背景技术
卵巢的主要功能是排卵和分泌女性激素,在女性一生中掌控着几件重要的生理事件:青春发育、生儿育女、良好身材和美丽容颜的维持,其中卵巢颗粒细胞发挥了极其重要的作用。随着年龄增长,女性的卵巢功能在45-50岁左右时开始衰退,有些女性因为环境、心理或病理的因素发生卵巢早衰,随后出现一系列更年期症状,例如月经紊乱或绝经,情绪变化,肌肉松弛、弹性降低、没有光泽,继而衰老加速,身体免疫功能发生改变,心血管疾病、骨质疏松、生殖系统肿瘤和神经退行性疾病的发病率上升等,这一系列问题严重影响了女性的生活质量。因此,抗衰老或延缓衰老就成了众多女性追求的目标。
针对卵巢功能衰退传统的治疗方法就是激素替代治疗(HRT),虽然雌激素单独或雌孕激素联合应用可以改善女性更年期症状,预防骨质疏松等疾病的发生,改善生活质量,但是HRT的弊端也是不容忽视的。HRT已经证实的副作用有撤退性出血,增加卒中、血栓形成和栓塞的风险,引起水钠储留导致体重增加,对于HRT引起子宫内膜癌和乳腺癌的观点还存在争议。权衡利弊,美国FDA和欧洲EMEA都对HRT做出了严格的限定。
乌鸡是我国独特的药食同源鸡种,既可单独食用又可配药入复方,具有补肝肾,益气血,退虚热,治心腹痛等功效。乌鸡黑色素中含有Fe、Mg、Zn、Ca等多种金属元素,是吲哚型的不规则高聚物及吲哚类前体物质。乌鸡具有保养卵巢的作用,乌鸡因其特殊的营养滋补和药用价值,在医疗保健领域掀起了一股研究热潮。传统的制备方法对乌鸡骨中的小分子肽的提取较少,且含有大量的其他杂质,乌鸡多肽纯度较低。
发明内容
基于上述背景技术,本发明拟提出一种对女性尤其是更年期女性卵巢保养效果好的组合物。
本发明是通过以下技术方案实现的:
具有卵巢保养功效的组合物,由以下配方组成:乌鸡肽骨粉、纳豆、大豆肽粉、玛咖粉、人参、膳食纤维。
所述的具有卵巢保养功效的组合物,由以下配比按重量份数配比而成:乌鸡肽骨粉30-50份、纳豆10-30份、大豆肽粉5-15份、玛咖粉10-20份、人参1-5份、膳食纤维0.1-0.5份。
所述的乌鸡肽骨粉,由以下步骤提取得到:
(1)将乌鸡骨粉碎至100-150目;
(2)将破碎后的乌鸡骨粉按比例1:20-30的体积比加入纯化水,搅拌均匀后投入亚临界萃取釜中进行亚临界流体萃取,萃取压10MPa,萃取温度为140℃,萃取时间为40min;
(3)过滤得到萃取后的乌鸡骨粉,向超声仪中加入过滤后的粗粉,再加入8倍重量当量的浓度为65%乙醇,控制超声功率为200-300W、超声时间为120min;
(4)大孔树脂分离提纯:步骤(3)中得到的超声处理液,置于规格为1000mm×100mm的732型号强酸性阳离子-D201型号强碱性阴离子树脂进行大孔树脂吸附提取,控制温度为38℃,流速控制0.5-1ml/min,得到提取液;
(5)蒸干提取液得到乌鸡肽骨粉。

纳豆是以大豆为原料,经纳豆枯草芽孢杆菌发酵制成的即食豆制品。纳豆中含有较多的纳豆激酶,纳豆激酶是在纳豆发酵过程中由纳豆枯草杆菌产生的一种丝氨酸蛋白酶。纳豆激酶是一种丝氨酸蛋白酶在具有溶解血栓,降低血粘度,改善血液循环,软化和增加血管弹性的同时可以断裂乌鸡肽骨粉中的大分子蛋白的肽键,将乌鸡肽骨粉中的大分子多肽裂解成更容易吸收的小分子多肽。

