本发明涉及食品加工技术领域,尤其是一种抗氧化组合物及其应用。
背景技术
随着自由基研究的发展,自由基氧化损伤学说越来越被认可。皮肤作为自由基氧化应激的主要目标,是抵御外界环境以及皮肤自身产生的活性氧的重要生物屏障。自由基会诱发或者导致皮肤损伤和疾病,包括红斑、水肿、起皱、光老化、炎症、自身免疫反应、过敏、角质化异常、肿瘤病变甚至皮肤癌。皮肤成纤维细胞位于皮肤组织的真皮层,对维持细胞弹性和保持水分有重要的功能性作用,并且能够调节皮肤的生理和病理状态。
自由基清除剂和抗氧化剂能够减少活性氧的产生和脂质过氧化,对细胞和组织氧化损伤起到预防及保护作用,因而对机体健康有着重要的作用。目前,由于人们对于天然来源的抗氧化剂的偏好和对于化学合成抗氧化剂安全性的担忧使得天然抗氧化剂越来越受到人们的关注。天然植物来源的植物化学物质如多酚、黄酮等被报道具有突出的自由基清除能力和抗脂质过氧化能力。
皮肤的衰老与机体的衰老是同时进行的,皮肤老化是机体衰老的一部分。因为皮肤位于机体的最外层,更多地受到外源性因素的影响,因此皮肤的老化是内源性因素和外源性因素共同作用的结果。皮肤外源性的老化与紫外线的照射、风吹、吸烟、饮酒、与刺激物接触有关。而紫外线的长期照射是环境中促进皮肤老化的最主要因素,故外源性老化一般是指光老化。光老化在临床上及组织学上均有特点。临床表现主要是:暴露部位皮肤松弛、粗深皱纹、结节、皮革样外观,色素斑增多,毛细血管扩张,原有几何图形外观明细改变或消失,肤色常呈灰黄色,可发生各种良性、癌前期或恶性肿瘤。光老化皮肤在组织学主要表现为:真皮层炎性细胞浸润,弹性纤维增粗、排列紊乱或聚集成团,真皮层上层胶原退行性变明细、最后成为无定形物质;皮肤毛细血管扩张并扭曲,最后变得极为稀疏,黑素细胞灶性增生等。光老化皮肤中的许多改变,如粗细皱纹、皮肤粗糙及色素沉着等可以通过药物改善和逆转,治疗和预防皮肤的光老化是可能的。在皮肤皱纹与饮食结构关系的调查中发现,日常大量食用抗氧化食品的人群皮肤皱纹相对较少,进一步推测抗氧化食品中的某些物质的自由基清除能力也可能对皮肤健康产生直接的影响,包括预防与皱纹形成、皮肤变薄以及缺水有关的氧化效应。并陆续有研究不断证明的此推论的正确性。
发明内容
基于此,本发明的目的在于克服上述现有技术的不足之处而提供一种安全无毒,抗氧化、清除自由基性能好的抗氧化组合物。
为实现上述目的,本发明所采取的技术方案为:一种抗氧化组合物,包含以下重量份的组分:黑果腺肋花楸浓缩果汁粉0.1-10份和紫苏粉0.1-10份。

黑果腺肋花楸果实中富含花青素、酚酸、原花青素等多酚类物质。多酚类化合物可直接作为自由基清除剂,不但抑制促氧化作用酶的活性,而且增加抗氧化功能的活性。原花青素是植物中广泛存在的一大类多酚类化合物的总称,具有强抗氧化、消除自由基的作用,可有效消除超氧阴离子自由基和羟基自由基,也参与磷酸、花生四烯酸的代谢和蛋白质磷酸化,保护脂质不发生过氧化损伤;是强有力的金属螯合剂,可螯合金属离子,在体内形成惰性化合物;保护和稳定维生素C,有助于维生素C的吸收和利用。有人采用高效液相色谱法检测黑果腺肋花楸果、果汁及果渣的酚类化合物组成及抗氧化性,结果表明原花青素含量最高,占酚类化合物的45%~80%,这些都是小分子聚合物,在细胞壁中与多糖或蛋白质结合形式存在。
花青素,又称花色素,是自然界一类广泛存在于植物中的水溶性天然色素,是花色苷水解而得的有颜色的苷元。黑果腺肋花楸果实中富含花青素,其抗氧化指数非常高,是一种天然的“抗氧化剂”,能消除自由基,强身健体,保护细胞免受氧化破坏,延缓肌体衰老,促进身体排出毒素和重金属,因此黑果腺肋花楸果实被誉为“口服的化妆品”“超级食品”,被美国农业部评为最有价值的水果之一。黑果腺肋花楸中的花青素参与胶原蛋白的合成,能够保护胶原蛋白,改善皮肤弹性,从而减少或避免皱纹的产生。
