本发明涉及显色液技术领域,特别是涉及一种单组份TMB显色液。
背景技术
TMB显色液是一种以过氧化物酶为标记对象的免疫分析中常用的显色溶液。目前商品化的TMB显色液普遍只有1种过氧化物提供氧受体,有着保质期短,显色线性范围较窄等缺点。且传统的Elisa分析显色液是需要A、B两个配方,使用时再进行混合,操作繁琐。
发明内容
本发明要解决的技术问题是:现有的TMB显色液只有一种过氧化物提供氧受体,存在保质期短、显色线性范围较窄的缺点。
为了解决上述技术问题,本发明提供了一种单组份TMB显色液,按照重量份数计,包括以下组分:

优选的,所述TMB染色剂为3,3’,5’,5’-四甲基联苯胺或3,3’,5’,5’-四甲基联苯胺盐酸盐,所述3,3’,5’,5’-四甲基联苯胺盐酸盐的金属离子可以为钾或纳离子;所述过氧化物为过氧化乙酰丙酮、过氧化乙酸、过氧化尿素中的至少一种;所述缓冲剂为磷酸氢二钠和柠檬酸;所述螯合剂为EDTA;所述助溶剂为聚乙烯吡咯烷酮;所述溶剂为去离子水。
优选的,所述TMB染色剂的量为0.0450~0.0600份;所述过氧化物为0.0550~0.0750份;所述缓冲剂为3.6750~4.4900份;所述螯合剂为0.01300~0.01800份;所述助溶剂为1.8500~1.9000份;余量为溶剂。
优选的,所述过氧化物为过氧化乙酰丙酮、过氧化乙酸、过氧化钾的混合物,使用本发明的TMB单组份显色液,过氧化物酶能选择催化不同的过氧化物,其优先顺序依次为:过氧化尿素、过氧化乙酰丙酮、过氧化尿素;而三种过氧化物能够提供的供氧速度按照快慢顺序依次为:过氧化乙酸>过氧化乙酰丙酮>过氧化尿素,使得氧能够均匀且长时间释放,以形成不同的过氧化物供氧速度,最终达到灵敏显色的目的。

与现有技术相比,本发明具有以下优点:
1、本发明的单组份TMB显色液,采用多组分过氧化物提高TMB显色液的显色能力,限行范围的最高OD值可达到4.5,且在25℃条件下存储2年后,本底OD值仍然小于0.1;
2、本发明的单组份TMB显色液是一起配制成均质溶液,无需分成两个配方在使用时再混合,使用更加简单,显色效果更好。
具体实施方式
下面结合实施例,对本发明的技术方案进行清楚、完整的描述,显然,所描述的实施例仅是本发明的一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明的保护范围。
实施例1:本实施例提供一种单组份TMB显色液,包括0.6g的3,3’,5’,5’-四甲基联苯胺,16g柠檬酸,1L去离子水,0.2g过氧化乙酰丙酮,0.3g过氧化乙酸,0.3g过氧化尿素,36.8g磷酸氢二钠,0.185gEDTA,20g聚乙烯吡咯烷酮。将3,3’,5’,5’-四甲基联苯胺、柠檬酸依次溶于去离子水中,然后按先后顺序加入过氧化乙酰丙酮、过氧化乙酸、过氧化尿素、磷酸氢二钠、EDTA以及聚乙烯吡咯烷酮,搅拌至均匀,最后使用0.22μm的滤膜过滤除菌即得单组单组份TMB显色液,进行除菌分装。
实施例2:本实施例提供一种单组份TMB显色液,包括0.6g的3,3’,5’,5’-四甲基联苯胺盐酸盐,16g柠檬酸,1L去离子水,0.2g过氧化乙酰丙酮,0.3g过氧化乙酸,0.3g过氧化尿素,36.8g磷酸氢二钠,0.185gEDTA,20g聚乙烯吡咯烷酮。将3,3’,5’,5’-四甲基联苯胺、柠檬酸依次溶于去离子水中,然后按先后顺序加入过氧化乙酰丙酮、过氧化乙酸、过氧化尿素、磷酸氢二钠、EDTA以及聚乙烯吡咯烷酮,搅拌至均匀,最后使用0.22μm的滤膜过滤除菌即得单组单组份TMB显色液,进行除菌分装。
将本发明实施例1制备的单组份TMB显色液与市面上销售的单组份显色液以及传统双组份显色液进行显色效果实验对比,并将实验结果记录得到表1:

从表1中可以看出,常规ELISA实验的OD值对比中,本发明实施例1制备的单组份TMB显色液的OD值为2.52,明显大于市售的商品化单组份显色液1的OD值2.36、商品化单组份显色液2的OD值2.03,以及传统的双组份显色液的OD值1.88;在显色最大OD值的对比中,实施例1的显色最大OD值为4.5,均大于商品化单组份显色液1和2的显色最大OD值3.26、3.65,以及传统双组份显色液的显色最大OD值3.13;本发明实施例1在储存两年后的本底OD值仅为0.06,小于商品化单组份显色液1和2以及传统双组份显色液的本底OD值,对于显色效果实验结果的影响最小。

1.一种单组份TMB显色液,其特征在于,由以下组分组成,其中,各组分的质量浓度或质量为:
3,3’,5’,5’-四甲基联苯胺 0.6g/L;
过氧化乙酰丙酮 0.2g/L;
过氧化乙酸 0.3g/L;
过氧化尿素 0.3g/L;
磷酸氢二钠 36.8g/L;
柠檬酸 16g/L;
EDTA 0.185g/L;
聚乙烯吡咯烷酮 20g/L
去离子水 1kg。
2.一种单组份TMB显色液,其特征在于,由以下组分组成,其中,各组分的质量浓度或质量为:
3,3’,5’,5’-四甲基联苯胺盐酸盐 0.6g/L;
过氧化乙酰丙酮 0.2g/L;
过氧化乙酸 0.3g/L;
过氧化尿素 0.3g/L;
磷酸氢二钠 36.8g/L;
