Abstract
本发明公开了一种缓解肌肉流失的组合物及其制备方法,所述组合物包括以下重量百分比原料:动物蛋白40‑60%、植物蛋白20‑40%、β‑羟基‑β‑甲基丁酸盐HMB2‑5%、姜黄2‑5%、营养酵母粉0.1‑1.5%、表没食子儿茶素没食子酸酯EGCG0.5‑2.5%、维生素D0.0001‑0.001%、调味剂5‑15%和增稠剂1.5‑2.5%。本发明采用配比合理的蛋白质、HMB、抗氧化剂及维生素D,可针对老年人群和运动人群代谢特点及营养需求,特异性的补充肌肉营养,缓解肌肉流失;配方中经过调味剂调味处理并分装成小袋包装,易携带、口感细腻、甜度适中、适合目标人群长期服用。

技术领域:
本发明涉及缓解肌肉流失的组合物技术领域,尤其涉及一种缓解肌肉流失的组合物及其制备方法。
背景技术:
人体的肌肉有三种,即骨骼肌、心肌和平滑肌。心肌存在于心脏,平滑肌存在于内脏与血管,二者运动都不受自主控制、质量在成人中变化也不大。通常所指肌肉不包括心肌与平滑肌,单指骨骼肌。
肌肉在全身蛋白代谢中起核心作用,正常人的肌肉占体重的35%-45%。所谓肌肉流失,是指一段时间内,肌肉蛋白质分解多于合成,使得肌纤维的质量与大小,处于不断减少过程中。流行病学研究显示,人体的肌肉量随年龄的增长而不断地衰减。50岁以后,肌肉量平均每年减少1%-2%,60岁以后慢性肌肉丢失约30%,80岁后约丢失50%,当肌肉减少30%将影响肌肉的正常功能,导致走路不稳、行走困难,甚至跌倒、骨折。一旦骨折长期卧床,又会加重肌肉流失、骨量下降,严重影响生活质量。
肌肉衰减综合征就是一种伴随年龄增长的进行性骨骼肌量减少、伴有肌力或肌肉功能减退的综合性退行性病症。研究发现,肌肉衰减综合征是骨质疏松、骨关节炎等疾病发展的重要因素之一,肌肉衰减综合征已严重影响中老年人正常生活,会导致老年人丧失自理能力、伤残、住院;并且还增加罹患糖尿病、关节炎、骨质疏松症、心脏病等疾病的危险,最终使病死率增加。肌肉衰减综合症病因和发病机制十分复杂,目前认为与衰老、营养不良和缺少运动等相关,运动疗法和营养干预是目前公认防治肌肉衰减综合证的最有效手段。研究表明,经常从事运动的老年人较久坐活动量少的老年人有更为健康的骨骼肌。然而,老年肌肉衰减综合征患者多伴有其他慢性病,如心脑血管病、骨质疏松、关节退行性病变等,活动能力及强度大大受限。营养干预是防治肌肉衰减综合征的又一重要手段。
除了肌肉衰减综合征引起的肌肉流失外,日常生活中,还有一类人群特别担心肌肉流失,这类人群就是运动人群,特别是运动健身增肌人群。健身人群都知道增肌是一个非常难的过程,但其实保持肌肉更加困难,因为在不经意间辛苦训练的肌肉就会偷偷流失,有的人即使体重没有变化,但是整个人却不知不觉中从“肌肉型”变为“脂肪型”。所以对于健身人群来说,如何预防和缓解肌肉流失是非常重要的事情,这不仅需要合理的锻炼,更需要合理的营养补充。
