本发明公开了一种促进皮肤损伤快速修复的伤口敷料及其制备方法。该新型伤口敷料含有10%~20%聚谷氨酸,10%~20%阿拉伯胶,10%~20%丙三醇,20%~40%去离子水,0.5%~1%阿魏酸,1%~2%角蛋白(来自羊毛),4~6%十三肽,以及10~20%气雾剂。本发明制得液体伤口敷料具有良好的抗菌、抗炎和修复皮肤的性能,在损伤皮肤上可以快速成膜,阻隔细菌和外物的入侵,在保证伤口透气的同时还能为伤口保湿,为伤口处的皮肤再生创造了良好的环境,具有良好的生物相容性能和生物降解性能,可避免在换药时对伤口造成的二次创伤。

技术领域
本发明属于伤口敷料技术领域,涉及一种促进皮肤损伤快速修复的液体伤口敷料及其制备方法。
背景技术
皮肤是人体最大的器官,是人体抵御外界病原体、机械损伤、化学损伤、辐射损伤等的第一道物理屏障,并且还可以帮助人体调节体温,防止体液流失,参与各种代谢途径等,对人体十分重要。一旦皮肤出现损伤,不光人体对外界的抵抗能力会降低,还经常会影响人们的日常生活,并在愈合后留疤影响美观。
现代伤口敷料以为伤口处上皮细胞自由移动和良好氧合提供理想环境为目的,同时还要减少细菌的定植以及促进组织的修复。除了保护伤口以外,给伤口保湿会有助于伤口愈合,并且良好的伤口敷料还需要具备吸附多余渗出物、渗透气体和液体、保护伤口床免受机械损伤,不易黏连、易于更换等特性。许多研究表明,天然产物在制备伤口敷料方面具有很大优势,它们具有良好的吸湿性能,可有效将伤口湿度维持在一定范围内;对伤口粘附力较轻,可减少更换时对新生组织的破坏;具有抗菌活性以防止伤口二次感染;具有极佳的生物相容性且无毒无刺激。因此,开发一款基于天然产物的液体伤口敷料帮助伤口愈合、皮肤修复是人们所需要的。
发明内容
本发明以保护伤口、促进伤口愈合为目的,提供一种具有吸附多余渗出物、抑菌消炎、保湿透气、促进伤口愈合、皮肤修复功能的液体伤口敷料及其制备方法。
本发明的目的是通过以下技术方案实现的:
一种促进伤口愈合的液体伤口敷料,由下列质量分数的原料制成:
一种促进皮肤损伤快速修复的液体伤口敷料按照以下质量百分比原辅料制备而成:10%~20%聚谷氨酸,10%~20%阿拉伯胶,10%~20%丙三醇,20%~40%去离子水,1%~2%阿魏酸,0.5%~1%角蛋白(来自羊毛),4%~6%十三肽,以及10~20%气雾剂。
一种促进伤口愈合的液体伤口敷料,其步骤如下:
(1)溶液A的制备:在配制罐中加入配制总量70~80%的水,依次缓慢加入称量好的聚谷氨酸、阿拉伯胶、对羟基苯甲酸乙酯、对羟基苯甲酸丙酯,搅拌10~20min,获得溶液A;
(2)溶液B的制备:向溶液A加入丙三醇,搅拌10~15min,控制pH在5~6.5,加入称量好的角蛋白、阿魏酸以及十三肽,搅拌均匀后,获得溶液B;
(3)溶液C的制备:将溶液B加入高压均质机,加水至溶液总量,温度维持在40~50℃,高速均质5~7min至整个体系无颗粒状固体成分,冷却至室温,获得溶液C;
(4)成品的制备:对溶液C采用135℃高温瞬时灭菌后,获得促进皮肤损伤快速修复的液体伤口敷料,最后通过压力灌装加入气雾剂获得成品。
本发明选择新鲜蓝莓果为原料,提取蓝莓花青素,再加以辅料进行复配,这些原辅料能起到预防心脑血管的作用,具体功效如下:
1、聚谷氨酸: 聚谷氨酸是利用纳豆菌与谷氨酸进行微生物发酵而成的天然高分子聚合物,具有良好的水溶性、生物相容性和生物降解性,具有一定的抗菌和保湿性能,可以吸收大量伤口渗出液,并可以促进细胞的迁移以及增强细胞的粘附,从而促进伤口的愈合。另外,聚谷氨酸具有良好的成膜性和可塑性,可以帮助液体敷料在伤口处快速成膜。
