Abstract
The invention discloses a traditional Chinese medicine compound for improving growth performance and body immunity of broiler chickens and a preparation method and application thereof, and relates to the technical field of veterinary medicines. The traditional Chinese medicine compound comprises callicarpa nudiflora, purslane, houttuynia cordata, patchouli, mint, furnace soil and plant soot, and the mass ratio is 1.6-2.4: 0.8-1.2: 0.8-1.2: 0.8-1.2: 0.8-1.2: 0.7-0.9: 0.7-0.9, the medicines are combined, the effects of resisting bacteria and diminishing inflammation, clearing heat and drying dampness, cooling blood and detoxifying, stopping dysentery and checking diarrhea, nourishing yin and promoting the production of body fluid and enhancing immunity are achieved, the growth of cultured animals is promoted, and the feed has the advantages of low cost, safety, no drug resistance, environmental friendliness, no residue and no antibiotics. The broiler chicken can be effectively improved to improve the production performance and the immune function of the broiler chicken, so that the production efficiency of broiler chicken breeding is effectively improved; the preparation is simple, other adjuvants are not needed, and the requirement of large-scale use in the breeding industry can be met.

Background
Antibiotics are commonly used as a broad range of prophylactic agents and growth promoters for antibiotics in food animals and are the most cost effective means of maintaining feed efficiency and health in monogastric animals such as poultry by increasing growth rate, reducing mortality, and preventing disease. Due to the negative impact of synthetic drugs on poultry and consumer health and the rising cost of using drugs in poultry production, there is an urgent need to find functional replacement products. The european union has banned the use of antibiotics as growth promoters in 2006. The public announcement is also issued in rural areas of agriculture in China, and all varieties of growth-promoting drug feed additives except the traditional Chinese medicines are withdrawn from the 2020 by 1 month and 1 day.
The application of the traditional Chinese medicine in the feed or the additive thereof for preventing and treating the diseases of the cultured animals and promoting the growth is more and more extensive, and the non-resistant, green and healthy culture mode has wide development prospect. The traditional Chinese medicine contains various nutritional ingredients such as protein, amino acid, vitamin, polysaccharide and the like, can effectively improve the growth performance of animals and enhance the digestion and absorption functions; meanwhile, the compound immunopotentiator contains various effective immunocompetent components, promotes phagocytic function of macrophages and conversion rate of lymphocytes, is beneficial to inhibiting bacteria, and has obvious enhancement effect on cellular immunity and humoral immunity of organisms. Although the disease-resistant effect of the traditional Chinese medicine product developed by natural plants is hardly better than that of antibiotics and chemical synthetic medicines, the traditional Chinese medicine greatly reduces the risk of drug resistance and avoids the generation of super bacteria, and the traditional Chinese medicine in China also has the advantages of rich resources, low price, small toxic and side effects, no drug resistance, difficult residue and the like.
Therefore, a novel disease-resistant growth-promoting traditional Chinese medicine compound which is low in price, high in efficiency, safe, free of drug resistance, environment-friendly, free of residues and free of antibiotics is developed, and has an important application value in the animal breeding industry.

The invention also aims to provide a preparation method of the traditional Chinese medicine compound for improving the growth performance and the body immunity of the broiler chicken.

