本发明涉及一种含有菊芋叶提取液的组合物及其制备方法和应用,组合物包括:菊芋叶提取液、含锌化合物和含铜化合物的组合物、聚天门冬氨酸、复合氨基酸、葡聚烯糖、三氮唑核苷、植物免疫蛋白(疫霉菌属)提取液、玉米素发酵液、蜂蜡精提醇粉。制备方法为:(1)向菊芋叶提取液中依次加入聚天门冬氨酸和复合氨基酸;(2)加入柠檬酸,调节pH为4-5;(3)依次加入三氮唑核苷、葡聚烯糖和y-环糊精;(4)依次加入植物免疫蛋白(疫霉菌属)提取液、玉米素发酵液和蜂蜡精提醇液;(5)加入含锌化合物和含铜化合物的组合物,至完全溶解;(6)加入山梨酸钾和吐温。试验证明,本发明组合物显著降低黄瓜花叶病毒基因组复制的积累,继而降低病毒相关蛋白的表达,田间应用能够有效防治相应的植物病毒病。

技术领域
本发明涉及一种含有菊芋叶提取液的组合物及其制备方法和应用,尤其涉及含有菊芋叶提取液的组合物在防治苹果病毒病方面的应用,属于病毒病防治技术领域。
背景技术
近年来,受多种因素的影响,病毒病对植物的危害逐年加重,由次要病害上升为主要病害,已成为一种重要病害,主要表现在发病程度重,发病面积多,防治效果不理想,已成为制约产业发展的重要因素之一。目前世界上报道的仅苹果病毒病有39种,我国已检测到17种,造成我国病毒病的病原有病毒和类病毒两种。病毒病是系统性病害,一旦感病则终身带毒,最终导致树体衰弱死亡。
以苹果上病毒病为例,病毒病从症状上可以分为两大类:潜隐性病毒和非潜隐性病毒。潜隐性病毒顾名思义其在果树外观上没有明显的症状表现,但是会造成树体长势衰弱,枝条发芽率降低,嫁接不亲和,分枝能力降低,造成产量降低,最终导致树体死亡;非潜隐性病害症状明显,易于识别,症状表现在叶片和果实上。叶片上的症状多为花叶,也表现为小叶,极大影响果树光合作用,降低产量,严重时导致树体死亡,如图1所示。果实上症状表现多为花果,表面凹凸不平,丧失经济价值,如图2所示。
目前,苹果病毒病的防治措施主要通过预防和切断传播途径为主,辅助喷施植物免疫诱抗剂(蛋白类),可以激活树体的免疫相关通路,有一定治愈效果。而对于花果病,目前尚未有有效治愈药剂。
发明内容
本发明针对上述问题,提供了一种含有菊芋叶提取液的组合物及其制备方法和应用,试验证明,本发明组合物显著降低病毒基因组的积累,进而降低病毒蛋白的表达,能够有效防治植物病毒病。。本发明的技术方案如下:
一种含有菊芋叶提取液的组合物,包括以下组分:
菊芋叶提取液、含锌化合物和含铜化合物的组合物、聚天门冬氨酸、复合氨基酸、葡聚烯糖、三氮唑核苷、植物免疫蛋白(疫霉菌属)提取液、玉米素发酵液、蜂蜡精提醇粉。
优选的,所述含锌化合物选自乙酸锌或柠檬酸锌,所述含铜化合物选自乙酸铜或柠檬酸铜。
本发明组合物还包括辅料,所述辅料为y-环糊精、水杨酸钠、山梨酸钾和吐温80。
进一步的,本发明组合物,包括以下按重份数计的组分:菊芋叶提取液50-100份、含锌化合物和含铜化合物的组合物1-3份、聚天门冬氨酸1-3份、复合氨基酸5-15份、葡聚烯糖0.1-0.3份、三氮唑核苷5-15份、植物免疫蛋白(疫霉菌属)提取液5-10份、玉米素发酵液5-10份、蜂蜡精提醇液0.5-3份;y-环糊精10-15份、水杨酸钠0.5-1份、山梨酸钾和吐温80 0.5-1份;其中,含锌化合物和含铜化合物的重量比为1:99-99:1。

