Description
技术领域
本发明属于生物医药领域,涉及肿瘤差异表达基因及其应用。
背景技术
编码蛋白质的基因已经由学者通过人类基因组测序的方法检测出只占人类基因总数的1.5%左右(Stein,Lincoln DHuman genome:End of the beginning[J].Nature,2004,431(7011):915-916.)。LncRNA是一类由细胞内编码基因在正常转录过程中产生的无编码功能的并且总长度大于200个核苷酸的RNA分子。目前已经有超过50,000个人类的LncRNA被学者们发现且在细胞内受到详尽的网络调控,严格的内部结构保守是大多数LncRNA的特性(Xie C,Yuan J,Li H,et al.NONCODEv4:exploring the world of longnon-coding RNA genes[J].Nucleic Acids Research,2014,42(D1):D98-D103.)。大部分LncRNA是由RNA聚合酶II转录并且聚腺苷后酸化的,少部分LncRNA由RNA聚合酶III进行的转录。随着二代基因测序技术的不断开发,学者们发现在作用蛋白质和靶点上LncRNA复杂的二级结构具有很大的多样性和潜力(Chang H Y.Long noncoding RNAs:cellularaddress codes in development and disease.[J].Cell,2013,152(6):1298-1307.)。随着基因芯片技术的发展及应用,LncRNA广泛作用于肿瘤组织特异性差异表达。近年来越来越多的证据表明,LncRNA与多种疾病的产生与进展过程密切相关(Mercer T R,Dinger ME,Mattick J S.Long non-coding RNAs:insights into functions[J].NATURE REVIEWSGENETICS,2009,10(3):155-159.)。大量研究表明LncRNA在多种生物学过程中起着至关重要的作用,包括细胞增殖,代谢活动,分化,器官发生,计量补偿作用,基因转染功能和染色质重塑。
大量实验的结果表明,LncRNA作为早期诊断和治疗肿瘤以及判断预后的肿瘤分子标志物,可以通过促进或者抑制抑其他肿瘤相关基因的表达水平来参与恶性肿瘤的发生与发展过程。肝癌是我国成人恶性癌症中最常见的癌症之一。肝细胞癌预后差、致死率高的重要原因之一就是肝细胞癌发生、增殖与发展的相关分子机制目前仍然了解甚少。本申请立足于lncRNA与肝癌的研究,以期其能为肝癌的临床诊断和治疗提供新的途径。
发明内容
为了弥补现有技术的不足,本发明经过广泛而深入的研究,通过检测肝癌标本中lncRNA在肿瘤组织和正常组织的表达,发现其中具有明显表达差异的lncRNA,探讨其与肝癌的发生之间的关系,从而为肝癌的诊断及靶向治疗寻找更好的途径和方法。
为了实现上述目的,本发明采用如下技术方案:
本发明提供了用于检测基因标志物的一种或多种试剂在制备用于预测肝癌的产品中的用途,所述标志物包括AL161668.5。
进一步,所述试剂包括与来自AL161668.5的靶核苷酸序列能够特异性杂交的一种或多种寡核苷酸。
进一步,所述产品还包括从样本中分离核酸的试剂。
进一步,所述寡核苷酸可被检测地标记。
本发明提供了一种检测肝癌的试剂盒,所述试剂盒包括检测基因标志物的试剂,其中,所述标志物包括AL161668.5。

发明详述
本发明的发明人经过大量实验和反复研究,发现AL161668.5在肝癌组织中表达显著下调,为了探讨AL161668.5与肝癌的发生发展之间的相关性,通过增加AL161668.5基因表达来检测该基因对肝癌细胞的功能性影响。
本发明所用术语“差异表达”表示与第二种样品中相同的一种或多种本发明生物标志物的表达水平比较,经测定RNA的量或水平,在一个样品中本发明的一种或多种生物标志物的RNA和/或所述生物标志物RNA的一种或多种剪接变体表达水平的差异。差异表达可以如本文所述和本领域技术人员理解的方法确定。术语“差异表达”或“表达水平的变化”表示与第二种样品中给定生物标志物的可测定表达水平比较,经测定RNA的量,样品中给定生物标志物可测定表达水平的增加或降低。术语“差异表达”或“表达水平的变化”还可以表示与第二个样品群中生物标志物的可测定表达水平比较,样品群中给定生物标志物可测定表达水平的增加或降低。本文所用“差异表达”可以用给定生物标志物的表达水平相对于对照中给定生物标志物的平均表达水平的比率进行测定,其中比率不等于1.0。还可以用p值测定差异表达。当使用p值时,当p值小于0.1时生物标志物被鉴定为在第一和第二群体之间差异表达。更优选p值小于0.05。甚至更优选p值小于0.01。还更优选p值小于0.005。最优选p值小于0.001。当基于比率确定差异表达时,如果第一种和第二种样品中表达水平的比率大于或小于1.0,则RNA是差异表达的。举例来说,大于1.2、1.5、1.7、2、3、4、10、20的比率,或小于1的比率,例如0.8、0.6、0.4、0.2、0.1、0.05。在本发明的另一个实施方案中,如果第一群体的平均表达水平与第二群体的平均表达水平的比率大于或小于1.0,则核酸转录本是差异表达的。举例来说,大于1.2、1.5、1.7、2、3、4、10、20的比率,或小于1的比率,例如0.8、0.6、0.4、0.2、0.1、0.05。在本发明的另一个实施方案中,如果第一个样品中表达水平与第二个群体中平均表达水平的比率大于或小于1.0,例如包括比率大于1.2、1.5、1.7、2、3、4、10、20,或比率小于1,例如0.8、0.6、0.4、0.2、0.1、0.05,则核酸转录本是差异表达的。
“表达差异增加”或“上调”表示基因相对于对照而言,基因表达(以RNA表达测定)显示增加至少10%或更多,例如20%、30%、40%或50%、60%、70%、80%、90%或更多或1.1倍、1.2倍、1.4倍、1.6倍、1.8倍或更多。
“表达差异降低”或“下调”表示基因相对于对照而言,基因表达(以RNA表达或蛋白质表达测定)显示降低至少10%或更多,例如20%、30%、40%或50%、60%、70%、80%、90%或少于1.0倍、0.8倍、0.6倍、0.4倍、0.2倍、0.1倍或更少的基因。例如,上调基因包括与从正常个体分离的mRNA或蛋白质的表达相比,从以患有肝癌为特征的个体分离的血中mRNA或蛋白质表达水平增加的基因。例如,下调基因包括与从正常个体分离的血相比,从以患有肝癌为特征的个体分离的血中mRNA或蛋白质表达水平降低的基因。
在本发明中,术语“AL161668.5”位于14号染色体上,包括AL161668.5基因及其的同源物,突变,和同等型。该术语涵盖全长,未加工的AL161668.5,以及源自细胞中加工的任何形式的AL161668.5。该术语涵盖AL161668.5的天然发生变体(例如剪接变体或等位变体)。目前AL161668.5存在两个转录本,序列分别如ENST00000533984.1、ENST00000532213.2。一种代表性的AL161668.5的序列如ENST00000533984.1所示。
在本发明的实施方式中,本发明通过以下步骤来诊断肝癌:在来自个体的核酸样本中,检测与肝癌的诊断相关的基因标志物所对应的一个或多个核酸片段。在特定的实施方式中,检测对应于AL161668.5的核酸片段。
本文中使用的术语“片段”表示长度至少为约15、20、25、30、35、40、45、50、75、100、200、300、400、500、1000个核苷酸或更多核苷酸的多核苷酸。
本文中使用的术语“核酸”泛指:染色体的段;DNA、cDNA 和/或RNA的段或部分。核酸可以自最初与任何源分离的核酸样本(例如,与样本DNA或RNA分离、从样本DNA或RNA纯化、扩增、克隆或逆转录)获取或获得。
本文中使用的术语“寡核苷酸”表示由脱氧核糖核苷酸、核糖核苷酸或其任意组合构成的短聚合物。寡核苷酸的长度通常在大10个核苷酸和大约100个核苷酸之间。寡核苷酸优选地长度为15个核苷酸到70个核苷酸,最通常的是20个核苷酸到26个核苷酸。寡核苷酸可以用作引物或探针。
