本发明公开了一种黑青稞黑藜麦组合物、制备方法及其用途,其原料药包括以下重量份计的组分:黑青稞1~7份,黑藜麦1~10份,将原料药用乙醇和水提取后,组合物包括黑青稞提取物2~50份,黑藜麦提取物5~25份,进一步为黑青稞提取物10~20份,黑藜麦提取物12~15份。本发明组合物生物活性高,具有抗疲劳作用。

本发明属于医药领域,具体涉及一种黑青稞黑藜麦组合物、制备方法及其用途。
背景技术
青稞(Hordeumvulgare ssp.vulgare)是一种生长在高原低温低氧环境中的农作物,又称裸大麦,是禾本科大麦属作物。在世界许多国家和地区作为主要粮食作物已有几千年历史。青稞分为白青稞、黑青稞、墨绿色青稞等种类,其中黑青稞是西藏特有品种,喜马拉雅山北麓的山南地区措美县因气候、土壤特殊而盛产黑青稞,其脂肪、粗纤维和花青素含量高,具有很好的保健功能,深受食品加工企业和市场的青睐。
黑藜麦(Chenopodium quinoa willd),又叫南美藜,奎麦,金谷子等。原产于美洲安第斯山区,是印加土著居民的主要传统食物,由于具有独特丰富全面的营养价值,养育了印加民族,被称为粮食之母,也可代替婴幼儿奶粉食用。黑藜麦属藜木科,性平味甘,黑藜麦含有丰富的蛋白质,碳水化合物、脂肪、多种氨基酸、其中含有人体必需的全部9种氨基酸,比例适当且易吸收,还富含不饱和脂肪酸、花青素、类黄酮、β-葡聚糖、胆碱、甜菜碱、α一亚麻酸、矿物质钙、镁、磷、钾、铁、锌、硒、锰、铜等,维生素B族、叶酸、维生素E等化合物,膳食纤维含量高达7.1%,不含胆固醇,具有全面均衡补充营养,能够调节内分泌、促进儿童生长发育、预防疾病、减肥、通便、防癌抗癌、降血压、降血脂、降血糖、防治心脑血管疾病、辅助治疗慢性疾病等功效。
发明内容
本发明的目的在于提供一种黑青稞黑藜麦组合物、制备方法及其用途,生物活性高,具有提高抗疲劳和抗氧化的作用。
为了实现上述目的,本发明提供了以下技术方案:
一种组合物,其原料药包括如下重量份原料:黑青稞1~7份,黑藜麦1~10份。
进一步地,包括黑青稞1~3.3份,黑藜麦1~6.5份;
优选为包括黑青稞1~2.4份,黑藜麦1~1.7份;
更优选为包括黑青稞1份,黑藜麦1.7份。
本发明还提供了上述组合物的制备方法,包括如下内容:
(1)按重量份取原料药;
(2)取黑青稞提取物、黑藜麦提取物,制备产品即得。
黑青稞提取物包括醇提物和水提物,所述黑藜麦提取物包括醇提物和水提物;
进一步地,所述醇提物的醇选自甲醇、乙醇、丁醇、戊醇中的一种或几种,优选乙醇;进一步为20~100%v/v乙醇水溶液,优选35~95%v/v。
在本发明的具体实施方式中,黑青稞醇提物:黑青稞水提物质量比为1~1.5:1,优选1.5:1,黑藜麦醇提物:黑藜麦水提物质量比为0.2~2:1,优选1.4:1。
本发明还提供了一种组合物,包括以下重量份计的组分:黑青稞提取物2~60份,黑藜麦提取物5~30份,优选黑青稞提取物10~20份,黑藜麦提取物12~15份;
在本发明的具体实施方式中,所述黑青稞提取物为黑青稞醇提物和/或黑青稞水提物,所述黑藜麦提取物为黑藜麦醇提物和/或黑藜麦水提物;

本发明所述“可接受的辅料”是指包括任意不干扰活性成分的生物活性的有效性且对它被给予的宿主无毒性的物质。
本发明所述药学上可接受的辅料,是药物中除主药以外的一切附加材料的总称,辅料应当具备如下性质:(1)对人体无毒害作用,几乎无副作用;(2)化学性质稳定,不易受温度、pH、保存时间等的影响;(3)与主药无配伍禁忌,不影响主药的疗效和质量检查;(4)不与包装材料相互发生作用。

本发明具有如下有益效果:
(1)本发明提取方法步骤简单、简便快捷,可有效降低黑青稞、黑藜麦有关产品的成本,值得推广。
(2)本发明提供了一系列由黑青稞水提物和黑青稞醇提物、黑藜麦水提物、黑藜麦醇提物组成的组合物,并通过试验证明了组合物能够有效抗疲劳和抗氧化,可用于制备相关产品。

