Abstract
本发明属于试剂盒技术领域,尤其涉及一种神经元特异性烯醇化酶(NSE)测定试剂盒(磁微粒化学发光法)。包括:校准品、甲苯磺酰基磁珠偶联的神经元特异性烯醇化酶单克隆抗体、碱性磷酸酶偶联的神经元特异性烯醇化酶单克隆抗体。所述甲苯磺酰基磁珠偶联的神经元特异性烯醇化酶单克隆抗体的工作浓度为0.2‑1.0mg/ml。所述碱性磷酸酶偶联的神经元特异性烯醇化酶单克隆抗体的工作浓度为0.2‑2μg/ml。本发明神经元特异性烯醇化酶(NSE)测定试剂盒通过定向偶联的方法使得试剂盒批间差小、抗生物素干扰能力强、且线性范围大,成本低。

技术领域
本发明属于试剂盒技术领域,尤其涉及一种神经元特异性烯醇化酶(NSE)测定试剂盒(磁微粒化学发光法)。
背景技术
神经元特异性烯醇化酶(neuron-specific enolase,NSE)是烯醇化酶基因家族成员之一,烯醇化酶是由α、β、γ三个亚基组成的二聚体,其同工酶分为αα、ββ、γγ、αγ和αβ五种。α亚基主要存在于肝、肾等组织,故称为非神经系统的烯醇化酶(NNE);β亚基主要存在于骨骼肌和心肌,称为肌肉特异性的烯醇化酶(MSE);γ亚基主要存在于神经组织,γγ、αγ组成的同工酶为神经元和神经内分泌细胞特有,故命名为神经元特异性烯醇化酶。
血清和脑脊液中NSE的含量极少。近年来研究发现,在颅脑损伤后,部分神经元坏死、崩解,脑组织细胞膜的完整性受到破坏,可释放入细胞间隙和脑脊液中,同时血脑屏障的破坏和通透性改变,使某些蛋白成分易通过血脑屏障释放入血液和脑脊液中,导致颅脑损伤患者的血清和脑脊液中含量升高。与脑损伤程度一致,损伤越重死亡的神经元越多,致血脑屏障受损的程度越高,被释放入血及脑脊液的就越多。这为脑神经元损伤后检测的变化提供了一定的理论依据,并认为是评判颅脑损伤程度和预后的一项特异性敏感指标。
NSE被认为是监测小细胞支气管癌的首选标志物,60-81%小细胞支气管癌病例NSE浓度升高。尽管NSE浓度与转移部位或脑部转移没有相关性,但是与临床分期即疾病进展有很好的相关性。首个化疗周期开始后24小时NSE浓度有短暂的升高,原因是肿瘤细胞溶解。这种NSE浓度升高可持续1周或首个化疗周期结束时血清浓度迅速下降(治疗前浓度增加)。相反,对化疗无反应的患者NSE浓度持续升高或没有下降到参考范围。病情缓解期间,80-96%患者NSE浓度正常,而病情复发时NSE浓度升高。一些病例其1-4月潜伏期中NSE浓度升高,常为指数式升高(10-94天浓度翻倍),这与生存期有关。NSE可用于评估小细胞支气管癌患者的预后情况和疗效监测:诊断灵敏度93%,阳性预测值92%。
NSE在内分泌肿瘤中显示出很高的免疫活性,并且可以在血清中直接检出,因此可以用来监测肿瘤患者的病情进展,评价治疗效果。血清学检测比放射线检查、解剖学检查方便迅速,因此NSE作为肿瘤标记物在临床中的应用十分广泛。
国内现有的神经元特异性烯醇化酶(NSE)化学发光试剂盒与进口试剂盒比较,线性范围和灵敏度基本无差别。但是依然存在一些问题,导致国产试剂盒竞争力差,无法替代进口。例如批间差较大,供货不稳定,价格无优势,抗生物素干扰能力差等问题。目前行标对NSE试剂盒的批间差要求CV在15%以内,国内试剂盒的普遍水平在8%-10%之间。经研究发现批间差大的主要原因是由于NSE抗体标记碱性磷酸酶的过程具备很强的随机性,无法实现每次偶联都一致的情况。在免疫球蛋白抗体结构中有两个抗原结合位点,位于其高变区的N末端,也是其活性的重要组成部分。