2、阿拉伯胶:阿拉伯胶,来源于豆科金合欢树属的树干渗出物,即树皮在受伤时分泌出的一种黏性体液,用来保护伤口并促进伤口的愈合。阿拉伯胶独特的表面特性使它可以降低不同系统之间的界面张力,从而通过静电相互作用和氢键等增加材料的稳定性。阿拉伯胶具有抗氧化活性,在伤口局部使用可以防止皮肤中的脂质过氧化,并刺激伤口愈合;具有抗菌活性,可以抑制微生物在伤口的定植,防止伤口感染;具有极佳的亲水性,使它可以吸收伤口处过量的渗出液。
3、丙三醇:具有保湿、抗氧化的功能,可以改善角蛋白的溶解性,在酸性环境下促进蛋白质和多糖的交联,改善液体敷料成膜后遇水即溶的特性。
4、阿魏酸:阿魏酸属于酚类化合物,是天然的抗氧化剂,具有抗癌、抗炎、抗辐射、抗菌等生理活性,可以降低肿瘤坏死因子和诱导型一氧化氮合酶表达介导的炎症反应水平。另有研究表明,阿魏酸可通过调节血管内皮生长因子、血小板衍生生长因子和缺氧诱导因子-1α在体内外促进血管生成,还可以通过调节细胞周期蛋白D1和血管内皮生长因子来促进内皮细胞的增殖,达到对创面治疗的作用。
5、角蛋白:角蛋白是一种纤维状结构蛋白,其中半胱氨酸残基含量较高,占总氨基酸残基的7-20%。由于富含二硫键而具有良好的机械耐久性,并且基于角蛋白的材料可以与伤口周围环境相互作用加速伤口的愈合。另外,有研究表明,源于毛发的角蛋白中包含与细胞外基质蛋白相同的细胞粘附序列,能够支持细胞的附着和增殖。
6、十三肽: 十三肽是一种由微生物发酵获得的生物活性多肽,其氨基酸序列为Cys-Phe-Arg-Val-Cys-Tyr-Arg-Gly-Ile-Cys-Tyr-Arg-Cys,它的主要作用是可以抗菌消炎、促进创面愈合,修复皮肤并减少疤痕的形成。
本发明的特点是以聚谷氨酸、阿拉伯胶、阿魏酸、角蛋白、十三肽、丙三醇为主要原料按照合适的配比和加工方式制备出了一种可快速成膜的液体伤口敷料,具有抗菌消炎、促进愈合、修复皮肤、减少疤痕的优点,并且还方便使用。
具体实施方式
下面对本发明的技术方案作进一步的说明,但并不局限于此,凡是对本发明技术方案进行修改或者同等替换,而不脱离本发明技术的精神和范围,均应涵盖在本发明的保护范围中。
实施例1
一种促进皮肤损伤快速修复的液体伤口敷料的制备方法,其步骤如下:
1、溶液A的制备:在配制罐中加入配制总量70~80%的水,依次缓慢加入总配制溶液量10%的聚谷氨酸、18%的阿拉伯胶,以及总量为0.02%的对羟基苯甲酸乙酯和对羟基苯甲酸丙酯,搅拌10~20min,获得溶液A;
2、溶液B的制备:向溶液A加入15%的丙三醇,搅拌10~15min,控制pH在5~6.5,加入总配制溶液量1%的角蛋白、1%的阿魏酸以及5%的十三肽,加热到40~50℃,搅拌15~20min后,获得溶液B;
3、溶液C的制备:将溶液B加入高压均质机,加水至溶液总量,温度维持在40~50℃,高速均质5~7min至整个体系无颗粒状固体成分,冷却至室温,获得溶液C;
4、成品的制备:对溶液C采用135℃高温瞬时灭菌后,获得促进皮肤损伤快速修复的液体伤口敷料,最后通过压力灌装加入气雾剂获得成品。
实例2
一种促进皮肤损伤快速修复的液体伤口敷料的制备方法,其步骤如下:
1、溶液A的制备:在配制罐中加入配制总量70~80%的水,依次缓慢加入总配制溶液量15%的聚谷氨酸、13%的阿拉伯胶,以及总量为0.02%的对羟基苯甲酸乙酯和对羟基苯甲酸丙酯,搅拌10~20min,获得溶液A;
2、溶液B的制备:向溶液A加入10%的丙三醇,搅拌10~15min,控制pH在5~6.5,加入总配制溶液量1%的角蛋白、1.5%的阿魏酸以及4.5%的十三肽,加热到40~50℃,搅拌15~20min后,获得溶液B;
