Abstract
本发明提供一种具有皮肤美容和提升机体抗氧化能力的组合物,所述的组合物由中分子量银耳多糖1~2份和胶原蛋白肽6~10份组成,其中中分子银耳多糖的分子量10万~50万Da。通过动物和人体试食实验证明,中分子银耳多糖和胶原蛋白肽组合物,对皮肤美容和提升机体抗氧化能力具有协同增效作用,实现在皮肤美容和抗氧化时的低剂量、高效应,从而避免了现有口服美容产品配方复杂、成分种类多、制备工艺繁琐、服用携带不方便、成本高、效果一般等缺陷,实现产品配方简洁、功效强,符合市场发展趋势。
Description
技术领域
本发明涉及一种具有皮肤美容和提升机体抗氧化能力的组合物,特别涉及一种中分子量银耳多糖和胶原蛋白肽的组合物及其应用,属于食品保健技术领域。
背景技术
人们对美的追求一直孜孜不倦。近年来胶原蛋白肽类口服美容产品市场日益火爆,市场容量快速增长,反映了胶原蛋白肽类产品确实有一定的美容作用。皮肤的胶原蛋白主要位于皮肤的真皮层,含量约占真皮层的70%。目前,大量的人体试食实验表明,服用胶原蛋白肽或胶原蛋白水解物2.5g~10g/天,大约一个月以上服用周期可以改善皮肤的水分、弹性和减少皱纹。从剂量数据上看,每日服用剂量偏大。胶原蛋白肽或胶原蛋白水解物带有一定的腥味,因而需要较多的矫味剂、香精来掩盖腥味,从而导致产品配方复杂、成分多样,最终胶原蛋白肽/水解物类成品的服用量也较大,对消费者的体验不友好。同时,矫味剂、香精本身长期食用可能会对健康也有一定的风险,减少人工添加剂在产品中的用量是目前健康产品的发展趋势之一。
透明质酸是人体皮肤的组成成分之一,人体表皮和真皮层中占人体透明质酸总量一半以上。透明质酸可以吸附大量的水,皮肤的含水量与透明质酸含量紧密相关。透明质酸作为一种具有保湿、抗衰老功能的明星美容原料,在化妆品领域应用广泛,在保健食品领域偶有使用。根据中华人民共和国卫生部2008年第12号公告,透明质酸钠只能应用于保健食品中,而普通食品不允许使用。其口服作用效果也存疑。
随着年龄的增长、皮肤老化,人体皮肤中胶原蛋白和透明质酸的含量呈现下降的趋势,主要是由于内外因的综合作用打破了合成与降解的平衡,使得降解大于合成。
银耳多糖是一种酸性杂多糖,主要由甘露糖、木糖、半乳糖和葡萄糖醛酸组成。其分子结构与透明质酸分子有一定的相似性,因此又称为植物透明质酸。有动物实验表明,银耳多糖口服后能提高实验动物皮肤中透明质酸的含量、皮肤含水量,以及提高机体抗氧化酶的活性。然而,动物实验数据有效剂量≥200mg/kg·天,以小鼠与人等效关系推算人体服用的有效剂量需要达到1.2g/天以上,才具有改善皮肤功能和抗氧化效果。从商业应用而言,成本太高、服用量大、不方便,实际应用价值低。
此外,申请人注意到申请号2018101182081一种抗皱保湿、改善皮肤弹性的胶原蛋白复合饮品的发明,采用水解2型胶原蛋白、鲑鱼粉、银耳液、弹性蛋白、酪蛋白水解肽、浓缩果汁和多种维生素进行组合,具有抗皱保湿作用,然其组方和制备工艺复杂,成分多样;申请号2018101399869一种养颜健体延缓衰老胶原蛋白肽充剂及其制备方法,其组方有鱼鳞胶原蛋白肽、银耳多糖、芦荟多糖、蛹虫草多肽粉、花生胚芽粉、中药粉等,其制备方法和组方也都很复杂;申请号2018111564400一种胶原蛋白肽饮品,其组方包括胶原蛋白肽、弹性蛋白肽、银耳多糖、壳寡糖、乳酸钙、维生素C、浓缩果汁等,而主要功效成分是胶原蛋白肽;申请号2018109200514燕窝胶原肽饮品及其制备方法,组方包括胶原蛋白肽、樱桃汁、蓝莓汁、燕窝粉、壳寡糖、银耳多糖、γ氨基丁酸、维生素C等。这些发明组合物配方成分种类复杂,组方仅是各类有美容作用成分的堆砌,技术水平较低;同时,有些发明制备工艺繁琐复杂,导致以美容保健使用时食用量大、便捷性低、成本高,整体的功效机理作用不清楚,对消费者友好程度低。
