技术领域
本发明涉及植物饮料技术领域,是一种天山冷水藻饮液及其制备方法。
背景技术
天山冷水藻(Tian Shan Cold Water Algae)起源于古称“静海”的赛里木湖古海藻。石炭纪之末,二叠纪之初,现今欧亚板块的雏形已经形成,新疆已成为这一全球性巨型板块的组成部分,但在它的南侧,先后经历了古特提斯洋的闭合,新特提斯的打开与最终关闭和青藏高原的形成多期事件,最终促成了地堑湖——赛里木湖。
赛里木湖是中国罕见的冷水湖,每年冬天都会结冰。冰层有一米,温度很低。温带季风将大西洋的大量水汽带向东方,因为很远的距离,这些水在路上形成降雨和湖泊。当大西洋水漂流到新疆,不再向东推进,所以人们把赛里木湖视为“大西洋的最后一滴眼泪”。
赛里木湖的形成可追溯到七千万年前地球上发生的一次大变动。雄伟壮丽的喜马拉雅山经过地壳运动而从古海中拔地而起,随着这次造山运动,天山山区的地壳也发生了剧烈的褶皱、变质和断裂,形成了天山山脉的现状。在此期间塞里木湖便陷落成高山湖,这种恶劣的环境演变造就了如今的天山冷水藻。
天山山脉高山区极端环境使天山冷水藻具备了独一无二的细胞特性,天山冷水藻在牙膏行业中可作为很有价值的原料,同时,具备了很高的开发及市场价值。我们通过原地取样并筛查分离出了富含细胞生长因子(CGF),高活性钾离子(K+),胞外碱性多糖,多不饱和脂肪酸的天山冷水藻细胞。
从天山山脉的高山湖泊赛里木湖中分离出来的古海藻中的天山冷水藻细胞具有以下优点:
(1)其细胞中高含量的CGF,改善口腔环境,修复破损的牙龈组织细胞,具有预防牙周炎、牙龈出血的功能;
(2)其细胞中丰富的碱性多糖,能促进牙周上皮生长和组织修复,延长蛋白核小球藻有效成分在牙周袋和口腔粘膜上的停留时间,从而有效减少牙斑菌滋生,提高口腔免疫力,保持洁净的口腔环境;
(3)其细胞中所包含的高活性钾离子(K+)能有效缓解牙本质过敏,改善患者牙齿的整体舒适度;
(4)其不饱和脂肪酸,能有效降低血浆中胆固醇、甘油三酯、LDL-C(低密度脂蛋白胆固醇)、VLDL-C(极低密度脂蛋白胆固醇),组织血栓形成,防止牙周组织营养缺陷。
目前,采用天山冷水藻为原料制备植物饮料尚未见报道。

本发明提供了一种天山冷水藻饮液及其制备方法,克服了上述现有技术之不足,其营养成分丰富、具有增强机体免疫力,调血压、降血脂,抗氧化、抗疲劳等功能,是一款品质极好的保健植物饮料。
本发明的技术方案之一是通过以下措施来实现的:一种天山冷水藻饮液,按下述方法得到:第一步,将天山冷水藻细胞在BG11液体培养基中进行无菌培养,得到天山冷水藻细胞培养物;第二步,将天山冷水藻细胞培养物依次经过离心,脱盐处理后,得到含水量为70%至75%的天山冷水藻藻泥;第三步,向天山冷水藻藻泥中加水稀释并混匀,得到质量浓度为40g/L的天山冷水藻藻泥溶液,将天山冷水藻藻泥溶液进行低温纳米破碎,得到破碎液;第四步,将破碎液进行离心,收集上清液,向上清液中加水进行调配,得到天山冷水藻饮液。

本发明中,BG11液体培养基为自主改良过的培养基,相比现有的BG11液体培养基,将天山冷水藻细胞置于自主改良的BG11液体培养基中进行培养,其培养时间短,培养时间为7天至12天,生物量的增值较快,一般在7天左右生物量就达到了1×108cell/mL,自制的BG11培养基培养不含任何危化品,成本低,使用方便。
本发明采用将天山冷水藻(蛋白核小球藻)细胞置于管道式培养装置中,在温度为28℃至31℃,光照为8000Lux至10000Lux,14h:10h光暗比例的无菌条件下进行培养,在自主设计的特殊的光波长条件下进行培育,生物量增长速度和质量均较好,得到的培养物再通过膜分离法收集藻泥(藻体),藻泥(藻体)利用超纯水稀释后进行低温纳米破碎,得到细胞所含的内质物(原始活性成分),藻泥(藻体)在低温纳米破碎条件下,其破碎率可达到99.0%至99.9%,再将经过低温纳米破碎后的细胞内质物加水调配后,得到天山冷水藻饮液。本发明整个工艺过程均在无菌条件下进行,完全避免了天山冷水藻原料的污染,保证了100%纯度的天山冷水藻活性成分的供给。
