Description

Plant fermentation method and composition thereof
Technical Field
The invention relates to the technical field of microbial fermentation, in particular to a plant fermentation method and a composition thereof.
Background
With the growing concern of consumers on skin beauty and health in recent years and the continuous harsh, stressful and daily rise of environment, consumers increasingly know the importance of body and skin health. Microbial fermentation refers to a process of converting raw materials into products which are easily absorbed and utilized by the human body through a specific metabolic pathway by utilizing the decomposition action of microorganisms under appropriate conditions. The separation of active ingredients is promoted through the growth and metabolic activities of microorganisms, so that a new effect is generated, the medicinal value is enhanced, and the absorption and utilization of the active ingredients by an organism are promoted.
The plant and microorganism co-fermentation is to extract juice or extract plant raw materials and then add probiotics for fermentation, so that a large amount of effective active ingredients can be generated. With the continuous progress of research, a great number of scientific experiments report the application of the co-fermentation of plants and microorganisms in the directions of improving the medicinal effect, food and beverage, health care products, purifying pollution and the like.
However, the commonly used co-fermentation microorganisms with plants mainly include bifidobacteria, lactobacilli, yeasts, partial fungi, endophytic fungi and the like, and have poor skin conditioning effect, so that the increasing demands of consumers cannot be met.

The plant fermentation method comprises the following steps:
s1: inoculating a strain purchased from China center for industrial microorganism strain preservation management in a slant culture medium, and culturing at 30 ℃ for 36h to activate microspherical strains; inoculating the well-grown strains in a liquid culture medium, carrying out 150r/min in a UVB radiation environment, carrying out shake culture at 30 ℃ for 48h, screening microspherical strains capable of growing under the UVB radiation condition, calculating a growth curve as shown in figure 1, and facilitating later-stage further experiment culture of micrococcus with better UVB radiation resistance, wherein the preservation number of the microspherical strains is CICC 23810; the micrococcus screened out at this time has extremely strong radiation resistance, which means that the micrococcus has extremely strong oxidation resistance.
S2: screening S1 to obtain micrococcus in stationary phase, placing the micrococcus in liquid culture medium at 150r/min, and performing shaking table culture at 30 ℃ until logarithmic phase;
s3: pulverizing whole sunflower, adding 60% ethanol, ultrasonic extracting at 75 deg.C for 4 hr in ultrasonic tank, and rotary evaporating at 50 deg.C to obtain brown viscous substance to obtain sunflower extract;
s4: adding the sunflower extract obtained in S3 into the microspherical fungus culture solution in S2, fermenting and culturing, and culturing at 150r/min in a shaking table at 30 ℃ for about 48 h;
s5: and (2) centrifuging the fermentation culture solution obtained in the step (S4), filtering by using a plate frame, intercepting thalli on a filtering membrane (0.22 mu m) to obtain fermentation supernatant, adding activated carbon into the fermentation supernatant to adsorb impurities in the fermentation supernatant, removing peculiar smell, centrifuging by using a plurality of layers of absorbent gauze to remove carbon residue, and filtering the purified fermentation broth through the filtering membrane of 0.22 mu m to obtain the sunflower-micrococcus fermentation product.
The materials and equipment used in the above steps are: peptone, beef extract, sodium chloride, agar, pure water, whole sunflower, an ultrasonic instrument, a rotary evaporator, an autoclave, a biochemical incubator, a PH meter and the like.
Wherein the preparation and components of the culture solution are as follows:
slant culture medium, peptone 10.0g, beef extract 3.0g, sodium chloride 5.0g, agar 15.0g, and pure water 1000mL, with pH 7.2, 121 deg.C, and sterilizing for 20 min;
liquid culture medium, 10.0g of peptone, 3.0g of beef extract, 5.0g of sodium chloride and 1000mL of pure water, wherein the pH value is 7.2, the temperature is 121 ℃, and the sterilization is carried out for 20 min.
Because the skin conditioning effect of the single micrococcus fermentation product and the sunflower extract applied to the skin care product is poor, the sunflower-micrococcus fermentation product prepared by the method can degrade organic matters which cannot be directly utilized by a human body in the sunflower extract through the fermentation of the micrococcus, so that the organic matters can be better absorbed by the skin of the human body, and the sunflower-micrococcus fermentation product prepared by the method has stronger effect in the skin care product applied to the skin care product.
S6: the content of polypeptide, polyphenol and polysaccharide in the sunflower-micrococcus fermentation product is detected by preparing materials and equipment such as casein, sodium hydroxide, anhydrous copper sulfate, gallic acid, a forskolin phenol solution, sodium carbonate, glucose, concentrated sulfuric acid, phenol and a reduced glutathione detection kit, pure water, an ultraviolet spectrophotometer, a colorimetric tube, a 10mm cuvette, a water bath kettle and the like.

Figure BDA0002759875700000061
the polypeptide is one of peptide or peptide, and is a compound with a molecular structure between amino acid and protein. Amino acids are the smallest constitutional unit of proteins. An amino acid molecule is called an amino acid. Two amino acid molecules are dehydrated, condensed, connected and combined together to form the second peptide, and by analogy, the second peptide contains four amino acids and is called the fourth peptide; contains nine amino acids, named as Jiuzhanyun; peptides of less than 10 amino acids are generally called oligopeptides, peptides with a larger number of amino acids are called polypeptides, and peptides with more than 50 amino acids are called proteins. Because the varieties of the contained amino acids are different, the di-peptide and the tri-peptide also have different varieties, so that various peptides with different names and different functions appear. In short, amino acid constitutes peptide, peptide constitutes protein, but protein can not be absorbed and utilized by human body directly, but single amino acid and peptide chain separated from protein can be absorbed by human body, so that the polypeptide content in the sunflower-micrococcus fermentation product is higher, and the sunflower-micrococcus fermentation product has better absorption effect by human body.
Wherein the polyphenols are compounds found in vegetable foods with potential health promoting effects. It is found in some common plant foods, such as cocoa beans, popcorn, tea, soy beans, red wine, vegetables and fruits. The antioxidant function of polyphenol can prevent the chronic diseases; the plant polyphenol has the common properties of astringing the body, removing the body, resisting ultraviolet radiation, inhibiting the activities of the tyrosine and the peroxidase, and keeping various physiological activities, so that the plant polyphenol has various unique effects in the preparation of anti-aging and whitening cosmetics.
Wherein the chemical name of the reduced glutathione is N- (N-L-gamma-glutamyl-L-cysteinyl) glycine, the molecular formula is C10H17N3O6S, the molecular weight is 307.33, and the reduced glutathione is soluble in water, diluted alcohol, liquid ammonia and methyl formamide, but not soluble in alcohol, ether and acetone. It can be used for treating facial pigmentation, sweat stain and pigmentation caused by various reasons, and also has good antioxidant effect.
Therefore, the sunflower-micrococcus fermentation product prepared by the method has good functions of resisting oxidation, resisting aging, whitening, removing acnes, diminishing inflammation and repairing human skin, and can be better absorbed by a human body, which cannot be realized by a single sunflower extract.