当寡核苷酸和核酸对齐时,如果该寡核苷酸与该核酸的一部分具有至少50%的序列同源性,则该寡核苷酸对该核酸为“特异”的。对一核酸特异的寡核苷酸是这样的寡核苷酸:在合适的杂交条件或洗涤条件下,其能够与所关注的目标杂交且基本上不与未关注的核酸杂交。较高程度的序列同源性是优选的且包括至少75%、至少80%、至少85%、至少90%或至少95%的序列同源性。
本文中使用的术语“杂交”或“特异性杂交”指的是两个互补的核酸链在适当严格的条件下彼此退火结合。通常利用探针长度的核酸分子来进行杂交。核酸杂交技术在现有技术中是公知的。本领域的技术人员了解如何估计和调整杂交条件的严格度,使得具有至少所需程度的互补性的序列将稳定地杂交,而具有较低互补性的序列将不能稳定地杂交。本发明的多核苷酸可以是DNA形式或RNA形式。DNA形式包括cDNA、基因组DNA或人工合成的DNA。DNA可以是单链的或是双链的。
本发明的人AL161668.5核苷酸全长序列或其片段通常可以用PCR扩增法、重组法或人工合成的方法获得。对于PCR扩增法,可根据已公开的有关核苷酸序列,尤其是开放阅读框序列来设计引物,并用市售的cDNA库或按本领域技术人员已知的常规方法所制备的cDNA库作为模板,扩增而得有关序列。当序列较长时,常常需要进行两次或多次PCR扩增,然后再将各次扩增出的片段按正确次序拼接在一起。
一旦获得了有关的序列,就可以用重组法来大批量地获得有关序列。这通常是将其克隆入载体,再转入细胞,然后通过常规方法从增殖后的宿主细胞中分离得到有关序列。
此外,还可用人工合成的方法来合成有关序列,尤其是片段长度较短时。通常,通过先合成多个小片段,然后再进行连接可获得序列很长的片段。
应用PCR技术扩增DNA/RNA的方法被优选用于获得本发明的基因。用于PCR的引物可根据本文所公开的本发明的序列信息适当地选择,并可用常规方法合成。可用常规方法如通过凝胶电泳分离和纯化扩增的DNA/RNA片段。
本发明AL161668.5的促进剂,当在治疗上进行施用(给药)时,可促进AL161668.5的表达,从而抑制肝癌。
作为一种可选择的实施方式,所述促进剂为核酸构建体,其包括复制系统以及能够在给定的靶细胞内转录的序列。作为一种优选的实施方式,所述核酸构建体包含编码本发明的AL161668.5的核苷酸序列。
本发明还包括表达载体,其中包含所述的核酸构建体。
有多种合适的载体可用于表达本发明的AL161668.5基因,包括例如质粒载体、病毒载体如腺病毒载体、腺相关病毒载体、慢病毒载体等。这样的载体中不仅包含用于启动转录的启动子,还包含增强子、转录终止信号或其它表达调节序列。本发明的载体可以作为裸质粒DNA、与转染剂或靶递送材料相复合的复合物或者作为重组病毒载体而递送到宿主细胞内。表达载体的选择和构建是本领域普通技术人员所知晓的,并可参考一些因素进行,例如待转染的宿主细胞的状态或类型、期望的基因的表达时间和水平等。
此外,表达载体优选地包含一个或多个选择性标记基因,以提供用于选择转化的宿主细胞的表型性状,如真核细胞培养用的二氢叶酸还原酶、新霉素抗性以及绿色荧光蛋白(GFP),或用于大肠杆菌的四环素或氨苄青霉素抗性。
本发明的表达载体可通过转染载体递送到靶细胞或宿主细胞内,所述转染载体例如有脂质体、水凝胶、生物粘合性微球等。本发明的表达载体还可通过磷酸钙沉淀法、直接显微注射法、电穿孔法、微粒轰击法等递送到靶细胞或宿主细胞内。这样的方法也是本领域普通技术人员所熟知的。
本发明还包括一种宿主细胞,其中转化或转染了本发明的核酸构建体或表达载体。在一些实施方案中,所述宿主细胞是原核细胞。在另一些实施方案中,所述宿主细胞是真核细胞,例如原代细胞培养物、细胞系、酵母、完整器官或生物体的细胞群等。本发明提供了一种药物组合物,其中包含药学有效量的本发明的AL161668.5基因、核酸构建体、表达载体和/或宿主细胞以及药学上可接受的载体。“药学有效量”指的是在导入受试者后可增加AL161668.5水平至期望程度或者可实现期望效果的药物剂量。药学有效量可由治疗医师根据经验确定,并可能取决于多种因素,例如给药途径、制剂性质、疾病状况、受试者的体格、体重、年龄、性别等。
