The composition with anti-wrinkle effect has effects on secretion of matrix metalloproteinase (MMP-9, MDA, and GSH-Px) from HDF-alpha cell and HaCaT cell, and on secretion of glutathione peroxidase (GSH-Px) from cells in exponential growth phase of 5 × 104Each/mL suspension was inoculated into a 96-well plate, and 200. mu.L of complete medium was added to each well and cultured in a 5% CO2, 37 ℃ incubator. After the cells are subjected to drug administration and culture for 24h, supernatant is taken for experiment according to the instruction of the kit, and the supernatant is sequentially reacted with a biotin-labeled antibody, HRP-labeled streptavidin, a chromogenic substrate and stop solution. The blank wells were zeroed and the OD of each well was measured at a wavelength of 450nm within 15min after termination. In a HaCaT cell aging model, when the concentration of the medicine reaches 50 mu g/mL, the content of MMP-9 is 5.46 +/-0.21 ng/mL, the content of MDA is 1.27 +/-0.04 nmol/mL, and the content of GSH-Px is expressed as 41.62 +/-0.40 ng/mL (p is less than 0.01); in the HDF-alpha cell aging model, when the drug concentration reaches 50 mu g/mL, the MMP-9 content is 4.91 +/-0.13 ng/mL, the MDA content is 1.40 +/-0.01 nmol/mL, and the GSH-Px content is expressed as 44.28 +/-0.05 ng/mL (p is less than 0.01). In view of the above, it is desirable to provide,when the concentration of the composition reaches 50 mug/mL, the secretion levels of MMP-9 and MDA after radiation damage can be obviously reduced, the secretion level of GSH-Px is improved, the composition has a good protection effect on light damage of HaCaT cells and HDF-alpha cells, and can be applied to anti-wrinkle cosmetics.
In the process of skin aging, MMP-9, MDA and GSH-Px are important indexes of oxidative damage reaction.
MMP-9 mainly plays a role in degrading the content of collagen and elastin in the skin, and experiments show that the increase of MMP-9 secretion can cause the loss of collagen in the skin, the skin relaxation, the reduction of elasticity and the increase and the deepening of fine wrinkles, so that the skin shows the signs of aging. MDA is one of final products of membrane lipid peroxidation, and the high content and the low content of MDA can be used as one of indexes for examining the severity degree of the stress on cells. MDA can cause macromolecular cross-linking reactions in the tissue, damaging the dermal tissue. GSH-Px is an important enzyme for eliminating hydrogen peroxide and a plurality of organic hydroperoxides in vivo, and toxic hydrogen peroxide and lipid peroxides are reduced into nontoxic hydroxyl compounds by catalyzing glutathione, so that the structure and the function of a cell membrane are protected from being damaged and interfered by peroxides, and the GSH-Px is also one of the indexes of the anti-peroxidation capability of an organism.
The following is a content of the effect verification test example section.
1. Stability test
Test samples: examples 1 to 4 anti-wrinkle gel mask and anti-wrinkle cream
The test method comprises the following steps: centrifuging at 3000r/min for 30min, standing at room temperature, 45 deg.C and-20 deg.C for 24h, and observing whether there is oil-water separation phenomenon in appearance change.
The stability investigation result shows that no oil-water separation phenomenon exists and the appearance is unchanged.
2. Human body trial test
Test samples: examples 4-9 anti-wrinkle gel mask and anti-wrinkle cream
The test method comprises the following steps: folding several layers of gauze into a certain size (5cm × 5cm), sticking the tested object on the gauze, sticking it on the skin, covering a layer of oil paper or plastic film, and fixing with bandage. The test substance was removed after 24h, and the local skin reaction was observed at 0.5h, 24h, and 48h after removal, respectively.

Claims (10)

1. An anti-wrinkle effect composition is characterized in that: comprises ginsenoside, antler peptide, resveratrol, hydrolyzed collagen, sodium hyaluronate, superoxide dismutase and honey.
