Abstract

背景技术
高毒性重金属引发的环境和水体污染一直是人们关心的主要问题,它们可以通过空气、水体和土壤进入食物链严重危害生态系统。
砷是一种自然界中广泛存在的、具有类金属特性的元素。砷及其化合物已被世界卫生组织(WHO)下属的国际癌症研究机构和美国环保署(EPA)等诸多国际权威机构公认为致癌物。砷污染主要来自于农药、化肥、冶金、采矿、制药等行业的三废排放。砷在水环境中的主要存在形式是H3AsO3中的As(Ш)和H3AsO4中的As(V),其中,As(Ш)毒性最高,是As(V)毒性的100倍,慢性摄入或长期接触砷会使其大量沉积在人体皮肤、肺、膀胱、肝和肾中,引发肺癌、皮肤癌等多种癌变,同时还会导致心血管疾病和人体循环系统的损害。由于砷污染的进一步扩大,人类正面临严峻的砷中毒威胁,砷污染导致的以皮肤和内脏疾病为主的健康危害已在全世界范围内受到广泛关注。为此,美国环境保护署最近重新调整了饮用水中的砷含量从50μg/L到10μg/L。该变化促使更多的研究人员开发出能够高灵敏检测砷的技术以满足这一新的标准。
严重危害人类健康和环境的重金属还包括铅,铅污染主要来源于人类生活及化工生产等相关领域,涉及油漆、涂料、蓄电池、冶炼、电镀、餐具、燃煤、自来水管、化妆品、染发剂等产品。铅是一种潜在的神经毒素,也是普遍存在于自然界的重金属污染中毒性较大的一种。对环境和人体健康产生严重的影响,一旦进入人体将很难排除,直接损害人的脑细胞。即使暴露于非常低水平的铅离子浓度(血液中<100μg/L)下也会引起多种疾病,包括记忆力减退、易怒、贫血、肾损伤和智力低下等,造成各种神经毒性作用,引发神经系统、心血管系统损伤和发育障碍,特别是对于生长发育期的儿童来说,危害尤为巨大。铅可经呼吸吸入、食物和饮水摄入、经皮肤吸收。铅的毒性作用没有阈值性,即体内有铅便有毒,因此,“零血铅”已成为临床控制儿童铅中毒的目标。
除了砷和铅以外,金属汞也是一种具有严重毒性且分布广泛的重要环境污染物,即使在低浓度下也会对环境和人类构成严重威胁,造成严重的、永久的损害。Hg2+废水污染物是构成水体污染的重要组成部分,排入水中的无极汞污染物不能被生物降解,而是参与食物链循环,可被微生物转化为毒性更强的甲基汞,在生物体内大量富集后经过食物链(尤其是鱼类)进入人体,严重威胁着人类的健康,可造成中枢神经系统、内分泌系统及脑部的损害,此外,它可能还会破坏人体免疫系统,甚至导致死亡。为此,美国环境保护署提出了饮用水中汞的最大容许值为10nM。
因此,建立灵敏、准确、高效的可同时检测As3+、Pb2+、Hg2+的分析方法对于环境监测、食品安全和生物医药等领域至关重要。
目前,用于同时检测As3+、Pb2+、Hg2+的检测方法非常少,远远少于单个离子的检测方法。对于单一金属离子的检测,传统技术主要包括电感耦合等离子体质谱法(ICP-AES)、原子吸收/发射光谱法和极谱法等方法。但是,这些方法常常需要复杂的专业操作、耗时的分析和贵重的仪器设备等,限制了它们在资源有限的环境中的使用。由于荧光传感技术具有灵敏、简单、快捷等优点而得到日益广泛的研究。不可否认,文献报道的方法存在检测灵敏度不高、选择性不强等问题。为了克服这些缺点,迫切需要开发出既简单、灵敏、又经济、高效的测定方法来满足环境、生物、医药等领域对于As3+、Pb2+、Hg2+的同时检测的需求。
据文献报道,当金属离子进入错配的DNA碱基对时,金属离子和DNA碱基对之间会产生一种吸引力,将它们结合在一起。例如,Hg2+能够选择性地识别胸腺嘧啶碱基并形成强而稳定的胸腺嘧啶-Hg2+-胸腺嘧啶配位复合物。与汞离子类似,铅离子也能够特异性地和核苷酸发生作用,由于富含鸟嘌呤碱基的核酸分子对铅离子具有很强的亲和力,通过特异性的配位作用与铅离子结合而形成结构稳定的Pb2+-G-四链体,可以实现对铅离子高选择性、高灵敏的检测。对于As3+而言,可以与其核酸适配体特异性结合,但有关适配体的报道比较少见。
正是由于As3+对核酸适配体、Hg2+和Pb2+分别对富含胸腺嘧啶、鸟嘌呤碱基的功能核酸分子的亲和力高、特异性强的识别作用而形成结构稳定的金属离子-核酸分子配位复合物,该作用对于这3种金属离子的高灵敏、高特异性检测具有重要意义。本发明根据As3+、Hg2+和Pb2+的这一反应特性,将其分别与核酸适配体、富含胸腺嘧啶、鸟嘌呤的核酸生物分子相结合,巧妙地应用到具有中空、多孔结构的纳米材料领域,同时,为了进一步提高As3+、Hg2+和Pb2+的检测灵敏度,在生物识别技术的基础上,又结合了生物酶的剪切作用,使靶标As3+、Hg2+和Pb2+得到循环利用,最终使检测信号得到显著放大,为As3+、Hg2+和Pb2+的同时、高灵敏检测提供了新型、特异、高效的检测技术。
迄今为止,基于酶循环放大技术的、采用中空多孔纳米材料、利用碱基识别作用构建纳米胶囊-核酸生物分子复合物用于同时检测As3+、Hg2+和Pb2+的技术还未见文献报道。
发明内容
为了克服现有技术存在的不足,针对基于核酸外切酶循环放大技术与分子识别技术的纳米胶囊-核酸生物分子复合物用于同时检测As3+、Hg2+和Pb2+的技术未见报道,因此,本发明的第一目的:提出并构建一种新型的可同时检测As3+、Hg2+和Pb2+的基于核酸外切酶循环放大技术与分子识别技术的纳米胶囊-核酸生物分子复合物,具体是利用中空、多孔的纳米金作为纳米胶囊,分别筛选、设计并合成易于修饰、方便合成的、可被As3+、Hg2+和Pb2+识别的核酸生物分子,并将其分别组装到纳米胶囊表面,一方面作为堵孔材料用于封堵纳米胶囊的孔口,防止孔内物质的外泄;另一方面分别作为As3+、Hg2+和Pb2+的识别分子可与As3+、Hg2+和Pb2+发生特异性的识别反应,其中,Hg2+和Pb2+可分别形成T-Hg2+-T碱基对复合物、Pb2 +-G四链体复合物,在形成金属离子-核酸分子配位复合物的同时发生构象转化而脱离纳米胶囊表面,从而使封堵的孔口被打开,纳米胶囊内的染料分子得以释放,分离上清液,在一定波长的激发光照射下产生荧光信号,根据荧光发射信号的强弱实现对As3+、Hg2+和Pb2+的同时检测。在本发明中,为了进一步提高同时检测As3+、Hg2+和Pb2+的灵敏度,在分子识别的基础上,本发明又利用外切酶的剪切作用实现了荧光信号的循环放大。该循环放大技术利用了核酸外切酶可对具有双链结构的金属离子-核酸生物分子配位复合物进行剪切的作用,将As3+、Hg2+和Pb2+释放,释放出来的As3+、Hg2+和Pb2+能够再次与纳米胶囊表面的生物识别分子发生识别反应,然后再次被剪切……,循环往复,结果使As3+、Hg2+和Pb2+得到多次循环利用,更多的孔口被打开,释放出更多的孔内物质。正是由于剪切酶的作用,As3+、Hg2+和Pb2+被循环利用,使检测灵敏度得到显著提高。该技术可实现对于含量低至痕量的As3+、Hg2+和Pb2+样品的高灵敏、高选择性的同时检测。即便样品中同时含有极微量的As3+、Hg2+和Pb2+,也能够获得令人满意的检测结果。本发明的第二目的:提供一种同时检测As3+、Hg2+和Pb2+的基于核酸外切酶循环放大技术与分子识别技术的纳米胶囊-核酸生物分子复合物的制备方法。本发明的第三目的:提供一种同时检测As3+、Hg2+和Pb2+的方法。
本发明是通过以下技术方案实现发明目的的。本发明提出的可同时检测As3+、Hg2+和Pb2+的纳米胶囊-核酸生物分子复合物是以中空、多孔结构的纳米金作为纳米胶囊,利用其中空、多孔的结构特性,在其内部分别装载客体分子如荧光染料;为了实现对As3+、Hg2+和Pb2+的同时检测,分别筛选、设计并合成了3种核酸生物识别分子,通过对碱基种类及其数量的选择,确保其对且仅对目标金属离子As3+、Hg2+或Pb2+具有特异性的响应,而对其它任何共存离子和干扰离子没有响应。在此基础上,又对采用的3种荧光信号分子进行了相关的实验和筛选,为了克服同时采用的3种荧光分子之间的相互干扰和使用条件的限制,综合考虑溶解性好、荧光强、Stokes位移大、光学性质稳定、信噪比高、无光漂白、受环境影响小、生物相容性好等性能,最终选择采用的荧光染料优选罗丹明B、异硫氰酸荧光素FITC和荧光素Cy5。结果表明,这3种荧光信号分子的选择和使用成为本发明的关键条件之一。为了实现生物分子对As3+、Hg2+和Pb2+的特异性识别,同时也为了防止荧光信号分子的泄漏,本发明筛选并合成了可分别被As3+和Pb2+识别的生物分子,设计并合成了可被Hg2+识别的生物分子,并将它们分别组装到纳米胶囊表面用于封堵孔口,起到防止孔内荧光分子外泄的作用;其中,特异性识别As3+的碱基序列为:5’-GGTAATACGACT CAC TAT AGG GAG ATA CCA GCT TAT TCA ATTTTA CAG AAC AAC CAA CGT CGC TCC GGG TAC TCC TTC TTCATC GAGATAGTAAGT GCAATC C-3’(P1);富含鸟嘌呤碱基的特异性识别Pb2+的碱基序列为:5’-GGT GGT GGT GGT TGT GGTGGT GGT GG-3’(P2);可识别Hg2+的富含胸腺嘧啶的生物分子是经过特别设计并合成的、具有特定碱基长度的核酸生物分子,其碱基序列为:5’-TTT GTT TGT TGG CCC CCC TTC TTTCTTA-3’(P3);由于这3种序列对且仅对相应的金属离子有响应,因此,该检测体系能够完全避免相互干扰,确保同时检测的准确性和特异性。
作为可分别识别As3+、Hg2+和Pb2+的3种核酸生物分子是通过静电作用被分别组装到中空、多孔结构的纳米金表面,而纳米金表面则需要通过采用正电荷修饰剂进行预先修饰,优选采用地正电荷修饰剂是聚二烯丙基二甲基氯化铵。当靶标As3+、Hg2+和Pb2+存在时,被组装到纳米胶囊表面的核酸生物分子分别对As3+、Hg2+和Pb2+发生特异性的响应,3种离子分别与其相应的核酸生物分子特异性结合,使其构象发生改变,从纳米胶囊表面脱落,使封堵的孔口被打开,从而导致RhB、异硫氰酸荧光素FITC和Cy5从纳米胶囊内部被释放出来。对释放出的3种荧光染料分别给予相应波长的激发,均得到显著增强的3种染料的荧光信号。当加入不同浓度的As3+、Hg2+和Pb2+时,荧光信号与加入的离子浓度呈现良好的响应关系。因此,根据测得的荧光信号强度就可以实现对As3+、Hg2+和Pb2+的同时检测。