因此,开发高效应、低剂量的美容组合物具有良好的社会价值和经济价值。
发明内容
为解决现有美容产品服用剂量大、效果一般、便捷性低、技术水平低下等缺点,本发明提供一种通过协同增效作用实现低服用剂量、同时兼具皮肤美容和提升机体抗氧化作用的组合物。
为达到其目的,本发明的组合物由下述重量份配比组成:中分子量银耳多糖1~2份,胶原蛋白肽6~10份;
其中,中分子量银耳多糖分子量范围10万~50万Da。
本发明另一方面还提供上文所述的组合物在制备具有改善皮肤状态和延缓衰老产品中的应用。
与现有技术相比,本发明具有以下优点及有益效果:
本发明中分子量银耳多糖和胶原蛋白肽的组合物,在提高老化皮肤胶原蛋白、透明质酸含量,以及提升机体抗氧化功能上具有协同增效作用,从而实现美容、抗衰老的低剂量、高效应,提高消费者服用的便捷性、依从性、降低消费成本,对消费者友好。
本发明制得的中分子量银耳多糖和胶原蛋白肽组合物,由于起效剂量低,能通过常规制剂工艺和辅料,可以制成食品中的多种剂型(如片剂、粉剂、颗粒剂、口服液、饮料、果冻、糖果),制备工艺简单、每日服用量少、服用携带方便,符合养生健康产品配方简洁的原则,符合市场发展潮流趋势。
具体实施方式
下面结合实施例对本发明作进一步详细的描述,但本发明的实施方式不限于此。在未作说明的情况下,本发明涉及的原料与方法均为本领域的常规原料和方法。胶原蛋白肽为市售原料,购自罗赛洛公司,分子量分布范围2000Da~5000Da。中分子量银耳多糖采用专利申请号为2019108838683(一种中分子量银耳多糖的制备方法)的方法获得,经过高效凝胶渗透色谱分析分子量分布范围10万~50万Da。
实施例1
本发明实施例1提供的组合物由下述重量配比的组分制备而成:中分子量银耳多糖10g,胶原蛋白肽100g。经加入制粒成形辅料如羧甲基纤维素后,经压片得银耳多糖胶原蛋白肽片。
实施例2
本发明实施例2提供的组合物由下述重量配比的组分制备而成:中分子量银耳多糖10g,胶原蛋白肽60g。两种原料混合均匀后,加入适量60%乙醇进行制粒,热风烘箱干燥后制得银耳多糖胶原蛋白肽颗粒。
实施例3
本发明实施例3提供的组合物由下述重量配比的组分制备而成:中分子量银耳多糖20g,胶原蛋白肽60g。加入适量纯化水、甜味剂,搅拌至完全溶解,过滤后灌装杀菌,得银耳多糖胶原蛋白肽饮料。
实施例4
本发明实施例4提供的组合物由下述重量配比的组分制备而成:zhong分子量银耳多糖20g,胶原蛋白肽100g。加入适量纯化水、甜味剂,搅拌至完全溶解,过滤后灌装杀菌,得银耳多糖胶原蛋白肽口服液。
实施例5
本发明实施例5提供的组合物由下述重量配比的组分制备而成:中分子量银耳多糖15g,胶原蛋白肽80g。加入适量纯化水溶解,加入甜味剂、香精,搅拌溶解,然后加入至保温的卡拉胶溶液中,持续搅拌至分散均匀,冷却后得银耳多糖胶原蛋白肽果冻。
为考察组合物的应用效果,开展了人群和动物试验进行组合物功效验证实验:
实验1:皮肤美容功效实验
实验方法:按自愿原则选择女性受试者,年龄30岁-40岁,皮肤水分≤12。受试者排除标准:①妊娠或哺乳期妇女,过敏体质者;②合并有心脑血管、肝、肾、造血系统性疾病和精神病患者;③短期内服用与试验样品有关的物品,影响到对结果的判断者;④未按试验要求服用样品或中途脱离,无法判定功效或资料不全影响疗效或安全性判断者。符合纳入标准120名女性志愿者,随机分为对照组、实施例1组(S1)、实施例2组(S2)、实施例3组(S3)、实施例4组(S4)、实施例5组(S5)、中分子量银耳多糖组、胶原蛋白肽组、高分子量银耳多糖组、高分子银耳多糖胶原蛋白肽组共10组,每组12人,每日口服实验样品1次,持续30天。实验前试验组和对照组指标差异无统计学意义,实验前后测定皮肤水分、皮肤皱纹变化,数据分析采用统计软件处理。
各人群组样品组成和服用剂量见下表1。

