Abstract
本发明公开了一类基于竹荪蛋提取物的美容外用品或可食用产品及应用,属于生物医药领域。该产品由以天然竹荪蛋提取物为主体,复合具有延缓衰老、保湿、抗辐射功能的药食同源材料组合而成,各材料功能互补。本发明外用可预防或治疗血管异常,皮脂分泌过多相关的问题,同时可抗衰老、促愈合、保湿、抗过敏。内服可增强机体的抗氧化能力、改善肠道菌群、提高机体免疫,从而达到延缓衰老养颜美容的效果。此外,本发明原材料获取容易、制备方法简便,绿色安全,具有很好的应用前景和市场前景。

本发明属于生物医药制品技术领域,涉及一类基于竹荪蛋提取物的美容外用品或可食用产品及应用。
背景技术
随着年龄增大,环境污染加剧,皮肤逐渐老化,出现角质化、皱纹、斑点等衰老现象。美容的方式有很多,比如多喝水、锻炼身体、多吃水果和纤维素,但这些都是日常的基础调节方法,如果想在短期内快速有效的改善皮肤问题,需内调外敷多途径干预。
经常涂抹保湿抗氧化护肤品有助于修复肌肤,抵抗外界环境造成的不良影响,使皮肤保持柔软光滑。市场上的美容产品虽种类繁多,但是质量参差不齐,由于化学成分及重金属添加,尽管美白效果显著,却存在潜在的不可逆损伤风险。因此,急需高安全性和高有效性相结合的美容护肤产品。对人体无害的纯天然植物活性成分因其效用温和、不良反应小而得到大家的一致认可,抑制黑色素产生的绿色天然活性药物的开发将成为化妆品领域一个备受瞩目的课题。
竹荪是鬼笔科竹荪属中珍贵的食用菌,素有“菌中皇后”、“雪裙”仙子的美称。竹荪蛋是竹荪生长前期的胚胎,包含菌托、明胶体和胎竹荪。竹荪蛋营养价值很高。竹荪蛋作为一种生长在竹林的菌类胚胎外衣,菌肉肥硕、肉质滑嫩;含有维生素B2、大量蛋白质、多糖及锌、铁、磷、钙等多种微量元素,具有健脾益胃,益气补脑的作用,属菌类之上品。竹荪蛋中含有21种氨基酸,8种人体所必需氨基酸。现代研究表明,竹荪蛋中还含有活性多糖、总黄酮、凝集素等生物活性成分。竹荪多糖广泛存在于竹荪蛋子实体的细胞壁中,分子量14~380kDa,主要由葡萄糖(D-Glc)、甘露糖(D-Man)、半乳糖、木糖(D-Xyl)、L-岩藻糖等单糖组成,具有抗氧化、免疫调节、抑菌、护肝、抗肿瘤等多种生物活性,在医药(抗癌、抗菌、降血脂)与食品方面(防腐)等方面有极大的应用潜力。
皮肤衰老主要是由脱水、紫外辐射等因素诱导的自由基氧化造成。当前还未有将竹荪蛋提取物用于美白、抗衰老及防晒等方面产品开发。
发明内容
本发明的目的在于提供一类基于竹荪蛋提取物的美容外用品或可食用产品及应用
为了达到上述目的,本发明采用以下技术方案予以实现:
本发明公开了竹荪蛋提取物在制备美容美肤外用品或可食用产品中的应用,所述竹荪蛋提取物包括竹荪多糖提取物或竹荪蛋醇提物;
所述竹荪蛋多糖提取物制备方法如下:取竹荪蛋,清洗后打浆,加入竹荪蛋10倍体积的水,75℃~85℃浸提5~8小时,分离得到提取液,真空浓缩至原体积的1/3,活性炭脱色,3倍体积乙醇过夜沉淀,收集沉淀,水溶脱醇,真空浓缩,浓缩液120℃~140℃的温度下喷雾干燥,制得粉末状的竹荪蛋多糖提取物;
所述竹荪蛋醇提物的制备方法如下:竹荪蛋清洗后,以75%乙醇作为浸提试剂,料液比1:15,打浆30秒,75~80℃浸提3h,过滤,真空浓缩至1/5体积,乙酸乙酯萃取浓缩,得竹荪蛋醇提物。
