The chemical structure of the pueraria isoflavone is a bisphenol structure, and is a phytoestrogen which is most similar to the estrogen secreted by a human body on the molecular structure. On one hand, the compound can act on an estrogen receptor in vivo and produce an effect, and the effect is mild; on the other hand, the dynamic balance of estrogen in the body can be maintained: when the estrogen is deficient in the body, the pueraria isoflavone can play a role in supplementing the estrogen and has no obvious influence on the normal estrogen level; when the estrogen level in the body is too high, the pueraria isoflavone can prevent the estrogen from acting on a target organ too much, so that the estrogen level is kept relatively balanced; research shows that the pueraria isoflavone can improve hypofunction of ovaries caused by the fact that the estrogen level in the bodies of the aged rats is reduced to enhance the lipid peroxidation and reduce the activity of free radical scavenging enzymes. The present invention will be described in further detail with reference to specific examples, but the scope of the present invention is not limited to the examples.
The acne-removing composition disclosed by the invention applies the concepts of dialectical treatment and comprehensive treatment in the traditional Chinese medicine, adopts various plant extracts, and aims at all targets and links related to acne pathogenesis: excessive androgen secretion or/and over-expression of testosterone receptor; sebaceous gland exuberance and sebum accumulation in hair follicle; hyperkeratosis of hair follicle epithelium and poor excretion of sebum in hair follicle; fourthly, infection of propionibacterium acnes in hair follicles; local blood and lymph reflux disorder, hemoglobin and pigmentation; sixthly, local inflammatory mediator is excessively released; effectively blocks the pathophysiology process, thereby achieving the triple effects of controlling oil, removing acne and repairing acne marks.
The acne clinical manifestation severity classification standard is as follows:
mild (grade i): acne is a major skin lesion with small amounts of papules and pustules, with a total lesion count of less than 30.
Moderate (including II and III)
(stage II): has acne, moderate papules and pustules, and 31-50 total lesions.
(stage III): a large number of papules and pustules are occasionally associated with large inflammatory lesions, which are widely distributed. The total number of the lesions is 51-100, and the number of the nodules is less than 3.

Example 1

800 patients clinically diagnosed with acne were collected, wherein 420 male patients and 380 female patients were aged 18-30 years; patients were divided into 8 groups of 100 people each, with the acne being graded (similar numbers and proportions of mild, moderate and severe).

As can be seen from the results in table 2, the saw palmetto essential oil, the angelica ferulic acid and the pueraria isoflavone can be added according to a specific proportion to realize the best acne removing effect under the condition of the same basic formula; if only one or two of the above ingredients are selected, the effect is improved but the best effect is not achieved compared with the basic formula.
Example 2
In order to further illustrate that the saw palmetto essential oil, the angelica ferulic acid and the pueraria isoflavone have the acne-removing effect, animal experiments further verify that the saw palmetto essential oil, the angelica ferulic acid and the pueraria isoflavone have the acne-removing effect.
This example used a conventional method (Kligman method, a standard rabbit ear model established by the American dermatological society in 1989) to establish a rabbit ear acne model. The method specifically comprises the following steps: coating coal tar for 2cm × 2cm at the opening of ear canal at the inner side of external auditory canal of male rabbit for 2 weeks, 0.5ml each time, 1 time per day, and making it into experimental acne model; thereafter, acne was topically applied 2 times a day for a total of 4 weeks with cream 0.25ml, and the number of comedones was observed again to verify the therapeutic effect of the drug.
The rabbit ear acne is successfully established according to the conventional method, and 10 rabbits with moderate and severe acne are selected as a group; the experimental animals are divided into: the basic formula group, experimental group 1 (basic formula + saw palmetto essential oil + angelica ferulic acid + pueraria isoflavone), experimental group 2 (basic formula + saw palmetto essential oil), experimental group 3 (basic formula + angelica ferulic acid), experimental group 4 (basic formula + pueraria isoflavone), each group had 20 (n ═ 20), the time of ointment application was four weeks, and the experimental results are shown in table 3:

The experimental results were analyzed by X + -s, and the P was <0.01 in the experimental group compared with the basic formula group, and the comparison between the two groups was statistically different.
As can be seen from table 3: the basic formula is added with saw palmetto essential oil, angelica ferulic acid and pueraria isoflavone, and the treatment effect on the acne is better than that of other groups.
Analysis of the mechanisms of acne formation and treatment found that: the saw palmetto extract is locally used, so that the local skin androgen receptor and inflammatory reaction can be effectively inhibited, the secretion quantity and inflammatory reaction of local skin hair follicle endothelial sebaceous glands can be effectively reduced, the local hair follicle keratinization degree is reduced, and the clinical treatment effect of acne is improved. Local inflammation of hair follicles of acne causes local capillary constriction and blood circulation disturbance, and local red swelling and blood stasis; the saw palmetto achieves the effect of dredging local blood circulation by expanding local capillary vessels. The Serenoa repens extract is mainly fatty acid, and has high skin absorption rate and strong skin permeability when being used topically. The composition of the invention is added with the component for enhancing the skin permeability, so that the saw palmetto can effectively enter the dermis layer of the skin and fully exert the pharmacological effect thereof. At present, the domestic and foreign documents report that saw palmetto is mainly used for treating benign prostatic hyperplasia, and the using method and oral administration are the main; the invention discovers for the first time that the saw palmetto extract can be used for treating acne by local skin application and has the effect of preventing acne recurrence.
The factors such as sebaceous gland aggregation and Propionibacterium acnes infection cause aseptic and bacterial inflammatory reaction of the lesion local hair follicle, factors such as local inflammatory mediators and prostaglandin E aggravate local inflammatory reaction, multiple injuries of skin tissues are caused, and the factors are also one of the main reasons for forming scars locally. The angelica ferulic acid selected by the invention has pharmacological actions of resisting inflammation, resisting oxidation, inhibiting amino acid metabolism and the like, can directly resist the action of inflammatory mediators, reduces the release amount of prostaglandin E in inflammatory tissues, and inhibits inflammatory exudation, thereby inhibiting the subsequent inflammatory reaction.
Increased androgen levels or/and decreased estrogen levels in the body are one of the major pathogenesis of acne. The pueraria isoflavone is locally used, so that the local skin estrogen concentration can be increased, and the excessive sebaceous gland secretion caused by the overhigh local skin androgen level can be effectively resisted, so that the generation and the recurrence of the acne can be effectively inhibited from the root. The pueraria isoflavone can accelerate blood flow speed of local skin of acne, dilate capillary vessel, relieve vasospasm, inhibit platelet aggregation, repair endothelial cells, improve PGl2/TXA2 level and reduce catecholamine generation, thereby reducing blood viscosity, inhibiting thrombosis, accelerating local inflammation absorption, relieving tissue edema and eliminating local skin acne mark and pigmentation.
Example 3
The acne removing composition containing the serenoa repens essential oil is prepared from the following raw materials in parts by weight: 0.10 part of saw palmetto essential oil, 0.02 part of angelica ferulic acid, 0.60 part of pueraria isoflavone, 0.50 part of wheat protein extract, 3.00 parts of salicylic acid, 1.00 part of nicotinamide, 0.20 part of citric acid, 0.10 part of zinc gluconate, 3.00 parts of aloe vera essence freeze-dried powder, 0.05 part of patchouli essential oil, 0.01 part of chamomile essential oil, 3.00 parts of allantoin, 0.20 part of hyaluronic acid, 0.50 part of dipotassium glycyrrhizinate, 1.00 part of plant antibiotic composition and 0.50 part of arbutin.
The formula is prepared into cream by the following method, wherein the phase C is the composition:
(1) dispersing 0.016kg of xanthan gum and 0.16kg of polyacrylate crosslinked polymer-6 by using 1kg of glycerol and 0.6kg of 1, 3-butanediol, adding into 12.62kg of purified water, after complete dissolution, adding 0.08kg of aloe freeze-dried powder and 2.12kg of 1, 3-butanediol, stirring and dissolving, heating to 75 ℃ after the solution is completely dissolved and is semitransparent mucus, controlling the stirring speed to be 20rpm, and avoiding generation of a large amount of bubbles to obtain phase A.
(2) Transferring 0.54kg of cetostearyl alcohol, 0.2kg of SQE21 emulsifier, 0.4kg of dimethicone, 1kg of caprylic/capric triglyceride, and 0.6kg of C24 (ethylhexyl palmitate) into an oil phase pot, heating to 75 deg.C, and stirring while heating; the solution is white emulsion after being fully dissolved, and the heating time is controlled within 20 minutes to obtain a phase B.
(3) Slowly adding the phase B into the phase A at a constant speed under rapid stirring, continuously stirring for 15 minutes after homogenization, stirring at a speed of 20rpm, cooling to 40 ℃, accurately weighing 0.9kg of phase C raw material, adding, and continuously stirring for 10 minutes; stirring at low speed, filtering, and discharging to obtain the final product.
The using method comprises the following steps: applying on affected part, massaging gently for 2-3 times per day for three weeks, observing and collecting clinical results, and collecting and analyzing data.
In the embodiment, 100 patients clinically diagnosed as acne are randomly collected, wherein 55 male patients and 45 female patients are aged between 18 and 30 years; the effective rate of the acne-removing composition in the embodiment is 98%, wherein the effective rate is (the number of cured people plus the number of effective people)/the total number of people multiplied by 100%.
Typical case 1
Patient, female, 19 years old, long acne 4 years, oily skin. The acne is a severe mixed type acne with a large number of acne marks and pigmentation, and the acne is treated for a plurality of times in a beauty parlor before, and is often repeated and can not be cured all the time. The formula is prepared into cream by the following method, wherein the phase C is the composition:

The formula is prepared into cream by the following method, wherein the phase C is the composition:
