Abstract
The invention discloses a copper peptide-containing essence, which contains copper peptide, also innovatively combines ECTOIN and fibronectin, extends a repairing target point from the deep layer of skin to the surface layer of the skin, repairs damaged skin from inside to outside in an all-around way, has high safety and does not have the risk of excessive cell proliferation like EGF. According to the invention, a large number of experiments show that the optimal proportioning range is obtained, the effect is weak if the proportioning range is smaller than the range of the invention, the increase value of each index in skin experiments is small if the proportioning range is larger than the range, the cost is increased a lot, and the economical principle is not met, so that the invention is an economical, effective and optimal essence containing copper peptide for repairing skin.
Description
Technical Field
The invention belongs to the technical field of cosmetics, and particularly relates to essence containing copper peptide and a preparation method thereof.
Background
In China, more than 90% of women are troubled by skin problems every year, wherein the most common problems are dry and lack of water, large pores, dark circles, dark yellow faces, loose wrinkles, whelks and the like. The biggest cause of these results is from the deterioration of the external environment, such as environmental problems of air pollution, electric radiation, etc. For example, PM2.5 can cause great influence on skin, and personal habits can also affect skin state, and psychological pressure is more prone to cause skin problems than overeating, staying up in the night and the like.
Therefore, the skin problems are caused by the combination of internal and external factors, various skin problems are repaired, some skin care products are added with various sunscreens to resist the damage of ultraviolet rays to the skin aiming at external environmental factors, the sunscreens possibly bring intolerance to the skin and cause larger stimulus, and anti-allergic agents are selected aiming at internal factors, and hormone-dependent dermatitis can be generated and is undoubtedly more harmful to the skin if some people carelessly use cosmetics with different specifications; with the advancement of science and technology, polypeptide skin care becomes a new research hotspot, and many products containing copper peptides come out, wherein the copper peptides can repair the dermis layer of the skin, but cannot repair skin wounds immediately due to the damage of the stratum corneum caused by environmental invasion.
Disclosure of Invention
In view of the above, the technical problem to be solved by the present invention is to provide a copper peptide-containing essence composition, which contains copper peptide, ECTOIN (ECTOIN), fibronectin, etc., and can not only solve the side effects of sunscreen on skin, but also rapidly repair wounds, and finally repair damages of true epidermis of skin, thereby providing a high-efficiency and safe skin care product for consumers.
In order to solve the technical problems, the invention adopts the following technical scheme:
the essence containing copper peptide is innovatively matched with ECTOIN, fibronectin and barley seed extract besides the copper peptide, a repairing target point extends from the deep layer of the skin to the surface layer of the skin, the damaged skin is repaired from inside to outside in all directions, the safety is very high, and the risk of excessive cell proliferation is avoided like EGF.
Preferably, the weight percentages of the copper peptide, ECTOIN, fibronectin and barley seed extract in the essence are as follows:
0.05-0.5% of copper peptide, 0.05-5% of ECTOIN, 0.1-5% of fibronectin, 0.5-2% of barley seed extract and the balance of pure water.
The essence containing copper peptide also comprises a humectant.
Preferably, the moisturizers comprise sodium hyaluronate, butanediol, pentanediol and hexanediol, and the four moisturizers account for the following components in percentage by weight in the essence:
0.005-0.5% of sodium hyaluronate, 1-10% of butanediol, 1-10% of pentanediol and 0.2-5% of hexanediol.
The essence containing copper peptide also comprises a thickening agent.
Preferably, the thickening agent is xanthan gum, and the percentage content of the thickening agent in the essence is 0.01-0.5%.
The essence containing copper peptide also comprises a chelating agent.
Preferably, the chelating agent is disodium EDTA, and the percentage content of the disodium EDTA in the essence is 0.01-0.1%.
According to the invention, a large number of experiments show that the optimal proportioning range is obtained, the effect is weak if the proportioning range is smaller than the range of the invention, the increase value of each index in skin experiments is small if the proportioning range is larger than the range, the cost is increased a lot, and the economical principle is not met, so that the invention is an economical, effective and optimal essence containing copper peptide for repairing skin.
Copper peptide: copper is a trace element essential to human body and is distributed throughout tissues and organs of the whole body. In the human body, copper mainly acts in the form of copper-containing metalloenzyme and participates in a series of metabolic processes such as utilization of iron, hematopoiesis, synthesis of phospholipid, formation of collagen connective tissue and the like. Copper therefore has a major influence on the development and function of the blood system, the central nervous and immune systems, hair, skin and bone tissue. In 1973, Dr Loren Pickart who studied biochemistry and aging physiology discovered that young people contained large amounts of copper peptides, which are now known to activate hair growth factors and promote hair growth; can promote degradation of a large amount of collagen aggregates outside the scar, promote normal ECM generation of skin, promote growth of different skin cells, and has anti-inflammatory and free radical scavenging effects. The copper peptide used in the invention is solid powder, and the purity is more than 95%.
ECTOIN: tetrahydromethylpyrimidine carboxylic acid, Ectoin was found in 1985 in microorganisms living in Wadz El Natrun (Egypt desert) salt lakes, and is a substance for its self-protection and life-support. Amino acid derivatives belong to the group of the extreme molecules. The extreme microorganisms and plants are protected against the lethal and extreme environments such as saline lakes, hot springs, permafrost ice or deserts and the like. Ectoin can play a variety of skin care effects, and many studies confirm the protective properties of Ectoin against full spectrum white light. Ectoin, which does not act as a filter, does not absorb UV radiation, but protects cells and all other components of the skin by surrounding them with a protective layer of protein, protects Langerhans cells from damage or destruction, and also reduces the number of sun damaged cells caused by UVB radiation. This protective mechanism is even capable of protecting the skin from damage by UVA, UVB, infrared and visible radiation; can also provide long-acting moisture retention for skin; reducing wrinkles and increasing skin elasticity; protecting Langerhans cells, promoting the formation of heat shock proteins, and improving the self-defense performance of skin;
fibronectin: FN, Fibronectin, the Chinese name "Fibronectin", is a high molecular glycoprotein widely present in tissues and interstitial fluid, and plays an important role in promoting cell adhesion, migration, growth, proliferation and differentiation. FN consists of two peptide chains, has six functional regions, respectively binds to collagen, heparin, cells, fibers, etc., and has biological function of cell regeneration and repair.
Barley seed extract: the raw material is derived from food-grade barley, and the barley seed extract contains various amino acids available to human body, including glycine, proline and proline; leucine and other free amino acids can promote epidermal cell and connective tissue regeneration during wound healing, and can also act on nerve sense aspects such as burning heat, stabbing pain and the like related to sensitive skin to reduce inflammation symptoms.
Sodium hyaluronate: hyaluronic acid is a natural substance widely present in the human and animal bodies, and exhibits a variety of important physiological functions in the body with its unique physicochemical properties. Hyaluronic acid, like molecular sponge, can retain water with the weight 500-1000 times of the self weight, and has the functions of maintaining skin elasticity and lubricating joints. With the age, the cell metabolism slows down, and the hyaluronic acid synthesis ability of the human body gradually declines, which is characterized in that collagen fibers and elastic fibers in the skin cannot obtain enough moisture on the one hand, so that the skin loses elasticity and wrinkles appear. On the other hand, reduction of hyaluronic acid in joint synovial fluid causes joint inflammation such as joint pain and limited movement. The hyaluronic acid with the molecular weight of more than 100 ten thousand is used, has excellent lubricating property and film forming property, and forms a protective film and a water locking film on the surface layer of the skin to prevent the moisture in the skin from evaporating and maintain the luster and the tenderness of the skin;
butanediol: as moisturizers in cosmetics;
and (3) pentanediol: the humectant has excellent performance, has antiseptic effect, can be used for preparing a preservative-free product, and reduces allergy caused by the preservative;
hexanediol: the humectant has excellent performance, has antiseptic effect, can be used for preparing a preservative-free product, and reduces allergy caused by the preservative;
xanthan gum: it has unique rheological property, good water solubility, stability to heat and acid and alkali, and good compatibility with various salts, and can be used as thickener, suspending agent, emulsifier, and stabilizer;
disodium EDTA: it is a compound capable of reacting with Mg2+、Ca2+、Mn2+、Fe2+Divalent metal ion junctionThe chelating agent can remove the inhibition of heavy metal on enzyme or effective component.
