本发明提供一种防蚊消毒剂及制备方法,主要由醋酸氯己定、维生素B5、紫草提取物、薰衣草油、香茅精油、茶树油、维生素E、无水乙醇、甘油、吐温、水、亚甲基蓝组成,本发明无皮肤刺激性,适用于局部抑菌消毒,能有效地杀死皮肤上各种细菌,有害微生物。同时还可以有效防止蚊虫叮咬,可以缓解蚊虫叮咬后的不适,能够消除蚊虫叮咬后的瘙痒。

本发明提供一种皮肤损伤抑菌消毒剂及制备方法,能有效地杀死皮肤上各种细菌,有害微生物。并可有效防止蚊虫叮咬,能够消除蚊虫叮咬后的瘙痒,属于消毒剂技术领域。
背景技术
随着科学技术的发展,针对患者和接触患者人员的消毒已成为切断传染途径的必要措施。但是传统消毒剂通常具有一定毒性和残留。长期使用会对人和动物产生不同程度的伤害,并且对环境造成严重污染。所以,配制一种安全高效的消毒剂,对于预防干预环境卫生非常重要。特别是城市化速度加快造成的人群大量聚集,必然会使生命体不可避免地受到各种各样的环境影响,这些影响包括宇宙射线太阳光紫外线射线,寄居昆虫,环境中的微生物,病毒等。
国内临床常用的抑菌剂是醋酸氯己定。国内逾70年代初期开始将醋酸氯己定,尝试用于手术人员的泡手,消毒器械消毒等。
但是传统的醋酸氯己定溶液具有,配制困难,携带不便的缺点。因为常常配制成水溶液使用,用于人体时部分位置不易作用到,并且溶液容易挥发流失,抑菌时效性短。不适宜野外作业人群的使用。
高温高湿环境中滋生的蚊虫,给人类带来滋扰伤害和生命威胁,还会引起诸如影响睡眠,传播疾病等不利因素,尤其是叮咬过后引起的瘙痒。
瘙痒是皮肤或粘膜的一种引起搔抓欲望的不愉快的感觉。抑制减轻叮咬后的瘙痒不但可以减轻皮肤的不适,还可以减少瘙痒搔抓引起的皮肤损伤和继发性感染。
传统消毒剂酒精碘伏,对皮肤黏膜无损撞伤具有明显的消毒作用,尤其对皮肤创伤伤口具有消毒作用,但是由于具有一定刺激作用和残留,所以适用性较差。
综上所述,人们对安全无毒无副作用的皮肤粘膜抑菌消毒剂存在着极大的需求。尤其是处于高温高湿环境中的野外作业人群,室外活动人群。
本发明不但适用于局部抑菌消毒,较传统消毒,无刺激性和抑菌毒副作用的特点,还可以有效防止蚊虫叮咬,可以缓解蚊虫叮咬后的不适。
发明内容
人们对安全,无毒副作用的消毒液存在着极大的需求,尤其对于高温高湿环境中暴露于滋生蚊虫的野外作业人群、室外活动人群。对具有皮肤消毒,防蚊驱蚊,抑制瘙痒功能的产品有强烈的使用欲望。申请人经过大量的研究试验,成功地研制出一种防蚊消毒剂。
本发明所述的防蚊消毒剂的组合物。制备用于皮肤消毒,防蚊驱蚊,抑制瘙痒等用途。
本发明解决其技术问题所采用的技术方案是,提供一种防蚊消毒剂,其特征是:醋酸氯己定1~2份;维生素B5 2~4份;紫草提取物10~20份;薰衣草油5~10份;香茅精油1~2份;茶树油1~2份;维生素E 10~20份;无水乙醇 80~160份;甘油 60~120份;吐温80 10~20份;水800~1600份;亚甲基蓝0.008~0.016份;HPMC 20~40份混合均匀即得。

2)称量处方量甘油、无水乙醇、吐温80、紫草提取物、茶树油、香茅精油、薰衣草油、VE搅拌至溶解,配成醇相;
3)称取处方量HPMC,加入搅拌中的醇相,搅拌15分钟;
4)将水相经过过滤后加入搅拌中的醇相,持续15分钟;
5)搅拌混合样品经过-0.07Mpa真空乳化1小时,过滤灌装即为成品。
本发明积极效果在于本发明具有延缓释放驱蚊剂的效果,能在长时间内保持驱蚊效果,通过降低渗透速率,以达到驱蚊止痒而又不刺激皮肤的效果,避免了对皮肤产生刺激性的影响,提高了使用的舒适感,而且通过将亚甲基蓝与紫草提取的复配使用,改变产品色泽,更易于使用人群接受,同时亚甲基蓝提高了消毒成分的回收率,延长了产品的货架期,提高了杀菌效果。

结果:见表1。

结论:上述6个批号的消毒成分含量测定均超过80%。其配方中含有亚甲基蓝的处方,消毒成分含量均超过90%,实验结果显示,亚甲蓝成分对消毒成分回收率提高有明显促进作用。
试验2:皮肤消毒试验
一、器材
1、试验样品:20200313-2;
2、中和剂:D/E肉汤;
3、其他:灭菌棉签,含0.1%吐温80的PBS等。
二、方法
1、检测依据:中和剂鉴定试验和消毒剂对皮肤消毒现场试验按照卫生部《消毒技术规范》(2002版)2.1.1.5.5条和2.1.2.8条。
2、现场试验受试部位及数量:受试者左右前臂内侧中段皮肤。受试数量:30人次。
3、消毒处理方法:取样品原液涂擦在右手前臂内侧中段皮肤2次,2min后采样,结果见表2。

1、检测温度20℃,相对湿度53%;
2、样品处理:原样。
三、结果

仪器设备:GHP-9270N生化培养箱。
二、方法
检测温度21~23℃,相对湿度51%~53%。

四、结论
批号20200313-2原样作用20min对金黄色葡萄球菌的平均抑菌率为99.97%,有较强抑菌作用;对大肠杆菌的平均抑菌率为99.97%,有较强抑菌作用;对白色念珠菌的平均抑菌率为99.95%,有较强抑菌作用。
试验5皮肤刺激试验
动物:普通级新西兰兔3只,雄性。
环境条件:普通级动物房,室温 20℃~25℃;相对湿度41%~69%。
试验方法:
备皮:在染毒前24h,用除毛剂将家兔背部脊柱两侧腹部皮肤去掉,不得损伤皮肤。去毛面范围左右各不小于3cm×3cm。
染毒方法:采用同体左右侧自身对比法,将0.5g的样品直接均匀涂在面积为2.5cm×2.5cm 的左侧去毛完整皮肤上,然后用一层无刺激性的玻璃纸覆盖,再用无刺激胶布固定。右侧皮肤同法给予0.5ml氯化钠注射液作为阴性对照组。贴敷时间为4h,结束后用温水轻轻清洗染毒部位,并用吸水纸拭去残留水渍。每天涂抹一次,连续涂抹14d。
症状观察:在每次涂抹后24h 观察结果,按表7评分。为了便于受试物的涂抹和结果观察,必要时应剪毛。对照区的处理方法同试验区。

按下列公式计算每天每只动物平均积分(刺激指数),并以表8判定皮肤刺激强度。

结果:实验动物在染毒14天内染毒侧及对照侧皮肤未见任何皮肤红斑及水肿症状出现,每天每只动物染毒侧和对照测的平均积分均为0分。
结论:样品对家兔皮肤的刺激强度属无刺激性。
试验例6 本发明驱蚊效果实验
试验样品:批号20200313-2。
对照组样品:纯化水。
实验蚊虫:白纹伊蚊;饲养条件:温度(26±1)℃,相对湿度为(65±5)%,
实验方法:选取50位志愿者,随机分为10组,取人手背部5cm×5cm皮肤面积,掩盖其余皮肤,在其左手暴露的皮肤上涂抹0.25ml/cm2剂量的试验样品,涂抹后10min,将手伸进笼中停留100s,每间隔2 h测试1次,共测试4次,根据白纹伊蚊在手臂上的停留数,计算有效保护率(%),其中:有效保护率(%)=(对照组停落蚊虫数-实验组停落蚊虫数)/对照组停落蚊虫数×100%,结果见表10所示:

结果:由表10结果可知,试验样品4小时以内的有效保护率为81.2%,驱蚊效果显著。
试验7:有效成分稳定性试验
试验样品:醋酸氯己定;

试剂:乙腈,磷酸,磷酸二氢钾溶液
仪器:1,天平,2,pH计,3高效液相色谱仪,4稳定性试验箱SHH-500SD
色谱条件:流动相:磷酸二氢钾:乙腈=650:350;柱温,30℃、检测波长:254nm、进样量:10ul、色谱柱:C18柱(150mm×4.6mm ×5μm)、流量:1.0 ml /min。
试验样品:抑菌凝胶,批号20200310-1,20200313-2,20200313-2
放置条件:温度37℃,放置90d
结果:见表11。

结论:上述6个批号的消毒成分含有亚甲蓝成分含量下降率均未超过10%,实验结果显示,亚甲蓝成分对消毒稳定性有明显促进作用。

Claims (3)

1.一种防蚊消毒剂,其特征是:醋酸氯己定1~2份;维生素B5 2~4份;紫草提取物10~20份;薰衣草油5~10份;香茅精油1~2份;茶树油1~2份;维生素E 10~20份;无水乙醇 80~160份;甘油 60~120份;吐温80 10~20份;水800~1600份;亚甲基蓝0.008~0.016份;HPMC 20~40份混合均匀即得。
2.权利要求1所述的一种防蚊消毒剂,其特征是:醋酸氯己定1份;维生素B5 2份;紫草提取物10份;薰衣草油5份;香茅精油1份;茶树油1份;维生素E 10份;无水乙醇 80份;甘油60份;吐温80 10份;水800份;亚甲基蓝0.01份;HPMC 20份。
3.权利要求1或2所述的一种防蚊消毒剂,其特征是:防蚊消毒剂制备方法为:1)先在搅拌锅里加入去离子水,称量处方量亚甲基蓝溶液,加入醋酸氯己定、维生素B5搅拌至溶解,配成水相;2)称量处方量甘油、无水乙醇、吐温80、紫草提取物、茶树油、香茅精油、薰衣草油、VE搅拌至溶解,配成醇相;3)称取处方量HPMC,加入搅拌中的醇相,搅拌15分钟;4)将水相经过过滤后加入搅拌中的醇相,持续15分钟;5)搅拌混合样品经过-0.07Mpa真空乳化1小时,过滤灌装即为成品。