The mint is pungent and cool in flavor, enters lung and liver meridians, and has the effects of dispelling wind and heat and clearing head and eyes; patchouli is pungent in flavor and slightly warm in flavor, enters spleen, stomach and lung meridians, and has the effects of resolving turbidity with aromatics, regulating the middle warmer, relieving vomiting, relieving exterior syndrome and relieving summer-heat; the plant soot has been approved as a novel natural feed additive in 2020, can adsorb intestinal mycotoxin, inhibit bacterial reproduction and improve the digestive function of animals, is properly compatible with other medicaments, and is used for treating diseases such as wind-cold exogenous disease, spleen-stomach deficiency-cold, diarrhea, injury bleeding and the like of livestock and poultry; fulonggan, pungent and slightly warm in flavor, has the actions of warming middle energizer to stop bleeding, checking adverse rise of qi and stopping vomiting, and is commonly indicated for hematemesis, epistaxis, hematochezia, vomiting due to stomach cold, vomiting due to vomiting caused by cold of deficiency type, and vomiting of pregnancy.
The traditional Chinese medicine compound has the advantages of low cost, safety, no drug resistance, environmental protection, no residue and no antibiotic, and has the effects of resisting bacteria and diminishing inflammation, clearing heat and drying dampness, cooling blood and removing toxicity, stopping dysentery and checking diarrhea, nourishing yin and promoting the production of body fluid and enhancing immunity.
Repeated tests show that the most preferable scheme is that the weight ratio of callicarpa nudiflora, purslane, houttuynia cordata, patchouli, mint, furnace soil and plant soot is 2: 1: 1: 1: 1: 0.8: 0.8 the Wenchang chicken is tested by using the traditional Chinese medicine compound, and the results show that the performance of the broiler experimental group eating the feed added with the traditional Chinese medicine compound is better than or equal to that of the broiler control group eating the feed added with the positive control drug, namely the flos lonicerae and scutellaria baicalensis extract, in the aspects of average daily feed intake, average daily gain, total average weight, feed weight ratio, spleen index, bursa of fabricius index, thymus index, immunoglobulin IgG/IgM, antioxidant capacity and the like, and the traditional Chinese medicine compound does not contain antibiotics, can not cause drug resistance, can not expose a large amount of livestock and poultry and even human beings to infection risks caused by drug-resistant strains, and can not bring hidden troubles to food safety. And when the traditional Chinese medicine compound is added in 3g/kg (traditional Chinese medicine/feed), the traditional Chinese medicine compound is integrally superior to a broiler chicken control group of the feed of the positive control medicine, namely the wild flower scutellaria baicalensis extract, and a new thought can be provided for a non-antibiotic, green and healthy breeding mode.
Compared with the prior art, the invention has the following advantages and effects:
in the traditional Chinese medicine compound, the medicines are combined, so that the effects of resisting bacteria and diminishing inflammation, clearing heat and drying dampness, cooling blood and detoxifying, stopping dysentery and checking diarrhea, nourishing yin and promoting the production of body fluid and enhancing immunity are achieved, the traditional Chinese medicine compound has the effect of promoting the growth of cultured animals, has the advantages of low cost, safety, no drug resistance, environmental protection, no residue and no antibiotics, is superior to a positive control medicine, namely the wild flower scutellaria baicalensis extract on the whole, and provides a new thought for a non-antibiotic, green and healthy culture mode. In addition, the traditional Chinese medicine compound can effectively improve the broiler chickens, so that the production performance and the immune function of the broiler chickens are improved, and the production efficiency of broiler chicken breeding is effectively improved; the preparation is simple, other adjuvants are not needed, and the requirement of large-scale use in the breeding industry can be met.
Detailed Description
The present invention will be described in further detail with reference to examples, but the embodiments of the present invention are not limited thereto. The starting materials used in the examples of the present invention are conventionally purchased raw materials unless otherwise specified.
The extract of Scutellaria baicalensis Georgi used in the examples is from Tech technologies of Beijing, Taier, Inc., and is a new veterinary drug of three types disclosed in No. 2462 of Ministry of agriculture.
Example 1
1. Experimental Material
The traditional Chinese medicine compound composition comprises 20g of beautyberry, 10g of purslane, 10g of houttuynia cordata, 10g of patchouli, 10g of mint, 8g of furnace soil and 8g of plant soot, and the mass ratio is 2: 1: 1: 1: 1: 0.8: 0.8 weighing each raw material, crushing, drying in a hot air drying oven at 60 ℃, sieving with a 80-mesh sieve, uniformly mixing to obtain a traditional Chinese medicine compound, dividing into three groups, and uniformly mixing with the feed respectively to ensure that the addition amount of the traditional Chinese medicine compound in the feed is 1g/kg (traditional Chinese medicine compound/feed), 3g/kg (traditional Chinese medicine compound/feed) and 5g/kg (traditional Chinese medicine compound/feed). Positive control group (extract of Scutellaria baicalensis Georgi 0.5 g/kg): mixing the Scutellaria baicalensis Georgi extract with feed uniformly to make the mass ratio of 0.5g/kg (Scutellaria baicalensis Georgi extract/feed). Wherein the feed is the basic ration.
2. Experimental methods
Selecting 300 Wenchang chickens of 1 day age, dividing the Wenchang chickens into five groups (60 Wenchang chickens in each group, 3 repetitions in each group and 20 Wenchang chickens in each repetition), respectively taking blank control groups (only feeding the Wenchang chickens by using basic ration, the composition of the basic ration is shown in table 1), positive control groups (0.5 g/kg of extract of radix scutellariae of wild flowers), Chinese herbal compound groups 1(1g/kg), Chinese herbal compound groups 2(3g/kg) and Chinese herbal compound groups 3(5g/kg), weighing, calculating the average fasting weight of each group, respectively feeding for 28 days, and freely drinking water in the period to ensure good ventilation and proper temperature. After feeding for 28 days, recording feed intake, weighing on an empty stomach, and calculating average daily feed intake, Average Daily Gain (ADG), average body weight in the whole period and feed-weight ratio (F/G) of each group of test broilers; after the 28-day-old feeding test is finished, 6 chickens (2 chickens in each repeated group) are randomly extracted from each treatment group for wing vein blood sampling, a blood sample is centrifuged at 1200 Xg for 15 minutes, and a serum sample is collected and stored at-20 ℃; spleen, thymus and bursa of fabricius were collected, peripheral fat was removed and water was sucked off with filter paper, and then weighed and recorded, and immune organ index [ immune organ index (g/kg) ═ fresh weight (g) of immune organ/live weight (kg) ] was calculated. The content of serum immunoglobulin IgG and IgM was detected by enzyme-linked immunosorbent assay (ELASA) kit, which was purchased from Shanghai enzyme-linked Biotechnology Ltd. The kit is used for detecting the concentration or activity of MDA and T-AOC in serum, and is purchased from Nanjing to build a bioengineering institute.