为了进一步提高同时检测As3+、Hg2+和Pb2+的灵敏度,在特异性生物识别的基础上,本发明又利用核酸外切酶的剪切作用实现了荧光信号的循环放大。该循环放大技术利用了核酸外切酶可对具有双链结构的金属离子-核酸生物分子配位复合物进行剪切的作用,将As3+、Hg2+和Pb2+释放,而释放出来的As3+、Hg2+和Pb2+能够再次与纳米胶囊表面的生物识别分子发生识别反应,然后再次被剪切……,循环往复,结果使As3+、Hg2+和Pb2+得到多次循环利用,更多的孔口被打开,释放出更多的荧光物质,进一步实现荧光信号的显著增强,优选采用地核酸外切酶是外切酶ExoⅢ。
一种制备本发明提出的可同时检测As3+、Hg2+和Pb2+的基于核酸外切酶循环放大技术与生物分子识别技术的纳米胶囊-核酸分子复合物的制备方法,包括如下步骤:
(1)分别筛选、设计并合成3种核酸生物识别分子,确保其分别对且仅对目标金属离子As3+、Pb2+和Hg2+具有特异性的响应,而对其它任何共存离子和干扰离子没有响应;
(2)将磁珠用pH=7.4的Tris-HCl缓冲溶液清洗后与具有中空、多孔结构的纳米金载体溶液混合,加入聚二烯丙基二甲基氯化铵溶液,10-12h后磁分离,用pH=7.4的Tris-HCl缓冲溶液清洗;
(3)加入荧光分子Cy5溶液,10-12h后加入可识别As3+的核酸生物分子溶液,10-12h后磁分离,用pH=7.4的Tris-HCl缓冲溶液清洗,备用;
其中,所述的可识别As3+的核酸生物分子的碱基序列为5’-GGTAATACGACT CAC TATAGG GAG ATA CCA GCT TAT TCA ATT TTA CAG AAC AAC CAA CGT CGC TCC GGG TAC TCCTTC TTC ATC GAGATAGTAAGT GCAATC C-3’(P1),它和As3+的特异性结合产物能够被核酸外切酶剪切,释放出As3+;
(4)在(2)步产物中加入罗丹明B溶液,10-12h后加入可识别Pb2+的核酸生物分子溶液,10-12h后磁分离,用pH=7.4的Tris-HCl缓冲溶液清洗,备用;
其中,所述的可识别Pb2+的核酸生物分子的碱基序列为5’-GGT GGT GGT GGT TGTGGT GGT GGT GG-3’(P2),它和Pb2+的特异性结合产物能够被核酸外切酶剪切,释放出Pb2+;
(5)在(2)步产物中加入异硫氰酸荧光素FITC溶液,10-12h后加入可识别Hg2+的核酸生物分子溶液,10-12h后磁分离,用pH=7.4的Tris-HCl缓冲溶液清洗,备用;
其中,所述的可识别Hg2+的核酸生物分子的碱基序列为5’-TTT GTT TGT TGG CCCCCC TTC TTT CTTA-3’(P3),它和Hg2+的特异性结合产物能够被核酸外切酶剪切,释放出Hg2 +;
(6)将(3)步、(4)步和(5)步所得产物分别用pH=7.4的Tris-HCl缓冲溶液稀释后混合,磁分离,即可制得同时检测As3+、Pb2+和Hg2+的纳米胶囊-核酸生物分子复合物。
一种本发明提出的可同时检测As3+、Pb2+和Hg2+的方法,包括以下步骤:
(1)将含有As3+、Pb2+和Hg2+的溶液加入到本发明制备的纳米胶囊-核酸生物分子复合物中,加入剪切酶EXOⅢ,用pH=7.4的Tris-HCl缓冲溶液稀释,放入37℃摇床反应1-3h;