本发明的药物组合物可以配制成固体、半固体、液体或气体的形式,例如可以是片剂、胶囊、粉剂、颗粒、油膏、溶液、栓剂、注射剂、吸入剂等。这类制剂的制备方法是本领域普通技术人员所知晓的。
本发明的药物组合物可通过合适的给药途径施用给待处理/治疗的细胞、组织、器官和/或受试者,例如可以将药物组合物配制成合适的制剂形式而通过口服、口含、胃肠外、皮下、眼内、肌内、腹膜内、静脉内、经鼻、经直肠等方式给药。
为了促进本发明的AL161668.5基因、核酸构建体或表达载体递送到目的细胞、组织、器官或受试者内,可以将所述AL161668.5基因、核酸构建体或表达载体与运载体或赋性剂相混合。可应用的运载体或赋性剂包括盐水、缓冲液(如乙酸缓冲液、柠檬酸缓冲液、磷酸缓冲液等)、氨基酸、抗坏血酸、醇类、蛋白质、脂质体、甘油、山梨醇等。
作为一种可选择的实施方式,本发明又提供了一种预防、治疗或缓解AL161668.5基因相关疾病或病症、例如肝癌的方法,其包括对有此需要的个体施用本发明的促进剂。
在本发明中,术语“受试者”和“个体”可以互换使用,指的是在其中增加AL161668.5水平能取得有益效果的任何动物,优选脊椎动物,例如哺乳动物,其实例有人、马、牛、小鼠、大鼠等。
本发明的优点和有益效果:
本发明首次发现了肝癌的特征基因,所述特征基因在肝癌中表达下调。通过检测AL161668.5的表达水平,可以对肝癌以及肝癌的患病风险进行评估,具有较高的敏感性和特异性。同时,改变AL161668.5的表达水平,可以影响肝癌细胞的功能,从而为肝癌的治疗提供了新途径。

实施例1检测AL161668.5在肝细胞癌中的表达情况
1、样品收集
分别收集27例原发性肝细胞癌患者的癌组织及相对应的癌旁组织样本(距离肿瘤边缘≥5cm的肝组织),配对的癌组织经病理证实为肝细胞癌。标本于手术后内用大量生理盐水清洗干净后立即分装,经液氮冷冻后放入-80℃冰箱冻存。
2、RNA样品的制备及质量分析
使用TRIZOL法提取组织总RNA
1)用剪刀组织剪碎,加入1ml Trizol,振荡器上震荡1min;常温放置10min,使核蛋白体完全分解。
2)加入200μl三氯甲烷(氯仿),盖紧管盖,剧烈震荡15s,常温静置10min。
3)4℃,11000rpm离心15min。
4)将水样层转移到一个新的离心管中,加入500μl异丙醇;颠倒混匀后,常温静置10min。
5)4℃,11000rpm离心15min。
6)用枪小心吸走液体,留沉淀在管底,加入1ml 75%的乙醇,在振荡器上震荡5s,洗涤沉淀一次。
7)4℃,8000rpm离心5min。
8)将上清小心去掉,干燥沉淀10min,加入适量的水溶解沉淀10min。
9)检测RNA浓度,鉴定RNA的产量和纯度。
3、QPCR检测
1)引物设计
根据AL161668.5和GADPH的基因序列设计引物,具体引物序列如下:
AL161668.5基因:
正向引物为5’-AATCAATATAGGCTACAAG-3’(SEQ ID NO.1);
反向引物为5’-CACTACAGATAGATGAAC-3’(SEQ ID NO.2)。
GAPDH基因:
正向引物为5’-AATCCCATCACCATCTTCCAG-3’(SEQ ID NO.3);
反向引物为5’-GAGCCCCAGCCTTCTCCAT-3’(SEQ ID NO.4)。
2)逆转录反应
采用FastQμant cDNA第一链合成试剂盒(货号:KR106)进行lncRNA反转录,首先去除基因组DNA反应,在试管中加入5×gDNA Bμffer 2.0μl,总RNA 1μg,加Rnase Free ddH2O使总体积至10μl,水浴锅中42℃加热3min,再将10×Fast RT Bμffer 2.0μl,RT EnzymeMix 1.0μl,FQ-RT Primer Mix 2.0μl,RNase Free ddH2O 5.0μl,混合后加入上述试管中一起混合共20μl,水浴锅中42℃加热15min,95℃加热3min。
