Abstract
本发明的富含高稳定性叶酸和葡萄糖酸亚铁的孕妇营养包,属于孕妇营养补充食品的技术领域,特别涉及一种全程氮气介导下的富含叶酸微胶囊冻干粉和葡萄糖酸亚铁微胶囊冻干粉的孕妇营养包与其规模化生产关键技术,在高通量氮气与低温压缩空气双效介导下孕妇营养包的制备技术方面创新性突出。本发明高品质的补叶酸‑补铁产品经加速试验90天确证产品中叶酸和葡萄糖酸亚铁的平均衰减率均低于2.0%,有效避免生产、贮藏、销售环境中因光照、湿、热等环境因素所引起的叶酸分解和葡萄糖酸亚铁变色变味等品质劣变问题,而且降低孕妇营养包脂肪氧化哈败味产生速率和蛋白质劣变速率,为后续开展高品质孕妇营养包产品系列化加工提供技术基础。

技术领域
本发明属于孕妇营养补充食品创制的技术领域,涉及一种全程氮气介导下以叶酸微胶囊冻干粉和葡萄糖酸亚铁微胶囊冻干粉为强化因子的高品质孕妇营养包生产关键技术。
背景技术
(一)孕妇与叶酸强化
叶酸(Folic acid),又称碟酰谷氨酸,是一种维生素为维持细胞快速分裂生长所不可缺少的营养素,在人体中不能被合成,必须通过日常饮食伴随摄入,故世界卫生组织WHO建议孕妇每日叶酸(游离态)摄入量为400μg。
倘若孕妇叶酸缺乏,则会引起血浆中高半胱氨酸的浓度会提高,且DNA无法合成,导致细胞无法正常分裂,对胚胎个体发育构成严重危害,甚至会造成新生儿神经管畸形(NTD)等疾病的发生。孕期妇女需要科学摄入叶酸,以预防胎儿畸形如兔唇等的发病率,降低新生儿神经管缺陷发病率。基于此,孕妇营养补充食品叶酸强化问题倍受关注。
目前,国内外叶酸补充剂多以叶酸片、叶酸胶囊补充剂等药品形态存在。基于叶酸在生产、贮藏与流通环节中会因光照、受热和食物体系pH值等因素致使其被不同程度降解呈现出叶酸衰减等瓶颈问题。故高品质的孕妇强化叶酸产品市场需求越来越大,且以食品载体而非药品形态的高稳定性叶酸食品的市场需求就更加广阔。
通过查阅文献发现:于文龙等人2019年12月发表的《B族维生素微胶囊的制备及其缓释动力学研究》论文中,以维生素B1、维生素B2、烟酰胺、叶酸为芯材,以明胶和羧甲基纤维素钠(CMC)为壁材,采用微胶囊包埋与真空冷冻干燥技术制备混合B族维生素微胶囊,微胶囊包埋率为88.81%。刘欢于2015年发表的《食用叶酸的微胶囊化及高叶酸调和油的研制》中以乳清蛋白与麦芽糊精组合制得叶酸微胶囊乳化液,包埋率为44.71±0.66%;付晓俊于2018年发表的《乳清分离蛋白作为载体对多种活性成分的保护及其相互作用的研究》中研究了乳清分离蛋白(WPI)对叶酸光稳定性的影响,确证了混合蛋白WPI和其主要组分β-乳球蛋白,α-乳白蛋白均能延缓叶酸的光降解。上述三篇研究论文中并未阐明叶酸包埋效果与其控制叶酸衰减情况,而本申请专利是以孕妇营养补充食品为载体,以叶酸衰减率为重点约束指标,以鱼皮明胶和魔芋多糖为壁材的高稳定性叶酸微胶囊化技术手段研究与产品创制具有原始创造性和技术先进性。
(二)孕妇补铁与葡萄糖酸亚铁
铁是人体必需的微量元素之一,参与人体内血红蛋白和肌红蛋白的合成。世界范围内缺铁问题愈发严重,铁缺乏人口数量占总人口的四分之一左右,且以婴幼儿及女性占比较大。目前,许多国际组织和国家已经批准葡萄糖酸亚铁作为铁营养强化剂,并在营养素补充剂、特殊营养用途食品、食物强化中广泛应用,其优势主要体现在以下3个方面:
1.葡萄糖酸亚铁属于有机酸葡萄糖酸的亚铁盐类,具有易于吸收、生物利用率高,比其他铁补充剂对肠胃的刺激性小、补铁效果更好等优势。
2.葡萄糖酸亚铁一旦进入血液后,其二价铁离子立刻被氧化为三价铁离子,与血浆中转铁蛋白结合成血浆铁;在缺铁时,血浆铁转运率增加,吸收率也加速,使铁吸收量增加。葡萄糖酸亚铁是以转铁蛋白为载体转运到肝、脾、骨等组织,以铁蛋白形式贮存;在骨中,铁蛋白的铁结合为血红蛋白;在骨骼肌中,则成为肌红蛋白。
3.葡萄糖酸亚铁是水溶性亚铁,相比于其他水溶性铁补充剂伴随的铁味,葡萄糖酸亚铁口感更好,有焦糖气味、无涩味、安全性好,特别适合于强化乳饮料。
但是,葡萄糖酸亚铁的稳定性尚需被关注,需要避光、密闭或于相对湿度70%以下贮存;湿、热会引起其Fe供含量变化及变色;与其他补铁制剂副作用相近会有胃肠道不良反应,且在食品体系中容易引起食品色泽和风味变化,一定程度上限制了其应用。经文献查阅发现:王琳琳在2016年在学位论文《葡萄糖酸亚铁固体脂质纳米粒的制备及肌原纤维蛋白酶解多肽对其性质的影响》研究中采用多重乳状液的方法制备葡萄糖酸亚铁固体脂质纳米粒,当盐酸氨基葡萄糖的添加量与葡萄糖酸亚铁质量比为1:2时,葡萄糖酸亚铁最大包埋率为61.30%。但这篇文献报道存在葡萄糖酸亚铁为水溶性因子置于固体脂质纳米粒的保护体系,包埋率仅为61.30%,且不利于以水为冲调载体的营养粉包载体的补铁产品创制领域。但是,针对于葡萄糖酸亚铁光照不稳定性、摄入胃肠道内副作用、强化食品的感官品质与风味等技术瓶颈问题,目前高品质的补铁强化食品满足婴幼儿、孕产妇等特殊群体有机补铁制剂与营养补充品尚未被深入研究与开发。
综上,基于高稳定性的叶酸和葡萄糖酸亚铁强化产品的国内外市场需求量与日俱增,研究意义重大,经济效益和社会效益巨大。

申请发明的技术方案是:

3)所述的高纯度氮气与低温压缩空气双效介导下孕妇营养包的制备,是将富含高稳定性叶酸和葡萄糖酸亚铁的孕妇营养粉剂,经过氮气介导下的定量灌装与热封合处理、低温压缩空气介导下分割处理,制备出富含高稳定性叶酸和葡萄糖酸亚铁的孕妇营养包产品;经贮藏温度为37±2℃、相对湿度为75±5%,避免光线直射的条件下加速试验90天,确证产品中叶酸和葡萄糖酸亚铁的平均衰减率均低于2.0%;
所述的氮气介导下的定量灌装与热封合处理,是在氮气纯度为99.00~99.99%、氮气通气量为500~1500L/h,氮气通气压力为0.5~0.9MPa的氮气介导下进行全自动条形袋包或者枕包的定量灌装25±0.3g/袋和热熔封合,定量灌装精准度为99.9%以上,热融合温度为125~225℃;
所述低温压缩空气介导下分割处理,是将每袋装载量为25±0.3g且氮气残留量≥99.00%的条形包或枕包再迅速置于压缩空气洁净度为99.00~99.99%、通气量为100~500L/h、通气压力为0.5~0.9Mpa、通气温度为15~25℃环境下进行切割为独立包装,制得富含高稳定性叶酸和葡萄糖酸亚铁的孕妇营养包,产品的优品率≥99.99%,且氮气残留量≥99.00%,残氧量≤0.25%;