(2)磁分离,取上清液进行荧光检测,分别给予相应波长的激发,检测与As3+、Pb2+和Hg2+相对应的3种染料的荧光信号。
本发明的有益效果:本发明提出的可同时检测As3+、Pb2+和Hg2+的方法不但能够高效、经济地实现对As3+、Pb2+和Hg2+的同时检测,而且还在原有的基于可控释放放大的基础上,通过生物酶的剪切作用及靶标物的循环利用,最终实现了同时对3种荧光信号的循环放大与检测;尤为重要的是,本发明克服了传统技术缺陷,无需再对核酸生物分子进行任何荧光标记,完全避免了对核酸生物分子性能的影响;本发明通过简单高效地多重放大技术,使荧光信号从1:1模式转变为指数型放大模式,灵敏度得到显著增强。
结果表明,与现有技术相比,采用该体系同时检测As3+、Pb2+和Hg2+,可同时实现对As3+、Pb2+和Hg2+的高灵敏、高选择性的检测。而且,所采用的3种核酸生物识别分子分别对且仅对目标金属离子As3+、Pb2+和Hg2+具有特异性的响应,而对其它任何共存离子和干扰离子没有响应。当样品中仅含其中一种金属离子As3+、Pb2+或Hg2+时,该体系也能灵敏地检测出所含金属离子对应的荧光信号。因此,本发明提出的纳米胶囊-核酸生物分子复合物除了用于As3+、Pb2+和Hg2+的同时检测,也可用于仅含As3+、Pb2+、Hg2+或其中2种离子样品的单独或同时检测,所有这些检测体系均能获得高灵敏、高选择性的检测结果。
本发明提出的纳米胶囊-核酸生物分子复合物具有结构简单、易合成、性能优异、稳定、适用范围广、高效、灵敏等优点,并且不会受到其它常见干扰物质如Cd2+、Fe3+、Zn2+、Cu2+等金属离子的影响,同时,As3+、Pb2+和Hg2+相互之间也不存在干扰;具有高的特异性和选择性。实验结果表明,相比于传统的技术方法,采用本发明提出的纳米胶囊-核酸生物分子复合物对As3+、Pb2+和Hg2+进行了同时检测,显示出高灵敏度和优异的选择性,其中,As3+浓度在5.0×10-12~1.0×10-10mol/L范围内满足线性方程: Pb2+浓度在1.0×10-13~8.0×10-11mol/L范围内满足线性方程: Hg2+浓度在5.0×10-13~8.0×10-11mol/L范围满足线性方程:与文献值相比,本发明对As3+、Pb2+和Hg2+的同时检测在确保高特异性的前提下,灵敏度得到显著提高,提高10倍以上,同时,还获得了对3种离子更宽的线性检测范围。可以预见,本发明提出的纳米胶囊-核酸生物分子复合物及其制备方法和检测技术具有巨大的医学应用潜力和广阔的应用前景,将在重大疾病的早期诊断与治疗以及食品、生物医药、环境监测等领域发挥重要的作用。

以下是本发明涉及的具体实施例,对本发明的技术方案做进一步描述,但是本发明的保护范围并不限于这些实施例。凡是不背离本发明构思的改变或等同替代均包括在本发明的保护范围之内。
下面通过实施例具体地说明本发明,但本发明不受下述实施例的限定。
实验仪器:THZ-82A气浴恒温振荡器(金坛市医疗器械厂);F-4600荧光分光光度计(日立,日本);磁性分离架(天津倍思乐色谱技术开发中心)。
实验试剂:核酸外切酶ExoШ(Thermo Scientific,美国);罗丹明B(上海阿拉丁生化科技股份有限公司);异硫氰酸荧光素FITC、荧光素Cy5(Solarbio,北京索莱宝科技有限公司);聚二烯基丙二甲基氯化铵、硫酸汞、硝酸铅(上海阿拉丁生化科技股份有限公司);3-4μm巯基磁珠(天津市倍思乐色谱技术开发中心);亚砷酸钠母液、5’-GGTAAT ACG ACT CACTAT AGG GAG ATA CCA GCT TAT TCA ATT TTA CAG AAC AAC CAA CGT CGC TCC GGG TACTCC TTC TTC ATC GAGATA GTAAGT GCAATC C-3’(P1)、5’-GGT GGT GGT GGT TGT GGT GGTGGT GG-3’(P2)、5’-TTT GTT TGT TGG CCC CCC TTC TTT CTTA-3’(P3)(上海生工生物工程股份有限公司);Tris-HCl缓冲溶液pH=7.4,浓度为0.05M(上海阿拉丁生化科技股份有限公司)。
实施例1:
一种制备本发明提出的可同时检测As3+、Pb2+和Hg2+的、基于核酸外切酶循环放大技术与生物分子识别技术的纳米胶囊-核酸生物分子复合物的方法,包括如下步骤:
(1)分别筛选、设计并合成3种核酸生物识别分子,确保其分别对且仅对目标金属离子As3+、Pb2+和Hg2+具有特异性的响应,而对其它任何共存离子和干扰离子没有响应;
(2)用pH=7.4的Tris-HCl缓冲溶液清洗20μL磁珠后加入400μL具有中空、多孔结构的纳米金载体溶液,加入200μL 11.664mg/mL的聚二烯丙基二甲基氯化铵溶液,37℃10h后磁分离,用pH=7.4的Tris-HCl缓冲溶液清洗;
(3)加入荧光分子Cy5溶液2μL(终浓度1.0×10-5mol/L),用pH=7.4的Tris-HCl缓冲溶液稀释至100μL,37℃10h后加入10μL可识别As3+的核酸生物分子溶液(终浓度1.0×10-7mol/L),37℃10h后磁分离,用pH=7.4的Tris-HCl缓冲溶液清洗,备用;
其中,所述的可识别As3+的核酸生物分子的碱基序列为5’-GGTAATACGACT CAC TATAGG GAG ATA CCA GCT TAT TCA ATT TTA CAG AAC AAC CAA CGT CGC TCC GGG TAC TCCTTC TTC ATC GAGATAGTAAGT GCAATC C-3’(P1),它和As3+的特异性结合产物能够被核酸外切酶剪切,释放出As3+;
(4)在(2)步产物中加入罗丹明B溶液2μL(终浓度1.0×10-5mol/L),用pH=7.4的Tris-HCl缓冲溶液稀释至100μL,37℃10h后加入10μL可识别Pb2+的核酸生物分子溶液(终浓度1.0×10-6mol/L),37℃10h后磁分离,用pH=7.4的Tris-HCl缓冲溶液清洗,备用;
其中,所述的可识别Pb2+的核酸生物分子的碱基序列为5’-GGT GGT GGT GGT TGTGGT GGT GGT GG-3’(P2),它和Pb2+的特异性结合产物能够被核酸外切酶剪切,释放出Pb2+;
(5)在(2)步产物中加入异硫氰酸荧光素FITC溶液2μL(终浓度1.0×10-4mol/L),用pH=7.4的Tris-HCl缓冲溶液稀释至100μL,37℃10h后加入10μL可识别Hg2+的核酸生物分子溶液(终浓度1.0×10-6mol/L),37℃10h后磁分离,用pH=7.4的Tris-HCl缓冲溶液清洗,备用;
其中,所述的可识别Hg2+的核酸生物分子的碱基序列为5’-TTT GTT TGT TGG CCCCCC TTC TTT CTTA-3’(P3),它和Hg2+的特异性结合产物能够被核酸外切酶剪切,释放出Hg2 +;
(6)将(3)步、(4)步和(5)步所得产物分别用pH=7.4的Tris-HCl缓冲溶液稀释后混合,磁分离,即可制得同时检测As3+、Pb2+和Hg2+的纳米胶囊-核酸生物分子复合物;
所述的具有中空、多孔结构的纳米金材料按文献方法获得(W.Wang,C.Chen,X.Li,S.Y.Wang and X.L.Luo.Chem.Commun.,2015,51,9109–9112.)。
实施例2:
一种本发明提出的可同时检测As3+、Pb2+和Hg2+的方法,包括以下步骤:
(1)将30μL含有As3+、Pb2+和Hg2+的溶液加入到本发明提出的纳米胶囊-核酸生物分子复合物中,加入2.25μL EXOⅢ剪切酶(45U),用pH=7.4的Tris-HCl缓冲溶液稀释至100μL,混合均匀后,放入摇床反应2h,反应温度37℃,由于靶标物As3+、Pb2+和Hg2+的存在,被组装到纳米胶囊表面的核酸生物分子分别对As3+、Pb2+和Hg2+发生特异性的响应,3种离子分别与其相应的核酸生物分子特异性结合,使其构象发生改变,从纳米胶囊表面脱落,使封堵的孔口被打开,从而导致荧光物质Cy5、RhB和FITC分别从纳米胶囊内部被释放出来;与此同时,由于核酸外切酶ExoⅢ的剪切作用,将生成的具有双链结构的特异性结合产物剪切后释放出As3+、Pb2+和Hg2+,而释放出来的As3+、Pb2+和Hg2+能够再次与纳米胶囊表面的核酸生物识别分子发生识别反应,然后再次被剪切……,循环往复,结果使As3+、Pb2+和Hg2+得到多次循环利用,更多的孔口被打开,释放出更多的荧光物质,实现荧光信号的显著增强;
(2)磁分离,取上清液稀释至2.0mL进行荧光检测,对释放出的3种荧光染料分别给予相应波长的激发,均得到显著增强的3种染料的荧光信号;当加入不同浓度的As3+、Pb2+和Hg2+时,荧光信号与离子浓度呈现良好的响应关系;因此,根据测得的荧光信号强度就可以实现对As3+、Pb2+和Hg2+的同时检测;检测条件:用于As3+、Pb2+和Hg2+检测的激发波长分别是EXWL:649nm/550nm/488nm;发射波长分别是EMWL:653nm/580nm/520nm。
实验结果表明,采用该体系同时检测As3+、Pb2+和Hg2+,获得的线性检测范围分别为:5.0×10-12~1.0×10-10mol/L、1.0×10-13~8.0×10-11mol/L和5.0×10-13~8.0×10- 11mol/L;与文献值相比,本发明对As3+、Pb2+和Hg2+的同时检测在确保高特异性的前提下,灵敏度得到显著提高,提高10倍以上,同时,还获得了对As3+、Pb2+和Hg2+更宽的线性检测范围。由于本发明采用的3种核酸生物识别分子分别对且仅对目标金属离子As3+、Pb2+或Hg2+具有特异性的响应,而对其它任何共存离子和干扰离子没有响应,不会受到其它常见干扰物质如Cd2+、Fe3+、Zn2+、Cu2+等金属离子的影响,相互之间也不存在干扰;具有高的特异性和选择性。因此,本发明提出的纳米胶囊-核酸生物分子复合物不但能够同时检测As3+、Pb2+和Hg2+,还可用于仅含As3+、Pb2+或Hg2+样品的单独检测,均能获得高灵敏、高选择性的检测结果。