3. Results of the experiment
The statistical software SPSS 22.0 single-factor analysis of variance is used for carrying out difference significance analysis on test data among multiple groups of samples, all data are expressed as mean values +/-standard deviation, and the obtained test results are shown in tables 2-7.

As can be seen from table 2, compared with the blank control group, the positive control group (0.5 g/kg of scutellaria baicalensis extract) has a tendency of increasing the average daily gain of broiler chickens, the traditional Chinese medicine compound group 1(1g/kg) and the traditional Chinese medicine compound group 2(3g/kg) can significantly increase the average daily gain of broiler chickens (P <0.05), and the traditional Chinese medicine compound group 3(5g/kg) can significantly increase the average daily gain of broiler chickens (P < 0.01). The Chinese herbal compound group 3(5g/kg) can obviously improve the average daily gain (P is less than 0.05) of the broiler compared with a positive control group (0.5 g/kg) and the daily gains of the Chinese herbal compound group 1(1g/kg) and the Chinese herbal compound group 2(3g/kg) are also better than the positive control group (0.5 g/kg) to a certain extent.

As can be seen from Table 3, compared with the blank control group, the traditional Chinese medicine compound group 1(1g/kg) and the traditional Chinese medicine compound group 2(3g/kg) can obviously improve the whole-period average weight of the 28-day-old broilers (P is less than 0.05); compared with a positive control group (0.5 g/kg of the scutellariae barbatae extract), the traditional Chinese medicine compound group 3(5g/kg) can obviously improve the average weight of the broiler chickens at the age of 28 days (P is less than 0.05), and has very obvious difference (P is less than 0.01) compared with a blank control group.

As can be seen from table 4, compared with the blank control group, the material weight ratios of the positive control group (0.5 g/kg of the scutellaria baicalensis extract) and the compound traditional Chinese medicine group are not significantly different and show a decreasing trend, wherein the material weight ratio of the compound traditional Chinese medicine group 2(3g/kg) is decreased more, which indicates that the compound traditional Chinese medicine group 2(3g/kg) can promote the feed conversion rate of broiler chickens to a certain extent, and the effect of the compound traditional Chinese medicine group is better than that of the positive control group (0.5 g/kg of the scutellaria baicalensis extract).

Spleen, thymus and bursa of fabricius are organs which exert immune effect on poultry organisms, and the immune index of the organs can be detected to preliminarily measure the immune state of the organisms and the development condition of an immune system. As can be seen from Table 5, compared with the blank control group, the positive control group (0.5 g/kg of the extract of Scutellaria baicalensis Georgi) and the Chinese herbal compound groups all have certain improvements in thymus index, bursal index and spleen index, which belong to the increase in the normal range, reflect the enhancement of animal immune function to a certain extent, and are beneficial to promoting the organism to generate immune cells and improving the level of vaccine antibodies.

Immunoglobulin is an animal protein with antibody activity, is converted into an antibody after antigen stimulation, is the same as an immune organ index, and is an important index for measuring the immune function of animals. Wherein IgG is the highest content of immunoglobulin in serum, accounts for 75 percent, and is the main antibody generated by animals after natural infection and artificial active immunization; IgM is a high-performance antibody, the content of which is only second to that of IgG, and accounts for 10-12% of total serum Ig, and the IgM has strong bactericidal, bacteriolytic and agglutination effects. As can be seen from Table 6, compared with the blank control group, the positive control group (0.5 g/kg of Scutellaria baicalensis Georgi extract) has the advantages that the immunoglobulin IgG is remarkably increased, and the IgM is improved to a certain extent, so that the positive control group (0.5 g/kg of Scutellaria baicalensis Georgi extract) can improve the immunity of the broiler chicken. Observing the traditional Chinese medicine compound groups, the traditional Chinese medicine compound groups 1 and 2 can obviously improve the IgG level of the broiler chickens, the traditional Chinese medicine compound group 3 can greatly improve the IgG level of the broiler chickens, and similar results are reflected on immunoglobulin IgM, so that the traditional Chinese medicine compound groups can improve the immune function of organisms as same as a positive control group, and the traditional Chinese medicine compound groups have certain dosage tolerance, and the larger the dosage is, the more obvious the improvement effect is. In combination, IgG and IgM were equally or more quantitatively identical to or superior to that of the positive control group (Scutellaria baicalensis Georgi extract 0.5g/kg) starting from the traditional Chinese medicine compound group 2(3 g/kg).