NSE抗体与抗原分子的结合只由重链与轻链中不连续氨基酸参与的非共价键引起,换句话说,结合位点不是严格的由现行的氨基酸序列组成,而是由他们独特的三级结构而成。这些结合位点通过范德华力、离子键、疏水键、氢键等与特定抗原进行高亲和力的结合。抗体分子具有一系列可用于偶联的官能团,如赖氨酸的ε-氨基或末端氨基,天冬氨酸与谷氨酸的游离羧基等。由于这些官能团在抗体三级结构中的均匀分布,使得偶联反应随机低发生在抗体结构的任何位点,抗体的随机取向导致抗原结合位点被隐藏甚至占据,这样就降低了抗体的抗原结合活性。因此抗体的定向偶联成为人们研究的重点。
而生物素又称维生素H或维生素B7,是一种含硫水溶性维生素,广泛分布于动物及植物组织,也可人工合成。生物素是一种维持人体自然生长、发育和人体机能健康必要的营养素。应用于人体保健和疾病治疗生物素具有多种功效,广泛应用于头发、指甲和皮肤保养,控制体重,孕期保健等;作为治疗性药物,在临床应用于遗传性疾病、多发性生物素辅酶酵素缺乏症、多发性硬化(multiple sclerosis,MS)、甲状腺功能亢进、2型糖尿病、维生素D中毒等。随着补充生物素用量和浓度的不断增加,循环血液中的生物素对免疫检测产生了干扰和影响。若待测样本中存在游离生物素,就会干扰生物素-链霉亲合素免疫分析系统中链霉亲合素捕获目标分析物的能力,从而造成检测结果的假性升高或者假性降低,影响临床判断。生物素对免疫非竞争法的干扰机制以双抗体夹心法为例,若待测样本中存在游离生物素,其与链霉亲合素结合后,阻止部分夹心复合物中生物素化抗体与链霉亲合素结合,造成捕获信号减弱,结果假性降低。美国食品药物管理局(FDA)发布警示信息,提醒公众、医务人员、实验室工作人员和实验室检测试剂开发人员,生物素会严重干扰某些实验室检测,得出不准确的检测结果,且不容易被发现。患者通过膳食补充剂摄入高剂量生物素后,其血液或其他样品中的生物素可导致临床上实验室检测结果不准确。近年由于生物素干扰实验室检测导致的不良事件报告数量有所增加,其中包括1例死亡,这已引起广泛重试。而体外诊断试剂盒的抗生物素干扰能力也成为评估试剂盒性能的重要指标。目前国内外的试剂盒为了扩大线性范等指标采用生物素和链霉亲和素作为偶联标记物,而这样导致的其试剂盒对生物素的抗干扰能力变差,随着病人体内的生物素升高,测试结果而变得不准确。
中国专利(公开号CN102914650B)中公开了一种用于神经元特异性烯醇化酶定量检测的试剂盒。该试剂盒包括校准品、磁分离试剂(生物素化抗体偶联链霉亲和素磁珠)、酶反应物(碱性磷酸酶标记的NSE抗体)、稳定增强剂(抗异嗜性抗体)。然而该试剂盒采用了生物素和链霉亲和素磁珠体系,抗生物素干扰能力差,抗体非定向偶联碱性磷酸酶,成本高且批间差较大。
又如中国专利(公开号CN109187973B)中公开了一种用于检测神经元特异性烯醇化酶的化学发光免疫试剂盒。试剂盒A:校准品、磁颗粒反应液Ra1(链霉亲和素磁珠)、示踪结合物反应液Ra2(吖啶酯标记的NSE抗体)和捕获抗体反应液Ra3(生物素标记的NSE抗体)。试剂盒B:校准品、磁颗粒反应液Ra1(NSE抗体包被的磁珠)、踪结合物反应液Ra2(吖啶酯标记的NSE抗体)、项目稀释反应液Ra3。试剂盒C:校准品、磁颗粒反应液Ra1(NSE抗体包被的磁珠)、踪结合物反应液Ra2(吖啶酯标记的NSE抗体)。该试剂盒的线性范围做到了0-500ng/ml、灵敏度做到了0.09ng/ml,然而该试剂盒存在生产批间差异较大的问题。
发明内容
本发明的目的是针对现有技术中存在的上述问题,提供一种批间差小、抗生物素干扰能力强、且线性范围大,成本低,重复性好的神经元特异性烯醇化酶(NSE)测定试剂盒。
