Abstract
提供了一种RNA建库方法及其专用试剂盒。制备RNA文库的方法,包括如下步骤:提取RNA,进行打断;3'末端加尾;5'末端加接头,并与DNA探针混合物杂交;DNA探针混合物由若干条与预期去除的RNA反向互补的DNA探针组成;去除杂交链中的RNA并去除DNA;进行逆转录和PCR扩增,获得文库溶液。本发明结合了加polyA和5'Ligation。通过加polyA可以快速增加体系里RNA的含量,从而避免后续纯化损失。
