技术领域
本发明属于水产养殖饲料与繁育技术领域,具体涉及一种斑节对虾亲虾繁育专用配合饲料。
背景技术
斑节对虾(Penaeus monodon)通常被称为草虾(Grass shrimp)和黑虎虾(Blacktiger shrimp),隶属于节肢动物门(Arthropoda)、甲壳纲(Crustacea)、十足目(Decapoda)、对虾科(Penaeidae)、对虾属(Penaeus),是我国四大海水养殖虾类之一。近十年来,其年均产量约7万吨,在我国的主产区为广东、浙江、福建和海南。提升亲虾繁殖性能,培育优质苗种,是斑节对虾养殖业繁荣发展的关键环节之一。
营养是虾类性腺成熟和种苗质量的重要决定因素。沙蚕、牡蛎、鱿鱼、乌贼等天然饵料对亲虾性腺发育和繁殖具有促进作用,其实质是与其特异的营养组成密切相关。然而生物饵料在其应用过程中存在诸多问题:亲虾易被其携带的致病病原所感染;其质量和产量不稳定,易影响大批量亲虾催熟培育;生物饵料用量多、价格贵,制约养殖效益;投喂生物饵料易败坏水质,给亲虾培育带来管控难度与潜在风险。而亲虾人工配合饲料营养全面、运输使用方便、不携带致病病原,质量和产量相对稳定,前景广阔,经济效益可观。
发明内容
本发明的一个目的在于克服现有技术中的缺陷与不足,提供一种斑节对虾亲虾繁育专用配合饲料,该配合饲料诱食性好,易于被对虾消化吸收利用,可增强对虾免疫力及抗应激性能,能同时提升雌性与雄性斑节对虾亲虾的繁殖性能。
本发明的另一个目的在于提供所述斑节对虾亲虾繁育专用配合饲料的用途。
本发明的又一个目的在于提供所述斑节对虾亲虾繁育专用配合饲料的制备方法。
为了实现以上目的,本发明按照以下技术方案实现:
一种斑节对虾亲虾繁育专用配合饲料,其包括以下重量百分比的组分:鱼粉25%~40%、卤虫粉5.0%~9.5%、酶解鱼溶浆粉3%~6%、裂殖壶藻粉3%~9%、海带粉1%~3%、黑水虻虫粉2%~8%、虾壳粉2%~5%、贻贝粉6%~12%、海洋红酵母干粉2%~5%、面粉10%~15%、溶血卵磷脂1.5%~3%、ARA纯化油0.8%~1.4%、DHA纯化油1.5%、EPA纯化油1.5%、硬脂酸甘油三酯3.1%~3.7%、鱼油1%~2%、磷酸二氢钙1%~5%、维生素预混料0.9%~1.22%、矿物质预混料1.08%~1.4%。

另一方面,本发明还提供了一种用于促进斑节对虾繁育期生长性能的方法,其中,将根据本发明所述的斑节对虾亲虾繁育专用配合饲料饲喂所述斑节对虾。
本发明还提供了所述斑节对虾亲虾繁育专用配合饲料的制备方法,其包括以下步骤:
1)将除油类物料、矿物质预混料、维生素预混料之外的其它物料进行粉碎,达到95%过80目筛;
2)添加油类物料、矿物质预混料和维生素预混料进行二次混合,混合时间大于180秒;
5)制粒,调制温度90℃以上,挤压制粒;
6)制粒后进行后熟化、烘干、冷却,得成品。
本发明在精准营养配比的基础上,合理搭配了各原料的比例,极大满足斑节对虾亲虾对常规营养及微量营养素的需求的同时,提高了饲料的诱食性和消化利用率,且避免了原料的浪费;有效的改善了斑节对虾亲虾的繁殖性能,且减少了饲料造成的水体环境的污染。
本发明所用原料精选,性价比高,方便易得,且符合国家标准以行业标准的相关规定;所配饲料营养均衡、安全、卫生。