本发明的目的可通过下列技术方案来实现:一种神经元特异性烯醇化酶(NSE)测定试剂盒(磁微粒化学发光法),所述试剂盒包括:校准品、甲苯磺酰基磁珠偶联的神经元特异性烯醇化酶单克隆抗体、碱性磷酸酶偶联的神经元特异性烯醇化酶单克隆抗体。
甲苯磺酰基磁珠偶联的神经元特异性烯醇化酶单克隆抗体的工作浓度为0.2-1.0mg/ml。
碱性磷酸酶偶联的神经元特异性烯醇化酶单克隆抗体的工作浓度为0.2-2μg/ml。工作浓度过大造成原料浪费,影响成本。浓度过小,反应度较低,影响试剂性能。
在上述神经元特异性烯醇化酶(NSE)测定试剂盒中,所述的甲苯磺酰基磁珠偶联神经元特异性烯醇化酶单克隆抗体的制备方法包括:在促进剂的作用下,甲苯磺酰基磁珠与NSE抗体反应,磁珠表面的甲苯磺酰基与NSE抗体的伯胺基共价结合;然后加入封闭剂封闭处理得甲苯磺酰基磁珠偶联神经元特异性烯醇化酶单克隆抗。在该步骤中促进剂的作用在于提高抗体和磁珠的偶联效率,封闭处理为封闭多余的位点,降低磁珠的本底。
作为优选,每10mg甲苯磺酰基磁珠中加入0.01-0.2mg抗体。
进一步优选,甲苯磺酰基磁珠的粒径为1.0μm-3.0μm。
作为优选,甲苯磺酰基磁珠与NSE抗体反应前先更换缓冲液,缓冲液pH 5.0~9.5,清洗1~5次。
作为优选,促进剂的加入量是甲苯磺酰基磁珠与NSE抗体总质量的0.5-2倍。
作为优选,封闭剂的加入量为每10mg甲苯磺酰基磁珠中加入0.05-0.25ml封闭剂。
作为优选,加入封闭剂前后均在24-42℃下混悬反应6-24h。
进一步优选,封闭剂包括BSA,CE210,CE510中的一种或者几种。
更进一步优选,所述的甲苯磺酰基磁珠偶联神经元特异性烯醇化酶单克隆抗体的制备方法具体如下:
1)将粒径为1.0~3.0μm的甲苯磺酰基磁性颗粒更换缓冲液,缓冲液pH 5.0~9.5,清洗1~5次。
2)加入包被抗体与磁性颗粒的比例为0.01~0.2mg抗体:10mg磁性颗粒。
3)加入0.1~3mol/L硫酸铵按照磁性颗粒与抗体体积总和的0.5~2倍加入。
4)24~42℃条件下混悬反应4~24小时。
5)按照每10mg磁性颗粒,加入0.05~0.25ml含有BSA,CE210,CE510中的一种或者几种的封闭剂,24~42℃条件下混悬反应6~24小时。
6)更换为磁珠保存液后备用,甲苯磺酰基磁珠偶联神经元特异性烯醇化酶单克隆抗体的终浓度为10mg/ml。
使用时,用磁珠保存液进行稀释,稀释至0.2-1.0mg/ml。
进一步优选,磁珠保存液通过如下方法制备:将0.595g HEPES,1gBSA,0.8g氯化钠,0.1g吐温20,0.2gProclin300,1g庆大霉素加入到容器中,加入100ml纯化水搅拌均匀。
在上述神经元特异性烯醇化酶(NSE)测定试剂盒中,碱性磷酸酶偶联的神经元特异性烯醇化酶单克隆抗体的制备方法包括:
在还原剂的作用下,打开NSE抗体重链之间的二硫键,暴露出巯基,得到第一产物;
在SMCC的作用下,碱性磷酸酶的伯胺基与SMCC分子一端的NHS酯基团形成酰胺键,得到第二产物;
将第一产物和第二产物混合,第二产物中SMCC的另外一端与第一产物中暴露出巯基的NSE单链抗体通过双键的加成反应链接,得碱性磷酸酶偶联的神经元特异性烯醇化酶单克隆抗体。
作为优选,按NSE抗体体积每毫升加入0.5‰~2.5‰还原剂。
作为优选,所述的还原剂为三(2-羰基乙基)磷盐酸盐(TCEP),还原剂使用浓度为0.5-2mol/L。