The preparation method of the essence containing copper peptide comprises the following steps:
s1, adding xanthan gum, sodium hyaluronate and butanediol into a container, stirring uniformly, adding EDTA disodium and water, and stirring until the solid is completely dissolved;
and S2, adding the rest components into the step 1, and stirring until all the components are completely dissolved to obtain the essence.
Compared with the prior art, the invention has the beneficial effects that:
the invention contains copper peptide, ECTOIN (ECTOIN), fibronectin and other components, not only can solve the side effect of the sunscreen on the skin, but also can quickly repair the wound, and finally can repair the damage of the true epidermis of the skin, thereby providing a high-efficiency and safe skin care product for consumers.

In order to make the present invention more clear and intuitive for those skilled in the art, the present invention will be further described with reference to the accompanying drawings.
The essence containing copper peptide disclosed by the invention contains copper peptide, ECTOIN, fibronectin and barley seed extract, and the repairing target point extends from the deep layer of the skin to the surface layer of the skin, so that the damaged skin is repaired from inside to outside in all directions, the safety is very high, and the risk of excessive cell proliferation is avoided like EGF.
In addition, the water-based emulsion also comprises components such as a humectant, a thickening agent, a chelating agent and the like.
The essence containing copper peptide comprises the following components in percentage by weight:
the balance being pure water.
The invention will now be illustrated with reference to 4 examples and 9 comparative examples, each of which has the following composition and amounts as given in table 1 below.

Efficacy verification
Firstly, a testing instrument:
the Cutomer @ dual MPA 580, CK, Germany, the MPA system is a multi-functional analytical system that can simultaneously connect multiple probes, each with a different analytical window. The probe used for testing of the invention comprises: skin Water loss (TEWL) tester TM300, Cutomer MPA 580 (skin elasticity test probe)
II, experimental principle:
1. TEWL value, transdermal water loss (TEWL), measures the change in the vapor pressure of moisture adjacent to the skin surface. TEWL values represent the loss of water from the stratum corneum and are an important criterion for evaluating the barrier function of the stratum corneum. A lower TEWL value of the skin indicates a better barrier function of the skin, whereas the opposite is the worse. The efficacy of the moisturizing cosmetic can be evaluated by testing the TEWL value of the skin moisture loss, and the TEWL value can also be applied to allergic patch test, contact dermatitis, physical therapy, burn and monitoring of new tissues, so as to find out whether the protective function of the skin is damaged or not in time. The unit of TEWL is: g/hm3
2. The skin elasticity and skin elasticity testing principle is based on the suction and stretching principle, negative pressure is generated on the surface of tested skin to suck the skin into a specific testing probe, and the depth of the skin sucked into the testing probe is measured by a non-contact optical testing system. The test probe comprises a light emitter and a light receiver, the ratio of light (the ratio of emitted light to received light) is in direct proportion to the depth of the skin absorbed, so that a relation curve of the stretched length of the skin and the time is obtained, then the elasticity performance of the skin is determined through MPA software, the elasticity value is a ratio and is expressed by M, the maximum is 1, and the closer to 1, the better the elasticity of the skin is.