研究发现,营养素缺乏,特别是蛋白质摄入不足,可导致肌肉蛋白合成受阻,加速肌肉流失发展进程。足够的能量、蛋白质可促进肌肉蛋白质合成,并抑制其分解,从而增加肌肉质量。对于老年人,国际老年人膳食蛋白质需求研究小组建议,老年人每餐应摄入25~30g优质蛋白;老年慢性病患者在此基础上应适当增加摄入量。还应合理分配三餐蛋白质摄入量,以更好地促进肌肉蛋白的合成。而对于运动健身人群相对普通人而言,摄入的蛋白质量更高,主要因为以下四个方面:一是运动员拥有更高比例的去脂体重;二是剧烈运动会导致尿液中丢失的蛋白质更多;三是训练过程中,来自蛋白质的氨基酸供能占5%-15%,肌糖原消耗增多时,蛋白质供能会增加;四是运动会造成肌肉损伤,修复损伤组织需要更多的蛋白质。普通人为维持氮平衡所需要的蛋白质的摄入量大约为每日0.8g/kg,而运动人群需要的蛋白质摄入量是每日1.2~1.7g/kg。
亮氨酸被认为是吸收多种不同营养素所必需的,它可以通过加强肌肉蛋白质生产,减缓肌肉组织退化。β-羟基-β-甲基丁酸盐(HMB)是亮氨酸的代谢中间产物,人体大约每天可以合成0.25-1g左右的HMB。大量研究表明,直接补充HMB可抵抗肌肉蛋白质分解,增加蛋白质合成,减少体脂,具有明显的增肌强力作用。HMB与其它氨基酸配合补充,在医疗上已经得到应用。有训练史者服用HMB同样对力量增长有促进作用,许多健美运动员和力量型运动员,通过补充HMB使骨骼肌增大,提高运动成绩。
老年人体内氧化应激损伤可致骨骼肌代谢异常,最终导致老年人肌肉功能紊乱;补充适量的抗氧化剂可助老年人维持内环境稳定和线粒体功能,从而预防肌肉衰减综合征的发生和发展。大量运动和高强度运动也会导致体内氧化自由基成倍产生,同时高强度运动也消耗体内抗自由基物质,进而加剧集体氧化自由基的蓄积。因此,大运动量和高强度训练后,适当补充抗自由基物质可以减轻氧化自由基对身体功能的损害。
维生素D为固醇类衍生物,又称钙化醇、麦角甾醇、麦角骨化醇、“阳光维生素”、抗佝偻病维生素,主要用于维持骨骼的强壮,并通过不同途径影响骨骼肌代谢。肌细胞和神经系统细胞均有特异性维生素D受体,肌肉组织维生素D受体的表达随年龄增长而显著降低;同时随着年龄增长,肌肉强度和神经肌肉功能降低,与肌细胞中维生素D受体表达减少和受体周围维生素D缺乏直接相关。肌肉无力常是维生素D缺乏的表现,补充活性维生素可促进肌肉功能,使快肌Ⅱ型肌肉纤维数目与直径显著增加,显著增强肌肉力量和功能。补充维生素D对增加肌肉衰减综合征患者肌肉量十分重要。低维生素D水平的老年人,患肌肉衰减的风险率是正常维生素D水平者的5倍。
现有的技术中,有一些专门针对缓解肌肉流失和肌肉衰减综合征人群的药品和保健食品,但根据专利文献及非专利文献,还未发现同时根据老年人群和运动人群代谢特点和营养需求,综合补充蛋白质、β-羟基-β-甲基丁酸盐(HMB)、抗氧化剂及维生素D的固体饮料。