1.一种富含高稳定性叶酸和葡萄糖酸亚铁的孕妇营养包的制备方法,其特征在于,以大豆分离蛋白粉、乳清蛋白粉、葡萄糖全糖粉、叶酸微胶囊冻干粉、葡萄糖酸亚铁微胶囊冻干粉、醋酸维生素A、胆钙化醇、dl-α-醋酸生育酚、盐酸硫胺素、核黄素、盐酸吡哆醇、氰钴胺、烟酰胺、D-泛酸钙、L-乳酸钙、柠檬酸锌、硒蛋白、葡萄糖酸镁、麦芽糊精为原料,经过原料质量检验、高纯度氮气全程介导下孕妇营养粉剂的制备、高纯度氮气与低温压缩空气双效介导下孕妇营养包的制备,制得富含高稳定性叶酸和葡萄糖酸亚铁的孕妇营养包产品,优品率≥99.99%,氮气残留量≥99.00%,残氧量≤0.25%;经贮藏温度为37±2℃、相对湿度为75±5%、避免光线直射的条件下加速试验90天确证产品中叶酸和葡萄糖酸亚铁的平均衰减率均低于2.0%;
1)所述的原料质量检验,是指全部原料按照GB 31601-2015《食品安全国家标准孕妇及乳母营养补充食品》中要求进行质量检验合格备用;其中大豆分离蛋白粉质量要求蛋白含量为90%以上,无胰蛋白酶抑制因子,无凝血酶抑制因子;
2)所述的高纯度氮气全程介导下孕妇营养粉剂的制备,是将质量检验合格的全部原料,经氮气介导下过筛与配料处理、氮气介导下气力混合处理,制得富含高稳定性叶酸和葡萄糖酸亚铁的孕妇营养粉剂;
所述的氮气介导下过筛与配料处理,是将全部原料依次在氮气纯度为99.00~99.99%、通气量为200~500L/h的氮气介导下经过振动筛过筛处理,振动筛筛网目数控制在60~120目;再依次称量蛋白含量为90%以上的无胰蛋白酶抑制因子和无凝血酶抑制因子的大豆分离蛋白粉24.000~45.333kg、乳清蛋白粉1.579~26.842kg、葡萄糖全糖粉6.000~16.000kg、叶酸含量为0.00108~0.00192kg的微胶囊冻干粉、葡萄糖酸亚铁含量为0.382~0.612kg的微胶囊冻干粉、醋酸维生素A 0.0197~0.0344kg、胆钙化醇0.0092~0.0192kg、dl-α-醋酸生育酚0.0269~0.0672kg、盐酸硫胺素0.0043~0.0151kg、核黄素0.0384~0.1344kg、盐酸吡哆醇0.0070~0.0198kg、氰钴胺0.0077~0.0230kg、烟酰胺0.0390~0.0877kg、D-泛酸钙0.0213~0.0639kg、L-乳酸钙10.475~20.949kg、柠檬酸锌0.219~0.422kg、硒蛋白0.0672~0.132kg、葡萄糖酸镁12.448~27.026kg,剩余为麦芽糊精合计120kg;
所述的氮气介导下气力混合,是将称重配好的全部原料进行氮气介导下的气力混合处理,氮气纯度为99.00~99.99%、氮气通气压力为0.3~0.6MPa、氮气通气量为200~1500L/h,气力混合温度为20~25℃,气力混合时间为200~300秒,混吹时间为1~6秒,混吹间隔为1~3秒,制得富含高稳定性叶酸和葡萄糖酸亚铁的孕妇营养粉剂;
3)所述的高纯度氮气与低温压缩空气双效介导下孕妇营养包的制备,是将富含高稳定性叶酸和葡萄糖酸亚铁的孕妇营养粉剂,经过氮气介导下的定量灌装与热封合处理、低温压缩空气介导下分割处理,制备出富含高稳定性叶酸和葡萄糖酸亚铁的孕妇营养包产品;经贮藏温度为37±2℃、相对湿度为75±5%,避免光线直射的条件下加速试验90天,确证产品中叶酸和葡萄糖酸亚铁的平均衰减率均低于2.0%;