本发明饲料避免了生物饵料的一系列缺点,可有效替代生物饵料对亲虾进行营养强化,且降低了饵料系数,提升了养殖效益。
本发明选用的卤虫粉,其原料卤虫属丰年虫科,蛋白质含量高达60%,且富含多种必需氨基酸、类胡萝卜素、鱼虾味觉诱引剂等。本发明选用的酶解鱼溶浆粉,相对于鱼粉拥有更多易溶解于水的蛋白质、小肽和游离氨基酸等,其直接应用于饲料中可有效保持鱼蛋白水解物新鲜度和鱼体特殊活性成分,维护原料鱼体蛋白质和油脂新鲜度、保持海水鱼类原料对养殖动物生长的优势。本发明选用的裂殖壶藻粉,其原料裂殖壶藻富含油脂、蛋白质及碳水化合物等营养成分,分别约占细胞干重的56.7%、12.4%、25.3%。ω-3长链多不饱和脂肪酸含量丰富,尤其是DHA占总油脂的30~50%。研究表明,饲料中同时添加裂殖壶藻与VE可协同促进对虾雄虾精荚的再生,并提高精子数量。本发明选用的黑水虻虫粉,其原料黑水虻繁殖快、生物量大、吸收转化率高、虫体资源含量高、易饲养,且生命力顽强,是一种可资源化生产的昆虫。黑水虻虫体富含蛋白质、氨基酸、脂类、维生素、矿物质及一些活性物质。
本发明选用的海洋红酵母,含有丰富的蛋白质、脂类、肝糖等营养物质,同时富含类胡萝卜素、虾青素、消化酶类及维生素等多种活性物质,尤其是虾青素产量高。虾青素具有促生长、抗氧化、清除自由基、提高机体免疫和抗肿瘤的功能。另外,本发明添加的胆固醇既满足了亲虾对胆固醇的需求,提高了产卵量,又能显著提高虾青素的利用效率。
本发明着重提高了斑节对虾亲虾配合饲料中花生四烯酸(ARA)的含量,花生四烯酸作为一种重要的n-6LC-PUFA,同DHA和EPA相比,其在海水养殖动物营养中常处于被忽视的地位,但是,近年来众多研究发现,ARA代谢过程中形成的多种高生物活性的类烯酸物质,能调节一系列重要的生理代谢,且ARA在促进对虾发育和产卵方面起着关键作用。
本发明以苯磷硫胺取代了常规饲料中维生素B1的使用。苯磷硫胺是维生素B1的脂溶性衍生物,改善了水溶性维生素B1生物利用度低的缺点。适量的苯磷硫胺可提高饲料的诱食性、改善对虾的生长性能和对氨毒性的抵抗能力。
本发明添加的蛋白酶可将大分子蛋白水解成寡肽或氨基酸,提高饲料蛋白利用率。可显著改善对虾生长,提高对虾肝胰腺蛋白酶活性。本发明添加了分离自褐藻的岩藻聚糖(又叫多糖硫酸酯),具有抗凝血、免疫调节、抗肿瘤等生物活性。本发明添加了壳聚糖季铵盐,壳聚糖是唯一的碱性多糖,壳聚糖季铵盐克服了壳聚糖溶解性不足的缺点,且壳聚糖季铵盐分子量小、比壳聚糖更易溶于水,其抗菌性能优于壳聚糖及其它壳聚糖衍生物。辣椒是我国重要的经济农作物,在全国各地广泛种植。辣椒碱具有增强对虾免疫力、改善对虾生长性能的潜力,而且具有添加剂量少、添加成本低的优势。
本发明所选用的饲用益生菌种为丁酸梭菌,其作为厌氧芽孢杆菌,不受胃酸、胆汁酸等影响;丁酸梭菌产生的丁酸、乳酸,能够促进动物肠道中乳酸菌、双歧杆菌等有益菌群的增殖与发育,抑制肠道内有害菌和腐败菌的生长、繁殖,纠正肠道菌群紊乱,调节肠道微生态平衡,减少肠毒素的产生;丁酸梭菌在动物肠道内产生多种消化酶、多种维生素以及丁酸等,对动物具有营养保健作用;丁酸、乙酸、氢气等丁酸梭菌的代谢产物,能够减轻肠道炎症,加速肠道损伤修复。丁酸梭菌作为饲用益生菌,集芽孢杆菌与乳酸菌的优点于一身,既具有优良的生产性能,又具有耐热、易贮藏、适应动物肠道微生态环境(厌氧、酸、胆盐)的特性,对于减少当前动物生产中饲用抗生素的滥用、减少药物残留与保障动物健康方面有着重要的意义。
