本发明涉及食品加工领域,本公开了一种双参肽功能性固体饮料及其制备方法,该产品由胶原蛋白肽、人参肽粉、海参肽粉、赤藓糖醇、黄精粉、枸杞粉、维生素B、维生素B6、牛磺酸、食用香精组成;其制备方法包括预混、制粒、干燥、总混。本发明配方在现代研究的活性成分中,添加了中药成分,实现了现代与传统医药的结合,从多种角度共同起到提高免疫体、显著抗疲劳、显著抗氧化、显著调血糖、显著调血脂、显著解酒护肝、显著提高抗氧耐受力、显著清咽功能、显著增强改善性功能的作用,配方合理,功效确切。本发明复合物的制备工艺简单、稳定,且制备获得的颗粒复合物易成型,产品质量稳定可控,服用方便,易长期服用,无不良适应症。

本发明涉及食品加工领域,尤其涉及一种双参肽功能性固体饮料及其制备方法。
背景技术
功能饮料是指通过调整饮料中天然营养素的成分和含量比例,以适应某些特殊人群营养需要的饮品,不以治疗疾病为目的,具有调节机体功能作用。目前较为常见的功能性饮料大多为可直接饮用的液体功能性饮料,液体功能性饮料包装精美、饮用方便,但是液体的占用空间与重量相对固体较大,且成品的液体功能性饮料长时间存放会变成恒温,无法即时的根据需要进行温度的调整,相比而言,固体功能性饮料不仅携带方便,还能根据需要选择引用时的水温,有着很大的发展前景。
人参,中华医药中最具传奇性的药材之一。《神农本草经》中记载:“人参,味甘微寒,主补五脏,安精神,定魂魄,止惊悸,除邪气,明目,开心益智。久服,轻身延年。因此成为传统中医“治虚劳内伤第一要药”。
海参,被称为八珍之首,长寿之王。《本草纲目》中记载:味甘咸,补肾,益精髓,摄小便,壮阳疗痿,其性温补。海参作为一种营养价值极高的滋补品食用,含有高比例的蛋白质,丰富的维生素和对人体有益的多种微量元素,营养成分的种类高达50多种,食用海参对身体有多种益处,健脑益智、生血养血、预防心血管疾病、增强人体免疫力等等。
市场上固体饮料产品多种多样,品质参差不齐,一方面产品在食补温和调理作用上效果不明显,另一方面产品在药物活性成分刺激作用下易出现不良适应症等副作用。因此,双肽功能性固体饮料的研制具有很大的开发潜力。
发明内容
本发明目的在于针对现有固体饮料产品的技术不足,提供一种服用方便、温和不刺激,提高免疫体、显著抗疲劳,且具有显著缓解体力疲劳功能的双参肽功能性固体饮料及其制备方法。
为达到以上目的,本发明采取的技术方案是:
一种双参肽功能性固体饮料,由以下成分按重量百分比组成:胶原蛋白肽40-55%、人参肽粉12-20%、海参肽粉12-20%、赤藓糖醇2-3%、黄精粉5-8%、枸杞粉5-8%、维生素B0.03-0.1%、维生素B60.03-0.1%、牛磺酸0.03-0.1%、食用香精0.1-1%。
进一步的,所述海参肽制备步骤如下:
S1:海参处理,将去除内脏的海参,粉碎至10-20目,2~5%的食盐水清洗1-2次,滤除水溶液后加入5-8倍量90%乙醇浸泡18-24h,滤除乙醇溶液后用纯净水清洗1-2次,再加入4-8倍量0.4%-0.6%NaOH溶液浸泡15-20h,滤除NaOH溶液后用纯净水清洗1-2次;然后将处理后海参粗粉进行微波干燥,进一步粉碎至80-100目,备用;
S2:海参酶解,将海参粉与重量百分比浓度为2~5%的冰糖水溶液按重量比1:3~5的比例混合后放入打浆机内高速搅拌成海参匀浆液,再将海参匀浆液控制温度为45~55℃,并加入pH为7-8的0.01-0.03M的磷酸缓冲液以及复合碱性动物蛋白酶,复合碱性动物蛋白酶的添加量为海参匀浆液重量的3~5%,酶解35~50分钟,得海参酶解液;
S3:灭酶,将海参酶解液放入90~100℃水浴灭酶8~15分钟, 然后过滤除掉固体杂物,得灭酶海参匀浆液;
S4:干燥,将上述收集海参匀浆液浓缩至质量分数35-40%,喷雾干燥,制得海参肽粉。
进一步的,S2中所述复合碱性动物蛋白酶为胰蛋白酶和风味蛋白酶,两者用量比例为4:3。
进一步的,所述人参肽制备步骤如下:
S1,人参预处理,取3-5年以上老参,清洗去泥及其它杂质,自然晾干后打碎过40-50目筛,得人参粗粉;
S2,低温浸泡浸提,将上述人参粗粉,加入10-15倍量水0-4℃ 浸泡12-18h,第一次真空浸提 8-12h,收集滤液,再向药渣中加入5-10倍量水,第二次真空提取4-6h,收集滤液,合并两次真空浸提滤液,真空环境为 130-170mmHg,浸提温度25-35℃,浸提搅拌转速为60-100rpm;
S3,浓缩、醇沉:浸提滤液过300-400目滤布得滤液,滤液静置2h无沉淀物,在70℃以下真空浓缩,滤液浓缩至相对密度为1.2;浓缩液冷却至室温,向浓缩液中缓慢加入2-4倍量95%乙醇并充分搅拌,静置8-15h,离心,收集静置后醇不溶沉淀得人参多糖提取物;
S4,复溶脱醇精制:将上述醇不溶沉淀加等体积50℃水溶解,搅拌,并在酒精回收装置中蒸馏脱乙醇;
S5,再浓缩:将上述复溶脱醇溶液在70℃以下真空浓缩,滤液浓缩至含水量为30%-40%,得人参匀浆液;
S6:干燥,将上述浓缩人参匀浆液进行微波干燥后得块状固体提取物,经物理粉碎后粉末过80-100目筛,得人参肽粉。
一种双参肽功能性固体饮料的制备方法,具体的步骤如下:
(1)预混:按组分依次将胶原蛋白肽、人参肽粉、海参肽粉、赤藓糖醇、黄精粉、枸杞粉投料于高速湿法混合制粒机,搅拌桨转速350-420转/分钟,制粒刀转速2000-2800转/分钟,干料预混300-500s;
(2)制粒:加入预混干物料9%-15%倍量的95%乙醇,搅拌桨转速200-240转/分钟,制粒刀转速1500-1800转/分钟,混合制粒30-60s;
(3)干燥:过20-24目筛,于防爆热风循环干燥箱 50-60℃干燥60-80min,过16-18目筛整粒,至细粉率小于5%,得干燥颗粒混合物;
(4)总混:将步骤(3)所得干燥颗粒混合物、维生素B、维生素B、牛磺酸、食用香精按组分比投入三维混料机中混合30-40min得双参肽功能性固体饮料。
胶原蛋白肽是一种细胞外蛋白质;由二个或二个以上氨基酸组成蛋白肽;人体的吸收是以肽的方式进行,食用蛋白肽的吸收利用率可达100%;胶原蛋白是细胞外基质中最重要的组成部分。胶原蛋白肽是胶原蛋白的水解产物,富含羟脯氨酸,由2个以上氨基酸构成,分子量要求在10000以下的大于90%,吸收性能好,是一种食品原料。
海参肽主要是胶原肽类,还含有神经肽、糖肽以及抗菌肽等多种活性肽。小分子多肽具有多种功效成分,如降血压、降血脂、抗动脉粥样硬化、抗疲劳、抗菌、提高免疫力、延缓衰老和抗氧化等诸多生理功能。