所述的氮气介导下的定量灌装与热封合处理,是在氮气纯度为99.00~99.99%、氮气通气量为500~1500L/h,氮气通气压力为0.5~0.9MPa的氮气介导下进行全自动条形袋包或者枕包的定量灌装25±0.3g/袋和热熔封合,定量灌装精准度为99.9%以上,热融合温度为125~225℃;
所述低温压缩空气介导下分割处理,是将每袋装载量为25±0.3g且氮气残留量≥99.00%的条形包或枕包再迅速置于压缩空气洁净度为99.00~99.99%、通气量为100~500L/h、通气压力为0.5~0.9Mpa、通气温度为15~25℃环境下进行切割为独立包装,制得富含高稳定性叶酸和葡萄糖酸亚铁的孕妇营养包,产品的优品率≥99.99%,且氮气残留量≥99.00%,残氧量≤0.25%。
2.根据权利要求1所述的一种富含高稳定性叶酸和葡萄糖酸亚铁的孕妇营养包的制备方法,其特征在于,所述的叶酸微胶囊冻干粉,是以叶酸为芯材,按照壁材种类与其质量比为鱼皮明胶:魔芋多糖=1~2:1,壁材质量浓度为3~6%,壁芯质量比为1.5~2.5:1进行叶酸微胶囊化,经真空冷冻干燥制得叶酸微胶囊冻干粉,叶酸包埋率可达90%以上。
3.根据权利要求1所述的一种富含高稳定性叶酸和葡萄糖酸亚铁的孕妇营养包的制备方法,其特征在于,所述的葡萄糖酸亚铁微胶囊冻干粉,是以葡萄糖酸亚铁为芯材,按照壁材种类与其质量比为鱼皮明胶:魔芋多糖=1.2~1.6:1,壁材质量浓度为2~4%,壁芯质量比为1.2~2.0:1进行葡萄糖酸亚铁微胶囊化,经真空冷冻干燥制得葡萄糖酸亚铁微胶囊冻干粉,葡萄糖酸亚铁包埋率可达85%以上。
4.根据权利要求1所述的一种富含高稳定性叶酸和葡萄糖酸亚铁的孕妇营养包的制备方法,其特征在于,包括以纯度为99.00~99.99%和通气量为200~500L/h的氮气介导下进行过筛与配料、氮气介导下气力混合两个处理步骤。
5.根据权利要求1所述的一种富含高稳定性叶酸和葡萄糖酸亚铁的孕妇营养包的制备方法,其特征在于,所述的氮气介导下气力混合处理方法,气力混合处理过程在氮气纯度为99.00~99.99%、氮气通气压力为0.3~0.6MPa、氮气通气量为200~1500L/h的氮气介导下进行,且气力混合温度为20~25℃,气力混合时间为200~300秒,混吹时间为1秒~6秒,混吹间隔为1秒~3秒。
6.根据权利要求1所述的一种富含高稳定性叶酸和葡萄糖酸亚铁的孕妇营养包的制备方法,其特征在于,所述的氮气介导下的定量灌装与热封合处理方法,定量灌装与热封合处理均要在氮气纯度为99.00~99.99%、氮气通气量为500~1500L/h,氮气通气压力为0.5~0.9MPa的氮气介导下进行。
7.根据权利要求1所述的一种富含高稳定性叶酸和葡萄糖酸亚铁的孕妇营养包的制备方法,其特征在于,所述的低温压缩空气介导下分割处理方法,压缩空气洁净度为99.00~99.99%、通气量为100~500L/h、通气压力为0.5~0.9Mpa、通气温度为15~25℃环境下进行切割为独立包装。
8.一种富含高稳定性叶酸和葡萄糖酸亚铁的孕妇营养包,其特征在于,其使用如权利要求1-7任意一项所述的富含高稳定性叶酸和葡萄糖酸亚铁的孕妇营养包的制备方法,营养包中叶酸和葡萄糖酸亚铁稳定性高,经加速实验90天证明营养包中的叶酸和葡萄糖酸亚铁平均衰减率均低于2.0%。