进一步地,所述美容美肤外用品包括凝胶面膜、竹荪蛋多糖提取物-环糊精包合物凝胶和竹荪蛋多糖提取物精华液。
进一步地,所述可食用产品包括养肤茶包和速溶饮料。

将以上原料在室温下加水形成凝胶,制得竹荪多糖提取物-环糊精包合物凝胶;

与现有技术相比,本发明具有以下有益效果:
本发明公开了竹荪蛋提取物在制备美容美肤外用品或可食用产品中的应用,竹荪蛋提取物包括竹荪蛋多糖提取物和竹荪醇提物,其中竹荪蛋多糖在体内体外多种抗氧化体系中表现出还原能力和捕获自由基等抗氧化能力,还可以改善SOD活性、显著降低机体组织脂褐素的含量,高度的持水保湿能力。实验结果表明,本发明的外用产品可预防或治疗血管异常,皮脂分泌过多相关的问题,同时可抗衰老、促愈合、保湿、抗过敏。内服产品能够增强机体的抗氧化能力、改善肠道菌群、提高机体免疫,从而达到延缓衰老养颜美容的效果。此外,本发明原材料获取容易、制备方法简便,绿色安全,因此具有很好的应用前景和市场前景。

下面结合附图对本发明做进一步详细描述:
1、竹荪蛋多糖提取物制备及性能测定
竹荪蛋清洗后打浆,加10倍体积水,75℃~85℃浸提5~8小时,分离得到提取液,真空浓缩至原体积的1/3,活性炭脱色,3倍体积乙醇过夜沉淀,收集沉淀,水溶脱醇,真空浓缩,浓缩液120℃~140℃的温度下喷雾干燥,获得竹荪蛋多糖提取物细粉。
面膜精华液的制备:以质量百分比计,包括竹荪蛋多糖提取物12%,透明质酸2%,卡波姆0.5%,寡肽-1 3%,六肽-10 5%,玫瑰精油2%,丙二醇1%、竹荪蛋醇提物2%,乳化剂0.5%,余量为水。
竹荪蛋多糖提取物具有吸水性、成膜性;透明质酸具有保湿特点;寡肽-1,六肽均为美容肽,对损伤皮肤具有修复功效;玫瑰精油促进各功能性物质吸收,其天然香味给乳液带来宜人的芳香;竹荪蛋醇提物能抑菌,防腐。
采用体外法,以甘油(质量分数w=1%)、去离子水为对照,研究竹荪蛋提取物的保湿性能。竹荪蛋提取物的平均失水率均低于对照去离子水,略低于甘油水溶液(质量分数w=1%),由此证明此种竹荪蛋多糖提取物具有良好的保湿效果。
采用紫外分光光度法,测定防晒性能。将竹荪蛋多糖提取物用50%乙醇稀释后,以50%乙醇作为空白对照,测定各样品在UV-C区(200~280nm),UC-B区(280~320nm)和UV-A区(320~400nm)的平均透光率,并换算成平均紫外线吸收率。同样方法测定面膜精华液的紫外线吸收率。结果参见图1,图1中线条从上到下分别为10%竹荪蛋多糖提取物(蓝色线条)、5%竹荪蛋多糖提取物(红色线条)、50%含竹荪蛋多糖提取物的面膜精华液(绿色线条)以及50%不含竹荪蛋多糖提取物的面膜精华液(紫色线条),可以看出竹荪蛋多糖提取物及不同复配样品,在280~400nm内的平均紫外吸收率基本都在80%以上,大部分的UVB和UVA均可被吸收,说明竹荪蛋多糖提取物及复配产品具有广谱防晒作用。
研究竹荪蛋多糖提取物和面膜精华液的抑制羟自由基能力、抑制超氧阴离子自由基能力。结果表明,竹荪蛋多糖提取物抑制羟自由基能力为2.19(U/L),面膜精华液抑制羟自由基能力为1.18(U/L);竹荪蛋多糖提取物抑制超氧阴离子自由基能力为29.46(U/L),面膜精华液抑制超氧阴离子自由基能力为8.70(U/L)。
测定竹荪蛋多糖提取物稀释后对酪氨酸酶的抑制率,证明了当质量分数为1%时竹荪蛋提取物就可以有效的抑制酪氨酸酶的活力,最高可达88.61%。
以上实验结果说明竹荪蛋多糖提取物具有良好的保湿、紫外线防护、抗氧化及美白效果。
2、可食美容产品制备
本研究在探讨竹荪多糖的吸湿保湿性、抗氧化性、美白活性的基础上,将其作为生物活性物质,开发系列可食美容产品。
1)养肤茶包
将竹荪蛋多糖提取物、柿叶、甘草组合在一起,制成茶包,具有健脾渗湿、清热润肤等功效,常服可增加血管弹性,减少面部皱纹。适合扁平疵、面部紫瘫等慢性皮肤病患者的保健美容。
配方:竹荪蛋多糖提取物5g、柿叶粉10g、甘草粉15g。
精选优质的原材料,80~100℃烘干后研磨成粉末,装入透气性良好的无纺布袋中,布袋开口处密封,即可得到成品。饮用时,将茶包置于80~90℃开水中冲泡5~10分钟即可。
配方中,柿叶为柿科柿属植物柿Diospyros kaki Thunb.的新鲜或干燥叶,味微甘、苦,性凉。能清热止咳,止血,生津。柿叶在医疗与养生保健领域均有应用,柿叶可以制茶,日本民间素有饮柿叶茶的习惯。现代研究表明,柿叶中含胡萝卜素和丰富的维生素,以嫩叶含量较高又含黄酮苷、鞣至、酚类、香豆精类、挥发油。柿叶茶可以作为一种健身饮料,沏水后清香扑鼻,无毒,利尿,经常饮用能增进机体的新陈代谢、利小便、通大便、净化血液、使机体组织细胞复苏。柿叶黄酮能明显抑制由TNF诱导的大鼠血管平滑肌细胞凋亡信号调节激酶I蛋白的表达,可治疗血管疾病。柿叶提取物有降低链脲佐菌素致糖尿病小鼠模型的血糖水平、改善胰岛素抵抗、调节脂代谢紊乱的作用,同时还有明显的抗氧化和增强糖尿病小鼠免疫功能和显著改善实验糖尿病小鼠的胰岛素抵抗等作用。从柿叶中所得5种三萜类化合物可抑制由刺激引起的超氧化物的生成和酪酸磷酸化。柿叶对金黄色葡萄球菌、卡他球菌有一定抑制作用,也具有止血作用。柿叶中多酚类物质具有高效清除自由基的作用。柿叶对酪氨酸酶的抑制效果明显。柿叶干燥后粉碎,加10倍体积75%乙醇75℃提取1~2小时,2次。过滤、合并滤液,浓缩,乙酸乙酯萃取,取乙酸乙酯相,减压浓缩,真空冷冻干燥,得柿叶黄酮。
甘草,为豆科多年生草本植物甘草、胀果或光果甘草的干燥根及根茎。性味甘、平,归心、肺、脾、胃经,具有益气补中、祛痰止咳、缓急止痛、调和药性及清热解毒的功效。主要含有甘草酸、甘草甜素等三帖皂苷和甘草素等多种黄酮类成分。甘草提取物被广泛用于各类化妆品中。其所含的黄酮类成分能有效降低酪氨酸酶活性,减慢皮肤中黑色素的形成,同时又可调理皮肤,缓解皮肤的紧张状态甘草酸有表面活性剂样作用,具有广泛配伍性,与其他活性剂同用,可加速皮肤对它们的吸收甘草提取物还具有防晒、止痒以及生发护发作用。
2)速溶饮料
以质量百分比计,取竹荪多糖提取物10%~20%,加入0.1%~0.5%蛋白糖、0.5%~2%柠檬酸、0.2%~0.3%复合香精、25%~40%麦芽糊精和5%~10%β-环糊精,喷雾干燥制成颗粒。使用时,颗粒能够速溶于水中,配制成速溶饮料。
3、外用美肤产品1)凝胶面膜
以质量百分比计,取魔芋精粉3%,甘油6%,竹荪蛋多糖提取物0.2%,山梨醇2%,余量为水,在室温条件下将上述原料加水形成凝胶。该产品无毒、绿色、无刺激,具有抗氧化、保湿的功效。
2)竹荪提取物-环糊精包合物凝胶
柿叶黄酮1.0%,β-环糊精6%,0.2%卡波姆,竹荪多糖提取物5%,透明质酸钠2%,余量为水,将原料在室温条件下加水形成凝胶。
该产品能有效的清除面部氧自由基和促进细胞新陈代谢,具有抗氧化、舒展皱纹,淡化细纹的功效,促进受损肌肤修复,强化补水的效能;同时,具有抑菌,抑制皮脂腺分泌,缓解痤疮症状的作用。
3)面膜精华液
以质量百分比计,包括竹荪多糖提取物12%,透明质酸2%,卡波姆0.5%,寡肽-13%,六肽-10 5%,玫瑰精油2%,丙二醇1%、竹荪蛋醇提物2%,乳化剂0.5%,余量为水。
竹荪多糖提取物具有吸水性、成膜性;透明质酸具有保湿特点;寡肽-1,六肽均为美容肽,对损伤皮肤具有修复功效;玫瑰精油促进各功能性物质吸收,其天然香味给乳液带来宜人的芳香;竹荪蛋乙醇提取物能抑菌,防腐。复合后该产品无毒、绿色、无刺激,同时具有美白、抗糖化、抗衰老、保湿、调整皮脂腺分泌等功效,可长期使用。
上述5类产品所用所有材料均为食品级安全材料,可同时内服外用。
4、本发明制得的美容产品性能检测及安全评估
1)抑菌性测试
以大肠杆菌为试验菌种,测试抗菌效果。将所制备的活性样品稀释后,浸入滤纸,在37℃下平板固体培养,培养18h后观察抑菌效果。参见图2,图中样品(3)指凝胶面膜,样品(4)指竹荪提取物-环糊精包合物凝胶,样品(5)指面膜精华液,这3类外用产品抑菌环宽度分别为3mm,3mm,2.5mm,结果表明,本申请的三种外用产品均已达到良好的抗菌效果。
2)体外保湿实验
实验中选用0.9cm×5.5cm大小的医用透气胶带,并将其贴在玻璃板上。取样品0.198g均匀涂敷在医用透气胶带表面(相当于40mg/cm2),放入湿度为31%(CaCl2饱和溶液来控制湿度),温度为25℃的干燥器中,定时称量,并计算保湿率,结果如图3所示,表明本发明制得的的3类外用产品均表现出很好的保湿性能。
3)保湿功效证明试验
选取30位年龄在22-45周岁女性志愿者,志愿者面部有不同程度的脱皮、干燥紧绷的症状。使用上述本发明制得的3类外用产品(每个产品10例),连续使用2周。检测标准:(1)明显改善:基本解决面部脱皮、皮肤干燥紧绷的症状;(2)轻微改善:对面部脱皮、皮肤干燥紧绷的症状有所减轻;(3)无改善:使用前后无差别。结果表明,凝胶面膜和竹荪提取物-环糊精包合物凝胶对所有受试者均有轻微改善作用,对80%受试者有明显改善作用。产品精华液对90%受试者有轻微改善作用。
4)稳定性测试
将上述三个外用样品分别在-20℃、4℃、室温和48℃四个条件下循环,每个时间点持续12小时(连续变换四个温度条件)、光照条件下放置8周,料体颜色和形态都保持稳定。
5)皮肤安全测试
将上述制得的3类外用产品,每个选择20名左右18-60岁、非高度敏感体质的志愿者作为受试对象,进行皮肤刺激性检测。在上肢大臂处的不同部位选取面积大概为5×5cm2的皮肤区域,用棉棒沾取酒精擦拭干净,同时始终要保持受试部位的清爽与干燥,且避免接触其它外用制剂(如其他化妆护肤产品或药用试剂)。
将实验物(凝胶面膜、竹荪提取物-环糊精包合物凝胶、精华液)0.3-0.5g/mL,剪取5×5cm2左右面积大小的被精华液饱和的无纺布面贴型面膜)每天2次均匀地涂敷于受试部位,膏霜和液体状态产品的厚度大概为1mm左右,面膜制剂涂敷20min后洗净。在整个实验过程中,志愿者的实验部位都需要保持清洁及干燥,24小时后观察皮肤反应。如此操作连续7天后进行观察。证实对皮肤无刺激,不会引发红斑、脱屑、刺痛及灼热等不良反应。
6)使用感官测试
将本发明制得的3类外用产品,每个请20位25~50岁女性志愿者面部连续使用4周后,结果如图4-1和图4-2所示,明显发现肌肤更为白皙、通透度和水嫩饱满度提升。

所述竹荪蛋醇提物的制备方法如下:竹荪蛋清洗后,以75%乙醇作为浸提试剂,料液比1:15,打浆30秒,75~80℃浸提3h,过滤,真空浓缩至1/5体积,乙酸乙酯萃取浓缩,得竹荪蛋醇提物。

5.一种竹荪蛋多糖提取物-环糊精包合物凝胶,其特征在于,以质量分数计,原料包括3%~10%的竹荪蛋多糖提取物,0.09%~1.2%的柿叶黄酮,0.9%~12%的β-环糊精、0.02%~0.2%的卡波姆和0.3%~1.5%的透明质酸钠,余量为水;
将以上原料在室温下加水形成凝胶,制得竹荪多糖提取物-环糊精包合物凝胶;
所述竹荪蛋多糖提取物制备方法如下:取竹荪蛋,清洗后打浆,加入竹荪蛋10倍体积的水,75℃~85℃浸提5~8小时,分离得到提取液,真空浓缩至原体积的1/3,活性炭脱色,3倍体积乙醇过夜沉淀,收集沉淀,水溶脱醇,真空浓缩,浓缩液120℃~140℃的温度下喷雾干燥,制得粉末状的竹荪蛋多糖提取物。
6.一种竹荪蛋多糖提取物精华液,其特征在于,以质量分数计,原料包括10%~15%的竹荪蛋多糖提取物、1%~3%竹荪蛋醇提物、1%~4%的透明质酸,0.005%~0.05%的卡波姆、2%~6%的寡肽-1、3%~7%的六肽-10、1%~2%的玫瑰精油,0.5%~1.5%的丙二醇和0.2%~0.8%的乳化剂,余量为水;
所述竹荪蛋多糖提取物制备方法如下:取竹荪蛋,清洗后打浆,加入竹荪蛋10倍体积的水,75℃~85℃浸提5~8小时,分离得到提取液,真空浓缩至原体积的1/3,活性炭脱色,3倍体积乙醇过夜沉淀,收集沉淀,水溶脱醇,真空浓缩,浓缩液120℃~140℃的温度下喷雾干燥,制得粉末状的竹荪蛋多糖提取物;
所述竹荪蛋醇提物的制备方法如下:竹荪蛋清洗后,以75%乙醇作为浸提试剂,料液比1:15,打浆30秒,75~80℃浸提3h,过滤,真空浓缩至1/5体积,乙酸乙酯萃取浓缩,得竹荪蛋醇提物。
7.一种养肤茶包,其特征在于,以质量份数计,包括:竹荪蛋多糖提取物5份,柿叶粉10份、甘草粉15份;
所述竹荪蛋多糖提取物制备方法如下:取竹荪蛋,清洗后打浆,加入竹荪蛋10倍体积的水,75℃~85℃浸提5~8小时,分离得到提取液,真空浓缩至原体积的1/3,活性炭脱色,3倍体积乙醇过夜沉淀,收集沉淀,水溶脱醇,真空浓缩,浓缩液120℃~140℃的温度下喷雾干燥,制得粉末状的竹荪蛋多糖提取物。
8.一种速溶饮料,其特征在于,该速溶饮料是以质量分数计,将竹荪蛋多糖提取物10%~20%,加入0.1%~0.5%蛋白糖、0.5%~2%柠檬酸、0.2%~0.3%复合香精、25%~40%麦芽糊精和5%~10%β-环糊精,喷雾干燥制成颗粒;
所述竹荪蛋多糖提取物制备方法如下:取竹荪蛋,清洗后打浆,加入竹荪蛋10倍体积的水,75℃~85℃浸提5~8小时,分离得到提取液,真空浓缩至原体积的1/3,活性炭脱色,3倍体积乙醇过夜沉淀,收集沉淀,水溶脱醇,真空浓缩,浓缩液120℃~140℃的温度下喷雾干燥,制得粉末状的竹荪蛋多糖提取物。