酚酸是一类含有酚环的有机酸。具体包括单羟基苯甲酸(对羟基苯甲酸酯,对羟基苯甲酸甲酯、对羟基苯甲酸乙酯),双羟基苯甲酸(龙胆酸、原儿茶酸)和三羟基苯甲酸(没食子酸、均苯三酚酸)。三羟基苯甲酸(特别是没食子酸)是可水解丹宁酸的组分之一。这种物质不但可以杀菌,还可以使体味芬芳,醒脑提神,美白肌肤。在黑果腺肋花楸中检测到的酚酸主要是绿原酸和新绿原酸,两者比例大约是1∶1,除此之外还检测到少量的隐绿原酸。酚酸在果汁中含量高于果渣,因其具有较好的水溶性广泛存在于细胞液泡中。
紫苏中富含酚类物质,主要有咖啡酸、迷迭香酸、阿魏酸等,紫苏叶中迷迭香酸含量较多,紫苏提取物中的酚类物质大多是迷迭香酸,是一种水溶性物质,具有抗氧化和抗菌的作用。紫苏中的黄酮类物质和酚类物质具有抗氧化活性,它们含有酚羟基,可以清除人体内的自由基和超氧化物。迷迭香酸抗氧化活性比维生素C高,具有调节还原性谷胱甘肽和胞内抗氧化酶的含量。有研究发现紫苏提取物中迷迭香酸有清除超氧阴离子自由基和羟自由基的作用,在试验范围内迷迭香酸清除羟自由基的能力比抗坏血酸高,对人类养颜美容、防衰老、强身健体起着重要作用。
优选地,所述的抗氧化组合物,包含以下重量份的组分:黑果腺肋花楸浓缩果汁粉0.1-5份和紫苏粉0.1-5份。进一步优选地,所述的抗氧化组合物,包含以下重量份的组分:黑果腺肋花楸浓缩果汁粉2份和紫苏粉2份。
发明人经过大量创造性试验,探究出了抗氧化组合物中黑果腺肋花楸浓缩果汁粉和紫苏粉的最佳比例范围,在这个范围内,抗氧化的效果最好。
本发明所用抗氧化组合物主要原料黑果腺肋花楸(野樱莓)浓缩果汁粉为市售,购买自崴达国际股份有限公司,紫苏粉为市售,购买自德国Vital Solutions公司。
此外,本发明提供了所述的抗氧化组合物的制备方法,包括如下步骤:将黑果腺肋花楸浓缩果汁粉和紫苏粉按配方混合均匀,得到所述抗氧化组合物。
本发明提供的制备方法工艺简单,不存在危险操作,操作方便,并且节省能源。
进一步地,本发明提供了所述的抗氧化组合物在食品中的应用。
另外,本发明还提供了一种包含所述的抗氧化组合物的食品。

其中,实施例1-6所述食品口服液体饮料的制备方法,包括如下步骤:按配方称量各组分,加饮用水溶解完全,搅拌均匀后用分装机进行分装为50ml/瓶,制得本发明抗氧化食品的液体饮品。口服推荐量为每日1次,每次1支,本品适宜于睡前服用。
实施例
同时,本申请设置对比例,对比例1与实施例2相比,不含黑果腺肋花楸(野樱莓)浓缩果汁粉,其余组分、重量份及制备方法完全相同;
对比例2与实施例2相比,不含紫苏粉,其余组分、重量份及制备方法完全相同;对比例3为安慰剂。
试验例1色差测试
测试过程:选择100名35-45岁女性性作为志愿者进行功能性饮料的效果测试,测试人群选择工作环境一致、工作时间一致,时间起2020年9月15日,截止2020年12月15日,合计三个月,100名志愿者均分为7组,分别试用实施例的产品和对比例,每人每天饮用1支,连续饮用3个月。产品试用期间,所有志愿者的饮食习惯均保持一致。
测试方法:手持式色差仪,前后两个时间点测试额头、脸颊、脖子、手背,取差值的L值平均值。测试结果如下表1所示:

备注:L值表示从白色到黑色的平衡,纯黑色为0,纯白色为100,L值越大,越靠近白色,L值越小,越靠近黑色。
结果表明测试组在三个月的体验周期内,肤色变白,测试时间选测秋季,相对日照温和,工作环境为室内,时间规律,测试条件下,对照组肤色L值有较小的增加,测试组L值增加,对比大多数人有肉眼可见的增白效果。
抗氧化实验
待测试样:实施例1-6和对比例1-3的食品口服液体饮料;
试剂:1,1-二苯基-2-三硝基苯肼(DPPH)、无水乙醇;
