本发明涉及护肤品技术领域,具体涉及一种蔷薇花活性提取物及其应用。具体技术方案为:一种蔷薇花活性提取物,所述活性提取物中含有的活性成分为GeminA,和/或,GeminB,所述Gemin A的质量百分含量≥4.0%,和/或,所述Gemin B的质量百分含量≥0.8%。其活性成分具有良好的酪氨酸酶抑制活性,可以在美白护肤品和果蔬保鲜剂中作为酪氨酸酶抑制剂使用;也可以在酪氨酸酶抑制剂中加入Gemin A,和/或,Gemin B或多苞蔷薇花作为其活性成分使用。

本发明涉及护肤品技术领域,具体涉及一种蔷薇花活性提取物及其应用。
背景技术
众所周知,过多的紫外线辐射会导致人体产生大量的自由基和活性氧,进而造成机体的氧化损伤。人体内存在过多的活性氧会促进酪氨酸酶催化L-酪氨酸发生一系列复杂反应最终形成黑色素,适量的黑色素能保护人身安全,但在人体内大量累积则会引起色素斑及恶性黑色素瘤等一系列病症。因此,酪氨酸酶抑制剂在医药和美白化妆品等领域都有很好的应用潜力。
当前,随着物质生活水平的不断提高,人们更加注重皮肤的保养,对化妆品的功效提出更高的要求,如何安全地美白肌肤已成为人们基础护肤获得满足后更深层次的需求。目前,市场上广泛使用的黑色素生成抑制剂有龙胆酸、熊果苷、氢醌等,但都表现出一定的副作用。比如:长期使用氢醌可能会引起褐黄病和皮肤真皮弹性纤维变性,而熊果苷使用不当不仅不会抑制黑色素的产生,反而会增加黑色素的生成。所以,寻找高效安全的天然来源的酪氨酸酶抑制剂作为美白护肤品活性添加剂已成为化妆品行业发展趋势。
同时,酪氨酸酶又被称为多酚氧化酶,广泛存在于各种水果和蔬菜中,是导致果蔬发生酶促褐变的主要酶类,严重影响果蔬的风味、营养和外观。而酪氨酸酶抑制剂可以有效地防止果蔬褐变,是水果保鲜的重要手段之一。酪氨酸酶还是各种微生物生命活动所必需的酶,抑制酪氨酸酶的活性还能抑制腐烂菌的生长,达到保鲜防腐的目的,且不影响食品的风味及口感。早期,含硫化合物被广泛应用于食品保鲜领域,但因硫残留对人体健康造成一定的负面影响而被限制使用。现在,在食品保鲜中被广泛使用的酪氨酸酶抑制剂有半胱氨酸、抗坏血酸和柠檬酸等,安全性较高。
因此,寻找新的具有潜在价值的植物原料并从中提取具有酪氨酸酶抑制作用的活性成分对开发新的安全、高效的美白护肤品和果蔬保鲜剂具有重要的意义。
发明内容
针对现有技术的不足,本发明提供了一种蔷薇花活性提取物,具有优良的酪氨酸酶抑制活性,可用于美白护肤品,和/或,果蔬保鲜剂添加剂。
为实现以上目的,本发明通过以下技术方案予以实现:
本发明公开了一种蔷薇花活性提取物,所述活性提取物中含有的活性成分为Gemin A,和/或,Gemin B,所述Gemin A的质量百分含量≥ 4.0%,和/或,所述Gemin B的质量百分含量≥0.8%。

优选的,将蔷薇花浸膏溶于水中,通过大孔吸附树脂,先以纯水洗脱5~10个柱体积,再以体积分数为20~30%的乙醇溶液进行洗脱5~ 10个柱体积,然后将洗脱液在40~50℃下进行减压浓缩去除乙醇后即得活性提取物。

本发明从多苞蔷薇花中提取出一种多苞蔷薇花活性提取物,其中活性成分GeminA和Gemin B的质量分数分别不低于4.0%和0.8%。同时,该多苞蔷薇花活性成分具有良好的酪氨酸酶抑制活性,可以在美白护肤品和果蔬保鲜剂中作为酪氨酸酶抑制剂使用;也可以在酪氨酸酶抑制剂中加入Gemin A,和/或,Gemin B或多苞蔷薇花作为其活性成分使用。本发明制备的多苞蔷薇花活性提取物为天然来源,具有安全无毒、无致畸、无致癌、无致敏性等优点,在酪氨酸酶抑制活性测试中,多苞蔷薇花活性提取物和活性成分Gemin A在60μg/mL的测试浓度下,对酪氨酸酶的抑制率大于50%。
具体实施方式
下面将结合本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
若未特别指明,实施举例中所用的技术手段为本领域技术人员所熟知的常规手段。
实施例1不同蔷薇花粗提物的制备及化学成分分析
取各蔷薇花(在野外采集的川滇蔷薇、绢毛蔷薇、峨嵋蔷薇、西北蔷薇、多苞蔷薇和黄蔷薇)的干燥花瓣各50g,粉碎后置于不同三角瓶中,分别加入500mL 50%乙醇水溶液,于50℃条件下提取10h,过滤得各蔷薇花提取液,提取液分别在50℃条件下减压浓缩得各蔷薇花粗提物,依次命名为CDQWH-1A,JMQWH-1A,EMQWH-1A,XBQWH-1A,DBQWH-1A和 HQWH-1A。以分离获得的Gemin A和Gemin B作为对照品,采用反相液相色谱对上述6种蔷薇花粗提物进行分析,多苞蔷薇花粗提物中Gemin A 和Gemin B的质量分数依次为1.1%和0.2%,其它5种蔷薇花均未检测到Gemin A和Gemin B。
实施例2
将50g多苞蔷薇花粉碎,以10倍量50%乙醇水溶液在50℃条件下浸泡10小时,共浸泡3次,合并提取液,在50℃条件下减压浓缩干燥得浸膏16.8g。浸膏悬浮于水中,先以石油醚-乙酸乙酯(体积比1:1) 混合溶剂萃取3次脱除其中的脂溶性成分,再以正丁醇萃取3次,合并正丁醇萃取液,在50℃条件下减压浓缩得正丁醇萃取物5.1g。正丁醇萃取物溶于水中并通过HPD-100A大孔吸附树脂,以纯水洗脱10个柱体积,然后以25%乙醇水溶液洗脱10个柱体积,合并25%乙醇水溶液的洗脱液,在50℃条件下减压浓缩得富含目标活性成分的活性提取物2.3g;将该活性提取物进行反相C18制备色谱(色谱柱规格为5×25cm,乙腈:水=14:86(体积比),流速为50mL/min,检测波长为280nm)分离得目标化合物Gemin A(153mg)和Gemin B(38mg),化学结构分别如下所示:

实施例3
将200g多苞蔷薇花粉碎,以10倍量35%乙醇水溶液在50℃条件下提取10小时,提取3次,合并提取液,在40℃条件下减压浓缩得浸膏62g。将该浸膏溶于2L纯水中,通过HPD-100A大孔吸附树脂,先以纯水洗脱8个柱体积,然后以30%乙醇水溶液洗脱10个柱体积,合并 30%乙醇洗脱液,在45℃条件下减压浓缩至溶液无明显醇味,将溶液冷冻后低温真空干燥得9.1g活性提取物(编号DBQWH-1)。以分离获得的 Gemin A和Gemin B作为对照品,采用反相液相色谱对该活性提取物进行分析,Gemin A的质量分数为5.2%,Gemin B的质量分数为1.1%。
实施例4
将200g多苞蔷薇花粉碎,以8倍量50%乙醇水溶液在40℃条件下提取10小时,提取3次,合并提取液,在50℃条件下减压浓缩得浸膏 58g。将该浸膏溶于2L纯水中,通过HPD-100A大孔吸附树脂,先以纯水洗脱5个柱体积,然后以20%乙醇水溶液洗脱10个柱体积,合并20%乙醇洗脱液,在40℃条件下减压浓缩至溶液无明显醇味,将溶液冷冻后低温真空干燥得8.8g活性提取物(编号DBQWH-2)。以分离获得的Gemin A和Gemin B作为对照品,采用反相液相色谱对该活性提取物进行分析, Gemin A的质量分数为4.8%,Gemin B的质量分数为1.0%。
实施例5不同蔷薇花提取物的酪氨酸酶抑制活性
实验材料:实施例1制备的蔷薇花粗提物、实施例3和实施例4制备的活性提取物。
实验方法:将待测试样品与左旋多巴(终浓度1.25mM)混合,然后加入酪氨酸酶(终浓度25U/mL)开始反应,设定3个重复孔,设置阳性对照(α-熊果苷),在室温下反应5分钟,然后用酶标仪测定波长为490nm处的OD值。酪氨酸酶抑制率的计算公式如下:
酪氨酸酶抑制率(%)=(1-样品OD490nm/实验对照孔OD490nm)×100%
具体结果见下表1所示,结果显示多苞蔷薇花提取物活性优异,在同等质量浓度下,其活性优于阳性对照α-熊果苷,而其它5种蔷薇花提取物活性一般或较差。通过进一步分离分析发现多苞蔷薇花活性提取物富含鞣质类化合物,而其它几种蔷薇花提取物主要含有黄酮类成分,几乎未见鞣质类化合物。活性实验表明多苞蔷薇花中鞣质类化合物酪氨酸酶抑制活性好,为活性成分。

Claims (8)
2.根据权利要求1所述的一种蔷薇花活性提取物,其特征在于:所述蔷薇花为多苞蔷薇花。
3.一种酪氨酸酶抑制剂,其特征在于:含有多苞蔷薇花或其活性提取物中的活性成分,所述活性成分包括权利要求1~2任一项所述的蔷薇花活性提取物。
4.一种果蔬保鲜剂,其特征在于:含有多苞蔷薇花或其活性提取物中的活性成分,所述活性成分包括权利要求1~2任一项所述的蔷薇花活性提取物。
5.一种如权利要求1~2任一项所述的蔷薇花活性提取物的提取方法,其特征在于:采用乙醇提取和减压浓缩的方式提取得到蔷薇花浸膏;然后将蔷薇花浸膏经过大孔吸附树脂进行洗脱后,减压浓缩得到含有活性成分的活性提取物;再将活性提取物进行反相制备色谱分离得到蔷薇花活性成分。
6.根据权利要求5所述的一种蔷薇花活性提取物的提取方法,其特征在于:将蔷薇花浸膏溶于水中,通过大孔吸附树脂,先以纯水洗脱5~10个柱体积,再以体积分数为20~30%的乙醇溶液进行洗脱5~10个柱体积,然后将洗脱液在40~50℃下进行减压浓缩后即得活性提取物。
7.一种蔷薇花活性提取物的应用,其特征在于:包括权利要求1~2任一项所述的蔷薇花活性提取物或权利要求5~6任一项所述的提取方法所提取的蔷薇花活性提取物,所述GeminA,和/或,GeminB作为酪氨酸酶抑制剂在美白护肤品中的应用。
8.一种蔷薇花活性提取物的应用,其特征在于:包括权利要求1~2任一项所述的蔷薇花活性提取物或权利要求5~6任一项所述的提取方法所提取的蔷薇花活性提取物,所述GeminA,和/或,GeminB作为酪氨酸酶抑制剂在果蔬保鲜剂中的应用。