作为优选,第一产物和第二产物的质量比为(3-5):1。
作为优选,第一产物和第二产物混合后还包括加入氯化镁溶液。
进一步优选,氯化镁溶液加入的体积为反应总体积的1‰-5‰。
更进一步优选,所述碱性磷酸酶偶联的神经元特异性烯醇化酶单克隆抗体的制备方法具体如下:
1)、将1mg抗体用透析或PD-10脱盐柱更换为无氨基及巯基缓冲液,用浓缩管浓缩到2~4mg/ml;
2)按抗体体积每毫升加入0.5‰~2.5‰还原剂,室温反应10~30分钟,还原剂浓度为0.5-2mol/L;
3)加入与还原剂相同体积的1M pH7.3的甘氨酸,室温反应5~10min,使用PD-10脱盐柱更换缓冲液,浓缩到2~4mg/ml备用;
4)将0.25mg碱性磷酸酶用透析或PD-10脱盐柱更换为无氨基及巯基缓冲液,用浓缩管浓缩到2~4mg/ml;
5)称取SMCC,用DMF溶解到5~8mg/ml,按碱性磷酸酶体积2.5‰~7.5‰加入,室温反应10~30分钟;
6)加入与SMCC溶液相同体积的1M pH7.3的甘氨酸,室温反应10~20min,使用PD-10脱盐柱更换缓冲液,浓缩到2~4mg/ml备用;
7)将活化后的抗体和活化后的碱性磷酸酶混合,加入体积为反应总体积的1‰~5‰毫升的0.1~0.5M氯化镁溶液,2~8℃反应8~20小时,纯化后,2-8℃保存备用。
使用时,采用酶缓冲液对碱性磷酸酶偶联的神经元特异性烯醇化酶单克隆抗体进行稀释,稀释至0.2~2μg/ml。
酶缓冲液通过如下方法制得:将0.595g HEPES、1g BSA、0.8g氯化钠、0.5ml 1mol的氯化镁、0.1ml 0.1mol的氯化锌、0.2g Proclin300、1g庆大霉素加入到容器中,加入100ml纯化水搅拌均匀。
在上述神经元特异性烯醇化酶(NSE)测定试剂盒中,校准品稀释液包括HEPES、牛血清白蛋白、猪皮明胶、海藻糖、蔗糖、聚乙二醇20000、甲基纤维素、RPE2520、表面活性剂S7、Proclin300,庆大霉素。高蛋白浓度可防止神经元特异性烯醇化酶吸附,海藻糖和蔗糖能够干扰神经元特异性烯醇化酶的重折叠过程。聚乙二醇20000可以增加溶剂水表面张力而导致神经元特异性烯醇化酶优先水化,从而增加稳定性。甲基纤维素和猪皮明胶是很有效的增稠剂和稳定剂,可使神经元特异性烯醇化酶溶液能够形成均匀的悬浊液,保持活性的状态。两种防腐剂的加入大大低降了溶液张菌的几率。以此缓冲液配制的NSE校准品经过37℃加速破坏和2-8℃长期保存实验验证均可以稳定保存,提高了试剂盒的稳定性。
校准品的制备具体为:配制校准品稀释液:将0.595g HEPES,5g牛血清白蛋白,1g猪皮明胶,3g海藻糖,2g蔗糖,1g聚乙二醇20000,0.5g甲基纤维素,0.1g RPE2520,0.1g表面活性剂S7,0.2g Proclin300,1g庆大霉素,放入玻璃容器中,加入100ml纯化水搅拌均匀。用校准品稀释液将NSE抗原稀释到系列浓度。
本发明的神经元特异性烯醇化酶(NSE)定量检测试剂盒,利用全自动发光免疫分析仪(DXI800)对NSE校准品、进行检测,绘制标准曲线;然后测试血清样本,根据样本的发光值,计算出神经元特异性烯醇化酶的浓度。
使用本发明试剂盒定量检测神经元特异性烯醇化酶(NSE)的方法为,向仪器反应管中依次加入10μl样本,50μl的甲苯磺酰基磁珠偶联的神经元特异性烯醇化酶单克隆抗体工作液,50μl的碱性磷酸酶偶联的神经元特异性烯醇化酶单克隆抗体工作液,孵育10分钟,进行磁分离清洗,然后加入底物发光液,测定反应管中的发光值。仪器软件会根据内置曲线自动计算出对应的浓度值。所有反应均由全自动仪器自动操作,无需人工操作方便快捷。