Thirdly, testing the population: each group had 20 persons, 9 groups, 18-50 years old volunteers, healthy skin, no history of skin disease and allergy.
Fourthly, testing conditions: the subject was allowed to stand for 30 minutes at room temperature 20. + -. 2 ℃ and relative humidity 50. + -. 5% before testing the relevant data
Fifthly, an experimental process: the experiment is divided into 9 groups, and the testers in groups 1-3 are respectively smeared on the left face of the example 1 and the right face of the comparative examples 1, 4 and 5; group 4 subjects applied example 2 to the left face and comparative example 2 to the right face; group 5 subjects applied example 3 to the left face and comparative example 3 to the right face; subjects in groups 6-9 were left-face rubbed on example 4 and right-face rubbed on comparative examples 6, 7, 8, 9 2 times daily on the face for 4 weeks, tested with TM300 and MPA 580 and recorded TEWL and M values.
Sixth, experimental results
1. Skin transdermal water loss Rate (TEWL) test
The test set the subjects' TEWL at week 0 to TEWL0 and week 4 to TEWL4 with the TEWL variance being Δ TEWL, which is TEWL4-TEWL0, with the Δ TEWL being negative and the lower the number, indicating less moisture loss from the skin after four weeks, i.e. better barrier function of the repaired skin, the healthier the skin, table 2 shows the average Δ TEWL values for 20 people per group.

The experimental results show that:
from the experimental results, it can be seen that the Δ TEWL value, example 1> example 2> example 3, shows that as the addition amount of the active ingredient in the essence of the present invention increases, the Δ TEWL value becomes smaller, i.e. the less the moisture of the skin is lost, which means that the barrier function of the stratum corneum of the skin is more complete, the skin is not easy to lose moisture, and the skin is not easily affected by the external environment to cause the skin sensitivity problem. The comparative examples 1, 4 and 5 have delta TEWL values higher than that of example 1, compared with example 1, and thus show that the essence prepared by removing any one of copper peptide, ECTOIN and fibronectin from the essence of the present invention has poor skin repairing effect, and the more skin moisture is lost, the skin looks healthy and moist without the essence of the present invention. The delta TEWL value of the comparative example 2 is larger than that of the example 2, which shows that the effect of repairing skin is not as good as that of the essence of the invention when the repairing component in the composition of the invention is replaced by the known component oligopeptide-1 with better repairing performance; the delta TEWL value of comparative example 3 is greater than that of example 3, indicating that replacing ECTOIN and barley seed extract in the essence of the invention with beta-glucan, a known moisture retention and anti-allergy ingredient, lost moisture faster than the essence of the invention, indicating that the effect of repairing skin is not as good as that of the essence of the invention. From the values of example 4 and comparative examples 6 to 9, it is found that the effect of copper peptide + ECTOIN + fibronectin + barley seed extract on the skin barrier function was much better than that of the composition of the present invention obtained by simply adding any one of the components when the amounts of the active ingredients were the same, indicating that the composition and the combination of the components of the present invention are excellent in the effect of repairing the skin. For a more intuitive view of the results of this experiment, see the delta TEWL histogram of fig. 1.
2. Skin elasticity (M) test
In the test, the M of the test subject at the 0 th week is set as M0, the M at the 4 th week is set as M4, the M change value is delta M, the delta M is M4-M0, the delta M is a positive number, and the higher the value is, the more the elasticity of the skin is increased after four weeks, namely, the better the elasticity of the skin is, the more the contents of collagen and elastin in the dermis are increased, the better the repairing effect on the skin is. Table 3 shows the average Δ M values for 20 persons per group.

