Description
Technical Field
The invention belongs to the technical field of toothpaste, and particularly relates to a refreshing and refreshing chewing gum composition, a chewing gum toothpaste and a preparation method thereof. The present invention has been developed primarily for use in oral care compositions. It will be appreciated, however, that the invention is not limited to this particular field of application.
Background
The oral environment is the most daily one in human life, and oral problems can affect the whole body health. With the continuous development of science and technology, the continuous improvement and perfection of technological equipment, various types of toothpastes come out in succession, and the quality and the grade of products are continuously improved. At present, the variety of the toothpaste is developed from single clean toothpaste into multifunctional toothpaste with various varieties, complete efficacies and hundreds of brands, and the requirements of consumption levels at different levels are met.
Halitosis is odor and peculiar smell emitted from oral cavity or other air-filled cavities such as nose, nasal sinuses and pharynx, which affects normal interaction and communication of people and affects social image and mental health of people. A recent survey shows that the incidence rate of halitosis in Chinese people reaches 27.5%, and the incidence causes of halitosis belong to 80-90% of patients with intra-oral factors. Oral halitosis is mainly caused by substances such as sulfur-containing amino acids, polypeptides and proteins, which are decomposed by bacteria in oral cavity, and volatile sulfides and fatty acids with peculiar smell are generated. Therefore, refreshing breath has become the most desirable oral experience for consumers.
For halitosis of oral origin, it can inhibit halitosis and refresh breath by eliminating pathogenic microorganisms and toxins produced thereby and rapidly and directly eliminating odor compounds in oral cavity.
The exhaled gas of a person contains a plurality of trace Volatile Sulfides (VSCs), and the amounts of hydrogen sulfide, methyl mercaptan and dimethyl sulfide in the VSCs are directly related to the intensity of halitosis. These VSCs are generally associated with human health and metabolic processes and are generally classified as oral and non-oral, with 90% of them originating from the oral cavity. Data related to prevalence of halitosis in some countries or regions of recent years. Research reports that when oral microecology is out of balance, the proportion of anaerobes producing halitosis in oral cavity is increased, and the ozone decomposes sulfur-containing protein to generate VSCs (including hydrogen sulfide, methyl mercaptan, dimethyl mercaptan and the like). For example, patent application No. CN931060702 discloses synergistic cooling effects of menthol, menthylamide, menthone glycerol ketal, etc. and its simple application. The invention patent with the publication number of CN 102512337B mentions that the use of polyvinylpyrrolidone, polypropylene copolymer and the like with chlorohexidine, dodecyl dimethyl benzyl ammonium chloride and cetyl pyridinium chloride prolongs the bacteriostasis time.
In the above-disclosed techniques, the use of bacteriostatic agents is not considered in terms of the microecological balance in the oral cavity, and in the use of cooling agents, it has been studied that the combined use of cooling agents can produce a synergistic effect to enhance the cooling effect, but is deficient in the duration of cooling and the degree of moistening of the oral cavity.

Effect example 2
The experiment of the inhibition ring of the chewing gum toothpaste prepared in application example 3 on Porphyromonas gingivalis
The experimental principle is as follows: according to the sterilization ring inhibition test of 2002 edition 'Disinfection technical Specification' 2.1.8.2 of Ministry of public health, the negative control product is the matrix toothpaste without the chewing gum component. And judging the bacteriostatic ability according to the size of the bacteriostatic ring.
Preparing a bacterial suspension: 3 turbidity bottles were prepared, and 4mL, 8mL, and 10mL of phosphate buffered saline PBS (0.03mol/L, pH7.2) were added for further use. Taking out the well-grown porphyromonas gingivalis plate from the culture bag, scraping half of the strain into a 4mL PBS turbidity tube, shaking uniformly, adding liquid paraffin for sealing, and taking the liquid as mother liquid. Using a pipette to transfer 100 mu L of mother liquor to 8mL of PBS for shaking up each time, adjusting the concentration of the bacterial liquid on a turbidimeter, firstly using a turbidity bottle containing 10mL of PBS for zero calibration, and then measuring the turbidity until the turbidity is 0.5MCF (1.5 multiplied by 10) and the concentration is lower than the original value8cfu/mL)。
Wherein the Porphyromonas gingivalis is Porphyromonas gingivalis ATCC33277, which is purchased from Shenzhen Asekeridae Biotech Ltd.
Bacterial liquid coating: marking on a blood agar plate, sucking 100 mu L of 0.5MCF bacterial suspension onto the blood agar plate by using a pipette, uniformly coating by using a sterile cotton swab, and drying at room temperature for 5min for later use.
Preparing a toothpaste flat plate to be tested: a small hole of about 6mm in diameter was dug out in a triangle on a blood agar plate, toothpaste was evenly squeezed into a sterile syringe, 50mg of toothpaste was injected into the small hole through a needle, and three different samples were injected on each plate. Three plates were repeated. Placing the flat plate in an anaerobic culture bag, placing an aerogenic bag and an oxygen indicator in a constant-temperature incubator at 37 ℃, measuring the diameter of an antibacterial ring by using a vernier caliper after 48-72 hours, wherein the larger the diameter of the antibacterial ring is, the stronger the antibacterial effect is, and the diameter of the antibacterial ring is larger than 7mm, so that the plate is judged to have the antibacterial effect; if the diameter of the antibacterial ring is less than or equal to 7mm, the product is judged to have no antibacterial effect.
Number 1 is a chewing gum toothpaste 3 made by the formula of the chewing gum of application example 3 of the present invention; number 2 is control toothpaste 1 (blank EGCg chewing gum composition, the rest proportion is the same as application example 3) prepared in application example 3, number 3 is control toothpaste 2 (blank biotoli chewing gum composition, the rest proportion is application example 3) prepared in the chewing gum formulation of application example 3 of the present invention; no. 4 is negative control toothpaste, no chewing gum composition is contained, and the rest proportion is the same as that of application example 3.
TABLE 4 antibacterial ring of chewing gum for Porphyromonas gingivalis
Group of | Parallel sample 1 | Parallel sample 2 | Parallel sample 3 | Average |
No. 1 chewing gum toothpaste 3 inhibiting bacteria ring mm | 25 | 26.5 | 26 | 25.8 |
Number 2 control toothpaste 1 antibacterial ring mm | 20 | 19.5 | 18 | 19.2 |
Number 3 control toothpaste 2 bacteriostatic ring mm | 15.6 | 16.1 | 15.3 | 15.7 |
Number 4 negative control toothpaste antibacterial ring mm | 4.2 | 3.5 | 4 | 3.9 |

FIC index in the experiment is 0.375, which shows that Biotolife and EGCg have synergistic effect.
Effect example 4
Test for efficacy of oral malodor reduction using the chewing gum prepared in example 3.
The test method comprises the following steps: 60 subjects suffering from oral halitosis who met the inclusion and non-exclusion criteria were selected and randomly divided into two groups, 30 persons each for the test group and the control group, with the test sample being a breath toothpaste 3 produced according to the invention using the breath toothpaste formulation of example 3, and the negative control toothpaste being a toothpaste without a breath composition.
During the test, the corresponding toothpaste is squeezed on a unified standard toothbrush for 2cm respectively, and the tooth brushing time is guaranteed for 3 min. The subject tested the concentration of VSCs in the oral cavity on the 1 st time prior to brushing the teeth in mouth; testing the concentration of VSCs in the oral cavity at time 2 min post brushing; testing the concentration of VSCs in the oral cavity at 60min post-brushing 3 rd time; testing the concentration of VSCs in the oral cavity at the 4 th time 120min after brushing; the following day the concentration of VSCs in the oral cavity was tested at 5 th 60min post brushing. The concentration of VSCs in the oral cavity was tested 60min 6 th time after brushing on day three; the concentration of VSCs in the oral cavity was tested 60min after brushing on day four, at time 7. Fifth day VSCs concentration in the oral cavity was tested 60min post brushing at 8 th time.
The testing instrument uses a Halimeter breath measuring instrument. The method adopts an electrochemical principle to measure the concentration of Volatile Sulfides (VSCs) in oral gas as an index for measuring the oral gas level of a patient. Before use, the zero setting is carried out by using ambient air, during measurement, a subject breathes with the nose, slightly opens the mouth, places the gas collecting pipe mouth at the position 0.5cm above the junction of the middle and the rear third of the tongue, and reads the numerical value on the display screen. Each examination was repeated 3 times and the results averaged and the results are shown in table 7.
TABLE 7 results of the measurement of halitosis-alleviating effect of the toothpaste
Group of | Test group | Control group |
Breath value ppb before tooth brushing | 502 | 512 |
Breath value ppb at 10 minutes after tooth brushing | 383 | 467 |
Breath value ppb at 60 minutes after tooth brushing | 402 | 471 |
Breath value ppb at 120 minutes after tooth brushing | 454 | 513 |
The second day of breath value ppb | 380 | 460 |
Third day breath value ppb | 370 | 450 |
Fourth day breath value ppb | 356 | 440 |
Fifth day breath value ppb | 350 | 454 |
Results and discussion: as can be seen from Table 7, after the toothpaste is used, the breath value of each group is reduced to a certain extent, which indicates that the breath can be kept fresh to a certain extent after the toothpaste is used for brushing teeth, but the breath value of the test sample toothpaste is reduced to a large extent, the mouth can still keep fresh after 2h, the breath value of the common toothpaste is reduced to a small extent, and the breath value of the mouth is basically restored to the original state after 2 h. The breath value of the tested toothpaste is obviously reduced after five consecutive days, while the common toothpaste is basically kept at the same level.
The cool and refreshing chewing gum provided by the invention can effectively remove the peculiar smell in the oral cavity and refresh the breath after being used by a testee. Remarkably reduces halitosis, and maintains the clinical curative effect of the chewing gum. Repeated experiments show that the oral cavity odor-eliminating ointment has obvious effects on gingival inflammation, red swelling and bleeding and oral cavity odor.