进一步的,上述组合物的制备方法,具体步骤如下:
(1)将菊芋叶提取液加温至40-50℃,恒温;加入聚天门冬氨酸,搅拌均匀后,再边搅拌边加入复合氨基酸,搅拌至完全溶解,获得混合物A;
(2)向混合物A中缓慢加入柠檬酸,调节液体pH为4-5,持续搅拌;
(3)在搅拌状态下,依次缓慢加入三氮唑核苷和葡聚烯糖,搅拌至完全溶解,停止加温;边搅拌边加入y-环糊精,溶解后持续搅拌30-90min;
(4)稳定冷却至15-25℃,搅拌状态下依次加入植物免疫蛋白(疫霉菌属)提取液、玉米素发酵液和蜂蜡精提醇液(主要成分为正三十烷醇);
优选的,所述植物免疫蛋白(疫霉菌属)提取液中植物免疫蛋白的含量为0.1%(质量分数)、玉米素发酵液中玉米素含量为200ppm;
(5)搅拌状态下,加入含锌化合物和含铜化合物的组合物,至完全溶解;
(6)搅拌状态下加入山梨酸钾和吐温,搅拌均匀,且完全溶解,即得防治病毒病的组合物。
进一步的,所述菊芋叶提取液的制备方法:
鲜菊芋叶100份洗净粗粉碎,加入到预先加入5份乙酸乙酯的装有350-400份去离子水的搅拌反应釜中,常温下搅拌浸泡3天,甩干,捞出,再加入新鲜菊芋叶100份浸泡3天,浸提3次;过滤浸清液后加入水杨酸钠5份,即得菊芋叶提取液。
进一步的,本发明还包括组合物在防治病毒病的应用方法,兑水喷雾使用,每千克水中含本发明组合物0.5-15ml。
本发明与现有技术相比具有以下优点:
本发明相较于现有技术中的产品对病毒病具有非常好的防治效果,室内试验证明,本发明组合物明显降低病毒基因组复制的积累同时降低蛋白的表达,减轻植物病毒病的症状表型;本发明组合物对苹果花脸病毒病的矫正防效达到83.3%,而常规对照区防效为72%;已发生葡萄病毒病的植株经过喷施本发明组合物后,病毒病症状较对照明显改善,已经停长的生长点开始抽芽生长,新生成叶片平展,无花叶畸形现象,且植株可以正常进行光合作用。

实施例1:一种用于防治病毒病的组合物及其制备方法
组合物包括以下组分:菊芋叶提取液60份、含乙酸铜和乙酸锌的组合物2份、聚天门冬氨酸2份、复合氨基酸10份、葡聚烯糖0.2份、三氮唑核苷10份、植物免疫蛋白(疫霉菌属)提取液10份、玉米素发酵液5份、蜂蜡精提醇液1份;y-环糊精10份、水杨酸钠0.5份、山梨酸钾和吐温800.5份;其中,乙酸铜和乙酸锌的重量比为1:1。
上述组合物的制备方法,具体步骤如下:
(1)菊芋叶提取液的制备:鲜菊芋叶100份洗净粗粉碎,加入到预先加入5份乙酸乙酯的装有370份去离子水的搅拌反应釜中,常温下搅拌浸泡3天,甩干,捞出,再加入新鲜菊芋叶100份浸泡3天,浸提3次;过滤浸清液后加入水杨酸钠5份,即得菊芋叶提取液:
(2)将菊芋叶提取液加温至45℃,恒温;加入聚天门冬氨酸,搅拌均匀后,再边搅拌边加入复合氨基酸,搅拌至完全溶解,获得混合物A;
(3)向混合物A中缓慢加入柠檬酸,调节液体pH为4.5,持续搅拌;
(4)在搅拌状态下,依次缓慢加入三氮唑核苷和90%葡聚烯糖,搅拌至完全溶解,停止加温;边搅拌边加入y-环糊精,溶解后持续搅拌80min;
(5)稳定冷却至20℃,搅拌状态下依次加入植物免疫蛋白(疫霉菌属)提取液、玉米素发酵液和蜂蜡精提醇液(主要成分为正30烷醇);
所述植物免疫蛋白(疫霉菌属)提取液中植物免疫蛋白的含量为0.1%(质量分数)、玉米素发酵液中玉米素含量为200ppm;
(6)搅拌状态下,加入乙酸铜和乙酸锌,至完全溶解;
(7)搅拌状态下加入山梨酸钾和吐温,搅拌均匀,且完全溶解,即得防治病毒病的组合物。
实施例2:一种用于防治病毒病的组合物及其制备方法
组合物包括以下组分:菊芋叶提取液80份、含柠檬酸锌和柠檬酸铜的组合物3份、聚天门冬氨酸1份、复合氨基酸13份、葡聚烯糖0.1份、三氮唑核苷7份、植物免疫蛋白(疫霉菌属)提取液7份、玉米素发酵液8份、蜂蜡精提醇液2份;y-环糊精12份、水杨酸钠0.8份、山梨酸钾和吐温80 0.7份;其中,柠檬酸锌和柠檬酸铜的重量比为1:3;
上述组合物的制备方法,具体步骤如下:

(5)稳定冷却至25℃,搅拌状态下依次加入植物免疫蛋白(疫霉菌属)提取液、玉米素发酵液和蜂蜡精提醇液(主要成分为正30烷醇);
所述植物免疫蛋白(疫霉菌属)提取液中植物免疫蛋白的含量为0.1%(质量分数)、玉米素发酵液中玉米素含量为200ppm;
(6)搅拌状态下,加入乙酸铜和乙酸锌,至完全溶解;
(7)搅拌状态下加入山梨酸钾和吐温,搅拌均匀,且完全溶解,即得防治病毒病的组合物。
实施例3:一种用于防治病毒病的组合物及其制备方法
组合物包括以下组分:菊芋叶提取液90份、乙酸锌和柠檬酸铜的组合物3份、聚天门冬氨酸3份、复合氨基酸14份、葡聚烯糖0.3份、三氮唑核苷8份、植物免疫蛋白(疫霉菌属)提取液6份、玉米素发酵液8份、蜂蜡精提醇液1份;y-环糊精11份、水杨酸钠1份、山梨酸钾和吐温80 1份;其中,乙酸锌和柠檬酸铜的重量比为2:1。
上述组合物的制备方法,具体步骤如下:
(1)菊芋叶提取液的制备:鲜菊芋叶100份洗净粗粉碎,加入到预先加入5份乙酸乙酯的装有350份去离子水的搅拌反应釜中,常温下搅拌浸泡3天,甩干,捞出,再加入新鲜菊芋叶100份浸泡3天,浸提3次;过滤浸清液后加入水杨酸钠5份,即得菊芋叶提取液:
(2)将菊芋叶提取液加温至40℃,恒温;加入聚天门冬氨酸,搅拌均匀后,再边搅拌边加入复合氨基酸,搅拌至完全溶解,获得混合物A;
(3)向混合物A中缓慢加入柠檬酸,调节液体pH为4,持续搅拌;
(4)在搅拌状态下,依次缓慢加入三氮唑核苷和90%葡聚烯糖,搅拌至完全溶解,停止加温;边搅拌边加入y-环糊精,溶解后持续搅拌60min;
(5)稳定冷却至20℃,搅拌状态下依次加入植物免疫蛋白(疫霉菌属)提取液、玉米素发酵液和蜂蜡精提醇液(主要成分为正30烷醇);
所述植物免疫蛋白(疫霉菌属)提取液中植物免疫蛋白的含量为0.1%(质量分数)、玉米素发酵液中玉米素含量为200ppm;
(6)搅拌状态下,加入乙酸铜和乙酸锌,至完全溶解;
(7)搅拌状态下加入山梨酸钾和吐温,搅拌均匀,且完全溶解,即得防治病毒病的组合物。
试验例1:室内试验
黄瓜花叶病毒(CMV,Cucumber mosaic virus)是目前已知的寄主范围最广的植物病毒之一。本试验采用本发明组合物喷施接种CMV的植株,以测定本发明组合物对CMV的防治效果。试验设3个处理,健康植株、对照组(接种CMV未喷施本发明物)和处理组(接种CMV后喷施本化合物),每个处理重复3次。喷施28天后,对照组植株产生轻微花叶和严重皱缩,株高明显降低;处理组叶片逐渐伸展,无花叶症状,如图3所示。
利用Northern blot和Western blot分别检测对CMV的RNA和蛋白的积累水平的影响,系统叶片总RNA的Northern blot检测结果表明,喷施28天后显著降低CMV基因组的积累水平,如图4所示;系统叶片CP蛋白的Western blot检测结果表明,喷施化合物28后,显著降低CMV CP的表达水平。
综上所述,喷施本发明组合物显著降低CMV基因组复制的积累同时降低蛋白的表达,能够有效防治黄瓜花叶病毒病。。
试验例2:本发明组合物对苹果病毒病的防治
1.供试作物
苹果——长富2号,晚熟条红红富士品种。
2.试验地点
试验安排在烟台市牟平区宁海街道城北村。苹果园乔化种植,果实套袋管理,株行距3米×4米,树龄25年,黏壤土,往年苹果花脸病毒病较重,管理水平中等偏上。
3.试验设计和安排
试验设试验区、常规对照区和空白对照区三个处理,试验区喷施本发明组合物,常规对照区喷施市场上购买植物免疫蛋白,其中各选择往年苹果花脸病毒病较重的苹果树4株作为试验区和常规对照区,选择往年苹果花脸病毒病较重的苹果树3株作为对照区,各处理苹果树随机选择。
4.试验时间和方法
在苹果盛花期,使用背负式喷雾器喷施本发明组合物100倍液、植物免疫蛋白100倍液,重点喷施部位为苹果花器;每株数喷施药液3公斤。
5.调查方法
于苹果摘袋后10天左右,苹果着色后,调查发病情况。调查方法:每个处理调查3株树,每株数按东、西、南、北、中五个方向随机调查40个果实,共200个果实,统计病果数,并分别记载病级,分析防治效果。
病果分级标准:共分5级。
0级,无病果;
1级,有商品价值,凹凸或花脸面积占整个果实面积5%以下;
3级,有商品价值,凹凸或花脸面积占整个果实面积6%-10%;
5级,无商品价值,凹凸或花脸面积占整个果实面积10%以上;
结果与分析由表1可见,苹果树喷施本发明组合物后,苹果花脸病毒病显症率较空白对照显著下降,病果率仅有14%,其中病果具有商品价值的占9%,如图5所示,仅有5%的病果没有商品价值。常规对照区病果率25%,其中病果具有商品价值的占13.3%,如图5所示。本发明组合物对苹果花脸病毒病的矫正防效达到83.3%,常规对照区防效为72%,处理区防治效果显著优于常规对照区。