发明内容:
本发明的目的在于提供一种缓解肌肉流失的组合物及其制备方法,从而克服上述现有技术中的缺陷。
为实现上述目的,本发明提供了一种缓解肌肉流失的组合物,包括以下重量百分比原料:动物蛋白40-60%、植物蛋白20-40%、β-羟基-β-甲基丁酸盐HMB2-5%、姜黄2-5%、营养酵母粉0.1-1.5%、表没食子儿茶素没食子酸酯EGCG0.5-2.5%、维生素D0.0001-0.001%、调味剂5-15%和增稠剂1.5-2.5%。

所述动物蛋白选用乳清蛋白、牛奶蛋白、分离乳清蛋白、肉类蛋白、鱼类蛋白或禽类蛋白。
所述植物蛋白选用大豆蛋白、豌豆蛋白、小麦蛋白、大米蛋白、燕麦蛋白、花生蛋白、玉米蛋白或油菜蛋白。
一种缓解肌肉流失的组合物的制备方法,包括以下步骤:
(1)按照以下重量百分比称取原料备用:动物蛋白40-60%、植物蛋白20-40%、β-羟基-β-甲基丁酸盐HMB2-5%、姜黄2-5%、营养酵母粉0.1-1.5%、表没食子儿茶素没食子酸酯EGCG0.5-2.5%、维生素D0.0001-0.001%、调味剂5-15%和增稠剂1.5-2.5%;
(2)将步骤(1)的原料分别经过20-40目筛网过筛处理后备用;
(3)将步骤(2)过筛后的物料分多次进行初步混合;
(4)将初步混合的物料投入到混合机内继续混合,直至总混合物料无肉眼可见杂质、抱团等异常情况;
(5)将步骤(4)混合后的物料再次经过20目筛网过筛处理;
(6)将过筛后的物料分装成20g/袋的小包装即可。
所述步骤(1)-(6)均在D级洁净生产区、环境温度18-25℃、相对湿度RH≤50%条件下进行。
所述步骤(4)混合机的混合频率调节为10-20r/min,混合30-40分钟。
所述分装时选用铝箔复合膜或者聚乙烯药品包装用复合膜进行包装。
具体的原料中β-羟基-β-甲基丁酸盐HMB是人体内氨基酸正常代谢的产物。在力量运动时进行补充可减少肌肉蛋白质的分解,并可增加肌肉体积,增强肌力;在耐力运动时可促进脂肪分解和保护细胞膜,有助于提高耐力和降低体脂;
姜黄为姜科植物姜黄Curcuma longa L.的干燥根茎,冬季茎叶枯萎时采挖,洗净,煮或蒸至透心,晒干,除去须根,具有抗氧化、降血脂、抗肿瘤、抗炎、抗病原微生物、利胆、终止妊娠及光效应等作用;
营养酵母粉是理想的营养源,它含有丰富的蛋白质、必需氨基酸、B族维生素、矿物质和膳食纤维,而糖、胆固醇和脂肪比较少,因此非常适合现代人食用;
表没食子儿茶素没食子酸酯EGCG分子式为C22H18O11,是绿茶茶多酚的主要组成成分,是从茶叶中分离得到的儿茶素类单体,具有抗菌、抗病毒、抗氧化、抗动脉硬化、抗血栓形成、抗血管增生、抗炎以及抗肿瘤作用;
维生素D(简称VD)是一种脂溶性维生素,乃环戊烷多氢菲类化合物,一组结构上与固醇有关,维生素D的主要功用是促进小肠粘膜细胞对钙和磷的吸收。肠中钙离子吸收需要一种钙结合蛋白,1,25-二羟基维生素D3可诱导此蛋白合成,促进Ca2+吸收,又可促进钙盐的更新及新骨生成,也促进磷吸收与肾小管细胞对钙、磷的重吸收,故可提高血钙、血磷浓度,有利于新骨生成和钙化;
调味剂是指改善食品的感官性质,使食品更加美味可口,并能促进消化液的分泌和增进食欲的食品添加剂。食品中加入一定的调味剂,不仅可以改善食品的感观性,使食品更加可口,而且有些调味剂还具有一定的营养价值。调味剂的种类很多,主要包括咸味剂(主要是食盐)、甜味剂(主要是糖、糖精等)、鲜味剂、酸味剂及辛香剂等;
增稠剂又称胶凝剂,是一种能增加胶乳、液体黏度的物质,用于食品时又称糊料。增稠剂可以提高物系黏度,使物系保持均匀稳定的悬浮状态或乳浊状态,或形成凝胶;大多数增稠剂兼具乳化作用。可分为天然和合成两大类。天然品大多数从含多糖类黏性物质的植物和海藻类制取,如淀粉、阿拉伯胶、果胶、琼脂、明胶、海藻胶、角叉胶、糊精等,通用明胶、可溶性淀粉、多糖衍生物等可用于化妆品;合成品有羧甲基纤维素、丙二醇藻蛋白酸酯、甲基纤维素、淀粉磷酸钠、羧甲基纤维素钠、藻蛋白酸钠、酪蛋白、聚丙烯酸钠、聚氧乙烯、聚乙烯吡咯烷酮等。
与现有技术相比,本发明的一个方面具有如下有益效果:
本发明采用配比合理的蛋白质、HMB、抗氧化剂及维生素D,可针对老年人群和运动人群代谢特点及营养需求,特异性的补充肌肉营养,缓解肌肉流失;配方中经过调味剂调味处理并分装成小袋包装,易携带、口感细腻、甜度适中、适合目标人群长期服用;制备过程中经过多次过筛、混合,制得的粉剂更均匀细腻;此外生产过程中均在D级洁净生产区,并在恒温很湿的条件下进行,可保证产品的洁净性。

实施例1:
一种缓解肌肉流失的组合物,包括以下重量百分比原料:动物蛋白50%、植物蛋白30%、β-羟基-β-甲基丁酸盐HMB3%、姜黄3%、营养酵母粉0.5%、表没食子儿茶素没食子酸酯EGCG1.5%、维生素D0.0005%、调味剂10%和增稠剂1.9995%。
所述动物蛋白选用乳清蛋白、牛奶蛋白、分离乳清蛋白、肉类蛋白、鱼类蛋白或禽类蛋白。

(5)将步骤(4)混合后的物料再次经过20目筛网过筛处理;
(6)将过筛后的物料分装成20g/袋的小包装即可。
所述步骤(1)-(6)均在D级洁净生产区、环境温度18-25℃、相对湿度RH≤50%条件下进行。