2. The anti-wrinkle effect composition according to claim 1, wherein: the composition also comprises a thickening agent, a humectant, a surfactant, an emollient, a cosurfactant, a preservative, a pH regulator, an essence and deionized water.
3. The anti-wrinkle effect composition according to claim 1 or 2, wherein: the composition comprises, by mass, 0.1-1% of ginsenoside, 0.5-2% of antler peptide, 0.1-1% of resveratrol, 0.5-2% of hydrolyzed collagen, 0.1-1% of sodium hyaluronate, 0.5-2% of superoxide dismutase, 0.5-2% of honey, 0.4-2% of thickening agent, 10-20% of humectant, 2-10% of emollient, 10-25% of surfactant, 2-8% of cosurfactant, 0.1-0.5% of preservative, 0.2-0.6% of pH value regulator, 0.01-0.02% of essence and 60-80% of deionized water.
4. The anti-wrinkle effect composition according to claim 2 or 3, wherein: the thickener is a mixture of xanthan gum and carbomer.
5. The anti-wrinkle effect composition according to claim 2 or 3, wherein: the emollient is one of liquid paraffin, polydimethylsiloxane or polymethylphenylsiloxane.
6. The anti-wrinkle effect composition according to claim 2 or 3, wherein: the surfactant is any one or more of stearate, glyceryl monostearate, cetearyl alcohol or polyoxyethylene hydrogenated castor oil;
the humectant is any one or more of propylene glycol, glycerin or sodium hyaluronate;
the cosurfactant is polyethylene glycol 400, isopropanol or a combination of the two.
7. The anti-wrinkle effect composition according to claim 2 or 3, wherein: the preservative is any one or mixture of phenoxyethanol and methylparaben; the pH value regulator is triethanolamine; the hydrolyzed collagen is cod hydrolyzed collagen.
8. A skin care product containing an anti-wrinkle effect composition is characterized in that: 1-20% of an anti-wrinkle composition according to claims 1-7, comprising gel masks and creams.
9. A preparation method of a skin care product containing a composition with an anti-wrinkle effect is characterized by comprising the following steps: preparing the gel mask of claim 8, comprising, and in sequence,
step one, adding a humectant, a preservative and a thickening agent into deionized water, adding sodium hyaluronate, stirring to fully soak the mixture, standing for 12-15 hours at room temperature to fully swell and uniformly disperse the mixture to obtain a phase A;
step two, heating resveratrol, a surfactant, a cosurfactant and deionized water at 37 ℃ until the resveratrol, the surfactant, the cosurfactant and the deionized water are completely dissolved, and uniformly stirring to obtain a phase B;
and step three, adding the phase B into the phase A, stirring, simultaneously dripping a triethanolamine solution to adjust the pH value to 5.5-7, adding ginsenoside, antler peptide, hydrolyzed collagen, superoxide dismutase and honey, homogenizing, stirring, centrifuging to remove bubbles, and standing to obtain the gel mask.
10. A preparation method of a skin care product containing a composition with an anti-wrinkle effect is characterized by comprising the following steps: preparing the cream of claim 8, comprising, and in sequence,
step one, adding a humectant, a preservative and a thickening agent into deionized water, adding sodium hyaluronate, heating at 85 ℃ and stirring to obtain a phase A;
step two, mixing the surfactant and the emollient, heating and stirring at 85 ℃ until the surfactant and the emollient are completely dissolved, adding the pH value regulator, and uniformly stirring to obtain a phase B;
step three, heating resveratrol, a surfactant, a cosurfactant and deionized water at 37 ℃ until the resveratrol, the surfactant, the cosurfactant and the deionized water are completely dissolved, and uniformly stirring to obtain a phase C;
and step four, when the temperature is reduced to 75 ℃, adding the phase B into the phase A for homogenizing and stirring, then adding the phase C, when the temperature is reduced to 45 ℃, adding the ginsenoside, the antler peptide, the hydrolyzed collagen, the superoxide dismutase and the honey, homogenizing and stirring, discharging, and standing to obtain the cream.