试验例3:本发明组合物对葡萄病毒病的防治
1.供试作物
葡萄——阳光玫瑰,晚熟品种。
2.试验地点
试验安排在烟台市牟平区宁海街道城北村。葡萄篱架栽培,当年定植,管理水平中等偏上。
3.试验设计和安排
试验设试验区和对照区两个处理,其中选择定植后具有病毒病症状植株作为试验区(本发明组合物施药区),选择其中4株作为对照区,各处理随机选择。
4.试验时间和方法
在葡萄苗期,使用背负式喷雾器分别喷施本发明组合物和植物核糖核酸营养剂100倍液,重点喷施部位为生长点处。
结果与分析
葡萄病毒病表现为叶片畸形,小叶,不能进行光合作用,顶端生长点坏死,不能进行生长。喷施本发明组合物后,毒病症状较对照明显改善,表现在生长点开始生长,新生成叶片平展,无花叶症状,植株可以正常进行光合作用,如图6所示。

Claims (3)

1.一种含有菊芋叶提取液的组合物在防治苹果花脸病毒病的应用,其特征在于, 所述组合物包括以下按重量份数计的组分:菊芋叶提取液50-100份、含锌化合物和含铜化合物的组合物1-3份、聚天门冬氨酸1-3份、复合氨基酸5-15份、葡聚烯糖0.1-0.3份、三氮唑核苷5-15份、疫霉菌属植物免疫蛋白提取液5-10份、玉米素发酵液5-10份、蜂蜡精提醇液0.5-3份;γ-环糊精10-15份、水杨酸钠0.5-1份、山梨酸钾和吐温80 0.5-1份;其中,含锌化合物和含铜化合物的重量比为1:99-99:1;
所述菊芋叶提取液的制备方法:
鲜菊芋叶100份洗净粗粉碎,加入到预先加入5份乙酸乙酯的装有350-400份去离子水的搅拌反应釜中,常温下搅拌浸泡3天,甩干,捞出,再加入新鲜菊芋叶100份浸泡3天,浸提3次;过滤浸清液后加入水杨酸钠5份,即得菊芋叶提取液;
所述含锌化合物选自乙酸锌或柠檬酸锌,所述含铜化合物选自乙酸铜或柠檬酸铜。
2.根据权利要求1所述的应用,其特征在于,所述组合物包括以下按重量份数计的组分:菊芋叶提取液60份、含锌化合物和含铜化合物的组合物2份、聚天门冬氨酸2份、复合氨基酸10份、葡聚烯糖0.2份、三氮唑核苷10份、疫霉菌属植物免疫蛋白提取液10份、玉米素发酵液5份、蜂蜡精提醇液1份;γ-环糊精10份、水杨酸钠0.5份、山梨酸钾和吐温80 0.5份;其中,含锌化合物和含铜化合物的重量比为1:1。
3.根据权利要求1所述的应用,其特征在于,疫霉菌属植物免疫蛋白提取液中植物免疫蛋白的质量分数含量为0.1%、玉米素发酵液中玉米素含量为200ppm。