与实施例1不同的是,一种缓解肌肉流失的组合物,包括以下重量百分比原料:动物蛋白50%、植物蛋白30%、β-羟基-β-甲基丁酸盐HMB1%、姜黄5%、营养酵母粉0.5%、表没食子儿茶素没食子酸酯EGCG1.5%、维生素D0.0005%、调味剂10%和增稠剂1.9995%。
实施例4:
与实施例1不同的是,一种缓解肌肉流失的组合物,包括以下重量百分比原料:动物蛋白50%、植物蛋白30%、β-羟基-β-甲基丁酸盐HMB4%、姜黄2%、营养酵母粉0.5%、表没食子儿茶素没食子酸酯EGCG1.5%、维生素D0.0005%、调味剂10%和增稠剂1.9995%。
实施例5:
与实施例1不同的是,一种缓解肌肉流失的组合物,包括以下重量百分比原料:动物蛋白50%、植物蛋白30%、β-羟基-β-甲基丁酸盐HMB5%、姜黄1%、营养酵母粉0.5%、表没食子儿茶素没食子酸酯EGCG1.5%、维生素D0.0005%、调味剂10%和增稠剂1.9995%。

为验证本发明的功效,进行人群临床验证,其验证方法及验证结果如下:
(1)老年人群试验:在3个社区随机选取65~69岁男性、女性各30人,每人每日2次,每次20g服用本产品,六个月为一个使用周期,对试验对象的骨骼肌质量、肌力和功能性三个指标分别进行测试。
评判标准如下:
显效:指骨骼肌质量、肌力和功能性三个指标至少有两个指标改善;
有效:指骨骼肌质量、肌力和功能性三个指标仅一个指标改善;
无效:骨骼肌质量、肌力和功能性三个指标全部没有改善。
评判结果如附图1所示。
(2)健身人群:在健身房随机选取30位30-40周岁热爱健身的运动人群为试验对象,按照每次2次,每次20g补充本产品,三个月为一个使用周期,对试验对象进行骨骼肌质量进行测试。
评判标准如下:
有效:骨骼肌质量增加或维持;
无效:骨骼肌质量减少。
评判结果如附图2所示。
从试验结果看,本发明针对老年人群、健身人群均有较显著的缓解肌肉流失的作用,且配比中β-羟基-β-甲基丁酸盐HMB与姜黄为1:1时尤为明显;此外配方中经过调味剂调味处理并分装成小袋包装,易携带、口感细腻、甜度适中、适合目标人群长期服用。

1.一种缓解肌肉流失的组合物,其特征在于:包括以下重量百分比原料:动物蛋白40-60%、植物蛋白20-40%、β-羟基-β-甲基丁酸盐HMB2-5%、姜黄2-5%、营养酵母粉0.1-1.5%、表没食子儿茶素没食子酸酯EGCG0.5-2.5%、维生素D0.0001-0.001%、调味剂5-15%和增稠剂1.5-2.5%。
2.根据权利要求1所述的一种缓解肌肉流失的组合物,其特征在于:包括以下重量百分比原料:动物蛋白50%、植物蛋白30%、β-羟基-β-甲基丁酸盐HMB3%、姜黄3%、营养酵母粉0.5%、表没食子儿茶素没食子酸酯EGCG1.5%、维生素D0.0005%、调味剂10%和增稠剂1.9995%。
3.根据权利要求1所述的一种缓解肌肉流失的组合物,其特征在于:所述动物蛋白选用乳清蛋白、牛奶蛋白、分离乳清蛋白、肉类蛋白、鱼类蛋白或禽类蛋白。
4.根据权利要求1所述的一种缓解肌肉流失的组合物,其特征在于:所述植物蛋白选用大豆蛋白、豌豆蛋白、小麦蛋白、大米蛋白、燕麦蛋白、花生蛋白、玉米蛋白或油菜蛋白。
5.一种缓解肌肉流失的组合物的制备方法,其特征在于:包括以下步骤:
(1)按照以下重量百分比称取原料备用:动物蛋白40-60%、植物蛋白20-40%、β-羟基-β-甲基丁酸盐HMB2-5%、姜黄2-5%、营养酵母粉0.1-1.5%、表没食子儿茶素没食子酸酯EGCG0.5-2.5%、维生素D0.0001-0.001%、调味剂5-15%和增稠剂1.5-2.5%;
(2)将步骤(1)的原料分别经过20-40目筛网过筛处理后备用;