本发明天山冷水藻饮液深绿色液体,略带有清新绿藻味和果香味。营养成分丰富、具有增强机体免疫力,调血压、降血脂,抗氧化、抗疲劳等功能,是一款品质极好的保健植物饮料。

本发明使用的天山冷水藻细胞是取自于赛里木湖中的天山冷水藻。

本发明中,低温纳米破碎为现有公知的低温连续式破碎技术。
本发明的原料天山冷水藻(蛋白核小球藻)细胞具有以下特点:
(1)天山冷水藻细胞所富含的生长因子CGF有助于修复被破损的血脉内壁细胞,预防血液中沉淀物占到血脉内壁;
(2)天山冷水藻细胞所富含的碱性多糖有助于调节体液酸碱平衡,有利于恢复身体各器官的正常功能;
(3)天山冷水藻所富含的不饱和脂肪酸有助于降低血浆中胆固醇、甘油三酯、LDL-C(低密度脂蛋白胆固醇)、VLDL-C(极低密度脂蛋白胆固醇),有利于血栓预防,有利于防止心脑肾血管破损堵塞。
本发明天山冷水藻饮液具有以下特点:
(1)增强机体免疫力。天山冷水藻细胞含有的丰富细胞生长因子(CGF),在小鼠实验中,CGF被发现具有良好的免疫调节作用。它具有诱发干扰素,激发人体防御、免疫组织中的巨噬细胞、T 细胞和B 细胞的作用,能促进机体生长和体制的增强,并能强化免疫系统功能。蛋白核小球藻对身体虚弱的癌症患者以及放疗化疗后白细胞减少、免疫功能低下者具有良好的辅助作用;
(2)吸附体内毒素并排出体外。天山冷水藻细胞(蛋白核小球藻)中叶绿素含量高。叶绿素有很强的吸附毒素的作用,被誉为是肠、肾、肺血液中有害物质的“清道夫”。天山冷水藻细胞内丰富的叶绿素和植物纤维素,可排除体内化学毒素和重金属,有效保护肝、肾等排毒器官免受毒害,预防癌症发生;
(3)调血压、降血脂。天山冷水藻细胞内富含多不饱和脂肪酸,如亚油酸和γ- 亚麻酸等,具有调节血脂、降血黏等功效。这些天然而均衡的生物活性物质,可用于预防多种慢性疾病,构成人体健康的物质保障。因此天山冷水藻也可适合于高血脂症,防治动脉粥样硬化以及冠心病的辅助治;
(4)抗氧化、抗疲劳。天山冷水藻富含超氧化物歧化酶(SOD)和β- 胡萝卜素,尤其是富含叶黄素。这些物质具有很强的抗氧化能力,可以有效清除机体新陈代谢产生的各种自由基,长期服用可用于延缓衰老,改善心、肺功能。此外,蛋白核小球藻还富含多种维生素和微量元素,可快速补充人体缺乏的各种营养元素,促进消除疲劳、增强体力、辅助活化脑细胞;
(5)抗肿瘤。蛋白核小球藻细胞富含多种蛋白质,可以作为免疫激活剂,其抗肿瘤的机制可能为抑制致癌物的诱变性和基因毒性;增生核细胞;与宿主或受体一起作用;通过抗原专一性免疫;使T细胞增殖和活化等。小鼠体内体外实验表明,注射蛋白核小球藻热水抽提物后,癌细胞生长受到抑制,小鼠生存时间显著延长;
(6)抗感染、抗炎症。研究发现蛋白核小球藻粗提物对小鼠的消化性胃溃疡有显著的防治作用,能加强防溃疡形成的保护因子。临床试验证实,小球藻能够辅助治疗溃疡性结肠炎,加速伤口愈合。此外,研究发现蛋白核小球藻还可以抗巨噬细胞病毒的感染,减少感染病毒的复制,使器官免受病毒引起的组织病理学损害。
本发明实施例7至实施例9制备的天山冷水藻液饮营养成分丰富,具有增强机体免疫力,调血压、降血脂,抗氧化、抗疲劳等功能,以下是对本发明实施例9制备的天山冷水藻饮液的营养成分表、维生素和矿物质以及氨基酸含量进行考察。营养成分表、维生素和矿物质按照GB28050-2016《预包装食品营养标签通则》中规定的方法进行检测,氨基酸含量按照GB 5009.124-2016《食品安全国家标准食品中氨基酸的测定》中规定的方法进行检测。营养成分表如表1所示,维生素和矿物质如表2所示,氨基酸含量如表3所示,由表1、表2和表3可知,本发明实施例9制备的天山冷水藻饮液中的营养成分、维生素和矿物质以及氨基酸含量丰富,品质佳。
综上所述,本发明天山冷水藻饮液深绿色液体,略带有清新绿藻味和果香味。营养成分丰富、具有增强机体免疫力,调血压、降血脂,抗氧化、抗疲劳等功能,是一款品质极好的保健植物饮料。
以上技术特征构成了本发明的实施例,其具有较强的适应性和实施效果,可根据实际需要增减非必要的技术特征,来满足不同情况的需求。
Claims (9)
1.一种天山冷水藻饮液,其特征在于按下述方法得到:第一步,将天山冷水藻细胞在BG11液体培养基中进行无菌培养,得到天山冷水藻细胞培养物;第二步,将天山冷水藻细胞培养物依次经过离心,脱盐处理后,得到含水量为70%至75%的天山冷水藻藻泥;第三步,向天山冷水藻藻泥中加水稀释并混匀,得到质量浓度为40g/L的天山冷水藻藻泥溶液,将天山冷水藻藻泥溶液进行低温纳米破碎,得到破碎液;第四步,将破碎液进行离心,收集上清液,向上清液中加水进行调配,得到天山冷水藻饮液。
2.根据权利要求1所述的天山冷水藻饮液,其特征在于BG11液体培养基,原料包括氯化铵0.5g/L、三水合磷酸氢二钾0.04g/L、七水合硫酸镁0.075g/L、二水合氯化钙0.036g/L、柠檬酸0.006g/L、柠檬酸铁铵0.006g/L、乙二胺四乙酸二钠0.001g/L、乙酸铵2.4g/L、硼酸0.286g/L、一水合硫酸锰0.181g/L、七水合硫酸锌0.000222g/L、五水合硫酸铜0.000079g/L、四水合钼酸铵0.00039g/L和氯化钴0.000049g/L。
3.根据权利要求1或2所述的天山冷水藻饮液,其特征在于第一步中,培养条件:温度为28℃至31℃,光照为8000Lux至10000Lux,14h:10h光暗比例的无菌条件下进行,培养时间为7天至12天,当生物量达到1×108cell/mL后进行收集。
4.根据权利要求1或2所述的天山冷水藻饮液,其特征在于第二步中,离心时,转速为15000rpm至20000rpm,流速为180L/h至220L/h,离心后,收集沉淀物。
5.根据权利要求3所述的天山冷水藻饮液,其特征在于第二步中,离心时,转速为15000rpm至20000rpm,流速为180L/h至220L/h,离心后,收集沉淀物。
6.根据权利要求1或2或5所述的天山冷水藻饮液,其特征在于第二步中,脱盐处理为用水冲洗3次进行脱盐,每次冲洗后进行离心,收集沉淀物,离心时,转速为15000rpm至20000rpm,流速为180L/h至220L/h;或/和,第三步中,低温纳米破碎条件为在温度为3℃至5℃,280bar至320bar低频率条件下循环破碎3轮至4轮后,在温度为3℃至5℃,800bar至1200bar高频率条件下循环破碎5轮至6轮。
7.根据权利要求3所述的天山冷水藻饮液,其特征在于第二步中,脱盐处理为用水冲洗3次进行脱盐,每次冲洗后进行离心,收集沉淀物,离心时,转速为15000rpm至20000rpm,流速为180L/h至220L/h;或/和,第三步中,低温纳米破碎条件为在温度为3℃至5℃,280bar至320bar低频率条件下循环破碎3轮至4轮后,在温度为3℃至5℃,800bar至1200bar高频率条件下循环破碎5轮至6轮。
8.根据权利要求4所述的天山冷水藻饮液,其特征在于第二步中,脱盐处理为用水冲洗3次进行脱盐,每次冲洗后进行离心,收集沉淀物,离心时,转速为15000rpm至20000rpm,流速为180L/h至220L/h;或/和,第三步中,低温纳米破碎条件为在温度为3℃至5℃,280bar至320bar低频率条件下循环破碎3轮至4轮后,在温度为3℃至5℃,800bar至1200bar高频率条件下循环破碎5轮至6轮。
9.一种根据权利要求2至8中任意一项所述的天山冷水藻饮液的制备方法,其特征在于按下述方法进行:第一步,将天山冷水藻细胞在BG11液体培养基中进行无菌培养,得到天山冷水藻细胞培养物;第二步,将天山冷水藻细胞培养物依次经过离心,脱盐处理后,得到含水量为70%至75%的天山冷水藻藻泥;第三步,向天山冷水藻藻泥中加水稀释并混匀,得到质量浓度为40g/L的天山冷水藻藻泥溶液,将天山冷水藻藻泥溶液进行低温纳米破碎,得到破碎液;第四步,将破碎液进行离心,收集上清液,向上清液中加水进行调配,得到天山冷水藻饮液。