3、溶液C的制备:将溶液B加入高压均质机,加水至溶液总量,温度维持在40~50℃,高速均质5~7min至整个体系无颗粒状固体成分,冷却至室温,获得溶液C;
4、成品的制备:对溶液C采用135℃高温瞬时灭菌后,获得促进皮肤损伤快速修复的液体伤口敷料,最后通过压力灌装加入气雾剂获得成品。
实例3
一种促进皮肤损伤快速修复的液体伤口敷料的制备方法,其步骤如下:
1、溶液A的制备:在配制罐中加入配制总量70~80%的水,依次缓慢加入总配制溶液量20%的聚谷氨酸、13%的阿拉伯胶,以及总量为0.02%的对羟基苯甲酸乙酯和对羟基苯甲酸丙酯,搅拌10~20min,获得溶液A;
2、溶液B的制备:向溶液A加入10%的丙三醇,搅拌10~15min,控制pH在5~6.5,加入总配制溶液量0.5%的角蛋白、1.5%的阿魏酸以及5%的十三肽,加热到40~50℃,搅拌15~20min后,获得溶液B;
3、溶液C的制备:将溶液B加入高压均质机,加水至溶液总量,温度维持在40~50℃,高速均质5~7min至整个体系无颗粒状固体成分,冷却至室温,获得溶液C;
4、成品的制备:对溶液C采用135℃高温瞬时灭菌后,获得促进皮肤损伤快速修复的液体伤口敷料,最后通过压力灌装加入气雾剂获得成品。
Claims (7)
1.一种促进皮肤损伤快速修复的液体伤口敷料及其制备方法,其特征在于所述新型伤口敷料按照以下质量百分比原辅料制备而成:10%~20%聚谷氨酸,10%~20%阿拉伯胶,10%~20%丙三醇,20%~40%去离子水,1%~2%阿魏酸,0.5%~1%角蛋白(来自羊毛),4%~6%十三肽,以及10~20%气雾剂。
2.根据权利要求1所述的一种促进皮肤损伤快速修复的液体伤口敷料,其特征在于由以下原辅料制备而成:10%聚谷氨酸,18%阿拉伯胶,15%丙三醇,30%去离子水, 1%阿魏酸,1.5%角蛋白(来自羊毛),4.5%十三肽,以及20%气雾剂。
3.根据权利要求1所述的一种促进皮肤损伤快速修复的液体伤口敷料,其特征在于由以下原辅料制备而成:15%聚谷氨酸,13%阿拉伯胶, 10%丙三醇,35%去离子水, 1%阿魏酸,1%角蛋白(来自羊毛),5%十三肽,以及20%气雾剂。
4.根据权利要求1所述的一种促进皮肤损伤快速修复的液体伤口敷料,其特征在于由以下原辅料制备而成:20%聚谷氨酸,13%阿拉伯胶, 10%丙三醇,40%去离子水, 0.5%阿魏酸,1.5%角蛋白(来自羊毛),5%十三肽,以及10%气雾剂。
5.根据权利要求1所述的一种促进皮肤损伤快速修复的液体伤口敷料,其特征在于所述气雾剂选自丙烷、丁烷、异丁烷、二氟乙烷中的一种或几种。
6.根据权利要求1所述的一种促进皮肤损伤快速修复的液体伤口敷料,其特征在于可包含防腐剂:对羟基苯甲酸丙酯钠和对羟基苯甲酸乙酯钠中的一种或几种。
7.根据权利要求1-6任一权利要求所述的促进皮肤损伤快速修复的液体伤口敷料的制备方法,其特征在于所述方法步骤如下:
(1)溶液A的制备:在配制罐中加入配制总量70~80%的水,依次缓慢加入称量好的聚谷氨酸、阿拉伯胶、对羟基苯甲酸乙酯、对羟基苯甲酸丙酯,搅拌10~20min,获得溶液A;
(2)溶液B的制备:向溶液A加入丙三醇,搅拌10~15min,控制pH在5~6.5,加入称量好的角蛋白、阿魏酸以及十三肽,加热到40~50℃,搅拌15~20min后,搅拌均匀后,获得溶液B;
(3)溶液C的制备:将溶液B加入高压均质机,加水至溶液总量,温度维持在40~50℃,高速均质5~7min至整个体系无颗粒状固体成分,冷却至室温,获得溶液C;
(4)成品的制备:对溶液C采用135℃高温瞬时灭菌后,获得促进皮肤损伤快速修复的液体伤口敷料,最后通过压力灌装加入气雾剂获得成品。