下面对本发明的技术方案进行清楚、完整的描述,显然,所描述的实施例是本发明的一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域技术人员在没有做出创造性劳动的前提下所做出的的所有其他实施例,都属于本发明的保护范围。
实施例1
黑青稞提取物、黑藜麦提取物的制备
(1)黑青稞醇提物的制备:取干燥至恒重的黑青稞干粉分别加入95%v/v、85%v/v、75%v/v、65%v/v、55%v/v、45%v/v、35%v/v乙醇回流提取7次,过滤,药渣备用,合并提取液,回收乙醇,干燥,即得黑青稞醇提物,经测定,出膏率约为29.93%。
(2)黑青稞水提物的制备:取步骤(1)中的药渣,加水煎煮5次,过滤,合并滤液,浓缩,干燥,即得黑青稞水提物,经测定,出膏率约为20.18%。
(3)黑藜麦醇提物的制备:取干燥至恒重的黑藜麦干粉分别加入95%v/v、85%v/v、75%v/v、65%v/v、55%v/v、45%v/v、35%v/v乙醇回流提取7次,过滤,药渣备用,合并提取液,回收乙醇,干燥,即得黑藜麦醇提物,经测定,出膏率约为21.38%。
(4)黑藜麦水提物的制备:取步骤(1)中的药渣,加水煎煮5次,过滤,合并滤液,浓缩,干燥,即得黑藜麦水提物,经测定,出膏率约为15.37%。
上述出膏率可在5%范围内正常波动。
实施例2
取实施例1制备的黑青稞醇提物6份、黑青稞水提物4份、黑藜麦醇提物7份、黑藜麦水提物5份混匀后,即得本发明组合物。
实施例3
取实施例1制备的黑青稞醇提物12份、黑青稞水提物8份、黑藜麦醇提物7份、黑藜麦水提物5份混匀后,即得本发明组合物。
实施例4
取实施例1制备的黑青稞醇提物12份、黑青稞水提物8份、黑藜麦醇提物4份、黑藜麦水提物5份混匀后,即得本发明组合物。

取实施例1制备的黑青稞醇提物12份、黑青稞水提物8份、黑藜麦醇提物4份、黑藜麦水提物5份,加入适量微晶纤维素,混匀后,干燥,装胶囊,即得本发明组合物的胶囊剂。
实施例6抗疲劳作用的检测
取雄性ICR小鼠,重量20-22g,每组10只,随机分组,设对照组和不同配伍药物组,用于本发明各提取物的配比筛选。不同配比的黑青稞提取物与黑藜麦提取物组给药量为0.25mL/kg,15d,灌胃前将各组给药液体量统一稀释为0.01mL/g。对照组给予同体积蒸馏水,于末次给药30min后,在小鼠鼠尾根部装载5%体重铅条,放入水深30cm、水温25℃的水箱中游泳,记录小鼠自游泳开始至力竭的时间(力竭标准为小鼠沉入水面以下10s不能浮出)。

从表1的数据可以看出,黑青稞醇提物、黑青稞水提物、黑藜麦醇提物、黑藜麦水提物均能延长小鼠负重游泳时间,具有一定的抗疲劳作用,但延长时间较短;将黑青稞醇提物、黑青稞水提物、黑藜麦醇提物、黑藜麦水提物四者配伍后,小鼠负重游泳的时间均长于任意单一提取物的效果,说明黑青稞醇提物、黑青稞水提物、黑藜麦醇提物、黑藜麦水提物配伍后协同增效,提升了提取物的抗疲劳活性,其中,黑青稞醇提物:黑青稞水提物:黑藜麦醇提物:黑藜麦水提物=6:4:7:5时小鼠负重游泳的时间最长,可用于制备治疗预防和/或治疗相关疾病的产品。
实施例7氧化酶含量的检测
取重量20-22g的雄性ICR小鼠,每组10只,随机分组,分别为正常对照组、衰老模型组、维生素E组(阳性对照组)、不同配伍的药物组,用于验证本发明提取物的抗氧化作用。正常对照组对小鼠每天皮下注射等体积的生理盐水,其余各组以150mg(/kg·d)连续55d皮下注射D-半乳糖,制备小鼠亚急性过氧化损伤模型,通过行为学检测显示造模成功。首次注射D-半乳糖后次日开始按150g(/kg·d)灌胃给药,实验组灌胃相应剂量不同配比的黑青稞提取物与黑藜麦提取物组,阳性对照组以50mg(/kg·d)维生素E代替,正常对照组和衰老模型组给予饮用水。各组小鼠同室分笼饲养,自由进食。末次给药后禁食24h,摘眼球取血,小鼠脱颈处死。血浆于4℃低温3500r/min离心10min,分离血清,按照试剂盒说明书测定测定SOD水平和MDA水平。

