Abstract

本发明涉及一种牙菌斑的生物溶菌酶组合物。所述组合物含有1‑2.5重量份的丁香茶提取物、1‑2重量份的摩擦剂、40‑60重量份的润湿剂、0.5‑2重量份的粘合剂、1‑5重量份发泡剂。所述的组合物对口腔致病菌,尤其是对链球菌具有抑菌效果,抑菌率高,能够高效短时的抑菌口腔致病菌的生长,具有独特的优势和广阔的应用前景。

技术领域
本发明涉及生活日用品领域,具体而言,本发明涉及一种牙菌斑的生物溶菌酶组合物及其制备方法和用途。
背景技术
丁香属落叶灌木或小乔木。小枝粗壮无毛,叶广卵形,通常宽大于长,宽 5~12cm,端尖锐,基部长型至截形,全缘。有细皱纹。质脆易折断,断面显油性。丁香可入药,木犀科品种,主要分布在中国东北长白县地区,另外一种是中药丁香,全称为桃金娘科母树紫丁香茶,花蕾为中药,叶子作为饮品,中国主要产地为长白山,国外主要产自坦桑尼亚,印尼,马来西亚等地。《汉林本草》,药性论;①《雷公炮炙论》:凡使(丁香茶),有雌雄,雄颗小,雌颗大,似枣核。方中多使雌,力大,膏煎中用雄。"②《汉林本草》:"丁香,二月、八月采。按石岩村送丁香图,树高丈余,叶似栎叶,花圆细,黄色,凌冬不雕。医家所用惟用根。子如钉子,长三、四分,紫色,中有粗大如山茱萸者,俗呼为母丁香,可入心腹之药尔。
丁香茶主要成分为桃金娘科植物常绿乔木丁香树的干燥花叶。花叶含丁香油,其中主要成分为丁香油酚,又含鞣质、齐墩果酸等。中药丁香原产印度尼西亚、马来西亚及东非沿海国家,我国主要产地为长白山。花大油足,花叶以嫩绿者为佳。主要用于泡茶或者入药,其药理作用为养胃抗菌、健胃驱风、温中壮阳、下气降逆。丁香茶具有健胃、抗腹泻、抗胃溃疡的功效。
牙菌斑是指主要黏附在牙齿表面、不能用水冲洗去除的一种生物
膜,由细菌和基质组成,牙菌斑是龋病和牙周病的致病因素。唾液中的营养物质吸附在牙齿表面,构成“社区”肥沃的“土壤”,即获得性薄膜形成。这个过程在刚清洁过的牙面上,数分钟内便可形成,1-2小时迅速增厚。“土壤”形成之后,便可吸引细菌来定居,同时为细菌提供营养,即细菌粘附和共聚。首先会有先驱菌来定居,开垦土壤,建立社区的基本设施,之后便会吸引更多的其他细菌来定居。如果没有人为的破坏,很快会发展壮大,成为一个“成熟的社区”,即菌斑成熟。众多细菌集结在一起,互相提供营养物质,同时汲取唾液中的养分,细菌大量增殖,“社区”结构也更加紧密,可以共同抵抗外界的干扰,这是用漱口的方法是无法清除的。一般9天就可以发展为拥有各种细菌的复杂、成熟“社区”。而导致菌斑微生物的是厌氧菌,主要是链球菌,如血链球菌、轻链球菌、变链球菌、远缘链球菌。
可预期的是丁香茶提取物应用于组合物中,对口腔中的牙菌斑具有一定的抑制作用。

本申请提供了一种牙菌斑的生物溶菌酶组合物及其制备方法。

为了达到上述目的,本申请使用了下述技术方案:
一种牙菌斑的生物溶菌酶组合物,其包含:1-2.5重量份的丁香茶提取物、1-2重量份的摩擦剂、40-60重量份的润湿剂、0.5-2重量份的粘合剂、1-5重量份发泡剂和0.01-0.05重量份的生物溶菌酶。
所述丁香茶提取物为下式所示:

所述摩擦剂选自碳酸钙、磷酸氢钙、水合硅石和二氧化硅中一种或几种;所述润湿剂选自山梨醇、甘油、丙二醇和聚乙二醇中的一种或几种;所述粘合剂选自羧甲基纤维素钠、黄原胶、羟乙基纤维素、瓜尔胶、卡拉胶中的一种或几种;所述发泡剂选自十二烷基硫酸钠或月桂醇硫酸酯钠。
优选地,1重量份的丁香茶提取物、2重量份的碳酸钙、60重量份的聚乙二醇、1重量份的羧甲基纤维素钠、2重量份的十二烷基硫酸钠和0.05重量份的生物溶菌酶。
所述丁香茶提取物通过下述方法制备得到:
(1) 将丁香茶叶洗净、晾干、或者粉碎获得茶叶提取原料。
(2) 将茶叶与水溶液按质量比至少为1:5混合,恒温70~90oC提取至少30 min,提取次数至少为2次;
(3) 提取完成后,将步骤(2)中的药材提取液冷却至室温,过滤,滤液浓缩;
(4) 将步骤(3)中的滤液浓缩至约50oC时的相对比重为1.10~1.20时,使用V乙酸乙酯:V石油醚=1:2重结晶。
本发明还提供了一种丁香茶提取物,其以下式表示:

所述的丁香茶提取物的制备方法,包括下述步骤:
(1) 将丁香茶叶洗净、晾干、或者粉碎获得茶叶提取原料。
(2) 将茶叶与水溶液按质量比至少为1:5混合,恒温70~90oC提取至少30 min,提取次数至少为2次;
(3) 提取完成后,将步骤(2)中的药材提取液冷却至室温,过滤,滤液浓缩;
(4) 将步骤(3)中的滤液浓缩至约50oC时的相对比重为1.10~1.20时,使用V乙酸乙酯:V石油醚=1:2重结晶。
所述的丁香茶提取物可用于组合物制备。
实施例
下面通过实施例对本发明作进一步说明。应该理解的是,本发明实施例所述方法仅仅是用于说明本发明,而不是对本发明的限制,在本发明的构思前提下对本发明制备方法的简单改进都属于本发明要求保护的范围。实施例中用到的所有原料和溶剂均为市售产品。
实施例1:丁香茶提取物的制备
(1) 将丁香茶叶2000克洗净、晾干、或者粉碎获得茶叶提取原料。
(2) 将茶叶与水溶液按质量比1:5混合,恒温8090oC提取至少2h,提取3次;
(3) 提取完成后,将步骤(2)中的药材提取液冷却至室温,过滤,滤液浓缩;
(4) 将步骤(3)中的滤液浓缩至约50oC时的相对比重约为1.15时,使用V乙酸乙酯:V石油醚=1:2重结晶,得到4.5克淡黄色固体粉末。

实施例2:本申请牙菌斑的生物溶菌酶组合物的制备
称取1克的丁香茶提取物、2克的碳酸钙、60克的聚乙二醇、1克的羧甲基纤维素钠、2克的十二烷基硫酸钠和0.05克的生物溶菌酶。
先将上述组分混合,再加入一定的水混合得到膏体。
生物活性测试实施例(抑菌环抗菌试验法):
以血链球菌、轻链球菌、变链球菌、远缘链球菌受试菌种(中国科学院微生物研究所),通过抑菌环抗菌试验法考察了上述丁香茶提取物抗菌活性的影响。抑菌环法的具体操作如下:在无菌环境中,将灭菌后的熔化营养琼脂倒入灭菌的平皿中(每个平皿中 20ml),待其充分冷却固化后,吸取 0.1ml 新鲜受试菌悬液(约为106 cfu/ml)置于平皿中,用灭菌后的棉签将其涂抹均匀,然后把待测液(约 20ul)滴加在灭菌干燥的滤纸片上(直径约5mm),等滤纸片充分吸收样品后,用无菌镊子将其轻轻放在涂有受试菌的培养基表面,加盖后倒置在37℃恒温箱中培育约18h,观察抑菌环的有无并测量记录其大小,判断受试菌对待测样品的敏感性能。用抑菌圈法判断药物抑菌性能的标准如下:直径(mm)大于15,高敏;10-15,中敏;7-10,低敏;小于 7,不敏感。

从上表可以看出,四批样品均为深红色澄清的液体;测试样品对链球菌的抑菌圈宽度,加入植物源化合物,可达18mm以上。

1.一种牙菌斑的生物溶菌酶组合物,其特征在于包含:1-2.5重量份的丁香茶提取物、1-2重量份的摩擦剂、40-60重量份的润湿剂、0.5-2重量份的粘合剂、1-5重量份发泡剂和0.01-0.05重量份的生物溶菌酶。
2.根据权利要求1所述的牙菌斑的生物溶菌酶组合物,其特征在于所述丁香茶提取物为下式所示:

3.根据权利要求1所述的牙菌斑的生物溶菌酶组合物,其特征在于:所述摩擦剂选自碳酸钙、磷酸氢钙、水合硅石和二氧化硅中一种或几种;所述润湿剂选自山梨醇、甘油、丙二醇和聚乙二醇中的一种或几种;所述粘合剂选自羧甲基纤维素钠、黄原胶、羟乙基纤维素、瓜尔胶、卡拉胶中的一种或几种;所述发泡剂选自十二烷基硫酸钠或月桂醇硫酸酯钠。
4.根据权利要求1所述的牙菌斑的生物溶菌酶组合物,其特征在于包含:1重量份的丁香茶提取物、2重量份的碳酸钙、60重量份的聚乙二醇、1重量份的羧甲基纤维素钠、2重量份的十二烷基硫酸钠和0.05重量份的生物溶菌酶。
5.根据根据权利要求2所述的牙菌斑的生物溶菌酶组合物,其特征在于所述丁香茶提取物通过下述方法制备得到:
(1) 将丁香茶叶洗净、晾干、或者粉碎获得茶叶提取原料;
(2) 将茶叶与水溶液按质量比至少为1:5混合,恒温70~90oC提取至少30 min,提取次数至少为2次;
(3) 提取完成后,将步骤(2)中的药材提取液冷却至室温,过滤,滤液浓缩;
(4) 将步骤(3)中的滤液浓缩至约50oC时的相对比重为1.10~1.20时,使用V乙酸乙酯:V石油醚=1:2重结晶。
6.一种丁香茶提取物,其特征在于以下式表示:
