CRISPR/Cas9系统是从细菌和古生菌对抗外来病毒或质粒的适应性免疫系统发展而来,可通过特异结合并剪切DNA片段,有效帮助菌体抵御外源性DNA侵染,从而实现对目标位点的基因定点敲除、敲入以及基因修正等多种修饰。该系统包含三个基本结构:tracrRNA(trans-activating crRNA,tracrRNA)序列区、Cas(CRISPR associated system,Cas)基因序列区和CRISPR RNAs(crRNAs)。该系统包括Type I、Type II、TypeIII三种不同的类型,其中TypeI和TypeIII需要多种CRISPR(Cas蛋白)共同发挥作用,而TypeII型系统的Cas9有HNH和RuvC 2个核酸酶结构域,HNH核酸酶结构域剪切与crRNA互补的DNA链,RuvC结构域剪切非互补链,在基因组上产生双链断裂(DSB),降低脱靶概率,所以应用也最为广泛。CRISPR/Cas9系统的原理是CRISPR RNAs(crRNAs)通过碱基配对与tracr RNA(trans-activatingcrRNA)结合成双链RNA,crRNA与tracr RNA形成二元复合体后,与双链DNA特异位点以20nt的长度互补配对结合,继而引导Cas9核酸酶对结合位点进行切割。在完成切割后,细胞会以非同源性末端接合(non-homologous end joining,NHEJ)或同源重组修复(HomologyDirected Repair,HDR)的机制将断裂的DNA重新连接,在连接过程中完成对目标基因的人工敲除(Knock-out)或敲入(Knock-in)。CRISPR-Cas9技术因其高效性、准确性和便利性,已成功应用于多种生物基因的修饰与治疗。
发明内容
本发明人经过广泛而深入的研究,首次意外地发现基于CRISPR/Cas9基因敲除技术建立CNGA1基因敲除动物模型的方法,在CNGA1基因的第二个外显子设计两个间隔约100bp的靶点,靶率效率低,从而达到基因敲除效果。通过本方法得到的动物表现出明显的视网膜病变的生理特征和表型特征。在此基础上,发明人完成了本发明。
为了实现本发明的上述目的,特采用以下技术方案:
本发明涉及一种制备视网膜病变的非人哺乳动物模型的方法,包括:
使用Crispr-Cas系统将所述动物CNGA1基因第二个外显子的部分序列敲除;
其中所述Crispr-Cas系统包括sgRNA和Cas9酶,所述sgRNA包括对应的靶位点序列为SEQ ID NO:1和2所示的sgRNA1和sgRNA2。
可选的,所述基因敲除的对象为受精卵。
可选的,所述Crispr-Cas系统以sgRNA和Cas9 mRNA的形式转入所述受精卵。
可选的,所述Crispr-Cas系统转入所述受精卵的方法为显微注射。
可选的,所述方法还包括将处理后受精卵移植到假孕雌性动物体内并产出F0代,将正确敲除CNGA1基因的F0代动物与野生型动物交配获得F1代杂合子动物。
可选的,所述方法还包括将所述F1代杂合子动物自交获得F2代纯合子动物。
可选的,所述非人哺乳动物为啮齿类动物。
可选的,所述非人哺乳动物是小鼠(Mus musculus)。
可选的,所述方法不引入可稳定表达的外源基因。
本发明还涉及如上所述方法得到的动物在鉴别和/或测试药物中的用途;
所述药物用于预防和/或治疗的适应症为视网膜病变和/或治疗与视网膜病变相关的并发症。
基因敲除的关键在于靶点选择,作用于正确靶点可通过序列突变使基因功能缺失。通过分析动物CNGA1基因结构,在CNGA1基因第二个外显子序列设计sgRNA,挑选出敲除效率高的sgRNA1和sgRNA2。本发明在构建CNGA1基因敲除动物模型时,考虑了多方面因素,选择了更为简便、高效的CRISPR/Cas9技术。CRISPR/Cas9技术打靶效率高、设计便捷,步骤简单,对生物学领域的技术发展起到了深远的影响。本发明采用CRISPR/Cas9基因敲除技术,首次建立了CNGA1基因敲除的动物模型。选取了sgRNA1和sgRNA 2,本发明设计的sgRNA1-2及方法具有敲除效率高、可以不引入外源基因干扰的优点。
与现有技术相比,本发明的有益效果为:
1、本发明提供了一种通过CNGA1基因敲除动物模型模拟人CNGA1基因突变导致视网膜病变的方法。
2、本发明采用CRISPR/Cas9基因敲除技术,首次建立了CNGA1基因敲除的动物模型。实验结果表明:本发明设计的sgRNA1和sgRNA 2及方法具有敲除效率高、脱靶效率低、无外源基因干扰的优点。
3、本发明为研究CNGA1与遗传性视网膜病变、基因治疗等提供便捷、可靠、经济的动物模型。

本发明涉及一种制备视网膜病变的非人哺乳动物模型的方法,包括:
使用Crispr-Cas系统将所述动物CNGA1基因第二个外显子的部分序列敲除;
其中所述Crispr-Cas系统包括sgRNA和Cas9酶,所述sgRNA包括对应的靶位点序列为SEQ ID NO:1和2所示的sgRNA1和sgRNA2。
本发明涉及一种基于CRISPR/Cas9基因敲除技术建立CNGA1基因敲除动物模型的方法,此处的“基因敲除”仅指针对目的基因的Cas9,对目的基因基因组的特定外显子区域敲除,引发目的基因蛋白翻译读码框的移码或者蛋白翻译的提前终止,而非目的基因为其他DNA片段所取代,从而达到基因敲除的效果。
SEQ ID NO:1和2所对应的靶点,脱靶效率低,因而动物几乎不表现出除视网膜病变以外的其他明显症状,并且对动物寿命影响不显著。
在一些实施方式中,所述基因敲除的对象为受精卵。
在一些实施方式中,所述Crispr-Cas系统以sgRNA和Cas9 mRNA的形式转入所述受精卵。
在一些实施方式中,所述Cas9来自化脓性链球菌或肺炎链球菌。
在一些实施方式中,所述Crispr-Cas系统转入所述受精卵的方法为显微注射。
在一些实施方式中,所述方法还包括将处理后受精卵移植到假孕雌性动物体内并产出F0代,将正确敲除CNGA1基因的F0代动物与野生型动物交配获得F1代杂合子动物。
在一些实施方式中,所述方法还包括将所述F1代杂合子动物自交获得F2代纯合子动物。
非人哺乳动物非限制性地包括牛、马、乳牛、猪、绵羊、山羊、大鼠、小鼠、狗、猫、兔、骆驼、驴、鹿、貂、鸡、鸭、鹅、火鸡、斗鸡等。在一些实施方式中,所述非人哺乳动物为啮齿类动物。
在一些实施方式中,所述非人哺乳动物是小鼠(Mus musculus)。
小鼠品系可以选择BALB/c、C57BL、C3H/He、昆明小鼠、ICR、NIH、CFW、LACA、nude小鼠或Scid小鼠等,优选C57BL小鼠。
在一些实施方式中,所述方法不引入可稳定表达的外源基因。
在一些实施方式中,所述外源基因包括报告基因和/或标签基因。
根据本发明的再一方面,本发明还涉及如上所述方法得到的动物在鉴别和/或测试药物中的用途;
所述药物用于预防和/或治疗的适应症为视网膜病变和/或治疗与视网膜病变相关的并发症。
在一些实施方式中,所述视网膜病变为常染色体隐性遗传视网膜色素变性(ARRP)。
下面结合具体实施,进一步阐述本发明。应理解,这些实施例仅用于说明本发明而不用于限制本发明的范围。下列实施例中未注明具体条件的实验方法,通常按照常规条件,例如Sambrook等人,分子克隆:实验室手册(New York:Cold Spring Harbor LaboratoryPress,1989)中所述的条件,或按照制造厂商所建议的条件。除非另外说明,否则百分比和份数按重量计算。
实施例CNGA1-KO小鼠模型的构建及鉴定
一、材料
本实施例所用小鼠品系为:C57BL/6J,代孕母鼠品系为C57BL/6J,购自上海斯莱克实验动物有限公司,将小鼠随机分为对照组与实验组。
各种限制性内切酶、T4 DNA连接酶、Taq酶及PCR相关试剂购自NEB公司。常规化学试剂主要购自Sigma和上海化学试剂公司。胶回收Kit,质粒抽提Kit购于Qiagen公司,体外转录Kit购自Invitrogen公司。
引物等合成自苏州金唯智生物科技有限公司
二、方法
实验组按照以下方法进行实验:
1、在目标基因内含子中寻找合适的靶序列,为方便后期基因型鉴定,选择设计两个靶点,同时注射。
通过在线设计工具(http://crispr.mit.edu/)及gRNA的设计原则,评估小鼠CNGA1基因序列上得分较高的靶位点设计gRNA,靶位点序列为SEQ ID NO:1-SEQ ID NO:2。

上述基因序列中大写字母代表编码区,斜体大写部分代表非编码区,小写部分代表内含子区。
2、Cas9mRNA制备
将线性化及纯化DNA并与Cas9核酸酶体外转录成mRNA:纯化sgRNA至适合转基因注射的纯度。
预先在293T细胞中测试sgRNA编辑效率,结果如图2所示,sgRNA1和sgRNA 2同时使用敲除效率最高。
3、显微注射受精卵
使用显微注射仪器将上述sgRNA、Cas9mRNA按((25ng/uL和50ng/uL)比例混匀后,注射入体外受精小鼠受精卵的胞浆部分,构建形成特定的小鼠胚胎细胞(受精卵)。体外培养1-2小时后将存活的受精卵移植入假孕母鼠的输卵管,胚胎移植的小鼠出生即为F0代小鼠。
4、基因型鉴定
4.1、基因组DNA提取:
A、消化:小鼠出生约一周内,剪取0.5cm小鼠脚趾,放入1.5ml EP管中,稍离心后加500ul裂解液(配方:100mM Tris pH8.0,5mM EDTA pH8.0,0.5%SDS,NaCl 1.17g/100ml)、0.5ul蛋白酶K(浓度:20mg/ml,溶解在pH7.4,20mM Tris和1mM CaCl2中,50%甘油缓冲液,-20℃保存),混匀55℃水浴消化过夜;
B、异丙醇沉淀抽提DNA:
1)从水浴锅中取出离心管,在室温静置10-15min,使样品温度降至室温,将离心管颠倒混匀。13000rpm,室温离心15min。
2)吸取400μl上清至另一个新的离心管中。加入等体积的异丙醇,立即温和地上下翻转,充分混匀,此时会出现白色絮状沉淀,室温下12000rpm离心10min,弃上清。
3)往离心管中加入700μl冰冷的75%乙醇漂洗,温和地上下翻转混匀。12000rpm,室温离心5min,将上清液全部吸除。
4)将离心管倒置在吸水纸上,以吸干乙醇。风干后用无菌ddH2O,50ul溶解DNA,55℃溶解2h(如不立即使用,-20℃保存)。
5)检测DNA的浓度,取100-200ng的DNA用做PCR模板。
4.2、PCR鉴定:
针对靶点上下游约200~300bp区域分别设计正反向PCR引物。

注:如果序列复杂,可换其他的酶进行PCR。加入5%DMSO、GC enhancer等。
5、小鼠杂交(策略图如图1所示)
提取上述步骤将F0代小鼠的基因组DNA进行基因型鉴定,PCR扩增并将产物送测序;
将阳性小鼠与野生型异性小鼠交配获得F1代杂合子小鼠;对F1代小鼠进行鉴定;
F1代鼠鉴定
F1代23#,25#♀&WT,2018.9.19生,9.25剪1-9#。10.29生,11.5剪10-20#。
将繁育得到的KO杂合子与KO杂合子自交,得到F2代,从F2代中选出KO纯合子作为模型小鼠。
对F2代的部分PCR电泳图见附图2。
通过电泳图可以看出9号泳道为KO小鼠(KO-/-);1、7、8、11、12、13、15为WT;其余泳道为KO+/-。
三、结果
WT鼠与10周龄的CNGA1-KO小鼠的HE染色对比结果如图4所示:CNGA1-KO小鼠外核层及外丛状层明显萎缩。
WT鼠与5周龄的CNGA1-KO小鼠的ERG图如图5所示:CNGA1-KO小鼠外核层及外丛状层明显萎缩。
由上可知,通过本方法制备得到的小鼠已经发生了视网膜病变。
以上所述实施例的各技术特征可以进行任意的组合,为使描述简洁,未对上述实施例中的各个技术特征所有可能的组合都进行描述,然而,只要这些技术特征的组合不存在矛盾,都应当认为是本说明书记载的范围。
以上所述实施例仅表达了本发明的几种实施方式,其描述较为具体和详细,但并不能因此而理解为对发明专利范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进,这些都属于本发明的保护范围。因此,本发明专利的保护范围应以所附权利要求为准。

1.制备视网膜病变的非人哺乳动物模型的方法,其特征在于,包括:
使用Crispr-Cas系统将所述动物CNGA1基因第二个外显子的部分序列敲除;
其中所述Crispr-Cas系统包括sgRNA和Cas9酶,所述sgRNA包括对应的靶位点序列为SEQ ID NO:1和2所示的sgRNA1和sgRNA2。
2.根据权利要求1所述的方法,其特征在于,所述基因敲除的对象为受精卵。
3.根据权利要求2所述的方法,其特征在于,所述Crispr-Cas系统以sgRNA和Cas9 mRNA的形式转入所述受精卵。
4.根据权利要求2或3所述的方法,其特征在于,所述Crispr-Cas系统转入所述受精卵的方法为显微注射。
5.根据权利要求4所述的方法,其特征在于,所述方法还包括将处理后受精卵移植到假孕雌性动物体内并产出F0代,将正确敲除CNGA1基因的F0代动物与野生型动物交配获得F1代杂合子动物。
6.根据权利要求5所述的方法,其特征在于,所述方法还包括将所述F1代杂合子动物自交获得F2代纯合子动物。
7.根据权利要求1~3、5、6任一项所述的方法,其特征在于,所述非人哺乳动物为啮齿类动物。
8.根据权利要求7所述的方法,其特征在于,所述非人哺乳动物是小鼠(Musmusculus)。
9.根据权利要求1~3、5、6、8任一项所述的方法,其特征在于,所述方法不引入可稳定表达的外源基因。
10.权利要求1~9任一项所述方法得到的动物在鉴别和/或测试药物中的用途;
所述药物用于预防和/或治疗的适应症为视网膜病变和/或治疗与视网膜病变相关的并发症。