本发明所选用的抗氧化剂为α-硫辛酸,其又称为二硫辛酸,是线粒体中经酶促反应由正辛酸转化而来的一种天然的二巯基化合物。α-硫辛酸分子结构中有一个二硫键,一旦进入细胞内部,其二硫键即被多种酶还原,生成二氢硫辛酸。α-硫辛酸和二氢硫辛酸在机体中的相互转化及再生,能够清除机体内几乎全部类型的自由基。α-硫辛酸可与自由金属离子螯合,能够缓解重金属中毒。二氢硫辛酸是一种强还原剂,能够将许多内源性的抗氧化剂(维生素C、维生素E、GSH)从氧化态转化为还原态。营养性抗氧化剂维生素E、维生素C和微量元素等可显著缓解氧化应激,但日粮中添加量过多会造成总日粮营养的不平衡以及营养物质间的拮抗。α-硫辛酸是唯一脂溶性和水溶性兼具的非营养性抗氧化剂,易被吸收,且能分布到机体的各部位发挥作用。饲料中适量添加α-硫辛酸可弥补营养性抗氧化剂的不足。
总体而言,本发明所述专用配合饲料专用于斑节对虾亲虾繁育期间使用,所添加的各成分协同增效,显著改善了斑节对虾亲虾的生长性能,显著促进了斑节对虾雌虾与雄虾的性腺发育,显著提高了亲虾的产卵量、产卵率、连续产卵率、受精卵孵化率、变态率及卵径。本发明所述斑节对虾亲虾繁育专用配合饲料,达到了鲜活饵料营养强化的效果,且避免了因投喂鲜活饵料而感染外源病源,污染水质的各种问题。
具体实施方式
为了能更清晰的理解本发明,以下将详细阐述本发明的具体实施方式,但本发明的实施方式并不限于此。在未作说明的情况下,本发明所采用的试剂、设备和方法均为本技术领域常规市购的试剂、设备和常规使用的方法。
一、饲料配方与制备
根据本发明实施例1至3的斑节对虾亲虾繁育专用配合饲料配方组成如下表1所示。表1中所有原料的用量单位均是克(g),按照每公斤对虾饲料中的添加量进行配置。对比例1是普通市售商业草虾饲料;对比例2是鲜活饵料(长吻沙蚕与杜氏枪乌贼的重量比为1:1)。

*所述纯化核苷酸混合物为纯化胞苷酸、腺苷酸、胸苷酸、鸟苷酸和核糖核酸以等重量比例复配而成。
实施例1至3的亲虾饲料的制备方法,具体步骤如下:
1)除油类物料、预混料混合物外,其它物料经粗粉碎进配料仓;
2)经计量配料后进行一次混合,混合时间大于90秒,后依次经细粉碎、超微粉碎,达到95%过80目筛;
3)超微粉碎结束后进行二次配料;
4)添加油类物料和预混料混合物进行二次混合,混合时间大于180秒;
5)经制粒系统制粒,调制温度90℃以上,挤压制粒;
6)制粒后进行后熟化、烘干、冷却,得成品。
二、亲虾与养殖管理
实验于2018年5月10日至10月10日在广东省湛江市东海岛恒兴863试验基地进行。选取4月龄、大小均匀、体质健壮、性腺未开始发育的斑节对虾。雌虾初始体重为(42.55±0.23)g,雄虾的初始体重为(39.68±0.28)g。
