In the vacuum concentration process, the vacuum concentration pressure is less than 1.60Mpa, the vacuum concentration temperature is less than or equal to 48 ℃, and the end point of the vacuum concentration process is that the Baume value of the second concentrated solution is 28-32. In this example, the active ingredients in the raw materials were reduced by low-temperature concentration.
In the solidification process, the solidification pressure is less than-0.97 Mpa, and the solidification temperature is 35-45 ℃. In this embodiment, the second concentrated solution is naturally solidified, the extract of the prepared traditional Chinese medicine composition is in a loose powder form with a certain particle size, no other auxiliary materials are required to be added, no granulation is required, and the extract of the traditional Chinese medicine composition can be directly used as a solid beverage-like pharmaceutical preparation, which is completely different from the traditional Chinese medicine granule. Specifically, the particle size of the extract is 0.1 mm-0.2 mm.
According to the embodiment of the invention, the raw materials of the traditional Chinese medicine composition are sequentially extracted, microfiltered, concentrated by a membrane and concentrated in vacuum, no auxiliary material is added, and finally, the raw materials are naturally solidified into loose powder with certain granularity directly under the environment of negative pressure, low temperature and drying, so that the extract is obtained.
The extract prepared by the embodiment is in a loose powder shape with certain granularity, can be directly brewed by water, the soup color after brewing is clearer than that of the traditional Chinese medicine granule, and meanwhile, the amount of the extract required by brewing is less than that of the traditional Chinese medicine granule. That is to say, the extract prepared by the embodiment can be used as solid beverage for production, can meet large-scale and standardized production, can be rapidly put on the market, and meanwhile, the solid beverage is free of decoction, can be dissolved after being mixed with water, and is convenient and rapid.
Of course, it will be understood by those skilled in the art that the extract of the above-mentioned Chinese medicinal composition can be made into other oral dosage forms, such as capsule, tablet, oral liquid or syrup, besides being directly used as granule.
The traditional Chinese medicine composition, the extract of the traditional Chinese medicine composition and the pharmaceutical preparation containing the extract are suitable for preventing new coronary pneumonia, and are particularly suitable for preventing new coronary pneumonia in young adults.
Example 1
This embodiment 1 provides a method for preparing an extract of a traditional Chinese medicine composition, which specifically includes the following steps:
step 1, pretreatment: 1 ton of total material, and the following weight ratio: 11.6% of honeysuckle, 11.6% of dandelion, 7.7% of houttuynia cordata, 7.7% of wrinkled gianthyssop herb, 7.7% of poria cocos, 7.7% of coix seed, 4.6% of dried orange peel, 4.6% of mint, 4.6% of platycodon grandiflorum, 23% of fresh reed rhizome, 4.6% of ginger and 4.6% of Chinese date.
Crushing the raw materials of dandelion, houttuynia cordata, agastache rugosus, poria cocos, mint, dried orange peel, platycodon grandiflorum and Chinese date, drying the raw materials for 0.5-1 h at 75-80 ℃ by using a dryer if the water content of the raw materials is large, crushing the crushed raw materials, directly crushing coix seed and honeysuckle, cutting fresh reed rhizome and ginger by using a cutting machine, and then crushing by a wet method, wherein the crushed fine powder can pass through a 30-mesh screen.
Step 2, wall breaking extraction: the method comprises the steps of adopting a 500L extraction tank, putting raw materials into the extraction tank in batches of about 20kg each time, adding purified water according to the proportion of 1:20, enabling the temperature of feed liquid to be 25-30 ℃, uniformly stirring, and then conveying the feed liquid to an extraction wall breaking machine set (extractor) for extraction, wherein the extraction time is 20-30 min.
Step 3, centrifugation: centrifuging the extracting solution, wherein the frequency of a main centrifugal machine is 42.0Hz, the frequency of an auxiliary machine is 32.0Hz, the feeding frequency is 14.0Hz to 18.0Hz, and 5 percent to 10 percent of pure water is additionally added in the centrifuging process.
Step 4, microfiltration: and (3) filtering the centrifuged feed liquid by using a ceramic membrane, wherein the operating pressure is lower than 0.28MPa, the temperature is less than or equal to 50 ℃, and 15-20% of pure water is additionally added in the microfiltration process.
And step 5, membrane concentration: the microfiltration clear liquid is subjected to membrane concentration, the operation pressure is lower than 1.60MPa, and the temperature is less than or equal to 48 ℃. And when the membrane concentrated feed liquid reaches 0.5-1.5 Baume or the concentration cycle time of the feed liquid exceeds 4 hours, receiving the feed liquid.
Step 6, vacuum concentration: and (3) carrying out vacuum concentration on the membrane concentrated solution, wherein the concentration temperature is 35-45 ℃, and the vacuum degree is lower than-0.095 MPa. And collecting when the concentrated extract is 28-32 Baume.
Step 7, natural solidification: naturally solidifying the concentrated extract at 30-45 deg.C under vacuum degree lower than-0.097 MPa.
Application example
According to the application example, the effects of the extract prepared in the embodiment 1 of the invention on clearing heat, dispelling wind, clearing lung, detoxifying, expelling pathogenic factors, tonifying spleen, eliminating dampness, relieving cough, reducing sputum and the like are researched through group comparison investigation.
In this application example, 300 persons were randomly invited to the experiment, and 300 persons were divided into 3 groups of 100 persons on average. The first group provides one dose of the product twice a day for each person, and each dose is one gram of the product; a second group provides twice daily, two doses per person, one gram of the product per dose; the third group provides the product twice a day for each person, three doses each time, and one gram of the product; for four weeks, the effects of clearing heat, dispelling wind, clearing lung, removing toxic substances, expelling pathogenic factors, removing dirt, invigorating spleen, eliminating dampness, relieving cough and reducing sputum of all experimental participants were investigated once a week, and the investigation results are shown in fig. 1.
Referring to fig. 1, ninety percent of the persons participating in the experiment considered that the product has better effects in clearing heat and dispersing wind, clearing lung and detoxifying, expelling pathogenic factors and removing dirt, strengthening spleen and eliminating dampness, relieving cough and reducing sputum, and the like, wherein the second group has the highest satisfaction degree in four surveys, that is, each person provides twice a day, two doses each time, and the administration mode of one gram of the product is the most economical and the best effect.

a membrane concentration step of performing membrane concentration on the ceramic membrane clear solution to obtain a first concentrated solution;
