本发明公开了口腔抑菌品技术领域,具体领域为口腔抑菌凝胶,壳聚糖为天然多糖甲壳素脱除部分乙酰基的产物,具有生物降解性、生物相容性、无毒性和抑菌等特性,其溶于偏酸性的液体中呈粘稠胶状,将其作为本发明中原材组成部分,对口腔起到抑菌作用的同时,使本发明口腔抑菌凝胶整体呈粘稠胶状,相比于制剂型抑菌剂更容易粘附于口腔粘膜上,使得其对口腔粘膜的抑菌效果更好,将苦参提取物、金银花提取物配合银离子使用,对口腔内黄色葡萄球菌、大肠杆菌和白色念珠菌起到抑制作用,其中对金银花提取物的提取方式在传统萃取的基础上,采用膜分离设备进行过滤使得金银花中的有效抑菌物质绿原酸在较大程度上得到保留,抑菌效果显著。

本发明涉及口腔抑菌品技术领域,具体领域为口腔抑菌凝胶。
背景技术
口腔是消化道的起始部分,前借口裂与外界相通,后经咽峡与咽相续。口腔内有牙、舌等器官。口腔的前壁为唇、侧壁为颊、顶为腭、口腔底为黏膜和肌等结构。口腔借上、下牙弓分为前外侧部的口腔前庭和后内侧部的固有口腔;当上、下颌牙咬合时,口腔前庭与固有口腔之间可借第三磨牙后方的间隙相通。口腔是细菌、病毒、真菌和其他外来抗原进入人体的主要途径之一,除了这一外源性负担外,还有大量的内源性微生物菌群寄生在口腔、牙齿和粘膜表面,微生物菌落集群,可以分为两大类,第1类由游离形式的细菌组成,主要存在于唾液中,第2类由菌落形式的细菌组成,主要存在于牙斑或粘膜表面,口腔中的细菌包括非致病菌和致病菌,其中金黄色葡萄球菌、大肠杆菌和白色念珠菌是比较典型的三类致病菌,现有的口腔抑菌剂,形态上不易粘附于口腔壁上,与口腔接触时间短,抑菌效果不佳,且现有口腔抑菌剂有效成分多为抗生素类,长期使用会导致口腔菌群失衡,如是中药类型,则有效的抑菌物质纯度低,抑菌效果不佳,为此提出一种口腔抑菌凝胶。
发明内容
本发明的目的在于提供口腔抑菌凝胶,以解决上述背景技术中提出的问题。
为实现上述目的,本发明提供如下技术方案:口腔抑菌凝胶,其原材料组成和相应重量百分比为:壳聚糖0.3%-1%、苦参提取物2%-4%、金银花提取物1%-3%、银离子0.01%-0.03%、其余为纯化水。
优选的,其原材料组成和相应重量百分比为:壳聚糖0.4%-0.8%、苦参提取物2.5%-3.5%、金银花提取物1.5%-2.5%、银离子0.015%、其余为纯化水。
优选的,其原材料组成和相应重量百分比为:壳聚糖0.5%、苦参提取物3%、金银花提取物2%、银离子0.015%、其余为纯化水。
口腔抑菌凝胶的制备方法,其制备方法包括如下步骤:
步骤1:苦参提取物的制备
选取药用植物苦参,将其烘干后进行粉碎处理,置于干燥容器中备用;
取苦参粉末,以乙醇为溶剂,将苦参添加至乙醇溶剂中,在水浴温度为70℃条件下加热,回流提取,离心、过滤,重复回流提取3-4次,合并上清液,在60℃水浴温度下对滤液进行减压浓缩,浓缩完毕后密封低温保存备用;
步骤2:金银花提取物的制备
将金银花加水煎煮,将煎煮液过滤后浓缩,浓缩液使用醋酸乙酯进行萃取,对萃取液进行减压回收,得到萃取物,在将萃取物采用膜分离设备进行过滤得到金银花提取物,密封低温保存备用;
步骤3:口腔抑菌凝胶的制备
将壳聚糖、苦参提取物、金银花提取物、银离子和纯化水在温度为40-60℃条件下搅拌,搅拌时间30-40min,搅拌物逐渐呈凝胶状,即制备出口腔抑菌凝胶。
优选的,所述乙醇溶剂的质量分数为50%。
优选的,所述膜分离设备是卷式超滤膜分离设备。
本发明的有益效果是:口腔抑菌凝胶,壳聚糖为天然多糖甲壳素脱除部分乙酰基的产物,具有生物降解性、生物相容性、无毒性和抑菌等特性,其溶于偏酸性的液体中呈粘稠胶状,将其作为本发明中原材组成部分,对口腔起到抑菌作用的同时,使本发明口腔抑菌凝胶整体呈粘稠胶状,相比于制剂型抑菌剂更容易粘附于口腔粘膜上,使得其对口腔粘膜的抑菌效果更好,将苦参提取物、金银花提取物配合银离子使用,对口腔内黄色葡萄球菌、大肠杆菌和白色念珠菌起到抑制作用,其中对金银花提取物的提取方式在传统萃取的基础上,采用膜分离设备进行过滤使得金银花中的有效抑菌物质绿原酸在较大程度上得到保留,在传统萃取方式中,绿原酸可溶于水,因此较难提取出来,采用膜分离设备进行过滤使得分子量较小的绿原酸得以分离提出,抑菌效果显著。

将0.3%壳聚糖、2%苦参提取物、1%金银花提取物、0.01%银离子和97.69%纯化水在温度为40℃条件下搅拌,搅拌时间30min,搅拌物逐渐呈凝胶状,即制备出口腔抑菌凝胶。

步骤2:金银花提取物的制备
将金银花加水煎煮,将煎煮液过滤后浓缩,浓缩液使用醋酸乙酯进行萃取,对萃取液进行减压回收,得到萃取物,在将萃取物采用膜分离设备进行过滤得到金银花提取物,密封低温保存备用;
步骤3:口腔抑菌凝胶的制备
将壳聚糖、苦参提取物、金银花提取物、银离子和纯化水在温度为40-60℃条件下搅拌,搅拌时间30-40min,搅拌物逐渐呈凝胶状,即制备出口腔抑菌凝胶。
5.根据权利要求4所述的口腔抑菌凝胶的制备方法,其特征在于:所述乙醇溶剂的质量分数为50%。
6.根据权利要求4所述的口腔抑菌凝胶的制备方法,其特征在于:所述膜分离设备是卷式超滤膜分离设备。