从表2的数据可以看出,单独使用黑青稞水提物、黑藜麦水提物时,虽然丙二醛(MDA)的含量下调了,但超氧化物歧化酶(SOD)含量却下降了,而黑青稞醇提物、黑藜麦醇提物单独使用时均能提高SOD的含量,且能下调MDA含量,但效果较弱;将黑青稞醇提物、黑青稞水提物、黑藜麦醇提物、黑藜麦水提物四者配伍后,小鼠血清中的SOD的含量均高于任意单一提取物或两两提取物使用的效果,MDA含量均低于任意单一提取物或两两提取物使用的效果,说明黑青稞醇提物、黑青稞水提物、黑藜麦醇提物、黑藜麦水提物配伍后协同增效,提升了提取物的抗氧化活性,其中,黑青稞醇提物:黑青稞水提物:黑藜麦醇提物:黑藜麦水提物=12:8:4:5时,抗氧化作用最强。

Claims (10)
1.一种组合物其特征在于,其原料药包括如下重量份原料:黑青稞1~7份,黑藜麦1~10份。
2.根据权利要求1所述的组合物,其特征在于,包括黑青稞1~3.3份,黑藜麦1~6.5份;优选为包括黑青稞1~2.4份,黑藜麦1~1.7份;更优选为包括黑青稞1份,黑藜麦1.7份。
3.权利要求1或2所述组合物的制备方法,其特征在于,包括如下内容:
(1)按重量份取原料药;
(2)取黑青稞提取物、黑藜麦提取物,制备产品即得;
所述黑青稞提取物包括醇提物和水提物,所述黑藜麦提取物包括醇提物和水提物;
进一步地,所述醇提物的醇选自甲醇、乙醇、丁醇、戊醇中的一种或几种,优选乙醇;
进一步为20~100%v/v乙醇水溶液,优选35~95%v/v。
4.根据权利要求3所述组合物的制备方法,其特征在于,黑青稞醇提物:黑青稞水提物质量比为1~1.5:1,优选1.5:1,黑藜麦醇提物:黑藜麦水提物质量比为0.2~2:1,优选1.4:1。
5.一种组合物,其特征在于,包括以下重量份计的组分:黑青稞提取物2~60份,黑藜麦提取物5~30份,优选黑青稞提取物10~20份,黑藜麦提取物12~15份;
进一步地,所述黑青稞提取物为黑青稞醇提物和/或黑青稞水提物,所述黑藜麦提取物为黑藜麦醇提物和/或黑藜麦水提物;
进一步地,所述醇提物的醇选自甲醇、乙醇、丁醇、戊醇中的一种或几种,优选乙醇;
进一步为20~100%v/v乙醇水溶液,优选35~95%v/v乙醇水溶液。
6.根据权利要求5所述组合物,其特征在于,包括黑青稞醇提物1~30份、黑青稞水提物1~30份、黑藜麦醇提物1~20份、黑藜麦水提物5~15份;
优选为包括黑青稞醇提物3~24份、黑青稞水提物2~24份、黑藜麦醇提物1~14份、黑藜麦水提物5~10份;更优选为黑青稞醇提物6~12份、黑青稞水提物4~8份、黑藜麦醇提物7份、黑藜麦水提物5份;
进一步地,黑青稞醇提物:黑青稞水提物质量比为1~1.5:1,优选1.5:1,黑藜麦醇提物:黑藜麦水提物质量比为0.2~2:1,优选1.4:1。
7.根据权利要求6所述的组合物,其特征在于,包括黑青稞醇提物6份、黑青稞水提物4份、黑藜麦醇提物7份、黑藜麦水提物5份;
或,包括黑青稞醇提物12份、黑青稞水提物8份、黑藜麦醇提物7份、黑藜麦水提物5份。
8.一种产品,其特征在于,包括权利要求1、2、5~8任一项所述的组合物;进一步地,还包括药学上可接受的辅料;进一步地,所述药学上可接受的辅料为微晶纤维素;进一步地,所述产品的剂型选自片剂、散剂、丸剂、胶囊剂、颗粒剂、溶液剂、糖浆剂、乳剂、混悬剂、膏剂、气雾剂、喷雾剂或酊剂,优选为胶囊剂。
9.根据权利要求8所述的产品,其特征在于,所述产品为抗疲劳产品。
10.权利要求1、2、5~10任一项所述的组合物在制备抗疲劳产品中的用途;进一步地,所述抗疲劳产品为提升体能的产品。