人参肽含有人参皂苷成分,对血压具有双向调节作用,而且可扩张血管,促进血液循环,从而降低血压,可防治高血压等疾病;人参肽还具有降低血清胆固醇的作用,而且还可降低脂肪在血管壁的沉积,从而达到降低血脂的作用,防治动脉粥样硬化、脑栓塞、心肌梗塞等心脑血管疾病;人参肽还可促进机体新陈代谢,促进体内酒精的排泄,可用于改善因醉酒导致的恶心呕吐、头晕等症状,还可防治酒精肝。
枸杞子的性味甘平,能够滋补肝肾、益精明目、补血养血。对于现代人来说,枸杞子最实用的功效就是抗疲劳和降低血压。《神农本草经》称枸杞子“久服坚筋骨,轻身不老,耐寒暑”。《本草汇言》赞之“使气可充,血可补,阳可生,阴可长”。
黄精,又名:鸡头黄精、黄鸡菜、笔管菜、爪子参、老虎姜、鸡爪参。为黄精属植物,具有补脾,润肺生津的作用。黄精性味甘甜,食用爽口。其肉质根状茎肥厚,含有大量淀粉、糖分、脂肪、蛋白质、胡萝卜素、维生素和多种其他营养成分,生食、炖服既能充饥,又有健身之用,可令人气力倍增、肌肉充盈、骨髓坚强,对身体十分有益。现代研究证明,黄精水提出液对伤寒杆菌、金黄色葡萄球菌、抗酸杆菌有抑制作用;对肾上腺素引起的血糖过高呈显着抑制作用;具有较强的抗疲劳、抗氧化和延缓衰老作用;有明显增加麻醉犬冠脉流量作用。
本发明的优点在于:
本发明配方在现代研究的活性成分中,添加了中药成分,实现了现代与传统医药的结合,从多种角度共同起到提高免疫体、显著抗疲劳、显著抗氧化、显著调血糖、显著调血脂、显著解酒护肝、显著提高抗氧耐受力、显著清咽功能、显著增强改善性功能的作用,配方合理,功效确切。
如果机体基础营养不足或不平衡,会导致系统紊乱,如机体缺乏维生素B1、B2或不足,会使人感到乏力,肌肉运动无力,耐力下降。而此时补充功能性活性物所得的成效甚微,基本没有意义且造成浪费。本发明复合物所含海参肽、人参肽分子量低易吸收利用,再结合胶原蛋白肽、赤藓糖醇、黄精粉、枸杞粉、维生素B、维生素B6、牛磺酸等活性提取物来提升机体活力,缓解体力疲劳效果显著,营养素作为机体健康的基础平衡,天然食源性活性物作为机体健康的加强提升,二者结合科学且富有成效,共同温补调理机体,使机体焕发原生活力。从现代营养学角度上来说,更符合逻辑;从传药食同补理论上来说,科学配伍能有效达到益气固本、强精健体,全面温补强身,机体活力充沛的目的。
本发明复合物的制备工艺简单、稳定,克服了物料固体制剂强引湿吸附性带来的技术难题,大大提高了生产效率,且制备获得的颗粒复合物易成型,生产连续性好,产品质量稳定可控,服用方便,适口性好,易长期服用,无不良适应症。
具体实施方式
为让本发明的上述和其他目的、特征和优点能更明显易懂,下面特举较佳实施例,作如下详细说明,这些实施例是为了说明而不是以任何形式来限制本发明。
实施例1:
一种海参肽的制备方法,具体步骤如下:
S1:海参处理,将去除内脏的海参,粉碎至120目,4%的食盐水清洗2次,滤除水溶液后加入5倍量90%乙醇浸泡24h,滤除乙醇溶液后用纯净水清洗2次,再加入5倍量0.4%NaOH溶液浸泡15h,滤除NaOH溶液后用纯净水清洗2次;然后将处理后海参粗粉进行微波干燥,进一步粉碎至100目,备用;
S2:海参酶解,将海参粉与重量百分比浓度为2%的冰糖水溶液按重量比1:3~5的比例混合后放入打浆机内高速搅拌成海参匀浆液,再将海参匀浆液控制温度为45℃,并加入pH为8的0.01-0.03M的磷酸缓冲液以及复合碱性动物蛋白酶,复合碱性动物蛋白酶的添加量为海参匀浆液重量的3%,酶解45分钟,得海参酶解液;