实验分为5组,分别投喂实施例1、实施例2、实施例3的配合饲料、商业草虾料(对比例1)和鲜活饵料(对比例2)。每个实验组三个平行,每个平行有20尾亲虾,雌雄亲虾分开饲养。实验开始前,对每尾亲虾的单侧眼柄套上眼标作为个体标记。营养强化阶段从2018年6月5日持续到2018年8月20日,共计76天,每天饱食投喂4次。营养强化阶段结束之后,摘除雌虾单侧眼柄进行促熟。繁殖产卵第一阶段继续投喂各组饲料,时间从2018年8月21日至9月10日;第二阶段所有处理组均投喂鲜活饵料沙蚕和乌贼,时间从2018年9月11日至10月15日。
实验过程中,养殖水温为27-28℃,盐度为28-30‰,pH为7.9-8.2,溶解氧≥5mg/L,总氨氮≤0.5mg/L,亚硝酸盐≤0.1mg/L。
三、交配管理和幼体培育
雌虾单侧眼柄摘除后,每日检查是否有性腺发育成熟即将产卵的个体;同时挑选同一处理组的雄虾(精荚呈白色且饱满凸出)进行人工授精。
受精产卵期间,产卵孵化用水为经消毒过滤没有余氯的新鲜海水,同时加入EDTA-2Na络合水中的重金属离子,并保持24小时不间断地微充气。
人工授精后,给雌虾做好标记待其产卵。亲虾产卵结束后,及时捞出并在2小时内完成计数。雌虾所产的卵继续留在孵化桶内,每隔1小时翻动1次,直至次日傍晚孵化出无节幼体并计数。孵化桶内存活下来的无节幼体在第三天傍晚左右变态成为蚤状幼体后计数统计。
四、样品采集
实验开始时,根据眼标个体标记称量每尾亲虾的初始体重。营养强化阶段结束后,称量全部的亲虾体重,计算营养强化后的增重率。同时各实验组每个平行各取3尾性腺发育完全的亲虾,取其血液、肝胰腺和性腺称重,并计算营养强化后期的肝胰腺指数和性腺指数。繁殖产卵阶段结束之后,同样测量全部的亲虾体重,计算繁殖产卵后的增重率。每个试验组的各个平行各取3尾性腺发育成熟的雌虾和精荚饱满的雄虾,同样取其血液、肝胰腺和性腺称重,计算繁殖产卵后期的肝胰腺指数和性腺指数。
人工授精后,统计各实验组产卵量、无节幼体数、蚤状幼体数、受精卵孵化率以及无节幼体变态率;计算各处理组的雌性亲虾产卵率、多次产卵率和平均产卵次数。取每组雌虾前10次产卵的卵子,待产卵结束后各取50粒左右的卵子,用光学显微镜测量其卵径。
五、实验数据的统计分析
增重率=(终末体重-初始体重)×100/初始体重
性腺指数=性腺重×100/亲虾体重
受精卵孵化率=各组无节幼体的计数平均数×100/各组同一亲虾个体产卵量的计数平均数
无节幼体变态率=各组蚤状幼体的计数平均数×100/各组相应的无节幼体的计数平均数
产卵率=产卵的雌虾数/受精产卵期间存活的雌虾总数×100%
多次产卵率=多次(两次及以上)产卵的雌虾数/产卵的雌虾数×100%
平均产卵次数=受精产卵期间各平行组总产卵次数/产卵的雌虾数
卵径=(长径+短径)/2
六、实验结果
不同处理组亲虾的生长情况如表2所示。

从表2可以看出,营养强化阶段结束之后,各实验组雌虾的增重幅度均高于雄虾;实施例1至5雌虾和雄虾的增重率都显著高于对比例1(商业草虾料)(P<0.05);实施例1、2、4、5雌虾增重率和雄虾增重率均与对比例2(鲜活饵料组)没有显著性差异(P>0.05),但实施例3雌虾和雄虾增重率显著高于对比例2(P<0.05)。
繁殖产卵阶段结束之后,各实验组雌虾的增重幅度均高于雄虾;实施例1至5雌虾和雄虾的增重率都显著高于对比例1(商业草虾料)(P<0.05);实施例1、2、5雌虾增重率与对比例2没有显著性差异(P>0.05),实施例1、2、4、5雄虾增重率与对比例2没有显著性差异(P>0.05),而实施例3、4雌虾增重率显著高于对比例2(P<0.05);实施例3雄虾增重率显著高于对比例2(P<0.05)。
