技术领域
本发明属于促睡眠保健品领域,具体涉及乳源活性肽CCL-3S在制备促睡眠产品中的应用。
背景技术
人的一生当中,睡眠约占生命的三分之一,睡眠良好是身心健康的主要标志。但随着现代社会的发展,人们的生活节奏普遍加快,各方面压力等造成失眠的现象越来越多。睡眠不足直接导致人们思维迟缓、头脑紧张、注意力不集中、行动迟缓、厌烦、忧虑、倦怠,对工作缺乏热情,使工作主动性与工作意愿下降,从而降低工作效率。长期以往,睡眠不足的人会感到特别的疲惫辛苦,情绪不稳定,暴躁、易怒,缺乏自制力,造成心理方面严重的问题。此外,失眠还会导致抵抗力下降,患病机率大。
临床上常用化学合成类镇静催眠药治疗失眠及其他疾病引起的睡眠障碍。此类药物在延长睡眠时间的同时,都会出现如头晕、乏力、困倦、注意力不集中等不同程度的副作用,甚至出现暂时性健忘和宿醉现象,并未真正起到改善睡眠质量的作用。因此,提供药效显著、副作用小、安全的改善睡眠作用的药物是医药卫生领域的一项重要任务。
大脑中存在着一系列由神经核团纤维和递质组成的睡眠、觉醒调节系统,彼此之间形成既有形态上联系又有功能上制约的神经网络。下丘脑内视前区及其邻近基底前脑神经元为睡眠发生系统。视前区的促眠神经元主要位于下丘脑腹外侧视前区(VLPO),对睡眠的产生和维持有着至关重要的作用。
牛奶具有促进睡眠的作用,睡前喝杯热牛奶更容易入睡。牛乳中含有多种生物活性多肽,但天然牛乳中活性多肽的含量较少,无法满足亚健康人们的需要。牛乳酪蛋白,是许多具有潜在生物活性物质的前体,通过蛋白酶的酶解作用,得到具有生物学功能的小肽,即乳源生物活性肽。这些小肽一般由2-20个氨基酸残基组成,能够对机体的消化系统、心血管系统、免疫系统等多方面产生积极的影响,具有调节神经系统、抗菌、抗氧化、抗高血压、免疫调节等生理活性。乳源肽的抗菌、抗氧化、抗高血压、免疫调节作用研究得比较多,而对乳源肽调节神经系统的研究较少。
因此,挖掘更多具有调节神经系统作用的促睡眠肽对于开发安全、高效的促睡眠产品具有重要的价值。
发明内容
为了克服上述现有技术的不足,本发明的首要目的是提供乳源活性肽CCL-3S在制备促睡眠产品中的应用。
本发明的第二个目的是提供一种促睡眠药品。
本发明的第三个目的是提供一种促睡眠功能性食品。
为实现上述目的,本发明所采用的技术方案为:
乳源活性肽CCL-3S在制备促睡眠产品中的应用,所述乳源活性肽CCL-3S具有如SEQ ID NO:1所示的氨基酸序列,所述乳源活性肽CCL-3S的分子量为1759.998Da。
本发明利用体外膜片钳技术从牛奶中分离鉴定出一种具有抑制神经细胞动作电位发放频率、能促进睡眠的乳源活性肽(命名为CCL-3S),通过放电数目和频率对其促睡眠活性进行评价,发现CCL-3S作用于细胞后,Vm的变化率为12.56±3.67%,与空白组比较,具有显著性抑制作用(p<0.05),能显著抑制神经细胞动作电位发放频率,可以作为药物开发及促睡眠保健品的原料,可充分利用乳产品资源,操作简单,为乳源多肽的高值化利用提供了新途径。
优选的,所述促睡眠产品包括但不限于促睡眠药品和促睡眠功能性食品。
优选的,所述促睡眠是抑制神经细胞动作电位发放频率。
本发明还提供了一种促睡眠药品,包括乳源活性肽CCL-3S,所述乳源活性肽CCL-3S具有如SEQ ID NO:1所示的氨基酸序列。
优选的,为提高药品的适用范围,还包括药学上可接受的载体和/或辅料,如酸味剂、调色剂、香味剂、甜味剂,或其组合。
优选的,所述促睡眠药品的剂型包括但不限于口服液、冲剂、胶囊剂、片剂、颗粒剂、丸剂、膏剂和注射剂。
本发明还提供了一种促睡眠功能性食品,包括乳源活性肽CCL-3S,所述乳源活性肽CCL-3S具有如SEQ ID NO:1所示的氨基酸序列。
优选的,为提高功能性食品的适用范围,还包括食品上可接受的载体和/或辅料,如稀释剂、赋形剂、填充剂、粘合剂、湿润剂、崩解剂、吸收促进剂、甜味剂、香味剂,或其组合。
优选的,所述促睡眠功能性食品的形式包括但不限于汤剂、饮料、糖果、口服液、胶囊剂、片剂和粉剂。

下面对本发明的具体实施方式作进一步说明。在此需要说明的是,对于这些实施方式的说明用于帮助理解本发明,但并不构成对本发明的限定。此外,下面所描述的本发明各个实施方式中所涉及的技术特征只要彼此之间未构成冲突就可以相互组合。
下述实验例中所使用的试验方法如无特殊说明,均为常规方法;所使用的材料、试剂等,如无特殊说明,为可从商业途径得到的试剂和材料。
实施例1乳源活性肽的筛选鉴定
按照申请号为201510915215.0,名称为“利用膜片钳技术筛选乳源中促睡眠肽粗提物的方法”的专利方法从牛乳中分离酪蛋白水解物,然后从中筛选得到一种乳源活性肽,命名为CCL-3S,经鉴定发现,该乳源活性肽的氨基酸序列为HQGLPQEVLNENLLR,分子量为1759.998Da。其中,酪蛋白水解物的高效液相色谱图如图1所示,乳源活性肽CCL-3S的高效液相色谱图和质谱图分别如图2和图3所示。
实施例2乳源活性肽的电生理-睡眠效果
急性分离大鼠下丘脑腹外视侧核区脑片,厚度约300-500um,采用电生理膜片钳技术,根据实施例1的乳源活性肽对大鼠下丘脑腹外侧视前区神经元的放电特征,判断该乳源活性肽的促睡眠活性。具体的试验方法参考申请号为201510915215.0,名称为“利用膜片钳技术筛选乳源中促睡眠肽粗提物的方法”的专利。其电生理-睡眠效果如图4所示。
根据图4的结果可知,该乳源活性肽作用细胞后,Vm的变化率为12.56±3.67%,与空白组比较,具有显著性抑制作用(p<0.05),能显著抑制神经细胞动作电位发放频率,促睡眠效果好。
综合实施例1和实施例2可见,本发明将成分复杂的酪蛋白水解产物采用简单的活性分离及纯化技术得到具有较好促睡眠效果的乳源活性肽,操作简单,分离效果好,制备的样品通过膜片钳技术进行促睡眠活性评价,将复杂的促睡眠活性物研究简化为借助膜片钳筛选促睡眠物质,操作简单,本发明提供的乳源活性肽可以作为药物开发及促睡眠保健品的原料,可充分利用乳产品资源,为乳源多肽的高值化利用提供了新途径。