The experimental results show that:
in order to more intuitively see the result of the experiment, the result can be seen in a delta M bar chart of fig. 2, and as can be seen from fig. 2, the delta M value, example 1 < example 2 < example 3, thereby illustrating that the increase value of the skin elasticity of the essence of the invention is increased along with the increase of the adding amount of the active ingredients, that is, the effect of increasing the skin elasticity is better when the proportion of each ingredient is increased within the concentration range of the invention, that is, the effect of repairing the skin matrix is better. The comparison of comparative examples 1, 4 and 5 with example 1 shows that the Δ M values are smaller than that of example 1, and thus it is demonstrated that the effect of increasing skin elasticity is deteriorated and the skin is not as plump and healthy as observed by naked eyes in the essence prepared by removing any one of copper peptide, ECTOIN and fibronectin from the essence of the present invention. The delta M value of the comparative example 2 is smaller than that of the example 2, which shows that the effect of repairing skin is not as good as that of the essence of the invention when the repairing component in the composition of the invention is replaced by the currently known oligopeptide-1 which is a relatively good repairing component; the Δ M value of comparative example 3 is less than that of example 3, indicating that the ability to increase skin elasticity is greatly diminished by replacing ECTOIN and barley seed extract in the serum of the invention with β -glucan, a known moisturizing and healing ingredient, indicating that its ability to increase the basal muscle mass is far less than that of the composition of the invention. From the values of example 4 and comparative examples 6 to 9, it is found that the average value of the increase in skin elasticity values by cupeptide + ECTOIN + fibronectin + barley seed extract is much higher than the value obtained by adding only one of the components when the amounts of the active ingredients are the same, indicating that the effect of the combination of the components of the present invention on the increase in skin elasticity is much better than that of the combination of the components of the present invention, indicating that the synergistic effect of the composition of the present invention is very excellent. For a more intuitive view of the results of this experiment, see the Δ M histogram of fig. 2.
3. Test Picture comparison
The effect of the essence of the invention in example 3 before and after 14 days of use of a patient with scald is compared, and the effect of the essence of the invention on the damaged skin can be seen more clearly in fig. 3, which shows that the effect of the essence of the invention on the damaged skin is very good.
The left graph in fig. 4 shows that the skin barrier of a patient suffering from hormone-dependent dermatitis is damaged, the face of the patient is red and itchy, and the skin is repeated repeatedly, and after 28 days of using the essence of the embodiment 3, the right graph in fig. 4 shows that the skin red and itchy is faded, no repetition occurs, and the skin cuticle returns to a healthy state, so that the essence of the invention has very excellent repairing and treating effects on the people with damaged skin barrier, and is safer and has no dependence compared with western medicines.
The embodiments described above are presented to enable those skilled in the art to make and use the invention. It will be readily apparent to those skilled in the art that various modifications to these embodiments may be made, and the generic principles described herein may be applied to other embodiments without the use of the inventive faculty. Therefore, the present invention is not limited to the embodiments described herein, and those skilled in the art should make improvements and modifications to the present invention based on the disclosure of the present invention within the protection scope of the present invention.
Claims (9)
1. An essence containing copper peptide, which comprises copper peptide and is characterized in that,
also comprises ECTOIN, fibronectin, and fructus Hordei vulgaris seed extract.
2. The essence containing copper peptide according to claim 1, wherein,
the copper peptide, the ECTOIN, the fibronectin and the barley seed extract are contained in the essence in percentage by weight:
0.05-0.5% of copper peptide, 0.05-5% of ECTOIN, 0.1-5% of fibronectin, 0.5-2% of barley seed extract and the balance of pure water.
3. The essence containing copper peptide according to claim 1 or 2,
the essence also comprises a humectant.
4. The essence containing copper peptide according to claim 3,
the moisturizer comprises sodium hyaluronate, butanediol, pentanediol and hexanediol, and the four moisturizers account for the following weight percentages in the essence:

9. The method for preparing the essence containing copper peptides according to claim 8, comprising the steps of:
s1, adding xanthan gum, sodium hyaluronate and butanediol into a container, stirring uniformly, adding EDTA disodium and water, and stirring until the solid is completely dissolved;