与现有技术相比,本发明试剂盒具有如下优点:
1.本发明试剂盒碱性磷酸酶偶联神经元特异性烯醇化酶单克隆抗体的方法是,通过一定浓度的还原剂,打开NSE抗体两条重链间的二硫键,将抗体由两条重链和两条轻链的结构变成,一条轻链和一条重链的结构。而碱性磷酸酶通过SMCC定向地偶联在抗体的两条重链中间的位置。因为重链间的二硫键在铰链区,远离抗原结合位点,即不堵塞抗原结合位点,也有效地解决了偶联的随机性导致的试剂盒批间差大的问题。对比常规偶联,这种定向偶联的方法不会导致抗体聚合,抗体和酶的活性不易受到破坏,可进一步提高试剂盒的稳定性。本发明试剂盒37℃加速破坏9天和2-8℃保存15个月,降幅都可保持在10%以内。
2.本发明试剂盒通过定向偶联也解决了生产成本高的问题。碱性磷酸酶是试剂盒内的重要物料,价格昂贵。而碱性磷酸酶在偶联过程中的使用率制约着试剂盒的成本。常规偶联碱性磷酸酶的利用率只有不到30%,本发明采用SMCC定向偶联,碱性磷酸酶的利用率提高到了95%以上,大大降低了试剂盒的成本。

实施例1
一种神经元特异性烯醇化酶(NSE)测定试剂盒,包括:校准品、甲苯磺酰基磁珠偶联的神经元特异性烯醇化酶单克隆抗体、碱性磷酸酶偶联的神经元特异性烯醇化酶单克隆抗体。
配制校准品稀释液:将0.595g HEPES,5g牛血清白蛋白,1g猪皮明胶,3g海藻糖,2g蔗糖,1g聚乙二醇20000,0.5g甲基纤维素,0.1g RPE2520,0.1g表面活性剂S7,0.2gProclin300,1g庆大霉素,放入玻璃容器中,加入100ml纯化水搅拌均匀。用校准品稀释液将NSE抗原稀释到0、5、20、50、200、500ng/ml浓度。
所述的甲苯磺酰基磁珠偶联神经元特异性烯醇化酶单克隆抗体的通过如下方法制得:
1)将粒径为1.0μm的甲苯磺酰基磁性颗粒更换缓冲液,缓冲液pH 5.0,清洗3次。
2)加入包被抗体与磁性颗粒的比例为0.1mg抗体:10mg磁性颗粒。
3)加入3mol/L硫酸铵,按照磁性颗粒与抗体体积总和的0.5倍加入。
4)24℃条件下混悬反应18小时。
5)按照每10mg磁性颗粒,加入0.1ml CE210,24℃条件下混悬反应6小时。
6)更换为磁珠保存液后备用,甲苯磺酰基磁珠偶联神经元特异性烯醇化酶单克隆抗体的终浓度为10mg/ml。
7)使用时,用磁珠保存液进行稀释,稀释至0.25mg/ml。
8)磁珠保存液配制通过如下方法制得:将0.595g HEPES,1gBSA,0.8g氯化钠,0.1g吐温20,0.2gProclin300,1g庆大霉素加入到容器中,加入100ml纯化水搅拌均匀。
碱性磷酸酶偶联的神经元特异性烯醇化酶单克隆抗体通过如下方法制备:
1)、将1mg抗体用PD-10脱盐柱更换为0.1mol的三乙醇胺缓冲液,用浓缩管浓缩到2~4mg/ml。
2)按抗体体积每毫升加入1‰浓度为1mol/L的TCEP,室温反应30分钟。
3)加入与还原剂相同体积的1M pH7.3的甘氨酸,室温反应10min,浓缩到4mg/ml备用。
4)将0.25mg碱性磷酸酶用PD-10脱盐柱更换为0.05mol的三乙醇胺缓冲液,用浓缩管浓缩到2mg/ml。
5)称取SMCC,用DMF溶解到5mg/ml,按碱性磷酸酶体积2‰加入,室温反应30分钟。
6)加入与SMCC溶液相同体积的1M pH7.3的甘氨酸,室温反应20min,浓缩到2~4mg/ml备用。