待测试样配制:分别取20mg实施例1-6和对比例1-3的食品口服液体饮料,用无水乙醇溶解并定容至250mL,备用;
DPPH的配制:准确称取20mg DPPH,用无水乙醇溶解并定容至250mL,备用;
仪器:紫外可见光分光光度计、恒温箱;
方法:将2mL经预配制的待测试样及2mL DPPH溶液至于具筛的试管中,至于恒温箱中37℃下恒温1h,以无水乙醇为空白对照,在517nm下测定各待测试样的吸光度A1,并按下式计算清除率:
DPPH清除率=[1-(A1-A2)/A0]×100%
其中,A0:2ml无水乙醇加2mL DPPH乙醇溶液的吸光度(对照试验);
A1:2mL待测试剂加2mL DPPH乙醇溶液的吸光度;
A2:2mL待测试剂加2mL无水乙醇溶液的吸光度(空白试验);
每个待测试样测三组平行样,求平均值,结果如下表2所示:

由表2可知,结果表明黑果腺肋花楸(野樱莓)浓缩果汁粉与紫苏粉相互协同,具有很好的抗氧化效果。
试验例2动物安全性毒理学评价实验
测试标准:取本发明实施例制得的抗氧化食品,按照卫生部《保健食品检验与评价技术规范》(2003年版)的评价方法进行动物安全性毒理学评价实验。
测试过程:取小白鼠90只(体重为20.0-25.0g),分成9组,每组10只,饥饿15小时左右,按每只每天50ml灌喂饮料,连续灌喂30天,分别观察小鼠的行为和体征,记录中毒表现及死亡情况。
实验结果:实验期间动物行动灵活,反应敏捷,被毛整洁,目光有神,眼鼻口无分泌物,进食饮水正常,生长发育良好,均未见明显中毒反应、无一死亡。表明口服液无急性毒性,饮用安全。
结果表明本发明经动物实验证实:安全无毒,不含兴奋剂,人体试食试验证明具有明显改善,无明显厌食、腹泻、乏力等副作用出现。
最后所应当说明的是,以上实施例仅用以说明本发明的技术方案而非对本发明保护范围的限制,尽管参照较佳实施例对本发明作了详细说明,本领域的普通技术人员应当理解,可以对本发明的技术方案进行修改或者等同替换,而不脱离本发明技术方案的实质和范围。
Claims (9)
1.一种抗氧化组合物,其特征在于,包含以下重量份的组分:黑果腺肋花楸浓缩果汁粉0.1-10份和紫苏粉0.1-10份。
2.如权利要求1所述的抗氧化组合物,其特征在于,包含以下重量份的组分:黑果腺肋花楸浓缩果汁粉0.1-5份和紫苏粉0.1-5份。
3.如权利要求2所述的抗氧化组合物,其特征在于,包含以下重量份的组分:黑果腺肋花楸浓缩果汁粉2份和紫苏粉2份。
4.一种如权利要求1-3任一项所述的抗氧化组合物的制备方法,其特征在于,包括如下步骤:将黑果腺肋花楸浓缩果汁粉和紫苏粉按配方混合均匀,得到所述抗氧化组合物。
5.一种如权利要求1-3任一项所述的抗氧化组合物在食品中的应用。
6.一种食品,其特征在于,包含权利要求1-3任一项所述的抗氧化组合物。
7.如权利要求6所述的食品,其特征在于,所述食品还包含辅料,所述辅料包含填充剂、酸味剂、稀释剂和增稠剂。
8.如权利要求6所述的食品,其特征在于,所述食品为口服液体饮料,包含以下重量份的组分:黑果腺肋花楸浓缩果汁粉0.1-10份、紫苏粉0.1-10份、黑果腺肋花楸浓缩汁0.5-10份、蔓越莓浓缩汁0.5-10份、浓缩苹果清汁4-8份、蜂蜜5-8份、针叶樱桃粉2-5份、赤藓糖醇7-10份、结晶果糖8-12份、柠檬酸0.1-0.2份、柠檬酸钠0.1-0.2份和黄原胶0.1-0.3份。
9.如权利要求8所述的食品,其特征在于,包含以下重量份的组分:黑果腺肋花楸浓缩果汁粉0.1-5份、紫苏粉0.1-5份、黑果腺肋花楸浓缩汁0.5-5份、蔓越莓浓缩汁0.5-5份、浓缩苹果清汁4-6份、蜂蜜5-7份、针叶樱桃粉3-5份、赤藓糖醇8-9份、结晶果糖8-10份、柠檬酸0.1-0.2份、柠檬酸钠0.1-0.2份和黄原胶0.1-0.3份。