S3:灭酶,将海参酶解液放入90℃水浴灭酶15分钟, 然后过滤除掉固体杂物,得灭酶海参匀浆液;
S4:干燥,将上述收集海参匀浆液浓缩至质量分数35-%,喷雾干燥,制得海参肽粉。
进一步的,S2中所述复合碱性动物蛋白酶为胰蛋白酶和风味蛋白酶,两者用量比例为4:3。
实施例2
一种人参肽的制备方法,具体步骤如下:
S1,人参预处理,取3-5年以上老参,清洗去泥及其它杂质,自然晾干后打碎过40目筛,得人参粗粉;
S2,低温浸泡浸提,将上述人参粗粉,加入15倍量水0-4℃ 浸泡18h,第一次真空浸提12h,收集滤液,再向药渣中加入10倍量水,第二次真空提取6h,收集滤液,合并两次真空浸提滤液,真空环境为170mmHg,浸提温度25℃,浸提搅拌转速为100rpm;
S3,浓缩、醇沉:浸提滤液过300目滤布得滤液,滤液静置2h无沉淀物,在70℃以下真空浓缩,滤液浓缩至相对密度为1.2;浓缩液冷却至室温,向浓缩液中缓慢加入2倍量95%乙醇并充分搅拌,静置8h,离心,收集静置后醇不溶沉淀得人参多糖提取物;
S4,复溶脱醇精制:将上述醇不溶沉淀加等体积50℃水溶解,搅拌,并在酒精回收装置中蒸馏脱乙醇;
S5,再浓缩:将上述复溶脱醇溶液在70℃以下真空浓缩,滤液浓缩至含水量为30%,得人参匀浆液;
S6:干燥,将上述浓缩人参匀浆液进行微波干燥后得块状固体提取物,经物理粉碎后粉末过100目筛,得人参肽粉。
实施例3
一种双参肽功能性固体饮料,由以下成分按重量百分比组成:胶原蛋白肽55%、人参肽粉12%、海参肽粉12%、赤藓糖醇3%、黄精粉8%、枸杞粉8%、维生素B0.03%、维生素B60.03%、牛磺酸0.1%、食用香精0.5%。
一种双参肽功能性固体饮料的制备方法,具体的步骤如下:
(1)预混:按组分依次将胶原蛋白肽、人参肽粉、海参肽粉、赤藓糖醇、黄精粉、枸杞粉投料于高速湿法混合制粒机,搅拌桨转速350转/分钟,制粒刀转速2000转/分钟,干料预混300s;

(4)总混:将步骤(3)所得干燥颗粒混合物、维生素B、维生素B、牛磺酸、食用香精按组分比投入三维混料机中混合30min得双参肽功能性固体饮料。
双参肽功能性固体饮料功效及安全性评价:
试验动物:健康12-16gBALB/c雄性幼鼠480只,动物房适应性喂养2天 后,分为3批,每批160只,每批随机分为16组,每组10只,其中一组作为阴性对照组,其余15组作为各供试物不同剂量试验组,第一批进行负重游泳实验,第二批进行血清尿素测定,第三 批进行血乳酸测定。屏障环境饲养。
阴性供试物:清洁级全价鼠颗粒饲料,蒸馏水。
灌胃剂量选择:每只小鼠每天灌胃量按0.2ml/10g.d计算,分别称取1g、2g、6g各供试物内容物,溶解于100ml蒸馏水中,分别制成低剂量0.2g/kg.bw,中等剂量0.4g/kg.bw,高剂量1.2g/kg.bw 灌胃物。每天经口灌胃1次,连续灌胃30天后测各项指标。实验期间各组小鼠正常摄食,自由饮水。
试验组设置
1阴性对照组:灌胃等剂量蒸馏水 ;
2低剂量组A:灌胃实施例1低剂量0.2g/kg.bw ;
3低剂量组B:灌胃实施例2低剂量0.2g/kg.bw ;