7)将活化后的抗体和活化后的碱性磷酸酶4:1质量比混合,加入体积为反应总体积的1‰的0.1M氯化镁溶液,2~8℃反应12小时,纯化后,2-8℃保存备用。
8)使用时,采用酶缓冲液对碱性磷酸酶偶联的神经元特异性烯醇化酶单克隆抗体进行稀释,稀释至0.5μg/ml。
其中,酶缓冲液通过如下方法制得:将0.595g HEPES、1g BSA、0.8g氯化钠、0.5ml1mol的氯化镁、0.1ml 0.1mol的氯化锌、0.2g Proclin300、1g庆大霉素加入到容器中,加入100ml纯化水搅拌均匀。
本发明的神经元特异性烯醇化酶(NSE)定量检测试剂盒,利用全自动发光免疫分析仪(DXI800)对NSE校准品、进行检测,绘制标准曲线;然后测试血清样本,根据样本的发光值,计算出神经元特异性烯醇化酶的浓度。
使用本发明实施例1试剂盒定量检测神经元特异性烯醇化酶(NSE)的方法为,向仪器反应管中依次加入10μl样本,50μl的甲苯磺酰基磁珠偶联的神经元特异性烯醇化酶单克隆抗体工作液,50μl的碱性磷酸酶偶联的神经元特异性烯醇化酶单克隆抗体工作液,孵育10分钟,进行磁分离清洗,然后加入底物发光液,测定反应管中的发光值。仪器软件会根据内置曲线自动计算出对应的浓度值。所有反应均由全自动仪器自动操作,无需人工操作方便快捷。
具体实验内容如下:
1.重复性:使用混合人血清样本1、样本2、样本3(三支样本分别为低、中、高值样本)和肿瘤通用伯乐质控品测试重复性,每例血清和质控测试连续测试10次,得到结果如表1所示。

从表1可以得出,本发明的神经元特异性烯醇化酶(NSE)测定试剂盒批内重复性可控制在2%以内。
2.天间精密度:使用混合人血清样本1、样本2、样本3(三支样本分别为低、中、高值样本)和肿瘤通用伯乐质控品测试天间精密度,样本分装冻存,每天早、中、晚各取一只测试一次,连续测试7天,每个样本共得到21个数据,得到结果如表2所示。

从表2可以得出,本发明的神经元特异性烯醇化酶(NSE)测定试剂盒天间精密度可控制在3%以内。
3.热加速稳定性:将试剂盒处于37℃加速破坏的环境,在第3天,第7天和第14天分别取出,测校准品发光值,得到结果如表3所示。

从表3可以得出,本发明的神经元特异性烯醇化酶(NSE)测定试剂盒在37℃加速破坏14天,降幅可控制在10%以内。
4.长期稳定性:将试剂盒处于4℃冰箱保存,在第3个月,第7个月,第12个月,第15个月分别取出测试校准品发光值,得到结果如表4所示

从表4可以得出,本发明的神经元特异性烯醇化酶(NSE)测定试剂盒在4℃冰箱保持15个月,降幅可控制在10%以内。
5.干扰样本测试,将NSE的样本分成三份,不加生物素,加入100ng/ml浓度的生物素,500ng/ml浓度的生物素。将三份样本分别用本发明试剂盒与一种进口罗氏生物素体系试剂盒及一种国产博奥赛斯生物素体系试剂盒做样本的添加回收,得到回收率结果如表5所示。

从表5可以得出,本发明的神经元特异性烯醇化酶(NSE)测定试剂盒抗生物素干扰强同比增加,测试结果更加准确。
6.批间差测试:连续偶联4个批次的甲苯磺酰基磁珠-神经元特异性烯醇化酶单克隆抗体、碱性磷酸酶-神经元特异性烯醇化酶单克隆抗体,生产成4个批次的试剂盒,使用混合人血清样本1、样本2、样本3(三支样本分别为低、中、高值样本)和肿瘤通用伯乐质控品测试,结果如表6所示。

从表6可以得知,本发明的神经元特异性烯醇化酶(NSE)测定试剂盒批间差小,供货更加稳定。
