Detailed Description
The invention will be further described with reference to the drawings and the detailed description, which are not intended to limit the invention in any way. The starting materials used in the examples of the present invention are conventionally purchased raw materials unless otherwise specified.
Example 1
1. Experimental Material
The traditional Chinese medicine compound group: according to the mass ratio of 1: 1: 1: 1: 1, weighing the raw materials of purslane, radix sophorae flavescentis, meadowrue root and rhizome, fuling and patchouli, crushing, drying in a hot air drying oven at 60 ℃, sieving with a 80-mesh sieve, uniformly mixing to prepare a traditional Chinese medicine compound, dividing into two groups, and respectively and uniformly mixing with feed to ensure that the addition amount of the traditional Chinese medicine compound in the feed is 1g/kg (traditional Chinese medicine compound/feed) and 3g/kg (traditional Chinese medicine compound/feed).
Antibiotic group (aureomycin 250 mg/kg): the aureomycin and the feed are mixed evenly to ensure that the mass ratio of the aureomycin to the feed is 250mg/kg (aureomycin/feed).
2. Experimental methods
Selecting 100 yellow-feathered broilers of 1 day old, dividing the broilers into four groups, namely a blank group (only feed), an antibiotic group (250 mg/kg) of aureomycin, a Chinese herbal compound group 1(1g/kg) and a Chinese herbal compound group 2(3g/kg), weighing, calculating the average empty stomach weight of each group, feeding the broilers for 42 days respectively, and freely drinking water during the period to ensure good ventilation and proper temperature. After feeding for 42 days, recording feed intake, weighing on an empty stomach, and calculating Average Daily Gain (ADG), total gain and feed-weight ratio (F/G) of the broilers in each group; quickly taking the middle section of the jejunum of 2-3 mm, immersing the middle section of the jejunum into prepared normal saline, rinsing off the content, adding 20mL of 4% paraformaldehyde phosphate buffer, fixing for 48h (keeping the pH value equal to 7.4), embedding by using conventional paraffin, preparing 10 continuous transverse sections (the thickness is 5 mu m), staining by conventional hematoxylin-eosin (H.E.), measuring the villus height and the crypt depth of the small intestine, and calculating the ratio of the villus height and the crypt depth; spleen, thymus and bursa of fabricius were collected, peripheral fat was removed and water was sucked off with filter paper, and then weighed and recorded, and immune organ index [ immune organ index (g/kg) ═ fresh weight (g) of immune organ/live weight (kg) ] was calculated.
3. Results of the experiment
The experimental results are shown in 1-5
As can be seen from Table 1, compared with the blank group, the antibiotic group (250 mg/kg of aureomycin), the traditional Chinese medicine compound group 1(1g/kg) and the traditional Chinese medicine compound group 2(3g/kg) have certain effects on daily weight gain of the broiler chickens, wherein the traditional Chinese medicine compound group 1(1g/kg) has more obvious effect on daily weight gain of the broiler chickens, and can improve the average daily weight gain of the broiler chickens.

As shown in Table 2, compared with the blank group, the antibiotic group (250 mg/kg of aureomycin), the traditional Chinese medicine compound group 1(1g/kg) and the traditional Chinese medicine compound group 2(3g/kg) have certain effects on increasing the weight of the broilers in the whole period, wherein the traditional Chinese medicine compound group 1(1g/kg) has more obvious weight increasing effect on promoting the growth of the broilers in the whole period, and the difference is obvious (P is less than 0.05).

As can be seen from Table 3, compared with the blank group, the weight ratio of the antibiotic group (250 mg/kg of aureomycin), the Chinese herbal compound group 1(1g/kg) and the Chinese herbal compound group 2(3g/kg) is obviously reduced, and the feed conversion rate is high. Wherein, the traditional Chinese medicine compound group 1(1g/kg) has more obvious effect of promoting the feed conversion of the broiler chickens (P is less than 0.05).

The absorption and utilization of the animal to the nutrient substances can be directly influenced by the quality of the intestinal mucosa structure, and the ratio of the villus height of the small intestine to the depth of the crypt is an important index for measuring the digestive absorption function of the small intestine. As can be seen from the observation of the data in the figure 1 and the analysis table 4, compared with the antibiotic group (250 mg/kg of aureomycin), the broiler chicken small intestine crypt depth of the traditional Chinese medicine compound group 1(1g/kg) and the broiler chicken small intestine crypt depth of the traditional Chinese medicine compound group 2(3g/kg) is reduced, and the villus height and the V/C value (small intestine villus height/small intestine crypt depth) are both improved to a certain extent, namely, the intestinal mucosa structure can be improved, the small intestine microvilli density is increased, the small intestine villus height is increased, the small intestine absorption area is increased, the intestinal mucosa crypt depth and the intestinal mucosa epithelial cell function are maintained, and the absorption and utilization of nutrient substances are facilitated, so that the growth of animals is promoted. Wherein, the Chinese herbal compound group 1(1g/kg) has the best effect of promoting digestion and absorption, the depth of crypt of small intestine is reduced, and the height of villus and the V/C value are both obviously improved (P is less than 0.05).
From the table 1-4, it can be seen that after the feed added with the traditional Chinese medicine compound is eaten, the weight increase, the feed conversion rate and the digestion and absorption functions of the broiler chickens are all obviously improved, and when the addition amount of the traditional Chinese medicine compound is 0.1% of the feed amount, the effect is optimal, and the increase of the broiler chickens can be obviously promoted.

Spleen, thymus and bursa of fabricius are organs which exert immune effect on poultry organisms, and the immune index of the organs can be detected to preliminarily measure the immune state of the organisms and the development condition of an immune system. As can be seen from Table 5, spleen index, thymus index and bursa of Fabricius index of the antibiotic group (250 mg/kg of aureomycin), the Chinese herbal compound group 1(1g/kg) and the Chinese herbal compound group 2(3g/kg) are all improved to a certain extent, which belong to the increase within the normal range, reflect the enhancement of animal immune function to a certain extent, and are beneficial to promoting the organism to generate immune cells and improving the level of vaccine antibodies. The spleen index and the thymus index of the traditional Chinese medicine compound group 1(1g/kg) are obviously increased, the bursa of Fabricius index of the traditional Chinese medicine compound group 2(3g/kg) is obviously increased (P is less than 0.05), and the feed added with the traditional Chinese medicine compound has the effect of obviously improving the cellular immunity and the humoral immunity of the broiler.
In conclusion, the performances of the broiler experimental group eating the feed added with the traditional Chinese medicine compound in the aspects of average daily gain, total gain, feed conversion rate, ratio of small intestine villus density to crypt depth, spleen index, bursa of fabricius index, thymus index and the like are superior to or equal to those of a broiler control group eating the feed added with antibiotics, and the traditional Chinese medicine compound does not add antibiotics, does not cause drug resistance, does not expose a large amount of livestock and poultry and even human beings to infection risks caused by drug-resistant strains, and does not bring hidden dangers to food safety, so the traditional Chinese medicine compound is superior to growth-promoting antibiotics on the whole, and can provide a new idea for a non-resistant, green and healthy breeding mode.
In addition, through repeated experiments of the inventor, when the mass ratio of the purslane, the radix sophorae flavescentis, the meadowrue root and rhizome, the furnace soil and the patchouli is 0.8-1.2: 0.8-1.2: 0.8-1.2: 0.8-1.2: 0.8-1.2, under the condition of the same total compound dosage, different proportions have little influence on experimental results, so the embodiment of the invention details that when the mass ratio is 1: 1: 1: 1: experimental results at 1.