3)QPCR扩增检验
用SuperReal PreMix Plus(SYBR Green)(货号:FP205),进行扩增,实验操作按产品说明书进行。
采用20μl反应体系:2×SuperReal PreMix Plus 10μl,正反向引物(10μM)各0.6μl,5×ROX Reference Dye△2μl,DNA模板2μl,灭菌蒸馏水4.8μl。每个样本设置3个平行管,所有扩增反应均重复三次以上以保证结果的可靠性。
扩增程序为:95°15min,(95℃10s,55℃30s,72℃ 32s)×40个循环。
4)cDNA模板浓度的筛选
将各样本cDNA混合后,以此为模板进行10倍梯度(10、100、1000、10000、100000倍)稀释,稀释后样品各取2μl作模板,分别用目的基因引物和内参基因引物进行扩增,同时在60-95℃进行融解曲线分析,根据扩增效率高和溶解曲线单峰原则进行模板浓度的筛选。
根据溶解曲线,可以看出,当cDNA的进行10倍稀释时,PCR的扩增效率较高,溶解曲线单峰比较好。
5)样品Real Time PCR检测
将各样品cDNA 10倍稀释后取2μl作模板,分别用目的基因引物和内参基因引物进行扩增。同时在60-95℃进行溶解曲线分析,通过融解曲线分析和电泳确定目的条带,2-ΔΔCT法进行相对定量。
4、结果
QPCR结果显示,以AL161668.5在对照组(癌旁组织)中的表达量作为基准定为1,与癌旁组织相比,AL161668.5在肝细胞癌组织中表达下调(图1),差异具有统计学意义(P<0.05)。
具体表达情况如表1所示,呈现AL161668.5显著下调的样本有26例,其中癌组织样本有19例,癌旁组织样本有7例。说明AL161668.5应用于肝细胞癌的诊断具有较高的特异性和敏感性。

实施例2AL161668.5的诊断效能的检测
1、数据收集
从TCGA数据库中下载lncRNA的表达谱数据,其中包括371例肝癌组织和50例癌旁组织。
2、ROC曲线分析
使用R语言中的pROC包分析lncRNA AL161668.5的受试者工作特征,计算二项精确置信空间,绘制ROC曲线。
3、结果
ROC分析结果如图2所示,从图中可以看出,AL161668.5作为检测变量具有较高的曲线下面积(AUC=0.863),最佳临界点为80.500,最佳临界点的特异性为0.940,敏感性为0.722。说明使用AL161668.5进行肝细胞癌的诊断具有较高的敏感性和特异性。
实施例3AL161668.5的功能验证
1、细胞培养
人肝癌细胞系HepG2购于上海细胞库,细胞系均以含10%胎牛血清和1%P/S的培养基DMEM在37℃、5%CO2的培养箱中培养。每天观察细胞生长情况,隔天换液。
2、转染
2.1过表达载体的构建
由上海吉凯基因设计并合成AL161668.5的过表达载体,将实验分为三组:对照组(HepG2)、阴性对照组(转染空载)和实验组(过表达AL161668.5)
2.2转染
按照Invitrogen公司的lipofectamine 2000转染试剂说明书进行转染。具体步骤如下:
1)将肝癌细胞系HepG2加入含10%胎牛血清的DMEM培养基并置于37℃、5%CO2的培养箱中培养。当细胞生长融合至80-90%的时,用PBS清洗3遍以去除细胞瓶中的血清成分,然后加入1-2mL的含0.25%的胰酶0.02%EDTA的消化液,当细胞变为单个细胞时加入1mL的完全培养基终止消化。
2)将细胞转移至离心管内,放入常温离心机,1000g/min离心5min收集细胞沉淀,用完全培养基将细胞重悬为1×105/mL,吸取500μL接种于24孔板内,放入37℃、5%CO2的培养箱中培养过夜。
3)在OPTI-MEM培养液中加入Lipofectamine 2000,室温孵育5min。
4)在OPTI-MEM培养液中加入过表达载体混合均匀。
5)将稀释好的3)和4)混合均匀,室温放置20min。
