Abstract

提供了一种RNA建库方法及其专用试剂盒。制备RNA文库的方法，包括如下步骤：提取RNA，进行打断；3'末端加尾；5'末端加接头，并与DNA探针混合物杂交；DNA探针混合物由若干条与预期去除的RNA反向互补的DNA探针组成；去除杂交链中的RNA并去除DNA；进行逆转录和PCR扩增，获得文库溶液。本发明结合了加polyA和5'Ligation。通过加polyA可以快速增加体系里RNA的含量，从而避免后续纯化损失。