T-AOC is used for evaluating the total antioxidant level formed by antioxidant substances, antioxidant enzymes and the like of the body; MDA is one of the most important products of membrane lipid peroxidation, and the damage of the membrane can be aggravated by the generation of MDA, and the degree of membrane lipid peroxidation is commonly known by the MDA. As shown in Table 7, compared with the blank control group, the total antioxidant capacity of each group of the Chinese herbal compound is higher than that of the blank control group, and the total antioxidant capacity of the group 3(5g/kg) of the Chinese herbal compound is significantly higher than that of the positive control group (P < 0.05). MDA of a positive control group (0.5 g/kg of the extract of the scutellaria baicalensis of the mountain flower) has a tendency of reducing, and the positive control group can reduce the oxidative damage of the oxidized oil to the organism or enhance the endogenous antioxidant capacity of the organism to a certain extent; MDA of each group of the traditional Chinese medicine compound also shows a descending trend, wherein the traditional Chinese medicine compound group 2(3g/kg) and the traditional Chinese medicine compound group 3(5g/kg) are remarkably reduced compared with a blank control group (P is less than 0.05). The results of T-AOC and MDA jointly show that the oxidation resistance of the traditional Chinese medicine compound group 2(3g/kg) and the traditional Chinese medicine compound group 3(5g/kg) is equal to or better than that of a positive control group (0.5 g/kg of the extract of the scutellaria baicalensis of the mountain flower).
In conclusion, the performance of the broiler experimental group eating the feed added with the traditional Chinese medicine compound in the aspects of average daily gain, total-period average body weight, material-to-weight ratio, spleen index, bursal index, thymus index, immunoglobulin IgG/IgM, antioxidant capacity and the like is superior to or equal to that of the broiler control group eating the feed added with the positive medicine (0.5 g/kg of the scutellaria baicalensis extract), and the traditional Chinese medicine compound does not contain antibiotics, does not cause drug resistance, does not expose a large amount of livestock and poultry and even human beings to infection danger caused by drug-resistant strains, the traditional Chinese medicine compound group 2(3g/kg) and the traditional Chinese medicine compound group 3(5g/kg) are equal to or superior to the positive control drug, namely the extract of the radix scutellariae barbatae, on the whole, so that more choices can be provided for clinical medication, and a new thought is provided for a non-resistance, green and healthy breeding mode.
In addition, through multiple experiments of the inventor, the inventor finds that the raw materials of the traditional Chinese medicine compound are weighed according to 20g of callicarpa nudiflora, 10g of purslane, 10g of houttuynia cordata, 10g of pogostemon cablin, 10g of mint, 8g of furnace soil and 8g of plant soot, and under the condition that the total compound dosage is the same, the influence of small change of the dosage of each medicine on the experimental result is very small, so the embodiment of the invention is described in detail as the experimental results of 16-24 g of callicarpa nudiflora, 8-12 g of purslane, 8-12 g of houttuynia cordata, 8-12 g of pogostemon cablin, 8-12 g of furnace soil, 7-9 g of furnace soil and 7-9 g of plant soot, and through dosage screening, the traditional Chinese medicine compound is considered to be uniformly mixed with feed according to 3g/kg, and the cost ratio of the medicines can be exerted to the maximum.
The above embodiments are preferred embodiments of the present invention, but the present invention is not limited to the above embodiments, and any other changes, modifications, substitutions, combinations, and simplifications which do not depart from the spirit and principle of the present invention should be construed as equivalents thereof, and all such changes, modifications, substitutions, combinations, and simplifications are intended to be included in the scope of the present invention.

The traditional Chinese medicine compound for improving growth performance and organism immunity of broiler chickens is characterized by comprising callicarpa nudiflora, purslane, houttuynia cordata, patchouli, mint, furnace soil and plant soot in a mass ratio of 1.6-2.4: 0.8-1.2: 0.8-1.2: 0.8-1.2: 0.8-1.2: 0.7-0.9: 0.7 to 0.9.
