Abstract
本发明公开了一种水中微生物检测用染色剂,由蒸馏水、醋酸、甲苯胺蓝、天然植物色素提取物、亚硫酸钠、二乙烯三胺、乙酸、聚乙二醇、辛基酚聚氧乙烯醚制成。与现有技术相比,本发明采用科学的配比及天然植物色素构成,针对于水中微生物着色力强,使得水中微生物容易在显微镜下观察,使得观察更清晰,提高水质检测精准度,具有推广应用的价值。

技术领域
本发明涉及一种生物检测染色剂,尤其涉及一种水中微生物检测用染色剂。
背景技术
水中常见的微生物有细菌、真菌、噬菌体、病毒、原生动物、藻类等。现有技术中,由于水中微生物颜色较为透明,在显微镜下观察不清晰,现有的染色剂对水中为生物的染色较淡,着色力差,导致观察难度提高,因此,需要一种针对于水中微生物的染色剂。
发明内容
本发明的目的就在于为了解决上述问题而提供一种水中微生物检测用染色剂。
本发明通过以下技术方案来实现上述目的:
本发明由蒸馏水、醋酸、甲苯胺蓝、天然植物色素提取物、亚硫酸钠、二乙烯三胺、乙酸、聚乙二醇、辛基酚聚氧乙烯醚制成。
优选的,按重量比,每份含所述蒸馏水92%、所述醋酸2%、所述甲苯胺蓝1%、所述天然植物色素提取物4%、所述亚硫酸钠0.2%、所述二乙烯三胺0.1%、所述乙酸0.3%、所述聚乙二醇0.2%、所述辛基酚聚氧乙烯醚0.2%。
优选的,所述天然植物色素提取物由黑加仑葡萄皮加等量重量的水进行超声提取后过滤得到的天然植物色素提取物。
本发明制作方法包括以下步骤:
S1:将蒸馏水加热至60℃;
S2:将醋酸、甲苯胺蓝加入到步骤S1中获得的溶液中搅拌,得到混合溶液;
S3:向S2中获得的溶液中加入亚硫酸钠、二乙烯三胺搅拌至淡蓝色;
S4:将天然植物色素提取物加入S3中获得的溶液中搅拌至溶解;
S5:将乙酸加入S4中获得的溶液中搅拌;
S6:向S5中获得的溶液中加入聚乙二醇、辛基酚聚氧乙烯醚搅拌至溶解,获得所述水中微生物检测用染色剂。
本发明的有益效果在于:
本发明是一种水中微生物检测用染色剂,与现有技术相比,本发明采用科学的配比及天然植物色素构成,针对于水中微生物着色力强,使得水中微生物容易在显微镜下观察,使得观察更清晰,提高水质检测精准度,具有推广应用的价值。

下面对本发明作进一步说明:
本发明由蒸馏水、醋酸、甲苯胺蓝、天然植物色素提取物、亚硫酸钠、二乙烯三胺、乙酸、聚乙二醇、辛基酚聚氧乙烯醚制成。
优选的,按重量比,每份含所述蒸馏水92%、所述醋酸2%、所述甲苯胺蓝1%、所述天然植物色素提取物4%、所述亚硫酸钠0.2%、所述二乙烯三胺0.1%、所述乙酸0.3%、所述聚乙二醇0.2%、所述辛基酚聚氧乙烯醚0.2%。
优选的,所述天然植物色素提取物由黑加仑葡萄皮加等量重量的水进行超声提取后过滤得到的天然植物色素提取物。
本发明制作方法包括以下步骤:
S1:将蒸馏水加热至60℃;
S2:将醋酸、甲苯胺蓝加入到步骤S1中获得的溶液中搅拌,得到混合溶液;
S3:向S2中获得的溶液中加入亚硫酸钠、二乙烯三胺搅拌至淡蓝色;
S4:将天然植物色素提取物加入S3中获得的溶液中搅拌至溶解;
S5:将乙酸加入S4中获得的溶液中搅拌;
S6:向S5中获得的溶液中加入聚乙二醇、辛基酚聚氧乙烯醚搅拌至溶解,获得所述水中微生物检测用染色剂。
实施例:
取常规待检测水备用。滴加所得水中微生物检测用染色剂约100μl。着染10~15min,染色同时在显微镜下观察染色情况,终止染色,于100×油镜下观察。显微镜观察结果显示:本实施例水中微生物检测用染色剂可使水中微生物清晰着染,但不能着染细胞核,染色封片后48h内,保持着清晰着染,但不能着染细胞核,显示更加锐利和清晰。

1.一种水中微生物检测用染色剂,其特征在于:由蒸馏水、醋酸、甲苯胺蓝、天然植物色素提取物、亚硫酸钠、二乙烯三胺、乙酸、聚乙二醇、辛基酚聚氧乙烯醚制成。
2.根据权利要求1所述的水中微生物检测用染色剂,其特征在于:按重量比,每份含所述蒸馏水92%、所述醋酸2%、所述甲苯胺蓝1%、所述天然植物色素提取物4%、所述亚硫酸钠0.2%、所述二乙烯三胺0.1%、所述乙酸0.3%、所述聚乙二醇0.2%、所述辛基酚聚氧乙烯醚0.2%。
3.根据权利要求1所述的水中微生物检测用染色剂,其特征在于:所述天然植物色素提取物由黑加仑葡萄皮加等量重量的水进行超声提取后过滤得到的天然植物色素提取物。
4.根据权利要求1所述的水中微生物检测用染色剂,其特征在于:制作方法包括以下步骤:
S1:将蒸馏水加热至60℃;
S2:将醋酸、甲苯胺蓝加入到步骤S1中获得的溶液中搅拌,得到混合溶液;
S3:向S2中获得的溶液中加入亚硫酸钠、二乙烯三胺搅拌至淡蓝色;
S4:将天然植物色素提取物加入S3中获得的溶液中搅拌至溶解;
S5:将乙酸加入S4中获得的溶液中搅拌;
S6:向S5中获得的溶液中加入聚乙二醇、辛基酚聚氧乙烯醚搅拌至溶解,获得所述水中微生物检测用染色剂。