本发明提出的纳米胶囊-核酸生物分子复合物具有结构简单、易合成、性能优异、稳定、适用范围广、高效、灵敏等优点;尤为重要的是,本发明克服了传统技术缺陷,无需再对核酸生物分子进行任何荧光标记,完全避免了对核酸生物分子性能的影响;本发明通过简单高效地多重放大技术,即在可控释放放大的基础上,通过生物酶的剪切作用及靶标物的循环利用,使荧光信号从1:1模式转变为指数型放大模式,灵敏度得到显著增强。
可以预见,本发明提出的纳米胶囊-核酸生物分子复合物及其制备方法和检测技术具有巨大的发展潜力和广阔的应用前景,将在重大疾病的早期诊断与治疗以及食品、生物医药、环境监测等领域发挥重要的作用。

Claims (5)
1.一种可同时检测As3+、Pb2+和Hg2+的方法,其特征在于包括以下步骤:
(1)将含有As3+、Pb2+和Hg2+的溶液加入到由3种核酸生物识别分子、3种荧光信号分子和具有中空、多孔结构的纳米金构建而成的纳米胶囊-核酸生物分子复合物中,用Tris-HCl缓冲溶液稀释,加入剪切酶EXOⅢ,放入37℃摇床反应1-3h;
(2)磁分离,取上清液进行荧光检测,分别给予相应波长的激发,检测与As3+、Pb2+和Hg2+相对应的荧光染料的荧光信号。

4.一种如权利要求1所述的可同时检测As3+、Pb2+和Hg2+的方法,其特征在于:所述的3种核酸生物识别分子是通过静电作用被分别组装到具有中空、多孔结构的纳米金表面。
5.一种如权利要求1所述的可同时检测As3+、Pb2+和Hg2+的方法,其特征在于:所述的纳米胶囊-核酸生物分子复合物的制备方法如下:
(1)分别筛选、设计并合成3种核酸生物识别分子,确保其分别对且仅对目标金属离子As3+、Pb2+和Hg2+具有特异性的响应,而对其它任何共存离子和干扰离子没有响应;
(2)将磁珠用pH=7.4的Tris-HCl缓冲溶液清洗后与具有中空、多孔结构的纳米金载体溶液混合,加入聚二烯丙基二甲基氯化铵溶液,10-12h后磁分离,用pH=7.4的Tris-HCl缓冲溶液清洗;
(3)加入荧光素Cy5溶液,10-12h后加入可识别As3+的核酸生物分子溶液,10-12h后磁分离,用pH=7.4的Tris-HCl缓冲溶液清洗,备用;
其中,所述的可识别As3+的核酸生物分子的碱基序列为5’-GGT AAT ACG ACT CAC TATAGG GAG ATA CCA GCT TAT TCA ATT TTA CAG AAC AAC CAA CGT CGC TCC GGG TAC TCCTTC TTC ATC GAG ATA GTA AGT GCA ATC C-3’;
(4)在(2)步产物中加入罗丹明B溶液,10-12h后加入可识别Pb2+的核酸生物分子溶液,10-12h后磁分离,用pH=7.4的Tris-HCl缓冲溶液清洗,备用;
其中,所述的可识别Pb2+的核酸生物分子的碱基序列为5’-GGT GGT GGT GGT TGT GGTGGT GGT GG-3’;
(5)在(2)步产物中加入异硫氰酸荧光素FITC溶液,10-12h后加入可识别Hg2+的核酸生物分子溶液,10-12h后磁分离,用pH=7.4的Tris-HCl缓冲溶液清洗,备用;
其中,所述的可识别Hg2+的核酸生物分子的碱基序列为5’-TTT GTT TGT TGG CCC CCCTTC TTT CTTA-3’;
(6)将(3)步、(4)步和(5)步所得产物分别用pH=7.4的Tris-HCl缓冲溶液稀释后混合,磁分离,即可制得同时检测As3+、Pb2+和Hg2+的纳米胶囊-核酸生物分子复合物。