具体实施方式
以下结合具体优选的实施例对本发明作进一步描述,但并不因此而限制本发明的保护范围。
实施例1
配方为:乌鸡肽骨粉40份、纳豆20份、大豆肽粉10份、玛咖粉15份、人参2份、膳食纤维0.1份。
乌鸡肽骨粉,由以下步骤提取得到:
(1)将乌鸡骨粉碎至100目;
(2)将破碎后的乌鸡骨粉按比例1:25的体积比加入纯化水,搅拌均匀后投入亚临界萃取釜中进行亚临界流体萃取,萃取压10MPa,萃取温度为140℃,萃取时间为40min;
(3)过滤得到萃取后的乌鸡骨粉,向超声仪中加入过滤后的粗粉,再加入8倍重量当量的浓度为65%乙醇,控制超声功率为200-300W、超声时间为120min;
(4)大孔树脂分离提纯:步骤(3)中得到的超声处理液,置于规格为1000mm×100mm的732型号强酸性阳离子-D201型号强碱性阴离子树脂进行大孔树脂吸附提取,控制温度为38℃,流速控制0.8ml/min,得到提取液;

实施例2

制备方式为:将上述配方破碎至200目,压片既得。
对比例1
配方为:乌鸡肽骨粉30份、纳豆10份、大豆肽粉5份、玛咖粉10份、人参5份、膳食纤维0.1份。其余制备步骤同实施例1。将实施例1-2对比例1所制的制剂进行测试。
昆明小鼠,36-38g,500只雌性(52周大),上海实验动物中心购买。室温18-22℃,相对湿度维持在60%-70%,光照12hr∶12hr(光∶暗),使用前适应环境3天。
药效学实验:
材料和方法:
实验材料:实施例1-2对比例1所制的制剂所制备的保健品。
实验动物:昆明种雌性小鼠(50周大)
给药方法:与食品一起喂食,每天早上8点一次。
小鼠生存实验:将500只50周大雌性小鼠随机分成5组,每组100只,第5组为空白对照。
实验方法:将雌性小鼠进行正常喂食第一组喂食实施例1所制备的产品。第二组喂食实施例2所制的的产品,依次类推,空白对照正常喂食,保证小鼠具有正常的生存环境,在相同的环境下连续喂食10周,观察记录死亡率,每周记录检测卵泡刺激素、促黄体生成素、孕酮的含量,1-3周算平均值,4-6周算平均值、7-10周算平均值,每组取平均值,结果见表1、表2。
表1(小鼠喂养10周内的死亡率%)
从上表1可以看出,实施例1、2采用了本发明的制剂的小鼠死亡率较低,衰老较慢。具有一定的抗衰老效果。
每周记录检测卵泡刺激素、促黄体生成素的含量,1-3周算平均值,4-6周算平均值、7-10周算平均值,每组取平均值结果见下表。

从表2、3可以看出服用本发明组合物的实施例1-2卵巢处于正常的较低水平,未服用的对比例1和对照组,小鼠的卵巢激素水平处于较高的衰老状态,衰老较快。实施例3未采用本发明中对乌鸡骨的提取方式得到的乌鸡多肽其防止卵巢衰老的效果不佳。
Claims (4)
1.一种具有卵巢保养功效的组合物,其特征在于,由以下配方组成:乌鸡肽骨粉、纳豆、大豆肽粉、玛咖粉、人参、膳食纤维。
2.根据权利要求1所述的具有卵巢保养功效的组合物,其特征在于,由以下配比按重量份数配比而成:乌鸡肽骨粉30-50份、纳豆10-30份、大豆肽粉5-15份、玛咖粉10-20份、人参1-5份、膳食纤维0.1-0.5份。
3.根据权利要求1所述的乌鸡肽骨粉,其特征在于由以下步骤提取得到:
(1)将乌鸡骨粉碎至100-150目;
(2)将破碎后的乌鸡骨粉按比例1:20-30的体积比加入纯化水,搅拌均匀后投入亚临界萃取釜中进行亚临界流体萃取,萃取压10MPa,萃取温度为140℃,萃取时间为40min;
(3)过滤得到萃取后的乌鸡骨粉,向超声仪中加入过滤后的粗粉,再加入8倍重量当量的浓度为65%乙醇,控制超声功率为200-300W、超声时间为120min;
(4)大孔树脂分离提纯:步骤(3)中得到的超声处理液,置于规格为1000mm×100mm的732型号强酸性阳离子-D201型号强碱性阴离子树脂进行大孔树脂吸附提取,控制温度为38℃,流速控制0.5-1ml/min,得到提取液;
(5)蒸干提取液得到乌鸡肽骨粉。
4.权利要求1所述组合物在女性卵巢抗衰老的应用。
