本发明涉及用于薄层色谱(“TLC”)的编码载体及其制备方法。
人们对现代分析方法的要求日益提高,例如在准确度、重复性、明确的归类以及分析结果的汇编方面。对于汇编的日增的要求通过诸如DIN—ISO9000、优异操作规程(GMP)或优异实验规程(GLP)得到满足。
相应地在TLC中需要对已制备好的薄层在使用其进行色谱分析时或分析后进行单个地标记或编码,以避免相互搞混。
这样的标记应有尽可能高的信息密度且应能很快施用在各薄层上或薄层中。尤其是它必须耐所有在TLC中常用的有机或无机溶剂、碱和酸(不仅以液体形式,而且还有通过蒸发的气相作用),并且不因此影响和干扰色谱显影及分离的结果。此外,标记不仅在日光下而且在UV—照射(如254和366nm)下是可识别的。
目前一般通过粘附标记或在载体层上刻划进行加标记的,这些方法只能部分满足要求或完全不能满足要求,此外非常繁琐。由于手工转印数据产生的转印错误使得这些转印数据的薄层不能使用。标记会脱落和丢失。
本发明的目的在于,制备标记过(编码的)TLC—载体,它不具备上述缺点或只带有很小程度的缺点。
令人惊奇地发现,所追求的效果是通过用激光照射载体来达到的,其中使用相应的模版(如数字和/或字母或用于条线代码的条线)或通过计算机控制激光。这样就产生永久性的标记,该标记在日光或UV—光下很好辩识。在借助计算机控制激光时则不需模版。
相应地,本发明的对象是用于TLC的、借助激光刻上编码特征的载体以及制备该载体的方法,其特征在于,用激光对通常用的TLC载体进行照射。
作为载体主是要玻璃板和薄片(如铝薄片或塑料薄片如聚对苯二甲酸酯(Polyterephthalat)),它涂有吸着剂且含有粘合剂以及必要时含荧光指示剂和/或其它添加物。
用作吸着剂的是所有常用于TLC的固定相,优选改性的或非改性的硅胶,此外例如纤维素、聚酰胺、硅藻土、氧化铝。
作为粘合剂优选使用聚合有机物,如聚丙烯酸或聚甲基丙烯酸及其酰胺。
作为激光源优选使用脉冲的或连续的CO2—激光(波长1.06μm)或Yag—激光(固体激光,波长10.6μm)。典型的照射时间为5—50毫微秒/信号。
所得到的编码可含任意的数据,例如制造公司的名字、文件号、批号、制造日期、单个独特的编号(连续编号)。
编码优选通过对含粘合剂的吸着剂层进行照射来进行。然而,也可能在载体玻璃板或已涂敷的塑料薄板的未涂敷的背面进行激光刻划。在用玻璃板的情况下,在照射过程中产生细的热应力裂缝(thermische Spannungsrisse),它在玻璃表面产生在可见光波长范围内可辩认的浑浊。
即使没有进一步的实施,本专业技术人员也可在最大的范围内利用上述说明。因此,优选实施例仅作为一种描述性公开,而不对本发明作出限制。

用脉冲CO2激光照射一已制备好的HPTLC色谱板(E.MerckKieselgel 60 F254,Art.—Nr.5642,100×200mm),其中波长为10.6μm,对于每个单个的信号照射时间分别为50毫微秒。在此使用一掩模,它覆盖了HPTLC板的大部分,除了一些用于所需信号的开孔外,移动板以使每次照射分别照射到需要信号的地方。实施例2