7.取100例血清样本,比较本发明实施例1的试剂盒与罗氏公司神经元特异性烯醇化酶试剂盒检测结果的相关性,样本测试相关性测试结果如图1所示。从图1中的结果显示,两个试剂线性回归方程Y=1.0136X+1.4965,相关系数R2=0.9886。可见本发明的神经元特异性烯醇化酶(NSE)测定试剂盒检测样本与罗氏公司产品结果基本一致,两者相关性良好。
综上所述,本发明神经元特异性烯醇化酶(NSE)测定试剂盒通过定向偶联的方法使得试剂盒批间差小、抗生物素干扰能力强、且线性范围大,成本低。
本处实施例对本发明要求保护的技术范围中点值未穷尽之处以及在实施例技术方案中对单个或者多个技术特征的同等替换所形成的新的技术方案,同样都在本发明要求保护的范围内,并且本发明方案所有涉及的参数间如未特别说明,则相互之间不存在不可替换的唯一性组合。
本文中所描述的具体实施例仅仅是对本发明精神作举例说明。本发明所属技术领域的技术人员可以对所描述的具体实施例做各种修改或补充或采用类似的方式替代,但并不会偏离本发明的精神或者超越所附权利要求书所定义的范围。
尽管对本发明已作出了详细的说明并引证了一些具体实施例,但是对本领域熟练技术人员来说,只要不离开本发明的精神和范围可作各种变化或修正是显然的。
Claims (10)
1.一种神经元特异性烯醇化酶(NSE)测定试剂盒,其特征在于,所述试剂盒包括:校准品、甲苯磺酰基磁珠偶联的神经元特异性烯醇化酶单克隆抗体、碱性磷酸酶偶联的神经元特异性烯醇化酶单克隆抗体。
2.根据权利要求1所述的神经元特异性烯醇化酶(NSE)测定试剂盒,其特征在于,所述甲苯磺酰基磁珠偶联的神经元特异性烯醇化酶单克隆抗体的工作浓度为0.2-1.0mg/ml。
3.根据权利要求1所述的神经元特异性烯醇化酶(NSE)测定试剂盒,其特征在于,所述碱性磷酸酶偶联的神经元特异性烯醇化酶单克隆抗体的工作浓度为0.2-2μg/ml。
4.根据权利要求1所述的神经元特异性烯醇化酶(NSE)测定试剂盒,其特征在于,所述的甲苯磺酰基磁珠偶联神经元特异性烯醇化酶单克隆抗体的制备方法包括:在促进剂的作用下,甲苯磺酰基磁珠与NSE抗体反应,磁珠表面的甲苯磺酰基与NSE抗体的伯胺基共价结合;然后加入封闭剂封闭处理得甲苯磺酰基磁珠偶联神经元特异性烯醇化酶单克隆抗。
5.根据权利要求4所述的神经元特异性烯醇化酶(NSE)测定试剂盒,其特征在于,每10mg甲苯磺酰基磁珠中加入0.01-0.2mg抗体。
6.根据权利要求4所述的神经元特异性烯醇化酶(NSE)测定试剂盒,其特征在于,所述促进剂的加入量是甲苯磺酰基磁珠与NSE抗体总质量的0.5-2倍。
7.根据权利要求4所述的神经元特异性烯醇化酶(NSE)测定试剂盒,其特征在于,所述封闭剂的加入量为每10mg甲苯磺酰基磁珠中加入0.05-0.25ml封闭剂。
8.根据权利要求1所述的神经元特异性烯醇化酶(NSE)测定试剂盒,其特征在于,所述碱性磷酸酶偶联的神经元特异性烯醇化酶单克隆抗体的制备方法包括:
在还原剂的作用下,打开NSE抗体重链之间的二硫键,暴露出巯基,得到第一产物;
在SMCC的作用下,碱性磷酸酶的伯胺基与SMCC分子一端的NHS酯基团形成酰胺键,得到第二产物;
将第一产物和第二产物混合,第二产物中SMCC的另外一端与第一产物中暴露出巯基的NSE单链抗体通过双键的加成反应链接,得碱性磷酸酶偶联的神经元特异性烯醇化酶单克隆抗体。
9.根据权利要求8所述的神经元特异性烯醇化酶(NSE)测定试剂盒,其特征在于,按NSE抗体体积每毫升加入0.5‰~2.5‰还原剂。
10.根据权利要求8所述的神经元特异性烯醇化酶(NSE)测定试剂盒,其特征在于,第一产物和第二产物的质量比为(3-5):1。
