得到的组合物溶液(0.25ml·d-1);样品组二给予每只小鼠对比例1制备得到的组合物溶液(0.25ml·d-1);样品组三给予每只小鼠对比例2制备得到的组合物溶液(0.25ml·d-1);样品组四给予每只小鼠对比例3制备得到的组合物溶液(0.25ml·d-1);样品组五给予每只小鼠实施例1提取步骤制备得到的提取液(0.25ml·d-1)。
6.空白组、模型组、各样品组小鼠继续喂养(给药)7d,第8日清晨取血其血尿酸水平。
结果如下表所示。

从上述结果可以看出,与空白组相比,模型组尿酸值显著升高,说明造模成功。各样品组与模型组相比较,小鼠血清尿酸值均有一定的下降,说明具有一定的降低尿酸作用。特别是其中实施例1制备产品的样品组一,小鼠血清尿酸值下降明显,具有显著性差异,改善高尿酸血症症状效果明显。
以上所述实施例的各技术特征可以进行任意的组合,为使描述简洁,未对上述实施例中的各个技术特征所有可能的组合都进行描述,然而,只要这些技术特征的组合不存在矛盾,都应当认为是本说明书记载的范围。
以上所述实施例仅表达了本发明的几种实施方式,其描述较为具体和详细,但并不能因此而理解为对发明专利范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进,这些都属于本发明的保护范围。因此,本发明专利的保护范围应以所附权利要求为准。
Claims (10)
1.一种缓解痛风的组合物,其特征在于,包括以下重量份的原料制备得到:
苋菜30-40份、玉米须5-10份、樱桃浓缩液20-30份、菊芋粉5-10粉、山芹3-5份。
2.根据权利要求1所述的缓解痛风的组合物,其特征在于,还包括:维生素C1-2份、赤藓糖醇10-15份、小球藻提取物1-3份、硫酸锌0.2-0.5份、氯化钾0.3-0.5份、维生素B60.01-0.05份。
3.根据权利要求1或2所述的缓解痛风的组合物,其特征在于,
所述樱桃浓缩液通过以下方法制备得到:选取樱桃,通过压榨设备破碎取汁,再经澄清及离心处理,去除粗纤维,得到樱桃汁,浓缩至原始体积的十分之一,即得到樱桃浓缩汁;
所述菊芋粉通过以下方法制备得到:以菊芋根状茎为原料,去除蛋白,胶质,粗纤维和矿物质,以水为溶剂进行提取,提取液进行离子交换,并以孔径为1-3μm的膜过滤,滤液喷雾干燥,得到菊芋粉;
所述小球藻提取物通过以下方法制备得到:取小球藻,以水为溶剂进行提取,得到小球藻悬浮液,加入果胶酶、纤维素酶、蛋白酶中的至少一种,经温育酶解处理,酶解液通过喷雾干燥,得到小球藻提取物。
4.根据权利要求3所述的缓解痛风的组合物,其特征在于,所述菊芋粉通过以下方法制备得到:过电渗析方法去除蛋白,胶质,粗纤维和矿物质,加入水后于50-60℃温度下提取1-2小时,提取液依次经过强酸阳离子交换树脂柱,强碱性阴离子交换树脂柱,弱碱性阴离子交换树脂柱,得到pH值为6.0-7.0且无色透明的洗脱液,将该洗脱液通过孔径为1-3μm的超滤膜过滤,滤液经浓缩喷雾干燥,即得;
所述小球藻提取物通过以下方法制备得到:取小球藻,以水为溶剂进行提取,得到小球藻悬浮液,按照果胶酶:纤维素酶:蛋白酶:小球藻为30000u:10000u:30000u:1g的量加入酶,在60-70℃pH7.5-8.5条件下酶解5-6h,酶解液通过喷雾干燥,得到小球藻提取物。
5.权利要求1-4任一项所述的缓解痛风的组合物的制备方法,其特征在于,包括以下步骤:
洗涤:选取原料中固体组分,进行洗涤;
提取:取上述洗涤后原料,加入水,进行微波提取,得到提取液,备用;
纯化:取上述提取液,离心后取上清液,以大孔树脂进行纯化,得到纯化液,备用;
混合:取上述得到的纯化液,加入配方中其它原料,混合均匀,即得。
6.根据权利要求5所述的缓解痛风的组合物的制备方法,其特征在于,所述洗涤步骤中,将原料中苋菜、玉米须和山芹,置于上下震动幅度为6±3m的水平运动震动筛上,进行动态喷淋清洗,振动筛水平运行速率3±1m/min,喷淋水流速8±2m/s。
7.根据权利要求54所述的缓解痛风的组合物的制备方法,其特征在于,所述提取步骤中,按照10±5ml水/1g原料的量加入水,浸泡后按照180±20W/kg的微波功率,在90-100℃条件下进行提取1±0.5h。
8.根据权利要求4所述的缓解痛风的组合物的制备方法,其特征在于,所述纯化步骤中,离心条件为:6000±1000r/min的转速离心30±10min,将所述上清液浓度调整为0.3±0.1g原料/ml,再上样至大孔树脂。
9.根据权利要求4所述的缓解痛风的组合物的制备方法,其特征在于,所述纯化步骤中,所述大孔树脂进行纯化的具体步骤如下:
以AB-8型大孔树脂进行纯化,上样速度为2±0.5BV/h,静置吸附2±1h,用浓度为0.5%-0.7%的盐酸溶液洗脱4-6BV,再用体积百分浓度为50±10%的乙醇洗脱4-5BV,收集洗脱液,将回收乙醇后的洗脱液依次用4±1倍量的乙酸乙酯和4±1倍量的水饱和正丁醇萃取3次,将得到的萃取液过滤,得到纯化液。
10.权利要求1-3任一项所述的缓解痛风的组合物在制备用于预防和/或治疗痛风的药物中的应用,或在制备用于缓解痛风的保健品中的应用。