不同处理组的亲虾性腺指数如表3所示。

从表3中可以看出,营养强化阶段结束之后,实施例1-5雌虾的性腺指数都显著高于对比例1(商业草虾料)(P<0.05),但与对比例2(鲜活饵料组)没有显著性差异(P>0.05);繁殖产卵阶段结束之后,实施例1-5雌虾和雄虾的性腺指数都显著高于对比例1(商业草虾料)(P<0.05),实施例1-5雌虾、实施例1、2、4、5雄虾的性腺指数与对比例2(鲜活饵料组)没有显著性差异(P>0.05),但实施例3雄虾的性腺指数显著高于对比例2。
不同处理组的亲虾产卵效果如表4所示。

从表4中可以看出,实施例1-5雌虾的产卵量、产卵率、平均产卵次数都显著高于对比例1(商业草虾料)(P<0.05),但与对比例2(鲜活饵料组)没有显著性差异(P>0.05);实施例1-5雌虾的连续产卵率均显著高于对比例1(商业草虾料)(P<0.05),实施例1、2、4、5与对比例2没有显著性差异(P>0.05),但实施例3显著高于对比例2(P<0.05)。

从表5可以看出,实施例1-5受精卵孵化率、变态率和卵径都显著高于对比例1(商业草虾料)(P<0.05),但与对比例2(鲜活饵料组)没有显著性差异(P>0.05)。
由以上实验结果可见,本发明提供的斑节对虾亲虾繁育专用配合饲料中,各原料组分协同增效,相比于非繁育专用的商业草虾料,显著改善了斑节对虾亲虾的生长性能,显著促进了斑节对虾雌虾与雄虾的性腺发育,显著提高了亲虾的产卵量、产卵率、连续产卵率、受精卵孵化率、变态率及卵径。本发明提供的斑节对虾亲虾繁育专用配合饲料达到了由沙蚕与乌贼组成的鲜活饵料的效果,部分实施例甚至优于鲜活饵料,具有显著优异的技术效果。

1.一种斑节对虾亲虾繁育专用配合饲料,其特征在于包括以下重量百分比的组分:鱼粉25%~40%、卤虫粉5%~9.5%、酶解鱼溶浆粉3%~6%、裂殖壶藻粉3%~9%、海带粉1%~3%、黑水虻虫粉2%~8%、虾壳粉2%~5%、贻贝粉6%~12%、海洋红酵母干粉2%~5%、面粉10%~15%、溶血卵磷脂1.5%~3%、ARA纯化油0.8%~1.4%、DHA纯化油1.5%、EPA纯化油1.5%、硬脂酸甘油三酯3.1%~3.7%、鱼油1%~2%、磷酸二氢钙1%~5%、维生素预混料0.9%~1.22%、矿物质预混料1.08%~1.4%。
2.根据权利要求1所述的斑节对虾亲虾繁育专用配合饲料,其特征在于包括以下重量百分比的组分:鱼粉32%、卤虫粉8%、酶解鱼溶浆粉3%、裂殖壶藻粉5%、海带粉1%、黑水虻虫粉7%、虾壳粉3%、贻贝粉12%、海洋红酵母干粉3%、面粉11.7%、溶血卵磷脂2%、ARA纯化油1.1%、DHA纯化油1.5%、EPA纯化油1.5%、硬脂酸甘油三酯3.4%、鱼油1%、磷酸二氢钙1.5%、维生素预混料0.9%、矿物质预混料1.4%。