从表2结果可知,各实施例配方组在提升皮肤水分含量上显著优于中分子量银耳多糖组、胶原蛋白肽组、高分子量银耳多糖组、高分子银耳多糖胶原蛋白肽组,同时也发现高分子量银耳多糖在提升皮肤水分作用方面弱于中分子量银耳多糖。而单一胶原蛋白肽服用剂量较低时,其改善皮肤水分的作用效果不明显,而当中分子量银耳多糖与胶原蛋白复配时,在较低的剂量条件下已经起到了改善皮肤水分的作用,说明了合适剂量的中分子量银耳多糖与胶原蛋白肽组合时,对皮肤水分含量改善具有协同增效作用,且这种协同作用与银耳多糖的分子量有极大的关系(由S4vs.高分子银耳多糖胶原蛋白肽结果对比可知)。

从表3结果可知,各实施例配方组在减少皮肤皱纹上显著优于中分子量银耳多糖组、胶原蛋白肽组、高分子量银耳多糖组、高分子银耳多糖胶原蛋白肽组。中分子量银耳多糖在减少皮肤皱纹方面强于高分子量银耳多糖。当中分子量银耳多糖与胶原蛋白复配时,在较低的剂量条件下对皮肤皱纹的改善作用已经很显著(p<0.01),说明了合适剂量的中分子量银耳多糖与胶原蛋白肽组合时,对皮肤减少皮肤皱纹具有协同增效作用,而且这种作用与银耳多糖的分子量有极大的关系。
实验2:动物抗氧化和减少胶原蛋白、透明质酸流失实验
实验方法:SPF纯系KM雄性小鼠,体质量(19±2)g。正常对照组小鼠腹腔注射生理盐水,其余各组每天按100mg/kg腹腔注射D-半乳糖连续注射8周建立衰老模型,每组10只。同时,除正常对照组外,其余各组灌胃按表4所示的药物剂量,8周后处死小鼠,取小鼠肝组织测定其中SOD(超验歧化酶)、CAT(过氧化氢酶)、GSH-Px(谷胱甘肽过氧化物酶)酶活及MDA(丙二醛)含量,取小鼠皮肤按试剂盒说明检测羟脯氨酸(胶原蛋白的特征氨基酸)、透明质酸含量。按统计学方法分析和处理数据,组间差异比较采用t检验。

实验结果:各组肝组织中SOD、CAT、MDA、GSH-Px指标见表5,小鼠皮肤中羟脯氨酸、透明质酸含量见表6。

从表5结果可知,衰老小鼠模型建立,各实施例组与模型组相比,SOD、CAT、GSH-Px酶活显著提高而MDA产物浓度显著下降(p<0.01),表明各实施例组具有极强抗氧化作用。此外,中分子量银耳多糖也能提升抗氧化酶活及降低MDA产物浓度(p<0.05),表明中分子银耳多糖也具有良好的抗氧化作用。而单独的胶原蛋白肽、高分子银耳多糖以及两者组合后,尽管各酶活具有一定的提升作用但不显著,表明抗氧化作用一般(p>0.05),且在CAT、MDA指标上与实施例组也有显著性差异。同时,表明银耳多糖活性的抗氧化活性与分子量有极大相关性,中等分子量活性强而高分子量则活性较低。进一步的,根据各实施例组药物的剂量及组成,与单一成分活性比较判断,中分子银耳多糖与胶原蛋白肽复合后在提升抗氧化酶活、降低MDA产物方面具有协同增效作用,可以实现低剂量高效应。

从表6结果可知,模型组小鼠皮肤中透明质酸、胶原蛋白含量极显著下降,表明衰老可导致透明质酸、胶原蛋白的流失。各实施例组方组合物可以显著减少透明质酸、胶原蛋白流失(p<0.01),中分子银耳多糖也具有明显的减缓流失作用(p<0.05),而高分子银耳多糖则作用不显著,由此提示银耳多糖分子量对活性有极大的影响。此外,单一胶原蛋白肽、高分子银耳多糖与胶原蛋白肽组合后的减缓透明质酸、胶原蛋白流失作用也较弱。根据各实施例组合物组成比例,以及与单一成分的活性相比较结果,可以判断中分子银耳多糖与胶原蛋白肽复合后,在减缓透明质酸、胶原蛋白流失速度上具有协同增效作用,其机制可能与提升抗氧化酶活有关。
以上所述,仅为本发明的较佳的具体实施例,但本发明的保护范围并不局限于此,任何熟悉本技术领域的技术人员在本发明揭露的技术范围内,根据本发明的技术方案及其构思加以等同替换或改变,都应涵盖在本发明的保护范围内。
Claims (3)
1.一种具有皮肤美容和提升机体抗氧化能力的组合物,其特征在于,所述组合由以下重量份组成:中分子量银耳多糖1~2份,胶原蛋白肽6~10份。
2.根据权利要求1所述的具有皮肤美容和提升机体抗氧化能力的组合物,其特征在于,所述中分子银耳多糖分子量范围10万~50万Da。
3.权利要求1-2任一项所述的组合物在制备具有改善皮肤状态和延缓衰老产品中的应用。