4低剂量组C:灌胃实施例3低剂量0.2g/kg.bw ;
5低剂量组D:灌胃实施例4低剂量0.2g/kg.bw;
6低剂量组E:灌胃实施例5低剂量0.2g/kg.bw;
7中剂量组A:灌胃实施例1低剂量0.4g/kg.bw ;
8中剂量组B:灌胃实施例2低剂量0.4g/kg.bw ;
9中剂量组C:灌胃实施例3低剂量0.4g/kg.bw ;
10中剂量组D:灌胃实施例4低剂量0.4g/kg.bw;
11中剂量组E:灌胃实施例5低剂量0.4g/kg.bw;
12高剂量组A:灌胃实施例1低剂量1.2g/kg.bw ;

试验方法及条件:
负重游泳实验:末次灌胃供试物30min后,置小鼠于30℃水中游泳,水深约30cm,水温25±1℃,鼠尾根部负重5%体重的铅块,记录小鼠自游泳开始至不能浮出水面的时间,作 为小鼠竭力游泳时间。
血清尿素测定:末次灌胃供试物30min后,置小鼠于30℃水中游泳90min,运动后安静60min拔眼球采血0.2ml左右,于4℃静置3小时后2000r/min离心15min,取血清20ul用全自动生化分析仪测定。
血乳酸测定:末次灌胃供试物30min后采血20ul,不负重在温度为30℃的水中游泳10min,分别于运动后0min和20min采血20ul,加入60ul溶血剂中,用乳酸分析仪测定。
结果分析:

由以上结果表明,本发明复合物中剂量组A、B,高剂量组A、B与对照组相比,小鼠负重游泳时间明显延长,差异显著;且中剂量组C、D、E,高剂量组C、D、E与对照组、中剂量组A、B,高剂量组A、B与对照组相比、相比,小鼠负重游泳时间明显延长,表明本发明固体饮料中各配伍组成具有协同作用。

由以上结果表明,本发明复合物中等剂量和高剂量具有明显加速清除疲劳小鼠血清尿素的作用。

注:血乳酸曲线下面积=5*(游泳前血乳酸值+3*游泳后0min血乳酸值+2*游泳后休息20min血乳酸值),与阴性对照组相比*P<0.05。
由以上结果表明,本发明复合物中等剂量和高剂量具有明显减少疲劳小鼠血乳酸增加的作用。
从以上试验结果可以看出,与阴性对照组相比本发明所述的复合物各剂量组连续灌胃小鼠30天,正常饲养生活状态良好,无不良现象;本发明的固体饮料的中等剂量(0.4g/kg.bw实施例3-5)和高剂量(1.2g/kg.bw实施例3-5)连续灌胃小鼠30天,能够显著延长小鼠负重游泳时间,加速运动后小鼠血清尿素的清除,减少小鼠运动后血乳酸的增加,且作用优于实施例1-2各剂量组;说明本发明所述的固体饮料具有缓解体力疲劳作用,显著提高抗氧耐受力,配伍合理,效果显著,且安全无毒副作用。
上述实施方案为本发明最佳的实施方案,但本发明的实施方案并不受上述实施方案的限制,其他的任何不违背本发明原理的条件下,可以通过改变参数的形式所产生的实施例,都包含于本发明的保护范围之内。
Claims (5)
1.一种双参肽功能性固体饮料,其特征在于,由以下成分按重量百分比组成:胶原蛋白肽40-55%、人参肽粉12-20%、海参肽粉12-20%、赤藓糖醇2-3%、黄精粉5-8%、枸杞粉5-8%、维生素B0.03-0.1%、维生素B60.03-0.1%、牛磺酸0.03-0.1%、食用香精0.1-1%。
2.根据权利要求1所述的一种双参肽功能性固体饮料,其特征在于,所述海参肽制备步骤如下:
S1:海参处理,将去除内脏的海参,粉碎至10-20目,2~5%的食盐水清洗1-2次,滤除水溶液后加入5-8倍量90%乙醇浸泡18-24h,滤除乙醇溶液后用纯净水清洗1-2次,再加入4-8倍量0.4%-0.6%NaOH溶液浸泡15-20h,滤除NaOH溶液后用纯净水清洗1-2次;然后将处理后海参粗粉进行微波干燥,进一步粉碎至80-100目,备用;
S2:海参酶解,将海参粉与重量百分比浓度为2~5%的冰糖水溶液按重量比1:3~5的比例混合后放入打浆机内高速搅拌成海参匀浆液,再将海参匀浆液控制温度为45~55℃,并加入pH为7-8的0.01-0.03M的磷酸缓冲液以及复合碱性动物蛋白酶,复合碱性动物蛋白酶的添加量为海参匀浆液重量的3~5%,酶解35~50分钟,得海参酶解液;
S3:灭酶,将海参酶解液放入90~100℃水浴灭酶8~15分钟,然后过滤除掉固体杂物,得灭酶海参匀浆液;
S4:干燥,将上述收集海参匀浆液浓缩至质量分数35-40%,喷雾干燥,制得海参肽粉。
3.根据权利要求2所述的一种双参肽功能性固体饮料,其特征在于,S2中所述复合碱性动物蛋白酶为胰蛋白酶和风味蛋白酶,两者用量比例为4:3。
4.根据权利要求1所述的一种双参肽功能性固体饮料,其特征在于,所述人参肽制备步骤如下:
S1,人参预处理,取3-5年以上老参,清洗去泥及其它杂质,自然晾干后打碎过40-50目筛,得人参粗粉;
S2,低温浸泡浸提,将上述人参粗粉,加入10-15倍量水0-4℃浸泡12-18h,第一次真空浸提 8-12h,收集滤液,再向药渣中加入5-10倍量水,第二次真空提取4-6h,收集滤液,合并两次真空浸提滤液,真空环境为 130-170mmHg,浸提温度25-35℃,浸提搅拌转速为60-100rpm;
S3,浓缩、醇沉:浸提滤液过300-400目滤布得滤液,滤液静置2h无沉淀物,在70℃以下真空浓缩,滤液浓缩至相对密度为1.2;浓缩液冷却至室温,向浓缩液中缓慢加入2-4倍量95%乙醇并充分搅拌,静置8-15h,离心,收集静置后醇不溶沉淀得人参多糖提取物;
S4,复溶脱醇精制:将上述醇不溶沉淀加等体积50℃水溶解,搅拌,并在酒精回收装置中蒸馏脱乙醇;
S5,再浓缩:将上述复溶脱醇溶液在70℃以下真空浓缩,滤液浓缩至含水量为30%-40%,得人参匀浆液;
S6:干燥,将上述浓缩人参匀浆液进行微波干燥后得块状固体提取物,经物理粉碎后粉末过80-100目筛,得人参肽粉。
5.一种双参肽功能性固体饮料的制备方法,其特征在于,具体的步骤如下:
(1)预混:按组分依次将胶原蛋白肽、人参肽粉、海参肽粉、赤藓糖醇、黄精粉、枸杞粉投料于高速湿法混合制粒机,搅拌桨转速350-420转/分钟,制粒刀转速2000-2800转/分钟,干料预混300-500s;
(2)制粒:加入预混干物料9%-15%倍量的95%乙醇,搅拌桨转速200-240转/分钟,制粒刀转速1500-1800转/分钟,混合制粒30-60s;
(3)干燥:过20-24目筛,于防爆热风循环干燥箱 50-60℃干燥60-80min,过16-18目筛整粒,至细粉率小于5%,得干燥颗粒混合物;
(4)总混:将步骤(3)所得干燥颗粒混合物、维生素B、维生素B、牛磺酸、食用香精按组分比投入三维混料机中混合30-40min得双参肽功能性固体饮料。