3.根据权利要求1所述的斑节对虾亲虾繁育专用配合饲料,其特征在于,所述维生素预混料由以下重量比例的组分组成:维生素A 0.01~0.05g,维生素D 0.015~0.03g,维生素E0.2~0.5g,维生素K 0.02~0.05g,维生素C酯0.5~1.5g,苯磷硫胺0.06~0.1g,核黄素0.02~0.05g,烟酸0.05~0.1g,泛酸钙0.05~0.1g,吡哆醇0.05~0.08g,生物素0.006~0.01g,肌醇0.2~0.5g,叶酸0.006~0.01g,维生素B12 0.003~0.009g,虾青素0.06~0.1g,L-肉碱盐酸盐0.1~0.15g,胆固醇0.7~1g,蛋白酶0.3~0.6g,褐藻岩藻聚糖0.3~0.6g,壳聚糖季铵盐0.2~0.4g,辣椒碱0.002~0.003g,纯化核苷酸混合物2.5~5g,丁酸梭菌0.5~1g,α-硫辛酸0.7~1.5g,脱脂米糠0.5~1g。
4.根据权利要求1所述的斑节对虾亲虾繁育专用配合饲料,其特征在于,所述维生素预混料由以下重量比例的组分组成:维生素A 0.03g,维生素D 0.02g,维生素E 0.35g,维生素K 0.03g,维生素C酯1.2g,苯磷硫胺0.07g,核黄素0.03g,烟酸0.06g,泛酸钙0.07g,吡哆醇0.063g,生物素0.008g,肌醇0.3g,叶酸0.009g,维生素B12 0.007g,虾青素0.08g,L-肉碱盐酸盐0.11g,胆固醇0.7g,蛋白酶0.4g,褐藻岩藻聚糖0.4g,壳聚糖季铵盐0.2g,辣椒碱0.003g,纯化核苷酸混合物2.96g,丁酸梭菌0.55g,α-硫辛酸0.75g,脱脂米糠0.6g。
5.根据权利要求3或4所述的斑节对虾亲虾繁育专用配合饲料,其特征在于,所述纯化核苷酸混合物为纯化胞苷酸、腺苷酸、胸苷酸、鸟苷酸和核糖核酸以等重量比例复配而成。
6.根据权利要求1所述的斑节对虾亲虾繁育专用配合饲料,其特征在于,所述矿物质预混料由以下重量比例的组分组成:硒代蛋氨酸0.002~0.006g,碘酸钾0.02~0.03g,氯化钾1~2g,氯化钠1~1.6g,氯化胆碱2.844~3.5g,硫酸镁1~1.6g,甘氨酸铜0.08~0.16g,硫酸亚铁0.2~0.5g,硫酸锰0.02~0.05g,蛋氨酸锌0.3~0.5g,蛋氨酸钴0.01~0.06g,沸石粉2~4g,甜菜碱0.5~1g。
7.根据权利要求1所述的斑节对虾亲虾繁育专用配合饲料,其特征在于,所述矿物质预混料由以下重量比例的组分组成:硒代蛋氨酸0.005g,碘酸钾0.028g,氯化钾1.6g,氯化钠1.5g,氯化胆碱3.5g,硫酸镁1.5g,甘氨酸铜0.13g,硫酸亚铁0.46g,硫酸锰0.046g,蛋氨酸锌0.46g,蛋氨酸钴0.051g,沸石粉3.72g,甜菜碱1g。
8.根据权利要求1至7任一项所述的斑节对虾亲虾繁育专用配合饲料用于促进斑节对虾繁育期生长性能的用途。
9.一种用于促进斑节对虾繁育期生长性能的方法,其特征在于,将根据权利要求1至7任一项所述的斑节对虾亲虾繁育专用配合饲料饲喂所述斑节对虾。
10.根据权利要求1至7任一项所述的斑节对虾亲虾繁育专用配合饲料的制备方法,其特征在于包括以下步骤:
1)将除油类物料、矿物质预混料、维生素预混料之外的其它物料进行粉碎,达到95%过80目筛;
2)添加油类物料、矿物质预混料和维生素预混料进行二次混合,混合时间大于180秒;
5)制粒,调制温度90℃以上,挤压制粒;
6)制粒后进行后熟化、烘干、冷却,得成品。