6)将混合液加入到无血清培养基培养细胞的24孔板中,轻轻晃动混匀,放入37℃、5%CO2的培养箱中培养6h后将细胞培养液更换为新鲜的完全培养基继续培养。
3、QPCR检测细胞中AL161668.5的表达水平
各组细胞转染培养48h后,使用Trizol法提取细胞总RNA,按照实施例1中的方法进行逆转录以及实时定量PCR检测。
4、CCK-8法检测细胞增殖能力
1)各组细胞转染后24h,用PBS清洗3次去除细胞内的血清成分,然后用胰蛋白酶将细胞充分消化,1000rpm离心5min收集细胞,弃去上清液并用1ml含10%胎牛血清的DMEM培养基重悬,细胞计数板对细胞进行计数。
2)用含有10%胎牛血清的DMEM培养基将细胞稀释至1×104/mL,然后向96孔板的每孔加入200μL细胞(每种细胞接种5个复孔)。
3)在96孔板四周未铺细胞的其他孔中加入200μL的PBS。
4)将接种好肿瘤细胞的96孔板放至37℃、5%CO2的细胞培养箱内培养72h后加入10μL CCK-8,放入培养箱内孵育1h。
5)将培养板取出,用酶标仪测量450nm波长的吸光度。
5、细胞迁移实验
1)各组细胞转染后24h,用PBS清洗3次去除细胞内的血清成分,然后用胰蛋白酶消化细胞,将细胞悬液转移至15mL离心管内,然后放入室温离心机内,1000rpm离心5min收集细胞。弃去上清然后用1ml不含胎牛血清的DMEM培养基重悬细胞,用细胞计数板对细胞进行计数。用不含胎牛血清的DMEM培养基将细胞稀释至1×106/mL,吸取1×105个细胞滴加入Transwell小室的上室。
2)向Transwell小室的下室加入600μL含10%血清的完全培养基,然后将上室放入孔板内,置于37℃、5%CO2的细胞培养箱内培养24h。
3)取出小室,轻轻弃去小室内的培养基,用PBS清洗2遍后放入75%的乙醇固定液中中固定15min。
4)用棉签将Transwell上室内细胞轻轻擦去,然后将小室置于结晶紫中染色15min。
5)取出小室,用PBS清洗3-5次去除多余细胞染液,然后在显微镜下进行观察。
6、细胞侵袭实验
将基质胶用无血清培养基1:20稀释,取100μL铺于小室内,4℃孵育过夜。剩余步骤同细胞迁移实验。
7、统计学分析
所有数据均为三次独立实验获得,按照Mean士SD显示。组间差异使用配对样本t检验分析,p<0.05时表示差异具有统计学差异。
8、结果
1)转染结果显示,以AL161668.5在对照组中(HepG2)的表达量作为基准定为1,转染结果显示,相比对照组,实验组在转染过表达AL161668.5的载体后AL161668.5的表达水平(4.36±0.743)显著上调(对照组vs实验组,P值=0.0159,*),而转染空载的阴性对照组中的AL161668.5的表达水平(0.937±0.0503)则无显著变化(对照组vs阴性对照组,P值=0.161,ns)
2)细胞增殖实验结果,相比阴性对照组的细胞增殖活性,实验组的细胞增殖活性显著降低(图3),差异具有统计学意义(阴性对照组vs实验组,P=0.0009,***),说明AL161668.5影响肝癌细胞的增殖,提示AL161668.5可作为分子靶标应用于肝癌的治疗。
3)细胞迁移和侵袭实验结果显示,相比迁移实验的阴性对照组,迁移实验的实验组的细胞穿膜数显著降低(图4A),差异具有统计学意义(阴性对照组vs实验组,P=0.0158,*),说明增加AL161668.5的表达水平可以降低细胞的迁移能力,提示AL161668.5可应用于肝癌转移的治疗;相比侵袭实验的阴性对照组,侵袭实验的实验组的细胞穿膜数显著降低(图4B),差异具有统计学意义(阴性对照组vs实验组,P=0.0166,*),说明增加AL161668.5的表达水平可以降低细胞的侵袭能力,提示AL161668.5可应用于肝癌浸润的治疗。
上述实施例的说明只是用于理解本发明的方法及其核心思想。应当指出,对于本领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以对本发明进行若干改进和修饰,这些改进和修饰也将落入本发明权利要求的保护范围内。
